Chondrogenic Differentiation of Porcine Skin-Derived Stem Cells with Different Characteristics of Spontaneous Adipocyte Formation

  • The purpose of this study is to confirm whether spontaneous adipocyte generation during chondrogenic induction culture affects the chondrogenic differentiation of porcine skin-derived stem cells (pSSCs). For this purpose, chondrogenic differentiation characteristics and specific marker gene expression were analyzed using cell lines showing different characteristics of spontaneous adipocyte formation. Of the four different lines of pSSCs, the pSSCs-IV line showed higher Oil red O (ORO) and glycosaminoglycan (GAG) extraction levels. Quantitative real-time polymerase chain reaction (qRT-PCR) analysis revealed that the levels of adipogenic markers peroxisome proliferator-activated receptor gamma 2 ($PPAR{\gamma}2$) and adipocyte Protein 2 (aP2) mRNAs were significantly higher in pSSCs-IV than those of the other pSSC lines (P<0.05). Among three chondrogenic markers, collagen type II (Col II) and sex determining region Y-box (Sox9) mRNAs were strongly expressed in pSSCs-IV (P<0.05), but not in aggrecan (Agg), which was significantly higher in pSSCs-II (P<0.05). These results demonstrate that the spontaneous adipocyte generation during chondrogenic differentiation has a positive effect on the chondrogenesis of pSSCs. More research is needed on the correlation between adipocyte generation and cartilage formation.

Comparison of Biomechanical Properties of Dura Mater Substitutes and Cranial Human Dura Mater : An In Vitro Study

  • Objective : The aim of this study was to investigate the biomechanical differences between human dura mater and dura mater substitutes to optimize biomimetic materials. Methods : Four groups were investigated. Group I used cranial dura mater (n=10), group II used $Gore-Tex^{(R)}$ Expanded Cardiovascular Patch (W.L. Gore & Associates Inc., Flagstaff, AZ, USA) (n=6), group III used $Durepair^{(R)}$ (Medtronic Inc., Goleta, CA, USA) (n=6), and group IV used $Tutopatch^{(R)}$ (Tutogen Medical GmbH, Neunkirchen am Brand, Germany) (n=6). We used an axial compression machine to measure maximum tensile strength. Results : The mean tensile strengths were $7.01{\pm}0.77MPa$ for group I, $22.03{\pm}0.60MPa$ for group II, $19.59{\pm}0.65MPa$ for group III, and $3.51{\pm}0.63MPa$ for group IV. The materials in groups II and III were stronger than those in group I. However, the materials in group IV were weaker than those in group I. Conclusion : An important dura mater graft property is biomechanical similarity to cranial human dura mater. This biomechanical study contributed to the future development of artificial dura mater substitutes with biomechanical properties similar to those of human dura mater.

  • 1차년도에 F9 EC cell의 분화유도 시스템을 확립한뒤 계속적으로 해양생물로 부터 분리한 여러가지 물질을 처리하여 세포의 형태 변화 및 분화 관련 유전자 즉 laminin Bl, type IV collagen, RAR-$\beta$ 의 발현을 조사하였다. 그 결과 다수의 물질이 F9 세포를 분화시키는데 효과가 있는 것으로 나타났으며, 효과있는 물질들에 대한 구조분석이 계속적으로 실시되고 있다. 뿐만 아니라 분화효과가 있는 물질중에서 그 구조적으로 특이한 성질을 가진 물질에 대하여 이 물질이 암세호의 분화에 미치는 기작에 대해 조사하였다. 구체적인 실험내용을 요약하면 다음과 같다. 1. Sesquicillin, an Extracellular Matrix Adhesion Inhibitor, Inhibits the Invasion of B16 Melanoma Cells In vitro

  • Tumor cell interaction with the extracellular matrix is defined as the critical event of tumor invasion that signals the initiation of a metastatic cascade. Sesquicillin has been identified as an inhibitor of melanoma cell adhesion to the components of the extracellular matrix (ECM) in cultured broth of fungal strain F60063. Sesquicillin strongly inhibited the adhesion of B16 melanoma cells to laminin, fibronectin, and typeIV collagen. It also inhibited B16 melanoma cell invasion of reconstituted basement membrane Matrigel in vitro in a dose-dependent manner. These results suggest that sesquicillin is a new class of nonpeptidic ECM adhesion inhibitor having anti-invasive activity.

