Solubilization of Rock Phosphates by Alginate Immobilized Cells of Pantoea agglomerans (Alginate에 고정화된 Pantoea agglomerans에 의한 인광석 가용화)

  • Pantoea agglomerans immobilized in alginate solubilized four different rock phosphates efficiently under in vitro conditions. The solubilization pattern differed according to the rock phosphate source, where maximum solubilization of Morocco and Tunisia rock phosphates (215.6 and $186.1mg\;P\;L^{-1}$) on 6 days, Israel rock phosphate ($60.98mg\;P\;L^{-1}$) and tricalcium phosphate ($132.3mg\;P\;L^{-1}$) on 10 days and China rock phosphate ($48.8mg\;P\;L^{-1}$) on 12 days after inoculation was observed. The shelf life of the immobilized bacteria immobilized beads stored in two different temperatures was studied for six months. Beads stored at both room temperature as well as cold storage ($4^{\circ}C$) were found equally good in supporting the bacterial population as well as phosphate solubilizing activity. P. agglomerans immobilized in alginate might be exploited for large scale biosolubilization of rock phosphates intended for fertilizer use.

  • The purpose of this study was to investigate the effect of ultrasonic treatment during probiotics production process on the shelf life and the survival rate. Once Lactobacillus plantarum and Bifidobacterium longum were cultured in edible culture medium, ultrasonic treatment was performed at 100 Hz for 1, 2, 5, 7, and 10 minutes. Ultrasonic-treated L. plantarum HY7715 and B. longum HY8001 were centrifuged, mixed with a cryoprotectant, and lyophilized. The prepared lactic acid bacteria powder was individually packaged. After 6 months, viable cell counts were measured separately under cold storage and room temperature. In vitro digestion experiments were performed to determine the survival rate at digestive tract. As a result, it was observed that shelf life and survival rate were increased compared to untreated control group.

  • Experiments have been performed to investigate biological characteristics of Diglyphus isaea (Walker), an ectoparasite of Liriomyza trifolii (Burgess). At $25^{\circ}C$, development time of D. isaea was estimated as 1.3 days for eggs, 4.5 days for larvae and 6.3 days for pupae. Adult longevity was 25.1 days, and a female adult could lay 305.3 eggs throughout its lifetime. Number of L. trifolii parasitized or killed by the wasp was 962.2 individuals/adult. Most of all parasitized or killed larvae were identified as third instar. With 20% honeyed water as food source, half of the adults could survive 180 days at $5^{\circ}C$, 150 days at $10^{\circ}C$, and 90 days at $15^{\circ}C$. After storing at $10^{\circ}C$ with absolute honey for 30 days, 70 days and 140 days, a female adult in room temperature laid eggs 190.8 individuals, 104.0 individuals and 89.4 individuals, respectively.

A COMPARATIVE EXPERIMENTAL STUDY ON THE SURFACE CHARACTERISTICS AND THE FITTNESS OF THE RESILIENT DENTURE LINES (탄성 의치상 이장재의 표면 특성 및 적합도에 관한 비교 실험 연구)

