Inhibitory Effects of Methanol Extracts from Korean Orysa sartiva and Coix lachryma-jobi var. ma-yuen on Mutagenicity and Growth of Human Cancer Cells (한국산 현미 및 율무 추출물에 의한 돌연변이 및 인체 암세포주 증식 억제 효과)

  • This study was carried out to determine the inhibitory effects of methanol extracts from Korean Orysa sartiva and Coix lachryma-jobi var. ma-yuen on mutagenicity using Ames test and growth of AGS human gastric adenocarcinoma and HT-29 human colon cancer cells. Both treatments of methanol extracts (5 mg/assay) from Orysa sartiva and Coix lachryma-jobi var. ma-yuen to Ames test system inhibited aflatoxin $B_1$ ($AFB_1$) induced mutagenicity by 76%. In case of N-methyl-N'-nitro-N-nitrosoguamidine (MNNG) induced mutagenicity, the methanol extracts (5 mg/assay) from Orysa sartiva and Coix lachryma-jobi var. ma-yuen showed 79% and 69% inhibitory rate, respectively and the inhibitory effect was a little stronger in Orysa sartiva Inhibitory effects of methanol extracts from Orysa sartiva. and Coix lachryma-jobi var. ma-yuen on the growth of AGS and HT-29 human cancer cells were increased as dose dependent patterns and the inhibitory effects on AGS and HT-29 cells were similar. The above results indicate that the consumption of these cereals, which contain many nutrients with good quality, may be recommended as potent functional foods for improving health.

Effect of the Seeds of Coix Lachryma-Jobi L. var. Ma-yuen Stapf. on the Proliferation of L1210 cells, a Leukemic cell-lines (율무가 백혈병세포주인 L1210 세포의 증식에 미치는 영향)

  • The purpose of this research was to investigate the effects of Coix Lachryma-Jobi L. var. Ma-yuen Stapf. on the proliferation of L1210 cells and immune cells. The hexane fraction of Coix Lachryma-Jobi decreased the proliferation of L1210 cells and induced DNA fragmentation of L1210 cells. Also, the fraction decreased the cell viability of murine thymocytes and splenocytes, but increased the phagocytic activity of murine peritoneal macrophages. In addition, fatty acids and fatty acid methyl esters decreased the proliferation of L1210 cells and induced DNA fragmentation of L1210 cells. The main components of the fraction are fatty acid and their derivatives. These results indicate that Coix Lachryma-Jobi has a cytotoxicity on tumor cells and immune cells.

Validation of Method Determining Coixol in Coix lachryma-jobi var. ma-yuen Roots Extract (율무근 추출물의 Coixol 성분 분석법 검증)

  • An high performance liquid chromatography (HPLC) analysis method was developed for standard determination of coixol as a functional cosmetic material in Coix lachryma-jobi var. ma-yuen roots extract. HPLC was performed on a $C_{18}$ Unison US column ($4.6{\times}250mm$, $5{\mu}m$ column) using a gradient elution of 0.1% (v/v) trifluoroacetic acid and acetonitrile at a flow rate of 1.0 mL/min at $30^{\circ}C$. The analyte was detected at 290 nm. The HPLC method was validated in accordance with the International Conference on Harmonization guideline of analytical procedures with respect to specificity, precision, accuracy, and linearity. The limit of detection and quantitation were 0.07 and 0.25 mg/mL, respectively. Calibration curves showed good linearity ($R^2$>0.9995), and the precision of analysis was satisfied (less than 0.29%). Recoveries of quantified compounds ranged from 98.36 to 100.30%. This result indicates that the established HPLC method is very useful for the determination of a marker compound in C. lachryma-jobi var. ma-yuen roots extracts.

Phytochemical constituents of Coix lachryma-jobi var. ma-yuen roots and their tyrosinase inhibitory activity (율무근의 식물화학적 성분 연구 및 Tyrosinase 저해 활성)

  • In the course of screening tyrosinase inhibitory activity, EtOAc-soluble fraction of Coix lachryma-jobi var. mayuen Stapf. (Gramineae) roots showed significant inhibition. Further fractionation of the EtOAc-soluble fraction resulted in six compounds, which were identified as (+)-icariol $A_2$ (1), zhepiresionol (2), 4-hydroxybenzaldehyde (3), trans-${\rho}$-coumaric acid (4), N-(2-hydroxy-4-methoxyphenyl)-2-hydroxyacetamide (5), and coixol (6). The chemical structures of these compounds were identified on the basis of spectroscopic methods (MS, 1D and 2D NMR) and comparison with literature values. Compound 1 was first reported from this plant. Also, this is the first time that the isolation of compound 5 has been reported from nature source. Among the isolated compounds, compounds 4 and 6 showed enzyme inhibitory activity, with $IC_{50}$ values of 6.5 and $62.4{\mu}M$, respectively, in comparison with these of positive control, arbutin.

