Preparation of Instant Powdered Soup Using Oyster Wash Water and Its Characteristics (굴 세척액을 이용한 인스턴트 분말 수프의 제조 및 특성)

  • To utilize oyster cannery processing waste water effectively, this study was carried out to prepare instant powdered soup using oyster wash water. Instant powdered soup from oyster hot-water extracts (HWE) was prepared by mixing oyster spray-dried hot-water extracts (15 g) with table salt (5 g), cream powder (19 g), milk replacer (12 g), wheat flour (20 g), corn flour (15 g), starch (5 g), glucose (7.5 g) and onion powder (1.5 g). In preparing instant powdered soup from oyster wash water (OWW), powder from oyster spray-dried wash water instead of the spray-dried hot water extracts, was added and other additives were added in proportion to those in the HWE. The OWW consists mainly of carbohydrates (71.1%). It was not different from the instant powdered soup from hot-water extracts. The volatile basic nitrogen, vaible cell counts, coliform group of instant powdered soup from oyster wash water contains 29.4 mg/100g, $4.6{\times}10^4\;CFU/g$, <18 MPN/100g, respectively and its water activity has 0.246. So it was a hygienically safe and conservable instant food. The main fatty acid of OWW was 16 : 0 and 18 : 1n-9. Its chemical score of protein was 59.4% and its main inorganic matter was iron. According to a sensory evaluation, in contrast to the HWE, the OWW had a slightly lower aroma but better taste. It was concluded from the above chemical and sensual evaluation that the oyster wash water can be used as a flavor enhancer for instant powdered soup.

Physicochemical and Microbiological Properties of Ginseng-Whey Beverages (인삼 유청음료의 이화학적 및 미생물학적 특성)

  • Ginseng-whey beverages were prepared with rennet whey, ginseng, sweetener, honey and Japanese apricot, inoculated with different strains of lactic acid bacteria or unfermented partly. The samples were stored at 4$^{\circ}C$ or 30$\pm$1$0^{\circ}C$ and then physicochemical and microbiological properties were investigated. The yield of whey was 78.8%. The pH-values reduced and acidities increased during the storage period. The contents of solid-substances, ash and lipid in ginseng-whey beverages were 7.90~8.20%, 0.62~0.66% and 0.16%, respectively. The protein contents of ginseng-whey beverages were 0.42~0.56% and the contents were not changed during the storage period. The lactose contents of fermented beverages were higher than those of unfermented beverages. During the storage period (1~5 weeks), the ranges of D(-) - and L(+)- lactic acid contents in fermented ginseng-whey beverages (17.3~156.1 mg/100g, 347.3~1894.2mg/100g) were higher than those of unfermented ginseng-whey beverages (6.2~82.8mg/100g, 7.1~885.5mg/100g). The contents of total saponin in unfermented sample and fermented sample (Lac. casei sub-sp. casei+Str. salivarius sub-sp. thermophilus) were increased during the storage period. But, those of the fermented sample(Lac. acidophilus+Lac. delbrueckii sub-sp. bulgaricus) were reduced. In the electrophoretic results of ginseng-whey beverages, an $\alpha$-lactalbumin and a $\beta$-lactoglobulin bands were shown apparently and there were no changes observed during the storage period. During the storage period (1~3 week) the coliform was not detected and total plate counts and psychrotrophs were increased according to the storage period.

Effect of Various Presoaking Treatments on the Microbial Contamination and Germination Ratio of Soybeans (다양한 침지처리가 대두의 미생물 오염 및 발아율에 미치는 영향)

  • The presoaking conditions of soybeans in various solution for decreasing microbial contamination and improving germination were investigated. Soybeans were treated with presoaking solutions (sterilized water, 0.1% grape fruit extract, 0.1% vitamin $B_1$ derivate solution AS5 and vitagen) at $25^{\circ}C,\;35^{\circ}C,\;45^{\circ}C\;and\;55^{\circ}C$ for $3\sim12$ hr and germinate at $25^{\circ}C$, 95%RH for 3 days. Total bacterial count and coliform count of soybeans were $6.0\times10^5CFU/g$ and $3.0\times10^5CFU/g$, respectively, and the most of presoaking treatments decreased the microbial contamination. The increase of presoaking time induces low germination ratio compared with unsoaked soybeans and the germination of soybeans was not detected at $55^{\circ}C$. The germination of soybeans was effected by presoaking solutions, temperature, and time. The optimal presoaking condition for decreasing microbial contamination and promoting initial germination was using grape fruit extract at $35^{\circ}C$ for 6 hr.

