Seed Germination Response to Temperature, Cold Stratification Period, and Gibberellin Treatment in Spiraea fritschiana

  • To improve the germination of Spiraea fritschiana seeds for mass propagation, we evaluated the effect of a range of temperatures, cold stratification periods, and gibberellic acid ($GA_3$) treatments on three germination characteristics. Final germination percentage (FGP) increased as the temperature for seed germination increased, up to $30^{\circ}C$, while the mean germination time (MGT) and the mean number of days to 30% germination ($T_{30}$) decreased when seeds were germinated at $25-30^{\circ}C$. The optimum germination temperature of S. fritschiana seeds is approximately $30^{\circ}C$ considering FGP, MGT, and $T_{30}$ together. FGP increased with the duration of cold stratification up to a period of 6 weeks, but declined after 8 weeks of cold stratification, as prolonged cold stratification can induce dormancy with a resultant decline in germination. Pretreatment with 6-8 weeks of cold stratification or soaking seeds in distilled water or $500mg{\cdot}L^{-1}$ $GA_3$ for 24 h accelerated and increased the germination of S. fritschiana seeds, regardless of temperature. However, further study might be required to evaluate the effect of $GA_3$ concentrations lower than $500mg{\cdot}L^{-1}$ on the promotion of germination in S. fritschiana seeds.

  • Low-temperature adaptation and protection for environmental stresses were studied in the gram-negative soil bacterium Bradyrhizobium japonicum 61A101c. B. japonicum was more resistant to alcohol, $H_2O_2$, heat and freezing following a pretreatment at $4^{\circ}C$, resulting in approximately 10 to 1,000 folds increased survival compared to mid-exponential-phase cells grown at an optimal temperature at $28^{\circ}C$. This phenomena relate to the cold shock protein expressed when cells are exposed to a downshift in temperature. To confirm the presence of cold shock protein genes in B. japonicum, a PCR strategy was employed using a degenerate primer set, which successfully amplified a putative csp gene fragment. Sequence analysis of the PCR product(200bp) revealed csp-like sequences that were up to 96% identical to csp gene of S. typhimurium.

Plant Regeneration from Anther Culture of Panax ginseng

  • The research concerned of the regeneration of plants from embryos obtained from anther cultures of ginseng (Panax ginseng C. A. Meyer). The aim was to determine the influence of the regeneration medium on the efficiency of the regeneration process. We conducted to determine the optimum conditions such as cold pretreatment, plant growth regulators and carbon sources on anther culture of P. ginseng. Highest callus formation rate was obtained when flower buds pretreated at $4^{\circ}C$ for 1 day. Among the treated growth regulators with various degrees of concentration in Murashige and Skoog's (MS) medium, 4.53 ${\mu}M$ of 2.4-dichlorophenoxyacetic acid and 4.44 ${\mu}M$ of 6-benzylaminopurine gives the most responsive callus with the frequency of 73.89% and 129.53 g of fresh weight. When we used 3-9% of sucrose and maltose among the different kinds and various concentrations of carbohydrates, callus was formed highest 67.29% in the medium with 3% of sucrose. Shoots induced from callus supplemented with 28.9 ${\mu}M$ of gibberellic acid and rooted in Gamborg's B5 medium supplemented with 14.7 ${\mu}M$ of indole-3-butyric acid.

Microspore-derived Embryo Formation in Response to Cold Pretreatment, Washing Medium, and Medium Composition of Radish (Raphanus sativus L.)

  • Cold pretreatment, washing medium and composition of nutrient media may have marked effects on microspore embryogenesis. When microspores isolated from radish (Raphanus sativus L. cv. Gwanhun) flower buds were washed with Nitsch & Nitsch (NLN) medium liquid medium containing $130g{\cdot}L^{-1}$ sucrose (NLN-13), yields of microspore-derived embryos were greater than when using B5 liquid medium containing $130g{\cdot}L^{-1}$ sucrose. Microspore viability is known to decrease rapidly with storage; however, in this experiment, microspore viability was maintained for 24 h at $4^{\circ}C$ without media. Among the various medium concentrations used ($0.25{\times}$, $0.5{\times}$, $1.0{\times}$, $2.0{\times}$, and $4.0{\times}$ NLN liquid medium), $0.5{\times}$ NLN liquid medium induced the most efficient formation of microspore-derived embryos. In addition, microspore-derived embryos yields were greater when microspores were cultured in $0.5{\times}$ NLN liquid medium supplemented with $0.25{\times}$, $0.5{\times}$, and $1.0{\times}$ NLN microelements, compared to medium not supplemented with microelements. In this study, the highest yield of microspore-derived embryos was observed when the microspores derived from flower buds were washed using NLN-13 liquid medium and then cultured on $0.5{\times}$ NLN liquid medium supplemented with $0.25{\times}$ NLN microelements, followed by incubation at $25^{\circ}C$ for 30 days.

