The Effects of Natural Food Additives on the Self-life and Sensory Properties of Shucked and Packed Pacific Oyster Crassostrea gigas (생굴(Crassostrea gigas)의 저장성 및 관능성에 대한 천연 첨가물의 효과)

  • We explored the efficiency of natural antibacterial agents used to enhance the self-life and sensory properties of shucked and packed Pacific oyster Crassostrea gigas, which are in high demanded. First, we screened natural resources exhibiting antibacterial activity against food spoilage and pathogenic bacteria. Of theses, ignited oyster shell powder (IOS) and the natural food preservative, lactic acid bacteria fermented powder (LBF), were selected for further study considering the efficacy, mass production, and cost. The addition of 0.1% IOS (W/V) and 0.5% LBF (W/V) to shucked and packed oyster optimally extended the shelf-life without affecting the sensory evaluation. The results obtained in this study will provide a clue to enhance self-life in raw oyster products.

Seasonal Variations of Acdity and Chemicstry of Precipitation in Iksan Area (익산지역 강수의 계절별 산성도와 화학성상)

  • Precipitation samples were collected by the wet-only sampling method in Iksan in the northwest of Chonbuk from March 1995 to February 1997. These samples were analyzed for the concentration of ion components, in addition to pH and electrical conductivity. The annual mean pH of precipitation was 4.8 and the seasonal trend of pH was shown to be low in Fall and Winter(4.5), middle-ranged in Spring(4.7) and high in Summer(5.0). The frequency of pH below 5.6 was about 71%. The seasonal pattern of pH frequency was found to be different in each season. In the case of the pH less than 5.0, the frequency was higher in Spring, Fall and Winter than in Summer, especially higher in Fall than in other seasons. The concentrations of analysed ions showed a pronounced seasonal pattern. However, major ion species for all seasons were $NH^+_4,;Ca^{2+};and;Na^+$ among cations and $SO^{2-}_4,;Cl^-;and;NO^-_3$ among anions. The major acidifying species appeared to be $nss-SO^{2-}_4;and;NO^-_3$, and the main bases responsible for the neutralization of precipitation acidity were $nss-Ca^{2+};and;NH^+_4$. The potential acidity of precipitation, pAi, was found to be between 3.0 and 5.0 for total samples, while the measured pH was approximately between 3.9 and 7.8. The seasonal trend of pAi showed a decreasing order: Summer (4.3), Winter(4.0), Spring and Fall(3.8). During the Fall, both pAi and pH were especially very low, which indicated that during this period the potential acidity of precipitation was high but the neutralizing capacity was low. For Spring, pAi was very low but pH was slightly high. This was likely due to the large amount of $CaCO_3$ in the soil particles transported over a long range from the Chinese continent that were incorporated into the precipitation, and then neutralized the acidifying species with its high concentraton.

Mineralogy and Chemical Composition of the Residual Soils (Hwangto) from South Korea (우리 나라 황토(풍화토)의 구성광물 및 화학성분)

  • The mineralogy and chemical composition of reddish to brownish yellow residual soils, so called "Hwangto" have been examined according to representative host rocks. The result of the study indicates that Hwangto consists of 40-80% clay minerals and various minerals such as quartz, feldspar, hornblende, goethite, and gibbsite. Clay minerals include kaolinite, halloysite, illite, hydroxy interlayered vermiculite (HIV), mica/vermiculite interstratifield mineral and chlorite. The mineralogical constituents and contents of Hwangto were different depending on the types of host rocks. Moreover, the Jurassic granitic rocks contain relatively more kaolin minerals, whereas the Cretaceous granitic rocks contain more HIV and illite. In addition, reddish Hwangto contains relatively more kaolinite and HIV, and yellowish Hwangto contains more illite and halloysite. It is suggested that feldspars and micas of host rocks were chemically weathered into illite, halloysite, illite/vermiculite interstratified minerals, and HIV, and finally into kaolinite. Compared with their host rocks, the major chemical compositions of Hwangto tend to contain more $Al_2O_3,\;Fe_2O_3,\;H_2O$ in amount and less Ca, Mg, and Na. Hwangto contains relatively high amount of trace elements, P, S, Zr, Sr, Ba, Rb, and Ce including considerable amount of Li, V, Cr, Zn, Co, Ni, Cu, Y, Nb, La, Nd, Pb, Th in excess of 10 ppm. Relatively high amount of most trace elements were detected in the Hwangto. The major and minor chemical compositions of the Hwangto were different depending on the types of host rocks. However, their difference was in the similar range compared with the compositions of host rocks.