Electrophoretic Characterization of Job′s tears (Yulmoo: Coix lachryma-jobi L. var. Ma-yuen stapf. & Yeomjoo: Coix lachryma-jobi L.) proteins (율무와 염주 단백질의 전기영동 특성)

  • This study was performed to examine the electrophoretic properties of Job's tears (Yulmoo: Coix lachryma-jobi L. var. Ma-yuen Stapf. & Yeomjoo: Coix lachryma-jobi L.) proteins. Albumins, globulins, gliadins and glutelins were extracted from the polished Yulmoo and brown Yeomjoo by the modified Osborne method. For a comparison, rice proteins were extracted and fractionated by the same method. The relative proportions of protein fractions were 17.4 : 19.6 : 55.2 : 7.7% in polished Yulmoo, 12.6 : 62.2 : 4.2 :21.0% in brown Yeomjoo and 14.2 : 57 4 : 0.77 : 27.8% in rice, in the order of albumis, globulins, gliadins and glutelins. Polyacrylamide gel electrophoresis (PAGE) and SDS-polyacrylamide gel electrophoresis (SDS-PAGE) were peformed to identify the subfractions of each protein fraction extracted from polished Yulmoo, brown Yeomjoo and rice. The electro-phoregrams of polyacrylamide gel electrophoresis showed that the same fractions of both polished Yulmoo protein and brown Yeomjoo protein had very similar electrophoretic patterns to each other respectively, but there were significant differences in the patterns between Job's tears proteins and rice proteins.

Tyrosinase-inhibitory and Radical Scavenging Activities from the Seeds of Coix lachryma-jobi L. var. ma-yuen [Roman] Stapf (율무의 tyrosinase 저해활성과 radical 소거효과)

  • The seeds of Coix lachryma-jobi L. var. ma-yuen(yullmoo) were investigated for the free radical scavenging and mushroom tyrosinase inhibitory activities. Brown yullmoo, which contains bran in the seed, has also been evaluated for comparison. After partitioning of the total methanol extracts of yullmoo and brown yullmoo, each organic layer exhibited mild or potent dose-dependent inhibition(about 20-80%) on 1,1-diphenyl picryl hydrazyl(DPPH) free radical with the concentration of $300\;{\mu}g/ml$ to $800\;{\mu}g/ml$. n-Butanol fraction of yullmoo showed the most potent scavenging effect on DPPH free radical. $IC_{50}$ of n-butanol fraction was $423\;{\mu}g/ml$, about six to seven fold higher concentration than standard BHT at the same inhibition rate. As for the tyrosinase inhibitory activity, all of the fractions including the methanol extract exhibited the similar activities even after partitioning. The chloroform fraction of brown yullmoo was found to show the most potent inhibitory activity with an $IC_{50}$ of $321.5\;{\mu}g/ml$.

Binding of Calcium and Iron by Job′s tears (Yulmoo: Coix lachryma-jobi L. var. Ma-yuen Stapf.) Total Dietary Fiber and Acid Detergent Fiber (율무 식이섬유의 칼슘, 철분과의 결합특성)

  • This study was performed to examine the mineral binding properties of total dietary fiber (TDF) and acid detergent fiber (ADF) from Job's tears (Yulmoo: Coix lachryma-jobi L. var. Ma-yuen Stapf.) bran. The endogenous Ca and Fe contents of dietary fiber extracted from Yulmoo bran were measured by atomic absorption spectrophotometer (AAS). Ca contents were 40, 118.6$\mu\textrm{g}$ Ca/g TDF and 131.6 $\mu\textrm{g}$ Ca/g ADF. Fe contents were 1,219.7 $\mu\textrm{g}$ Fe/g TDF and 277.5 $\mu\textrm{g}$ Fe/g ADF. The calcium binding capacities of dietary fiber at pH 5.0, 6.0, 7.0 were 50.77, 77.57, 86.64% for TDF, 40.27, 51.42, 40.90% for ADF, respectively. The iron binding capacity of dietary fiber at pH 5.0~7.0 was 95~97% for TDF and 85~95% for ADF.

