Mastitis Diagnostics by Near-infrared Spectra of Cows milk, Blood and Urine Using SIMCA Classification

  • Constituents of animal biofluids such as milk, blood and urine contain information specifically related to metabolic and health status of the ruminant animals. Some changes in composition of biofluids can be attributed to disease response of the animals. Mastitis is a major problem for the global dairy industry and causes substantial economic losses from decreasing milk production and reducing milk quality. The purpose of this study was to investigate potential of NIRS combined with multivariate analysis for cow's mastitis diagnosis based on NIR spectra of milk, blood and urine. A total of 112 bulk milk, urine and blood samples from 4 Holstein cows were analyzed. The milk samples were collected from morning milking. The urine samples were collected before morning milking and stored at -35$^{\circ}C$ until spectral analysis. The blood samples were collected before morning milking using a catheter inserted into the carotid vein. Heparin was added to blood samples to prevent coagulation. All milk samples were analyzed for somatic cell count (SCC). The SCC content in milk was used as indicator of mastitis and as quantitative parameter for respective urine and blood samples collected at same time. NIR spectra of blood and milk samples were obtained by InfraAlyzer 500 spectrophotometer, using a transflectance mode. NIR spectra of urine samples were obtained by NIR System 6500 spectrophotometer, using 1 mm sample thickness. All samples were divided into calibration set and test set. Class variable was assigned for each sample as follow: healthy (class 1) and mastitic (class 2), based on milk SCC content. SIMCA was implemented to create models of the respective classes based on NIR spectra of milk, blood or urine. For the calibration set of samples, SIMCA models (model for samples from healthy cows and model for samples from mastitic cows), correctly classified from 97.33 to 98.67% of milk samples, from 97.33 to 98.61% of urine samples and from 96.00 to 94.67% of blood samples. From samples in the test set, the percent of correctly classified samples varied from 70.27 to 89.19, depending mainly on spectral data pretreatment. The best results for all data sets were obtained when first derivative spectral data pretreatment was used. The incorrect classified samples were 5 from milk samples,5 and 4 from urine and blood samples, respectively. The analysis of changes in the loading of first PC factor for group of samples from healthy cows and group of samples from mastitic cows showed, that separation between classes was indirect and based on influence of mastitis on the milk, blood and urine components. Results from the present investigation showed that the changes that occur when a cow gets mastitis influence her milk, urine and blood spectra in a specific way. SIMCA allowed extraction of available spectral information from the milk, urine and blood spectra connected with mastitis. The obtained results could be used for development of a new method for mastitis detection.

  • Object When angiogenesis is excessive, Cancer, RA, Blindness, Psoriasis, Hemangioma, Diabetic retinopathy, Granulation, etc are induced. On the contrary, when it is insufficient, Stroke, Heart disease, Ulcer, Infertility, Scleroderma, artherosclerosis, delay of the wound recovery, etc occur. In recently, the methods which is control of abnormal angiogenesis are researching actively in relathion to anticancer research. This study is search for effective drugs which suppress this angiogenesis, in the ingredients of the herbs that invigorate and dispel blood stasis using to treat intravascular coagulation in the oriental herbal medicine. Methods We maked 80 % methanole extracts of Cnidii Rhizoma, Olibanum, Myrrha, Corydalidis Tuber, Curcumae Radix, Curcumae longe Rhizoma, Zedoariae Rhizoma, Salviae miltiorrhizae Radix, Polygoni cuspidati Rhizoama, Leonuri Herba, Persicae Semen, Carthami Flos, Trogopterorum Faeces, Achyranthis Bidentatae Radix, Manitis Squama, Eupolyphaga, Hirudo, Tabanus, Lycopi Herba, Artemisiae anomalae herba, Vaccariae Semen, Sappan Lignum, Gleditsiae Spina, Draconis Resina, Leonunari Semen, Selaginelliae Folium, Spatholobi Caulis, and these extracts were tested for MTT viabilaty test, BrdU incorporation, Tube foramtion assay on ECV304(immotalized human umbilical vein endothelial cell) at the concentration of $50{\mu}g/ml$, $100{\mu}g/ml$, $200{\mu}g/ml$, $400{\mu}g/ml$ Results All extracts except Draconis Resina have no cytotoxicity at the $100{\mu}g/ml$, and in BrdU incorporation test, proliferation rate were reduced below 60% at the concentaraion of $100{\mu}g/ml$ by Zedoariae Rhizoma, Sappan, Lignum Gleditsiae, Spina Draconis Resina Vaccariae Semen. Zedoariae Rhizoma Sappan Lignum Gleditsiae Spina Draconis Resina Vaccariae Semen Olibanum, Achyranthis Bidentatae Radix showed inhibition effects on tube formation of ECV304 at the concentration of $100{\mu}g/ml$. Conclusion At the concentration of $100{\mu}g/ml$ in which cytotoxicity is not found, Zedoariae Rhizoma, Sappan Lignum, Gleditsiae Spina, Vaccariae Semen showed the inhibition effect on proliferation and tubeformation of ECV304.