Evaluation of Microbiological Hazards of Hygiene by the Customers' Hands in University Foodservice Operation (대학급식소 고객의 손 위생에 대한 미생물학적 위해 평가)

  • The propose of this study was to evaluate the microbiological hazards of the customers' hands in university foodservice operation. A total of 190 customers' hands were sampled to test about aerobie plate counts, coliforms, E. coli and Staphylococcus aureus. The average number of aerobic plate counts was 3.11 log CFU/hand Ifrom 0.70 to 6.47 log CFU/hand) and that of coliform counts was 1.06 log CFU/hand (from not detected to 5.04 log CFU/hand). The mean level of aerobic plate counts (t=9.87, p<0.001) and coliforms (t=7.91, p<0.05) was significantly different by gender. E. coli was not detected, but Staphylococcus aureus was detected in 39 (20.5%) of 190 samples. The average of hand-washing frequency was 6.46 time per day. By the hand-washing frequency, the mean level of aerobic plate counts was significantly different (F=527, p<0.001), but coliforms was not significantly different. This study shows that it is needs to be given attention to hygiene handlers but also for customers in foodservice operations.

Sanitization of Commercial Powdered Products Using Gamma Irradiation (감마선 조사를 이용한 시판 분말원료의 위생화)

  • Microbiological populations and the sterility of commercial powdered products treated with gamma irradiation at 0~10 kGy were investigated before using them as ingredients for a non-cooked Saengsik product. We evaluated a total of 14 powdered products: 8 powdered cereals, 3 powdered tubers, and 3 powdered leafy vegetables. The total numbers of bacterial populations in non-irradiated powdered cereals, tubers, and leafy vegetables were 2.7~6.9, 5.6~6.0, and $5.3{\sim}6.8\;log\; CFU{\cdot}g^{-1}$, respectively. Moreover, coliform bacteria were not indicated in adlay, millet, germinated brown rice, soybean, and mulberry leaves powder within detection limit ($2.0\;log\; CFU{\cdot}g^{-1}$). The number of Bacillus cereus exceeded $3.0\;log\; CFU{\cdot}g^{-1}$ (the maximum limit for Saengsik products) in all samples, excluding perilla seeds, buckwheat, barley, oat, potato, and Jerusalem artichoke powder. However, a dose of 6 kGy of gamma irradiation reduced the microbiological populations in all samples, and all the powdered products met the microbial requirements for Saengsik products. Futhermore, it was confirmed that all microorganisms in the 9 powdered products, except fermented brown rice, sweet potatoes, and 3 leafy vegetables, were sterilized by 10 kGy of gamma irradiation.

  • 하천에서의 분변성대장균은 분변성 오염 정도를 나타내는 지표로서, 이 농도가 높을수록 오염된 하천수와의 접촉을 통한 호흡기, 소화기 및 피부 관련 질병의 발발 확률이 높다고 알려져 있다. 따라서 하천에서의 수영, 수상스키 등과 같은 입수형 친수활동을 할 때, 분변성대장균 농도가 농도 기준 이하인지를 확인하고 이러한 정보를 친수활동에 이용할 필요가 있다. 그러나 분변성대장균의 경우, 현재 자동수질측정망에서 측정되고 있는 다른 수질인자들과는 달리 실시간 측정이 불가능하다고 알려져 있다. 분변성대장균을 측정하는데 있어 최소 18시간 이상이 필요하며, 이러한 분변성대장균 측정 방식은 하천 이용자들이 안전한 친수활동을 영위하는데 있어 적절한 수질 정보를 제공하지 못한다. 그러므로 분변성대장균을 예측하는 모델을 개발하고, 이를 이용하여 실시간 분변성대장균 정보를 생성하여 하천 이용자들에게 제공할 필요가 있다. 본 연구에서는 친수활동이 활발하게 이루어지는 곳 중 하나인 북한강의 대성리 지점에 대해 데이터 기반 모델을 이용하여 분변성대장균을 예측하였다. 데이터 기반 모델은 물리 기반 모델에서 필요한 지형데이터나 비점오염원 등의 초기 오염물의 양에 대한 데이터를 필요로 하지 않고, 대신 독립변수로 사용되는 기상 및 수질데이터를 필요로 한다. 이러한 기상 및 수질데이터는 기존 기상관측소, 수질관측소에서 매일 자동으로 측정되기 때문에 데이터 기반 모델은 물리 기반 모델에 비해 입력데이터를 구성하기가 쉽다는 장점을 지닌다. 이러한 데이터 기반 모델 중 분류 모델은 회귀 모델과 달리 분변성대장균 농도가 일정 수질기준 이상을 넘는지를 바로 예측할 수 있다. 본 연구에서는 분류 모델 중 높은 예측력을 가진다고 알려진 랜덤포레스트(random forest) 기법을 이용하여 분변성대장균 예측 모델을 개발하였다. 분변성대장균 예측 모델은 주어진 기상 및 수질 조건에 대해 분변성대장균이 200 CFU/100ml가 넘는지를 예측하였다. 예측된 분변성대장균이 기준을 넘는 경우를 2등급, 넘지 않는 경우를 1등급으로 명명하였다. 모델을 개발하기 위하여 북한강 대성리 인근 측정소에서 2010년부터 2015년까지 측정된 기상 및 수질데이터를 수집하였다. 수집한 데이터를 훈련 및 검증데이터로 샘플링하였으며, 이 때 샘플링한 데이터가 기존 데이터가 가지고 있던 등급별 비율을 유지하기 위하여 층화샘플링을 하였다. 본 연구에서는 샘플링에 의한 불확실성을 줄이기 위하여 랜덤하게 50번 샘플링된 각각의 훈련데이터에 대해 모델을 개발하였다. 50개의 모델의 검증 결과를 종합한 결과, 전체 예측률은 0.139로 나타났다.