Purification and Characterization of Extracellular Proteinase Produced by Pseudomonas aeruginosa (Pseudomonas aeruginosa 세포질외 serine계열 단백질 분해효소의 정제 및 특성)

  • A serine proteinase of molecular weight 60 kd was purified from culture supernatant of P. aeruginosa using DEAE-Trisacryl M ion-exchange and AcA 54 gel filtration column chromatography, and the properties of serine proteinase were characterized. By means of SDS-polyacrylamide gel electrophoresis, the molecular weight of the enzyme was 55 kd. The optimal pH for the activity of purified enzyme was 7.5. The activity of the purified enzyme was completely inhibited by Di-isopropylfluorophosphate(DFP) and N-.alpha.-p-tosyl-L-lysine choloromethyl detone(TLCK) but not by other proteinase inhibitors such as E-64, pepstatin A, 1, 10-phenanthroline. The purified enzyme was capable of degrading type I and type IV collagen. Antisera obtained from hymans infected with Pseudomonas aeruginosa reacted to the purified serine proteinase in immunoblots. These results indicate that the purified enzyme is trypsin-like serine proteinase and this enzyme of P. aeruginosa may play an important role in tissue damage as a spreading factor and may be useful for serodiagnosis of Pseudomonas infections.

Relationship Between Vascularity and Other Remodeling Parameters in Asthmatic Airway (기관지천식환자에서 기도내 혈관분포정도(vascularity) 및 다른 기도 재구성 소견과의 연관성)

  • Background : The pathological features in asthmatic airway remodeling are diverse. The aim of this study was to examine the degree of airway vascularity in relation to the other remodeling parameters in asthmatics. Methods : Bronchial biopsies were done in 34 asthmatic patients, and 6 control subjects. The basement membrane thickness and the subepithelial thickness were measured in the hematoxylin-eosin stained tissue, and the degree of vascularity was measured using type IV collagen immunostaining. Results : 1) Compared to the control subjects, the asthmatics showed a significant increase in the basement membrane thickness ($6.92{\pm}2.01{\mu}m$ vs $9.67{\pm}2.84{\mu}m$, p<0.05) and the subepithelial thickness ($44.49{\pm}31.92{\mu}m$ vs $121.22{\pm}72.79{\mu}m$, p<0.05). 2) Compared to the control subjects, the asthmatics showed a significant increase in the vascular area per unit submucosal area ($4.51{\pm}2.13%$ vs $10.32{\pm}6.08%$, p<0.05). In addition, the number of vessels per unit submucosal area showed an increased tendency without statistical significance. 3) In the asthmatics, the number of vessels and the vascular area per unit submucosal area showed no correlation with the basement membrane thickness, the subepithelial thickness, the severity, the forced expiratory volume in 1 second($FEV_1$), and the methacholine provocative concentration 20($PC_{20}$). Conclusion : This study showed that vascularity was an important parameter in asthmatic airway remodeling but it was not related to the other remodeling parameters such as the basement membrane thickness and the subepithelial thickness. Each of these asthmatic remodeling parameters may have a different clinical significance. Therefore, further studies will be needed.

Impact of Low Versus Conventional Doses of Chemotherapy During Transcatheter Arterial Chemo-embolization on Serum Fibrosis Indicators and Survival of Liver Cancer Patients

  • Objectives: To explore the impact of low- vs conventional-dose chemotherapy via transcatheter arterial chemo-embolization (TACE) on serum fibrosis indicators and treatment efficacy of hepatocellular cancer patients (HCC). Materials and Methods: Patients fulfilling the eligibility criteria were assigned to TACE in Group A (with low-dose chemotherapy) or Group B (conventional-dose chemotherapy). Four serum fibrosis related indicators, hyaluronic acid(HA), human pro-collagen type-III (hPC-III), laminin (LN), and collagen type-IV(IV-C) before TACE were compared with the values 7 days after TACE. The response rate and survival time were also compared between the two groups. Results: Fifty patients with HCC were enrolled in this study, including 25 in Group A and 25 in Group B. No significant differences were detected between the two groups in the four indicators before TACE. After TACE, the value of the four serum indicators increased significantly in Group B. However, no significant differences regarding these four indicators were found in Group A after TACE. Significant differences were demonstrated between the two groups after TACE, but median survival time and 1 or 2 year overall survival rates did not differ (P>0.05). Conclusions: Low-, compared with conventional-dose chemotherapy exerts the same impact on the variation of fibrosis related indicators and has no influence on median survival time and survival rate after TACE in HCC patients.