Phosphorus Removal from Advanced Wastewater Treatment Process Using PAC (PAC를 이용한 하수의 고도처리공정에서의 인 제거)

  • To meet the reinforced discharge standards, effect of coagulant PAC (Poly aluminium chloride, 10.4% as $Al_2O_3$) on phosphorous removal in advanced wastewater treatment process (a modified $A^2$/O). 15 mg/L of PAC determined by jar-test was added to influent of settling basin in a modified $A^2$/O consists of anaerobic, anoxic, and oxic chamber which contains Bio-clod and porous polyurethane media. Performance of PAC was tested by supernatant after settling. The removal efficiencies of BOD, COD, TP (total phosphorus) and SP (soluble phosphorus) on biological process with PAC were 96.1%, 88.8%, 97.0% and 98.6%, compared with those on biological process without PAC were 95.4%, 72.4%, 71.6% and 59.5% respectively. 18.4% of TP and 39.1% of SP removal efficiency was increased, although increase of BOD and COD removal rate was not significant. Only PAC addition to influent of settling basin in $A^2O$ process can help total phosphorus removal to 0.13 mg/L with following discharge standard.

The Basic Study of Cheong-song Mineral Water for Tofu Coagulant (청송약수를 이용한 두부응고에 관한 기초연구)

  • This is the basic study of Cheong-song mineral waters(Dalgi and Sinchon) for a coagulant of Tofu. For comparison, $CaCl_2,\;CaSO_4,\;MgCl_2\;and\;MgSO_4$ are used as reference coagulants. The coagulability test was conducted by measuring the percentage transmittancy of filtered whey solution at 600 nm wavelength, in $80\;{^{\circ}C}$ and 10 mL of solution volume which composed of 2 mL of soy milk and coagulants. The coagulability test of mineral waters require more than an aliquot of 8 mL. Good result obtained by adding 0.4, 1, 0.8, 0.8 mL of reference coagulants and this is equivalent to the amount of 66.14, 232.83, 95.68, 78.88 mg of Ca and Mg respectively. The percentage transmittancy of filtered whey solution with added 8 mL of Dalgi and Sinchon mineral waters showed 50.74 and 58.52 respectively and the amount of mineral content of Ca and Mg equivalent to 2.41 and 2.48 mg respectively which is very lower value than the reference coagulants. This means mineral water has coagulation ability even in the low concentration.

Effect of a Pulsed Nd:YAG laser irradiation on human gingival tissues (파동형 Nd:YAG 레이저조사가 인체 치은조직에 미치는 영향)