  • The purpose of this investigation was to determine the surface characteristics and the fittness of the resilienct denture lines. Firstly, 50 samples ($2.0{\times}4.0{\times}0.3cm$) of 4 resilient lining materials (Molloplast B, Coe Super Soft, Mollosil, Coe Soft) and one conventional acrylic resin (K-33) were processed according to manufacture's direction and examined the surface characteristics by use of surface profilometer and scanning electron microscopy. Secondly, 50 identical maxillary casts were made and 50 denture bases were pro cessed of 4 resilient liners and one conventional acrylic resin and they were stored in the room temperature water bath of 1 day, 1 week, 2 weeks, 3 weeks, 4 weeks and 6 weeks after processing. The original casts were cut away 1 cm from the posterior border, the dentures were seated, and the existing space was measured at seven regions according to the storage time by use of the modified thickness guage. The results were as follows. 1. Surface roughness (Rz) were $4.00{\pm}1.60{\mu}m$ in Mollosil, $4.47{\pm}2.21{\mu}m$ in Molloplast B, $7.46{\pm}1.70{\mu}m$ in Coe Super Soft, $12.70{\pm}2.39{\mu}m$ in Coe Soft and $13.03{\pm}2.74{\mu}m$ in K-33. 2. The generation of porosity was far more active in cold-cured resilient liners (Coe Soft and Mollosil) than in heat cured resilient liners (Molloplast B, and Coe Super Soft) and conventional heat cured resin (K-33). 3. Denture bases showed the greatest discrepancy at the central portion of the posterior palatal border and the intimate contact in the buccal flange regardless of denture base materials. 4. When the denture bases were stored in the water for 1 day and 6 weeks after processing, the sum of average discrepancies in the seven regions of the denture base was the greatest in K-33 followed by Molloplast B, Mollosil, Coe Soft and Coe Super Soft but followed by Coe Soft, Molloplast B, Mollosil, Coe Super Soft in that order respectively. 5. There was not a significant difference (p>0.05) in Coe Super Soft, K-33 but there was a significant difference (P<0.01) in Molloplast B, Mollosil, Coe Soft at the amount of dimensional changes according to the storage time.

Bio-Green' Functional Water Supply Influences Mineral Uptake and Fruit Quality In Tsugaru Apples (‘바이오 그린’ 기능수 처리가 사과 쓰가루 품종의 무기성분 흡수와 과실품질에 미치는 영향)

  • Commercial Bio- Green(B.G.) functional water was manufactured through a series of processes : water - ultra-purification - adding catalysts - energy imprinting fermenting with energized water + zeolite and others + photosynthetic bacteria in fermenter longrightarrow filtering. Control(0), 5 or 10 liters per plant of B.G. functional water were supplied to the orchard soil under canopy of 10 year- old ‘Tsugaru’/M26 apple trees on March 20, May 20 and June 20, 1995, respectively. pH and content of Ca and Mg of orchard soil were increased by supply with B.G. functional water. However, P$_2$ $O_{5}$, K, and B contents were not influenced by the treatment. At harvest time soluble solid content of flesh tissue and anthocyanin of fruit skin were increased by the treatment. B.G functional water treatment showed higher root activities, and photosynthesis of leaves than that of control. Also B.G. functional water treatment enhanced Ca content in fruit skin and flesh tissues, whereas not affected N, K, and Mg contents. During storage at 4$^{\circ}C$ cold room, the more volume of B.G. functional water supply showed lower bitter pit symptom. Respiration and ethylene evolution in fruit decreased, while fruit firmness increased by the treatment during storage.

  • Peak cooling load of large buildings is generally greater than their peak heating load. Internal and solar heat gains are used fur selection of adquate equipment in large building in cold winter climate like Canada and even Korea. The cost of geothermal heat exchanger to meet the cooling loads can increase the initial cost of ground source heat pump system to the extend less costly conventional system often chosen. Thermal ice storage system has been used for many years in Korea to reduce chiller capacity and shift Peak electrical time and demand. A distribution system designed to take advantage of heat extracted from the ice, and use of geothermal loop (geothermal heat exchanger) to heat as an alternate heat source and sink is well known to provide many benifits. The use of thermal energy storage (TES) reduces the heat pump capacity and peak cooling load needed in large building by as much as 40 to $60\%$ with less mechanical equipment and less space for mechanical room. Additionally TES can reduce the size and cost of the geothermal loop by 1/3 to 1/4 compared to ground coupled heat pump system that is designed to meet the peak heating and cooling load and also can eliminate difficuties of geothermal loop installation such as space requirements and thermal conditions of soil and rock at the urban area.