The Effects of Coix Bran on Lipid Metabolism and Glucose Challenge in Hyperlipidemic and Diabetic Rats (율무겨 급여가 고지혈증 및 당뇨유발 백서의 지질대사와 당내성에 미치는 영향)

  • Even though coix (Coix lachryma-jobi L. var. ma-yuen) has many physiological effects, since it has been known to cause sterility, farmers avoid using coix bran as a forage for their livestock. Therefore, as the consumption of coix increases, coix bran, which is a by product of pounding process, becomes a serious issue of environmental problem. Present study examined the physiological effects of coix bran in normal and diabetic rats for its possible use as a functional material. The effects of coix bran supplementation on plasma and hepatic lipid profile were evaluated in rats fed one of the following diet for 12 weeks : chow diet ; chow-bran diet (chow diet+25% coix bran), high fat diet and high fat-bran diet (high fat diet+25% coix bran). Additionally, glucose challenge and lipid metabolism in streptozotocin-diabetic rats were also examined. In normal rats, consumption of coix bran remarkably reduced body weight gain in chow or high fat diet fed rats. Additionally, consumption of coix branreduced blood TG, TC and atherogenic index (26%, 24% and 72%, respectively) in chow diet fed rats. Liver TG and cholesterol concentrations were reduced (43% and 49%, respectively) in high fat fed rats by coix bran supplementation. In diabetic rats, fasting blood glucose level was reduced about 25% by coix bran consumption. Also, glucose challenge pattern was improved and resembled normal pattern : it reaches to peak 15~30 minutes after glucose administration and get back to fasting blood glucose level after 90 minutes. Plasma concentrations of TG were elevated in diabetic rats and were reduced to normal level by coix bran supplementation. Liver TG and cholesterol concentrations were also elevated in diabetic rats and reduced to normal level by consumption of coix bran. These results suggest that coix bran may have beneficial effects on blood lipid and glucose level in normal and diabetic rats.

Cytotoxicity and Multidrug -Resistance Reversing Activity of Extracts from Gamma-Irradiated Coix Zachryma-jobi L. var. ma-yuen Stapf Seed (감마선 조사된 율무종자의 세포독성 및 다제내성 극복활성)

  • This study was carried out to examine the effects of gamma irradiation on the cytotoxicity and multidrug-resistance reversing activity of methanol extracts from Coix lachryma-jobi L. var. me-yuen Stapf seed. The seed was irradiated with doses of 1, 4, 8, 16, 32 and 64 Gy of the gamma radiation, and then extracted by methanol. The extracts were examined for cytotoxicity on the human cancer cell lines, MCF-7 (human breast adenocarcinoma pleural effusion), Calu-6 (human pulmonary carcinoma) and SNU-601 (human gastric carcinoma) cells, and investigated for multidrug-resistance reversing activity using drug sensitive AML-2/WT and multidrug-resistant AML-2/D100 cells. The growth inhibitory activity of irradiated seed extracts on human cancer cell lines was higher than that of the control. In the case of Calu-6 cell line, the effect of cytotoxicity was observed in the extracts of 4, 8 and 16 Gy. $IC_{50}$ value in the MCF-7 cell line was measured in the only 8 Gy extract. And in the SNU-601 cell line as Calu-6, the effect of cytotoxicity was observed in the extracts of 4, 8 and 16 Gy. But the extracts of gamma-irradiated seed over 32 Gy showed little growth inhibitory effect against human cancer cell lines. In this result, 8 Gy extract had significant growth inhibitory in all human cancer cell lines $(Calu-6:\;633\;{\mu}g/mL,\;MCF-7:\;653\;{\mu}g/mL\;and\;SNU-601:\;683\;{\mu}g/mL)$. The extracts of 4, 8 and 16 Gy strongly potentiated vincristine cytotoxicity in AML-2/D100 cells. The reversal fold (RF) of 4, 8 and 16 Gy extracts was 1.7, 1.8 and 1.6, respectively. But their cytotoxicities to both sensitive AML-2/WT and resistant AML-2/D100 cells were in the same order of magnitude. These results indicate that the above samples would contain some principles which have cytotoxicity and multidrug-resistance reversing activity. Irradiation technology can be applied to promote physiological activities of medicinal plant seeds.