  • The purpose of this study was to determine the effect of a pulsed Nd:YAG laser irradiation on human gingival tissues. The patients, who were planned to be treated by clinical crown lengthening procedure and gingivectomy, were selected. All the patients received oral hygiene instruction, scaling and root planing at preoperation. The crest of gingival tissue on upper and lower anterior teeth was irradiated by a pulsed Nd:YAG laser(El. EN. EN060, Italy) with a fiber optic of 300 m in contact mode for 20 seconds. Gingival tissues were divided into 4 groups according to the laser power of 1.0W(10Hz, 100mJ), 2.0W(20Hz, 100mJ), 3.0W(30Hz, 100mJ) and 4.0W(40Hz, 100mJ). Immediately after the laser irradiation, the specimens were excised, fixed 10% neutral formalin, sectioned $4-6{\mu}m$ thick, stained by Hematoxylin-Eosin and Periodic Acid Schiff stain and observed under light microscope. The removed tissue depth and the coagulated layer depth due to a laser irradiation by a laser irradiation were measured on the microphotographs. The difference of measurements according to the different laser power was statistical1y analyzed by Kruskal Wallis Test with SAS program. The results were as follows : 1. In histologic findings of irradiated gingival tissues; a. In the irradiated gingival specimen with 1.0W laser power, some vesicles were observed in limited superficial layer of gingival epithelium. b. In the irradiated gingival specimen with 2.0W and 3.0W laser power, the epithelium was almost removed except for the traces of viable basal cell remnants at ret peg, and coagulation necrosis related with the thermal effect of laser was noted. c. In the irradiated gingival specimen with 4.0W laser power, complete removal of epithelium, partial removal of underlying connective tissue, and the coagulation necrosis of subjacent gingival tissue were shown. 2. The removed tissue depth was deeper in the irradiated specimens with higher power. There was a statistical significance in the difference of removed tissue depth between 1.0W group ($44.54{\pm}6.99um$) and 3.0W group ($99.75{\pm}6.64{\mu}m$), and between 1.0W group($44.54{\pm}6.99{\mu}m$) and 4.0W group($111.36{\pm}4.50{\mu}m$), and between 2.0W group($98.01{\pm}4.53{\mu}m$) and 4.0W group($111.36{\pm}4.50{\mu}m$)(P<0.05). 3. The coagulated layer depth was deeper in the irradiated specimens with higher power. There was a statistical significance in the difference of coagulated layer depth between 1.0W group($31.82{\pm}8.99{\mu}m$) and 3.0W group($55.99{\pm}20.94{\mu}m$), and between 1.0W group($31.82{\pm}8.99{\mu}m$) and 4.0W group($83.68{\pm}10.34{\mu}m$)(P<0.05). From this study, the results demonstrated that the effects of a pulsed Nd:YAG laser irradiation on gingival tissues seemed to depend on the laser power and that the irradiation with high power could be harmful to adjacent healthy tissue.

Effect of Korean Red Ginseng on Psychological Functions Patients with Severe Climacteric Syndromes : A Comprehensive Study from the Viewpoint of Traditional KAMPO-medicine and Western Medicine

  • Objective; Antistress effect of Korean red ginseng (RG) on postmenopausal women with severe climacteric syndrome (CS) were evaluated from the viewpoint of traditional KAMPO-medicine and Western medicine. Methods; All patients with CS were treated with daily oral administration of 6g RG for 30 days. Nine patients with CS were evaluated with the use of diagnostic scores for KI-deficiency (deficiency of vital energy) and OKETSU (blood stagnation) syndrome from the viewpoint of KAMPO-medicine. In the same patients with CS, peripheral blood levels of ${\beta}$-endorphin and total plasminogen activator inhibitor-1 (t-PAI-1) were measured before and after treatment with RG. In another group, 12 patients with CS, psychological test using CMI, STAI and SDS were performed from the viewpoint of Western medicine. Stress related hormones, such as ACTH, cortisol and DHEA-S in those 12 patients with CS were also measured before and after treatment with RG. Results; KI-deficiency score and OKETSU score in patients with CS were significantly (p<0.001) higher than those in patients without CS. After treatment with RG, both scores were markedly (p<0.001) decreased compared to before treatment with RG. ${\beta}$-endorphin levels in patients with CS were significantly (p<0.05) higher than those in patients without CS. Total PAI-levels in patients with CS were increased before treatment with RG. No significant difference, however, were observed between patients with and without CS. After treatment with RG, both levels of ${\beta}$-endorphin and total PAI-1 in patients with CS were significantly (p<0.001 and p<0.05, respectively) decreased compared to before treatment with RG. CMI and STAI scores in patients with CS were significantly (p<0.05) higher than those in patients without CS. SDS scores in patients with CS were also markedly (p<0.00l) higher than in those without CS. After treatment with RG, all scores decreased within normal range. DHEA-S levels in patients with CS were about a half of those without CS. Consequently, cortisol/DHEA-S (C/D) ratio was significantly(p<0.001) higher in patients with CS than in those without CS. Although the decreased DHEA-S levels were not restored to the levels in patients without CS, the C/D ratio decreased significantly (p<0.05) after treatment with RG. Conclusion; Reinforcement of vital energy and improvement of stagnant blood circulations by oral administration of RG were elucidated from the viewpoint of traditional KAMPO-medicine. From the viewpoint of Western medicine, effect of RG on postmenopsusal women with CS seemed to be brought about in part by not only an improvement of psychoneuroendocrine dysfunctions but also an amelioration of blood coagulation systems.