Difference in Freshness of Soybean Sprouts as Affected by $\textrm{CO}_2$ Concentration and Postharvest Storage Temperature (콩나물 재배과정 중 기체 조성과 수확 후 저장온도에 따른 선도 변화)

  • When soybean sprouts aye grown in the closed condition (where the ratio of $\textrm{O}_2$ and $\textrm{CO}_2$ is 7 : 3), amount of $\textrm{CO}_2$ is increased and $\textrm{O}_2$ is decreased with the passage of time. At the same time, the amount of ethylene is automatically increased. By increasing the concentration of ethylene gas up to 0.5-1.0 ppm in the growth room, the length of sprouts was restricted to 6-7 cm and the thickness of sprouts was increased to 2.70$\pm$0.30 mm. The production of good quality sprouts which were fat and short was possible without application of any growth regulators such as indole-3-acetic acid known to have accumulation problem in humane body. To maintain the freshness during the transportation and prevent sprouts from rotting and bad smell at market, cold storage at 2-$5^{\circ}$ and airtightness which will restrict photosynthesis and respiration (higher than $10^{\circ}$) are needed. The freshness of sprouts is depended on the increase of $\textrm{CO}_2$ and the depletion of $\textrm{O}_2$ in the package. When the sprouts were stored below 1$0^{\circ}C$ (preferably below 8$^{\circ}C$), the concentration of $\textrm{CO}_2$ in the package remained below 30% for more than 60 hours, which was possible to keep sprouts in freshness without any offensive odor, But sprouts were maintained at $13^{\circ}$ for more than 25 hours, the concentration of $\textrm{CO}_2$ increased over 30% and produced an offensive odor. The little amount of $\textrm{O}_2$ gas was existing for 30 hours at $5^{\circ}$ but it was disappeared completely within 7 hours over $10^{\circ}$ and the sprouts became rot and produced severe offensive odor.

Characterization of quality changes of whole super sweet corn (Zea mays saccharata Sturt.) during thermal sterilization for shelf-stable products (상온유통을 위한 가열살균 중의 통 초당옥수수의 품질변화 연구)

  • This study investigated the quality changes in whole super sweet corn during thermal processing to extend its shelf-life. To minimize the reduction of unique texture of whole sweet corn after the sterilization, the alcohol sanitation applied and the cold point of a whole corn ear was determined using a computer simulation. The cold point was located between the corn kernel and the cob. The microorganisms on the surface of sweet corn were reduced by more than 1 log CFU/g after alcohol sanitation, then the whole corn was treated to satisfy the degree of sterilization ($F_{121.1}=4$). The quality of sterilized sweet corn was compared with the control that was treated with steaming. The quality changes of sterilized sweet corn during storage were monitored for 9 months at $25^{\circ}C$. The hardness was maintained within 30% of its initial value. The minimum of hardness was $464.50{\pm}103.35g$ and maximum of hardness was $514.50{\pm}81.83g$. The differences in the sugar content among the samples were found, but the sugar content of corn kernel remained within 30% of the control, ranging from $28.83{\pm}1.05$ to $34.36{\pm}0.42%$. The yellowness was higher than that of control by 5%. The maximum value of yellowness was $34.36{\pm}0.42$. The general bacteria and molds and yeasts in corn kernel stored at $25^{\circ}C$ were not detected after 9 months of storage at $25^{\circ}C$. Therefore, in this study, we have demonstrated that the thermal sterilized method extends the shelf-life of whole sweet corn with minimizing its quality changes over 6 months in room temperature.