Microbiological Contamination Level and Radiation Sterilization in Disposable Kitchen Utensil (일회용 주방용품의 미생물 오염도평가 및 방사선 살균)

  • The purposes of this study were to assess the microbiological contamination level of various disposable kitchen utensils and evaluate the effectiveness of gamma irradiation as a sterilization process for the utensils. The 51 kinds in 17 groups of disposable kitchen utensils were tested for the enumeration of total aerobic plate count, coliforms, fungi and Salmonella. Generally, cell density of microorganisms in disposable kitchen utensils were lower than that of cooking utensils currently using in kitchen. The survivals of total aerobic plate counts, putrefactive bacteria, on the surface of the disposable utensils were ranged up to $10^3\;CFU/100\;cm^2$. Filamentous fungi were detected in 13 samples. Coliform bacteria were detected in two kinds of samples but Salmonella spp. was detected below detection limits in all the samples. The microorganisms survived on the surface of utensils were effectively eliminated by gamma irradiation process at 3 kGy.

Effect of Pediocin Treatment on Soybean curd Quality during Storage (Pediocin 용액침지가 두부의 저장성에 미치는 영향)

  • We investigated the effects of pediocin on physicochemical and microbial changes in soybean cut 9 days of storage at $10^{\circ}C$, in order to improve shelf-life. As the storage time of curd increased the pH of solutions treated by pediocin immersion did not vary greatly, whereas the pH of control curd decreased after 5 days of storage. Titratable acidity increased in non-treated curd after 5 days of storage, and also in cud immersed in pediocin solutions of 300 and 500 ppm, after 6 and 7 days of storage, respectively. A pediocin solution of 1,000 ppm the development of titratable acidity. Also, turbidity did not increase during storage of curd treated with a pediocin solution of 1,000 ppm. The bacterial count of the immersion solution was $10^{2.5}$ CFU/mL at the commencement of storage, remained stable for 5 days of storage, and then increased rapidly. Coliforms were detected in untreated curd after 2.5days of storage. In curd treated with 300, 500 or 1,000ppm of pediocin, the elapsed times to coliform detection were 3.5 days, 5 days and 7 days, respectively, It is thus possible to prevent the deterioration of soybean curd with pediocin treatment.

Quality Changes of Kongnamul Muchim (Cooked Soybean Spouts) Stored with Gamma-Irradiated Red Pepper Powder (감마선 조사된 고춧가루 첨가 콩나물 무침의 저장 중 품질의 변화)

  • This study investigated the microbiological and sensory characteristics of Kongnamul muchim stored with gamma-irradiated red pepper powder at $4^{\circ}C$. Total aerobic microbes in raw Kongnamul muchim were 5.72 log CFU/g in the red pepper powder and 2.40 log CFU/g in the garlic used during storage, but were not detected n other raw materials. Coliform bacteria and fungi were found, at 3.11 and 3.48 log CFU/g respectively, only n the red pepper powder. Microorganisms in Kongnamul muchim stored with gamma-irradiated (10 kGy) red pepper owder were not detected over 3 days of storage at $4^{\circ}C$. The pH, Hunter's color value, and sensory characteristics id not change significantly on storage. These results suggest that the addition of gamma-irradiated (less than 10 Gy) red pepper powder could improve the microbiological safety of Kongnamul muchim without changing desirable ensory characteristics.

Statistical Analysis of the Spatio-temporal Water Quality Characteristics of the Nakdong River (낙동강수계 수질의 시·공간적 특성에 대한 통계학적 분석)

  • Water quality is characterized by various complex factors. Therefore, a systematic understanding of water quality trends is required to carry out a proper evaluation. In this study, we analyzed the spatio-temporal water quality characteristics of the Nakdong River using five-year data from 2012 to 2016. Data was collected on the pH, DO, BOD, COD, SS, TN, TP, TOC, WT, EC, $NH_3-N$, $NO_3-N$, $PO_4-P$, Chl-a, rainfall, and total and fecal coliforms. A total of 38 water quality measurement stations, from Andong1 to Gupo, were considered. Statistical analyses including trend, cluster, and factor analyses were conducted to identify the dominant water quality components affecting the Nakdong River. The Nakdong River was spatially classified into three groups for up-stream (Andong1 to Sangju1), mid/up-stream (Donam to Dalseong), and mid/down-stream (Hwawonnaru to Gupo) data collection, and temporally into two groups for summer/fall (7~10), and the rest of the season (11~6) data. The water quality of the entire Nakdong River showed trends similar to the mid/down-stream section, which indicates the importance of water quality management in this section. Suspended solids, phosphorus, and coliform groups were established as important factors to be considered in the summer/fall season across the river, especially in the mid/down-stream section. Nitrogen and organic matter were identified as important factors to be considered in the rest of the season, especially in the mid/up-stream section. This study could help determine the water quality components that should be intensively monitored in the Nakdong River.