T-P Removal Efficiency According to Coagulant Dosage and Operating Cost Analysis (응집제 투입에 따른 인 제거 효율 및 운영비용 분석)

  • T-P removal efficiency was analyzed according to the metal to initial T-P ratio (mole basis) with respect to the samples from different WWTPs having various initial T-P and SS conditions. Also, operating costs were calculated based on the injected coagulant amount and the amount of sludge production. Most experiments were conducted by the standard jar-test protocol. Molar ratio of coagulant dose was varied considerably according to the initial SS concentration range in secondary clarifier effluent samples which had above 0.5 mg/L of initial T-P. Based on 90% T-P removal efficiency, results were: At the initial SS range of below 10 mg/L, Alum (8%) = 11 mol Al/mol P needed and PAC (17%) = 9.6 mol Al/mol P needed; At the initial SS range of above 10 mg/L, Alum (8%) = 3.9 mol Al/mol P needed and PAC (17%) = 3.2 mol Al/mol P needed.

The Physico-chemical and Sensory Properties of Milk with Water Soluble Chitosan (수용성 Chitosan을 첨가한 우유의 이화학적 및 관능적특성)

  • Attempts were made to evaluate possibilities of adding water soluble chitosan to milk for improving functionality of milk, and physico-chemical and sensory properties of chitosan added milk were studied. pH and acidity of milk with ethyl alcohol washed chitosan were close to those of control. Color and consistency of chitosan added milk were better with chitosan of lower molecular weight than with high molecular weight. Chitosan of high molecular weight resulted in increased consistency and browning of milk. Milk with less than 1.0% chitosan could be sterilized at $73^{\circ}C$ for 15 sec. with minimum protein coagulation and increase of consistency. Low molecular weight chitosan$(MW\;0.2{\sim}3\;kDa)$ accelerated the growth of Bifidobacterium bifidum, showing 10 times more cell population after 32 hrs incubation. Sensory tests showed that adding chitosan to the regular city milk resulted in significant difference in color and chemical off-flavor(p<0.05). When chitosan was added to coffee milk, there was no significant difference in sensory quality from control. The storage test showed that pH, acidity, consistency and color of coffee milk with 0.5% chitosan did not change markedly during 30 days storage at 0 and $5^{\circ}C$, but changed rapidly after 16days storage at $10^{\circ}C$. Bacterial counts increased when storage temperature was high, and the growth of bacteria was delayed in coffee milk with chitosan.