Acidic and steaming treatments of tteokbokki rice cake to improve its microbial and textural properties (떡볶이 떡의 미생물 안전성 및 식감 향상을 위한 유기산과 증숙 처리)

  • To improve the storage and firmness of Tteokbokki tteok Korean-style rice cakes with spicy sauce, steaming of the rice dough, soaking of the rice cake in acidic solution, and heat sterilization were conducted sequentially. The untreated control could be stored for 1 month under cold conditions, but this was extended to 10 months at room temperature ($15-30^{\circ}C$) after immersion in lactic acid solution (pH 4.0) for 20 min and heating to $100^{\circ}C$ for 30 min. The mechanical strength, which was related to firmness, was significantly increased to $11.4kgm/s^2$ compared with the untreated rice cakes ($8.8kgm/s^2$, p<0.05) and the overall descriptions of the sensory evaluation, including texture, was significantly higher than the control (p<0.05). The average size of the pores in the treated rice cakes was smaller than that of the conventional sample and the texture was improved as a result of decreased starch elution from smaller pores.

Biological Control of Soil-borne Diseases with Antagonistic Bacteria

  • Biological control has many advantages as a disease control method, particularly when compared with pesticides. One of the most important benefits is that biological control is an environmental friendly method and does not introduce pollutants into the environment. Another great advantage of this method is its selectivity. Selectivity is the important factor regarding the balance of agricultural ecosystems because a great damage to non target species can lead to the restriction of natural enemies' populations. The objective of this research was to evaluate the effects of several different bacterial isolates on the efficacy of biological control of soil borne diseases. White rot caused by Sclerotium cepivorum was reported to be severe disease of garlic and chive. The antifungal bacteria Burkholderia pyrrocinia CAB08106-4 was tested in field bioassays for its ability to suppress white rot disease. In field tests, B. pyrrocinia CAB08106-4 isolates suppressed white rot in garlic and chive, with the average control efficacies of 69.6% and 58.9%, respectively. In addition, when a culture filtrate of B. pyrrocinia CAB08106-4 was sprayed onto wounded garlic bulbs after inoculation with a Penicillium hirstum spore suspension in a cold storage room ($-2^{\circ}C$), blue mold disease on garlic bulbs was suppressed, with a control efficacy of 79.2%. These results suggested that B. pyrrocinia CAB08106-4 isolates could be used as effective biological control agents against both soil-borne and post-harvest diseases of Liliaceae. Chinese cabbage clubroot caused by Plasmodiophora brassicae was found to be highly virulent in Chinese cabbage, turnips, and cabbage. In this study, the endophytic bacterium Flavobacterium hercynium EPB-C313, which was isolated from Chinese cabbage tissues, was investigated for its antimicrobial activity by inactivating resting spores and its control effects on clubroot disease using bioassays. The bacterial cells, culture solutions, and culture filtrates of F. hercynium EPB-C313 inactivated the resting spores of P. brassicae, with the control efficacies of 90.4%, 36.8%, and 26.0%, respectively. Complex treatments greatly enhanced the control efficacy by 63.7% in a field of 50% diseased plants by incorporating pellets containing organic matter and F. hercynium EPB-C313 in soil, drenching seedlings with a culture solution of F. hercynium EPB-C313, and drenching soil for 10 days after planting. Soft rot caused by Pectobacterium carotovorum subsp. carotovorum was reported to be severe disease to Chinese cabbage in spring seasons. The antifungal bacterium, Bacillus sp. CAB12243-2 suppresses the soft rot disease on Chinese cabbage with 73.0% control efficacy in greenhouse assay. This isolate will increase the utilization of rhizobacteria species as biocontrol agents against soft rot disease of vegetable crops. Sclerotinia rot caused by Sclerotinia sclerotiorum has been reported on lettuce during winter. An antifungal isolate of Pseudomonas corrugata CAB07024-3 was tested in field bioassays for its ability to suppress scleritinia rot. This antagonistic microorganism showed four-year average effects of 63.1% of the control in the same field. Furthermore, P. corrugata CAB07024-3 has a wide antifungal spectrum against plant pathogens, including Sclerotinia sclerotiorum, Sclerotium cepivorum, Botrytis cinerea, Colletotrichum gloeosporioides, Phytophotra capsici, and Pythium myriotylum.