황산알미늄용액의 단판(單板) 전처리(前處理)에 따른 합판용(合板用) 요소수지(尿素樹脂) 도포량의 감소에 관한 연구(硏究) (The Study of the Veneer Pretreatment with Aluminum Sulfate Solution on the Reduction of Urea Formaldehyde Resin Spreading Content for the Manufacture of Plywood)

  • This study was carried out to examine the effect of pretreatment of veneer with aluminum sulfate solution for the manufacture of the kapur plywood on the reduction of urea resin spread. The results were as follows: 1. Specific gravities of plywoods made of the kapur veneers ranged from 0.83 to 0.84 and their moisture contents based on air dry weight showed from 10.6% to 11.4%. These met the Korean Industrial Standards. 2. In the case of dry shear strength for the 3rd class plywood, 40% reduction- spread met K S as well as normal urea resin spread. pH of the aluminum sulfate solution gave better result at pH 4.5 and pH 6 than at pH 3. For the manufacture of the 2nd class plywood, hot and cold water-wet shear strength show the best results at the following conditions; such as 20% reduction spread, pH 4.5 of aluminum sulfate solution and all veneer treatment.

Investigation into the mechanism of action of Moringa oleifera for its anti-asthmatic activity

  • In the present investigation, we studied the effect of alcoholic extract of Moringa oleifera (M. oleifera) seed kernels on various experimental models of bronchial asthma. Significant (P < 0. 05) increase in preconvulsion time was observed due to pretreatment with M. oleifera when the guinea pigs were exposed to either acetylcholine (Ach) or histamine aerosol. This bronchodilating effect of M. oleifera was comparable to ketotifen fumarate. Spasmolytic effect of M. oleifera was also observed by dose dependent inhibition of ideal contractions induced by Ach, 5HT, histamine and $BaCl_2$. Alcoholic extract of M. oleifera produced significant dose dependent protection by egg albumin and compound 48/80 induced mast cell degranulation. Pretreatment with alcoholic extract of M. oleifera also decreased carrageenan induced rat paw edema, which was comparable to that of standard diclofenac sodium. Minimum inhibitory concentration for alcoholic extract of M. oleifera was low as compared to cold-water extract and hot water extract when antimicrobial activity was tested against various respiratory pathogens like Escherichia coli (E. coli), Staphylococus aureus (S. aureus) and pseudomonas aeruginosa (P. aeruginosa). Our data suggest that antiasthmatic activity of M. oleifera seed kernels may be due to its bronchodilator, anti-inflammatory, mast cell stabilization and antimicrobial activity.

건조방법에 따른 삼투처리 단호박의 품질 특성 (Quality Characteristics of Osmotic Dehydrated Sweet Pumpkin by Different Drying Methods)

  • 단호박을 이용하여 새로운 가공제품을 개발하고자 여러가지 잇점을 줄 수 있는 삼투건조를 최적조건에서 처리하고 산업적으로 많이 사용되고 있는 열풍, 진공건조와 새로운 건조방법의 적용을 위해 선정한 냉풍건조를 행한 후 품질특성을 비교 평가하였다. 삼투처리 후 열풍, 진공, 냉풍건조된 단호박의 수분함량은 sucrose로 삼투처리한 경우가 모든 건조 방법에서 glucose 처리구보다 낮았으며, 건조방법에 따른 비타민 C의 함량은 glucose로 삼투처리 후 냉풍건조한 경우가 35.69 ㎎/100 g으로 가장 높게 측정되었다. 전반적으로 침지 용액에 따른 L, a, b값은 유사한 경향이었으며, 냉풍건조가 색도변화가 가장 작게 일어나 생단호박의 색상과 유사하였다. 건조 단호박의 표면구조는 삼투용액과 건조방법에 따라 상이하였는데, 삼투처리 후 냉풍건조된 단호박을 제외하고 열풍 및 진공건조된 단호박의 조직 손상이 크게 나타났다. 건조방법 및 건조시간에 따른 수분함량과 비타민 C함량을 살펴보면 수분함량의 경우 건조효율성을 고려할 때 sucrose로 삼투처리 후 진공건조한 단호박이 가장 우수하였으나, 비타민 C함량에서는 glucose로 삼투처리 후 냉풍건조한 단호박이 가장 양호하였다. 삼투용액에 따른 건조방법별 건조속도는 sucrose로 삼투처리한 단호박의 건조속도가 glucose 처리구보다 빠름을 알 수 있었다.

