The Analgesic Effect of Bee Venom Acupuncture and Its Mechanism on the Type II Collagen-Induced Arthritis Rats

  • Objectives : to evaluate the analgesic effect of bee venom acupuncture on Choksamni (ST36) in the collagen-induced arthritis rats and investigate the role played by serotonergic receptor subtypes (5-HT1a, 5-HT2a) in the antinociceptive effect of bee venom acupuncture in a thermal hyperalgesia test Methods : Experiments were performed on 5 week-aged 60 male Sprague-Dawley rats according to National Institute of Health guidelines and the ethical guidelines of the International Association for the Study of Pain (IASP). Arthritis was induced with arthrogenic collagen emulsion (Bovine type II collagen ${\mu}g$ with incomplete Freund's adjuvant $100\;{\mu}g$). The onset of arthritis was considered to be present when erythema and swelling were detected in at least one joint. The thermal hyperalgesia was evaluated weekly with tail flick test in the rats of severity grade 3 without any injury at tail and foot (including inflammation, ulceration, snap). In the fourth week after first immunization, the analgesic effect of bee venom acupuncture (Choksamni, ST36) was measured with consecutive tail flick latency after intraperitoneal injection of spiroxatrine (1mg/kg) and spiperone (1mg/kg). Results : Chronic inflammatory pain was induced as time elapsed after the immunization of arthrogenic collagen and the maximum value was reached from third to fifth week. Chronic inflammatory pain induced by CIA was inhibited by bee venom acupuncture on the left ST36. The analgesic effect of bee venom acupuncture was inhibited by intraperitoneal injection of 5-HT1a antagonist spiroxatrine and 5-HT2a antagonist spiperone. Conclusions : Therefore, a conclusion. that the analgesic effect of bee venom acupuncture in the chronic inflammatory pain is partially mediated by 5-HT1a and 5-HT2a receptors can be made.

낙석등(絡石藤)약침이 Collagen 유발 관절염에 미치는 영향 (Effect of Trachelospermi Caulis Herbal-acupuncture on the Collagen-induced Arthritis in Rats)

  • Objectives : This study was built to investigate the effect of Trachelospermi Caulis herbal-acupuncture on the Collagen-induced arthritis(CIA) in rats. Methods : Arthritis was induced by intradermal injection of Bovine type II collagen solution into base of tail. Experimental group were divided into 5 groups ; Normal(N) group, Control(C) group, Trachelospermi Caulis high(TH) group, Trachelospermi Caulis low(TL) group, Saline(S) group(n=7 for each group). Normal group was had no management. Control group was injected with Bovine type II collagen solution and taken no treatment. Trachelospermi Caulis high group was injected with Bovine type II collagen solution and taken high-intensity(10mg/kg) herbal-acupuncture treatment on $ST_{36}$. Trachelospermi Caulis low group was injected with Bovine type II collagen solution and taken low-intensity(5mg/kg) herbal-acupuncture treatment on $ST_{36}$. Saline group was injected with Bovine type II collagen solution and taken saline injection on $ST_{36}$. Body weight, paw edema volume and ankle joint thickness were measured during experimental day. On the last experimental day, we analyzed WBC count, TNF-$\alpha$ & IL-$1{\beta}$ concentration, c-fos immunohistochemistry and NADPH-d histochemistry for evaluating the effect of Trachelospermi Caulis herbal-acupuncture. Results : The results were as follows ; 1. In the change of paw edema volume, TH group only has significant difference compared with C group. 2. In the change of ankle joint thickness, TH group only has significant difference compared with C group. 3. In WBC count of serum, TH, TL groups have significant decrease compared with C group. 4. In TNF-$\alpha$ concentration of effusion, TH, TL groups have significant decrease compared with C group. and TH group has significant decrease compared with TL group. 5. In IL-$1{\beta}$ concentration of effusion, TH, TL groups have significant decrease compared with C group. and TH group has significant decrease compared with TL group. 6. In c-fos positive neurons of S1S2(cortex) region, TH, TL, S groups have significant decrease compared with C group. 7. In NADPH-d positive neurons of CPu(caudate putamen) region, TH, TL groups have significant decrease compared with C group. 8. In NADPH-d positive neurons of Tfp(transverse fibers of pons) region, TH, TL, S groups have significant decrease compared with C group. and TH group has significant decrease compared with S group. Conclusions : According to above results, we hope that Trachelospermi Caulis herbal-acupuncture may have the effect that decreases progression and development of CIA. And it can be suggested that Trachelospermi Caulis herbal-acupuncture may reduce the expression of c-fos and NOS.

금주사액약침(金注射液藥鍼)이 제2형 콜라겐 유도 관절염(關節炎)에 미치는 영향(影響) (The Effects of Aurothiomalate acupuncture on Type II Collagen-induced Arthritis)

  • Objective : The purpose of this study was to investigate the effects of Aurothiomalate acupuncture containing Sodium Aurothiomalate on Type II collagen-induced arthritic DBA/1 mice. Methods : The mice were divided into three groups, one control group and two test groups. The control group was injected with saline on Choksamni(ST36) and the two test groups were injected either with Aurothiomalate acupuncture into Choksamni(AT1 group), or with Aurothiomalate acupuncture into Hwando(G30, AT2 group). We measured the incidence of arthritis, the mean index of arthritis, the number of arthritic limbs for the clinical examination and evaluation. We also examined the changes in the level of TNF-$\alpha$ and IL-$1{\beta}$. Results : 1. The incidence of arthritis in mice of the AT1 and AT2 groups were significantly lower than that of the control group. 2. The mean indices of arthritis in mice of the AT1 and AT2 groups were significantly lower than that of the control group. 3. The mean number of arthritic limbs in mice of the AT1 and AT2 groups were significantly lower than that of the control group. 4. The levels of TNF-$\alpha$ of the mice in the AT1 and AT2 groups were significantly different from that of the control group. 5. The levels of IL-$1{\beta}$ of the mice in the AT1 and AT2 groups were not significantly different from that of the control group. Conclusion : These results indicate that the treatment with Aurothiomalate acupuncture could improve the clinical status of Arthritis by suppressing immune responses in the mice of Type II collagen-induced arthritic.