  • 축산폐수, 침출수 등의 고농도 폐수를 생물학적으로 처리할 경우 최종 방류수는 강한 색도를 띠며 고분자량의 유기물질을 다량 함유한다. 이는 생물학적으로 분해하기 어려운 유기성 복합체와 생화학적 반응에 의한 중간생성물로 색도를 띠는 천연유기물질(NOM)을 포함한다. 생물학적 처리수의 색도는 심미적인 불안감, 방류수역의 수질오염 및 공중보건상의 잠재적 위해성을 갖는다. 또한, 수자원 이용측면에서 정수처리공정에서의 약품투입량 증가와 특히, 소독부산물 생성이라는 잠재적 문제점이 뒤따른다. 따라서 이러한 문제점을 해소하기 위한 생물학적 2차 처리수의 후속처리가 요구되며, 실제로 난분해성 유기물과 색도를 제거하기 위한 흡착, 막 분리, 고급산화(AOP) 및 화학적 응집 등의 물리-화학적 공정에 대한 연구가 수행되어왔다. 특히, 화학적 응집은 무기응집제 또는 고분자중합체(Polymer)를 이용하여 콜로이드성 입자와 색도를 띠는 난분해성 유기물을 전기적 불안정화를 유도함으로서 흡착 및 응집과정을 통해 제거하는 공정으로 많은 연구자들에 의해 연구되어왔다. 그러나 난분해성 유기물과 색도제거는 대상원수의 성상과 화학적 특성 등에 따라 각각의 제거효율과 최적 운전조건이 상이하게 나타난다. 화학적 응집공정은 비교적 높은 제거효율을 보이지만, 운전 및 유지관리의 기술적 어려움, 경제적 비효율성 등으로 인하여 적용에 어려움을 겪고 있는 실정이다. 본 논문에서는 생물학적 혐기-호기성 공정에서 방류되는 축산폐수의 2차 처리수를 대상으로 화학적 응집에 의한 색도 및 난분해성 유기물의 제거거동을 고찰하였다. 대상 처리수의 $TCOD_{Cr}$ 농도는 평균 410 mg/L인 반면, $BOD_5$는 7-15 mg/L 범위로 난분해성 유기물을 다량 함유하고 있음을 알 수 있었다. 이에 황산알루미늄(Aluminium sulfate; $Al_2(SO_4){\cdot}14H_2O$)과 염화철(ferric chloride)의 무기응집제를 이용하여 자 테스트(jar test)를 수행한 결과, 동일한 응집제 주입량에서 염화철의 유기물 제거 효율이 높은 것으로 나타났다. 황산알루미늄과 염화철의 경우 각각의 응집제 주입율 5.85mM에서 89%, 7.03mM에서 97.5%의 최대 유기물 제거효율을 보여주었으며, 이 때 최종 pH는 4.0-5.6 범위이었다. 한편, 대상 원수 내의 콜로이드성 입자 또는 용존성 유기물의 작용기(functional group)는 일반적으로 음으로 하전 되어 있어 응집에 의해 잘 제거되지 않는 특성을 가지고 있다. 따라서 과량의 응집제를 주입하여 다가의 양이온성 금속염을 흡착시켜 전기적으로 중화시키고, 생성된 침전성 수화물 내에 포획 또는 여과시켜 제거하게 된다. 이 때, 금속염 수화종의 전하밀도가 응집효율에 영향을 주는 것으로 알려져 있는데, 다가의 양이온은 전기적 이중층(Double layer) 압축에 의한 불안정화를 향상시킬 수 있기 때문에다. 또한, 2가 금속염은 색도유발물질과 흡착하여 humate 또는 fulvate 등의 착화합물(complex)을 형성시켜 응집효율을 향상시킬 수 있다. 따라서 본 연구에서는 생물학적 2차 처리수의 화학적 응집처리에 있어서 알루미늄염 등의 다가이온 첨가가 응집에 미치는 영향을 관찰하고, 후속되는 플록형성 및 침전공정에 의한 제거효율을 비교, 평가함으로써 2차 처리수로부터 난분해성 유기물과 색도를 보다 효과적이고 경제적으로 제거할 수 있는 최적인자를 도출하고자 하였다.

Backgrounds of Studies Cerebrovascular accident. one of the three major causes of death among audults with cardiopathy and malignancy, has been on the increase in korea while it is on the decrease in European countries and Japan. Types of stroke undergo changes caused by prolongment of life expectancy. and social and economic variety. More patients of ischemic stroke show a tendency to increase now than those of hemorrhagic stroke in the past. Many clinical studies on medical cerebrovascular and oriental stroke of paralysis have been published. but few clinical studies on therapeutics of integrated oriental and western medicine are to be found. So I have made an attempt to study clinical observations and therapeutic responses of ischemic stroke under integrated oriental and western medicine. 2.Methods. Among the patients admitted into the clinic of Joong-Poong, Woo-Suk University Hospital from May 1. 1993 until April 30. 1994 those 56 patients who were diagnosed as ischemic stroke on Computed Tomography(CT) and showed no dubious symptom after examination of coagulation and bleeding time were classified into the following six steps and treated: l)diagnosis 2)emergency treatment 3)basic treatment 4)treatment of risk and provoking factors. and preceeding disease 5)complications and conservative therapy 6)rehabilitation. For a period of basic treatment both herb medication and urokinase therapy were applied at the same time. Intravenous injection has been given at a unit of 300.000 dosage a day as urokinase therapy during basic treatment. If they showed any dubious symptom in glucose tolerance test. fructose 500ml and urokinase 300.000 dosage were mixed and injected. In case of no symptom 5% DW 500ml was mixed with urokinase 300.000 unit. and injected at a speed of 15gtt per minute. 3. Results and Conclusions 1) The level of ambulation has been improved from 42.9% when admitted to 73.2% when discharged in the degree of recovery. The level of severe function disorder has been remarkably decreased from 55.4% when admitted to 19.6% when discharged. 2) The treatment effect on the basis of therapeutic response of clinical and subjective symptom shows as follows: 7.1% Excellent. 35.7% Good. 37.5% Effective. 10.7% Stationary. and 8.9% Aggravated. The total recovery above effective shows 80.3%. Judging from the above results I think it proper to develop the model of better preventing and treating ischemic stroke through effective therapeutic and clinical studies of integrated oriental and western medicine.