  • In order to compare and study the effects of Kamipalmijiwhang tang(加味八味地黃湯) & Kamigangwhalsokdantang(加味羌活續斷湯) on inhibition of osteoporosis induced by ovariectomy, the author administered Kamipalmijiwhangtang & Kamigangwhalsokdantang to the sample groups per orally before and after ovariectomy. The author observed the activities of cathepsin L, K and the contents of calcium, calcitonin, osteocalcin and phosphorus in the serum for the biochemical index, the contents of IL-6 in the serum and bone marrow for immunological index, the state of bone loss in the metaphysis of tibia by histological study, the expression of ELAM-1 &collagen IV by immunohistochemical analysis, the expression of fos by immunocytochemical analysis. The results were as follows: 1. The activities of cathepsin L, K in the serum had a significant inhibitive effects in all sample groups compared with the control group. The inhibitive effects of Sample A-1 & B-1 was better than those of A-2 & B-2 and the inhibitive effects of sample B-1 & B-2 was better than those of A-1 & A-2. 2. The content changes of IL-6 in the serum and bone marrow had a significant inhibitive effects in all sample groups compared with the control group. The inhibitive effects of sample A-1 & B-1 was better than those of A-2 & B-2 and the inhibitive effects of sample B-1 & B-2 was better than those of A-1 & A-2. 3. In the histological study of tibial metaphysis all the sample groups inhibited bone loss significantly compared with the control group and expressed trabecula shape of bone similar to the normal group. 4. The expression of ELAM-1 & collagen IV decreased a little bit in sample A-2. It also decreased to a similar degree in sample B-2 like normal group and disturbed itself. 5. The expression of fos was inhibited in all sample groups compared with control group. The inhibitive expression of sample B-1 & B-2 was more remarkable than those of A-1. & A-2. 6. The serum calcium had a significant increasing effects in an sample groups compared with control group. Each sample groups didn't have any significant difference. 7. The serum calcitonin had a significant increasing effects in sample A-1, A-2 and B-1 groups compared with control group. Each sample groups didn't have any significant difference. 8. The serum osteocalcin and phosphorus didn't have any significant difference in all sample groups compared with control group.

The Effect of Hair Growth and Distribution by Sophorae Radix, Panax ginseng, Salvia miltiorrhiza BUNGE Water Extracts (고삼, 인삼 및 단삼 혼합물에 의한 모발의 성장과 분포에 미치는 영향)

  • In this study, we investigated effects of Monegy (mixture of Sophorae Radix, Panax ginseng, Salvia miltiorrhiza BUNGE) on epilate-induced hair-loss in dorsal region of C57/BL6 mice and external structure of human hair. For morphological and histological analysis in scalp of epilate-induced hair-loss animal model, we utilized several microscopic techniques, such as confocal laser scanning microscopy (CLSM) and LAS 4000. Confocal analysis showed the distribution of FITC-conjugated Monegy and penetration depth compared with normal and control group. Furthermore, when Monegy was topically administrated onto a C57BL6 mouse, it penetrated very well. The fluorescence intensity was increased upto 205 and 113 folds compared to normal and control group, respectively. Also, area of fluorescence was increased to upto 255 to 127 folds compared to normal and control group. Broad scale area of fluorescence in dermis region was observed in the Monegy-treated mice. Furthermore, Monegy induced upto 75% hair repair against depilation. It might be promoted via the induction of growth factors in hair follicle.

Effect of Extracellular Matrix on the Growth Behavior of Corneal Endothelial Cells to Poly(lactic-co-glycolic acid) Film (각막 내피세포 성장 거동에 대한 락타이드 글리콜라이드 공중합체 필름과 세포외 기질의 효과)

  • Corneal endothelium is mono-inner cell layer of cornea and lay on Descmet's membrane which comprised of various proteins called extracellular matrix such as fibronectin, collagen, laminin, and proteoglycan, etc. In this study, we fabricated transparent poly(lactic-co-glycolic acid) (PLGA) film because PLGA is widely used for tissue engineering based on their properties. We investigated the behaviors of rabbit corneal endothelial cells (rCEnCs) on PLGA film surfaces coated with various cell-adhesive molecules like fibronectin, laminin, collagen type I and IV and FNC coating mix. The morphologic images, proliferation and adhesion assay, immunofluorescence for ZO-1 and $Na^+/K^+-ATPase$ and RT-PCR for expression of specific markers were conducted. These results showed that PLGA film plays a role as CEnC carriers in vitro and the cell-adhesive molecules give positive effects on the behaviors of rCEnC.