족삼리(足三里) 세신약침(細辛藥鍼)이 생쥐의 Collagen-induced arthritis에 미치는 영향 (A Study on the Effect of Herbal-acupuncture with Asari Herba Cum Radice solution at Joksamni(ST36) on Collagen-induced arthritis)

  • 세신약쇄(細辛藥鎖)이 Collagen-induced arthritis에 미치는 영향(影響)을 DBA/1J mouse를 이용한 실험(實驗)을 통하여 관찰(觀察)한 결과(結果), 다음과 같은 결론(結論)을 얻었다. 1. 세포독성(細胞毒性) 측정(測定) 및 TNF-${\alpha}$ 발현(發現) 억제효과(抑制效果) 1) 세신약침액(細辛藥鍼液) 1%의 농도(濃度)에서 가장 높은 세포생존률(細胞生存率)이 관찰(觀察)되었다. 2) 세행약침액(細幸藥鍼液) 1%의 농도(濃度)에서 TNF-${\alpha}$가 유의성(有意性)있게 감소(減少)하였다. 2. 관식염(關飾炎) 발병률(發病率)과 비장(脾臟)의 무게 관찰(觀察) 1) 세행약침(細幸藥鍼) I군과(群) 세행약침(細幸藥鍼) II군에(群)서 관절염(關節炎) 발병률(發病率)이 현저하게 감소(減少)하였다. 2) 세신약침(細辛藥鍼) I군과(群) 세졸약침(細卒藥鍼) II군의(群) 비장(脾臟) 무게가 대조체(對照諦)에 비하여 감소(減少)하였다. 3. 세신약침(細辛藥鍼)이 각종 싸이토카인, 면역 글로불린 및 Anti-collagen II에 미치는 영향(影響) 1) 혈청(血淸)내 IL-6, LNF-${\gamma}$, TNF-${\alpha}$는 세행약침(細幸藥鍼) I군과(群) 세행약침(細幸藥鍼)II군에(群)서 대조군(對照群)에 비하여 유의성(有意性)있게 감소(減少)하였다. 2) IgG, IgM은 세행약침(細幸藥鍼) I군과(群) 세행약침(細幸藥鍼)II군에(群)서 대조군(對照群)에 비하여 유의성(有意性)있게 감소(減少)하였다. 3) Anti-collagen II는 세행약침(細幸藥鍼) I군과(群) 세행약침(細幸藥鍼) II군에(群)서 대조군(對照群)에 비하여 유의성(有意性)있게 감소(減少)하였다 4. 세신약침(細辛藥鍼)이 관절(關節)의 조직학적(組織學的) 변화(變化)에 미치는 영향(影響) 1) Hematoxylin and eosin stain에서 세행약침(細幸藥鍼) I군과(群) 세행약침(細幸藥鍼)II군이(群) 연골(軟骨)의 파괴(破壞)와 활막(滑膜)의 증식(增殖)이 감소(減少)되고 정상군(正常群)에 류사(類似)한 구조(構造)로 회부(回復)되었다. 2) Masson's Tricho me stain에서 세행약침(細幸藥鍼) I군과(群) 세행약침(細幸藥鍼)II군의(群) 교원질섬유가 정상군(正常群)과 같이 유지(維持)되었다. 5. 세신약침(細辛藥鍼)이 면역세포(免疫細胞)의 발현(發現)에 미치는 영향(影響) 1) 림파절내(淋巴節內) 4CD3e^+$ 세포(細胞)와 $CDl9^+$ 세포(細胞)의 비율(比率), $CD4^+$ 세포(細胞)와 $CD8^+$ 세포(細胞)의 비율(比率)은 細幸藥鍼I군과(細幸藥鍼I群) 세행약침(細幸藥鍼)II군에(群)서 Control군에(群) 비하여 정상군(正常群)과 유사(類似)한 비율(比率)이 유지(維持)되었다. 2) 임파절내(淋巴節內) $CD69^+/CD3e^+$ 세포(細胞)와 $CDlla^+/CDl9^+$ 세포(細胞)는 세행약침(細幸藥鍼)I군과(群) 세행약침(細幸藥鍼)II군에(群)서 대조군(對照群)에 비하여 감소(減少)하였다. 3) 관절내(關節內) $CD11b^+/Gr-1^+$ 세포(細胞)는 세행약침(細幸藥鍼)I군과(群) 세행약침(細幸藥鍼)II군에(群)서 대조군(對照群)에 비하여 감소(減少)하였다. 이상(以上)의 모든 결과(結果)에 있어서, 세행약침(細幸藥鍼)II군보(群)다 예방(豫防)과 치료(治療)를 겸(兼)한 세행약침(細幸藥鍼)I군이(群) 싸이토카인 및 항체(沆體)의 감소(減少)와 면역세포(免疫細胞)의 발현(發現) 억제(抑制)에 있어서 탁월한 효과(效果)를 나타내었다.