갈색거저리(Tenebrio molitor) 유충의 냉장 저장 중 산화 안정성에 관한 연구 (Studies on Oxidative Stability of Tenebrio molitor Larvae During Cold Storage)

  • The purpose of this study was to evaluate the changes on the characteristics of the oxidative stability of Tenebrio molitor larvae during cold storage at $4^{\circ}C$. Pretreatment for T. molitor larvae was designed into three methods: raw (R), freeze-dried (F.D.), and pan-fried (P.F.). The water content of the raw sample (61.46%) was higher than those of other samples (F.D.: 5.02%, P.F.: 3.67%) and its high water content was expected to facilitate the oxidation of the raw sample. In our results, the peroxide value and the carbonyl value of all of the samples increased and the raw sample, after storage for 18 day, showed the highest value. The pan-fried sample had no significant increase in its lactic acid content, acid value, and thiobarbituric acid value; whereas those values were increased in the raw sample and the freeze-dried sample (p<0.05). The browning reaction was more progressed in the pan-fried sample than other samples at 0 day, but there was no significant change during the storage. The raw sample and the freeze-dried sample had their browning indexes increase with the increasing storage period (p<0.05). The pan-fried sample produced less oxidation products than the freeze-dried sample, indicating that the unheated sample was more susceptible to oxidation than the heated samples. In conclusion, heating treatment and low water content would be effective for improving the safety and stability of T. molitor larvae during cold storage.

Chlorella의 인산대사에 관한 연구 (Studies on the Phosphate Metabolism in Chlorella, with Special Reference to Polyphosphate)

Uniformly $^{32}P$-labeled Chlorella cells which were irradiated with Cobalt-60 gamma-rays of about 70, 000 $\gamma$ dose, were further grown in a standard "cold" medium ("hot".rarw."cold"), and some portions of the algae were taken out at the begining of, and at intervals during the culture, and subjected to analyze the contents of $^{32}P$- and total P in various fractions of the cell materials. Results obtained were compared with those of nonirradiated normal cells. 2. Amounts of phosphate in various fractions of the nonirradiated normal Chlorella cells were measured using uniformly $^{32}P$--labeled cells. Analysis of the $^{32}P$--labeled algal cells showed that the highest value in P-content was the fraction of RNA followed by those of lipid, polyphosphate "C" polyphosphate "B", DNA, nucleotidic labile phosphate compounds, polyphosphate "A" and protein. It was observed that content of total polyphosphates in a single Chlorella cell was almost equal to RNA-P content in the cell, and the amount of RNA-P was almost equal to ten times of DNA-P content. 3. When the $^{32}P$--labeled algae which were irradiated with gamma-rays were grown in a normal "cold" medium, phosphate contents in the fraction of DNA, nucleotidic labile phosphate compounds and protein decreased markedly, while the contents of phosphate in the fractions of polyphosphate "C" and potyphosphate "B" increased in comparison with those of unirradiated normal cells. So, it was considered that the pretreatment of above mentioned dose of gamma-ray inhibited DNA and protein synthesis from polyphosphate in Chlorella cells. 4. Proceeding the culture of $^{32}P$--labeled Chlorella in a "cold" standard medium, whose synthetic activity of DNA and protein from polyphosphate was disturded by gamma-ray irradiation, the amounts of $^{32}P$-in the fraction of polyphosphate "C" increased, in contrast with those of polyphosphate "B" fraction. Involvement of the spinal γ-aminobutyric acid receptor in the analgesic effects of intrathecally injected hypertonic saline in spinal nerve-ligated rats

  • Background: Hypertonic saline is used for treating chronic pain; however, clinical studies that aid in optimizing therapeutic protocols are lacking. We aimed to determine the concentration of intrathecally injected hypertonic saline at which the effect reaches its peak as well as the underlying γ-aminobutyric acid (GABA) receptor-related antinociceptive mechanism. Methods: Spinal nerve ligation (SNL; left L5 and L6) was performed to induce neuropathic pain in rats weighing 250-300 g. Experiment 1: one week after implanting the intrathecal catheter, 60 rats were assigned randomly to intrathecal injection with 0.45%, 0.9%, 2.5%, 5%, 10%, and 20% NaCl, followed by behavioral testing at baseline and after 30 minutes, 2 hours, 1 day, and 1 week to determine the minimal concentration which produced maximal analgesia. Experiment 2: after determining the optimal intrathecal hypertonic saline concentration, 60 rats were randomly divided into four groups: Sham, hypertonic saline without pretreatment, and hypertonic saline after pretreatment with one of two GABA receptor antagonists (GABAA [bicuculline], or GABAB [phaclofen]). Behavioral tests were performed at weeks 1 and 3 following each treatment. Results: Hypertonic saline at concentrations greater than 5% alleviated SNL-induced mechanical allodynia and had a significant therapeutic effect, while showing a partial time- and dose-dependent antinociceptive effect on thermal and cold hyperalgesia. However, pretreatment with GABA receptor antagonists inhibited the antinociceptive effect of 5% NaCl. Conclusions: This study indicates that the optimal concentration of hypertonic saline for controlling mechanical allodynia in neuropathic pain is 5%, and that its analgesic effect is related to GABAA and GABAB receptors.