Evaluation indicators for the restoration of degraded urban ecosystems and the analysis of restoration performance (훼손된 도시생태계 생태복원 평가지표 제시 및 복원성과 분석)

  • This study aims to analyze the effect of urban ecosystem restoration projects by evaluating the short-term restoration performance of the project sites, from both qualitative and quantitative evaluations. In this study, for the qualitative evaluation, we derived the evaluation frame from previous studies and literature. For the quantitative evaluation, the changes in ecological connectivity after the restoration project were described using landscape permeability and network analysis. In addition, changes in habitat quality after the restoration project were evaluated by using InVEST Habitat Quality Model. These evaluations were applied to the three natural madang (ecological restoration) projects and two ecosystem conservation cooperation projects. As a result, three categories, 10 indicators, and 13 sub-indicators were derived from literature as the evaluation frame for this study. In the case of quantitative evaluation of restoration performance, habitat quality increased by 45% and ecological connectivity by 37% in natural-madang, and habitat quality by about 12% and ecological connectivity by about 19% in ecosystem conservation cooperation projects. This implies that the ecological restoration project can increase the ecological connectivity and the habitat quality of degraded sites even in a short period of time by improving the land-cover and land use. The results by applying the evaluation frame indicated that ecological and environmental factors and the ecological functions were improved by the restoration works, even though the magnitude of performances were diverse depending on the specific evaluation items, project type, and site characteristics. This study clarified that the success of ecological restoration project should be assessed by both of the short-term and long-term goals, which can be achieved by the maintenance and sustainable management, respectively.

Pharmacokinetic Analysis of the Effect of Extrahepatic Cholestasis by Common Bile Duct Ligation on Hepatic Function in Rats (총수담관 결찰에 의한 간외 담즙분비정체가 흰쥐의 간기능에 미치는 영향에 대한 약물속도론적 분석)

  • In order to examine the effect of extrahepatic cholestasis induced by common bile duct ligation on the hepatic function, the pharmacokinetics of antipyrine and d-propranolol were investigated in rats. In addition, in an attempt to observe the degree of direct hepatic injury, light and electron microscopic observations and conventional pathologic test using serum were performed. Five days after common bile duct ligation, antipyrine(15 mg/kg) and d-propranolol(3 mg/kg) were intravenously administrated to the rats, respectively. The total clearances of antipyrine and d-propranolol were significantly(p<0.05) decreased. Because hepatic clearance of antipyrine poorly extracted by the liver and that of d-propranolol highly extracted by the liver are respectively dependent on the hepatic intrinsic clearance and the hepatic blood flow, it may be concluded that extrahepatic cholestasis following five days after common bile duct ligation decreased the hepatic intrinsic clearance and the hepatic blood flow. SGPT, SGOT, cholesterol, bilirubin(total bilirubin, direct bilirubin) and alkaline phosphatase were significantly increased(p<0.05). The proliferation of bile ducts was prominent, and degeneration and necrosis of hepatocytes were observed by light microscope. Also, ultrastructurally, bile canaliculi were containing the amorphous materials and losing microvilli, and SER and RER in hepatocytes were dilated and vacuolated.

Anti-Inflammatory Potential of Probiotic Strain Weissella cibaria JW15 Isolated from Kimchi through Regulation of NF-κB and MAPKs Pathways in LPS-Induced RAW 264.7 Cells

  • Probiotics are known to provide the host with immune-modulatory effects and are therefore of remarkable interest for therapeutic and prophylactic applications against various disorders, including inflammatory diseases. Weissella cibaria JW15 (JW15) has been reported to possess probiotic and antioxidant properties. However, the effect of JW15 on inflammatory responses has not yet been reported. Therefore, the objective of the current study was to evaluate the anti-inflammatory potential of JW15 against lipopolysaccharide (LPS) stimulation. The production of pro-inflammatory factors and the cellular signaling pathways following treatment with heat-killed JW15 was examined in LPS-induced RAW 264.7 cells. Treatment with heat-killed JW15 decreased nitric oxide and prostaglandin $E_2$ production via down-regulation of the inducible nitric oxide synthase and cyclooxygenase-2. In addition, treatment with heat-killed JW15 suppressed the expression of pro-inflammatory cytokines, interleukin $(IL)-1{\beta}$, IL-6, and tumor necrosis factor-${\alpha}$. The anti-inflammatory properties of treating with heat-killed JW15 were associated with mitogen-activated protein kinase signaling pathway-mediated suppression of nuclear factor-${\kappa}B$. These results indicated that JW15 possesses anti-inflammatory potential and provide a molecular basis regarding the development of functional probiotic products.

Optimization of Muffins Containing Dried Leek Powder Using Response Surface Methodology (부추 분말 첨가 머핀의 제조 조건 최적화)

  • The purpose of this study was to develop a muffin containing dried leek powder. The complete analysis was conducted using the Design Expert 7 program (Stat - Easy Co., Minneapolis, MN, USA). The leek muffins were produced with varying amounts of leek powder (A), sugar (B), and butter (C). According to response surface methodology (RSM), there were 16 experimental points, including two replicates. The leek muffin formulation was optimized using rheology and sensory analyses. For the results, a quadratic model was applied in determining lightness, volume, hardness, and sensory characteristics, including flavor, texture, and overall quality; redness and height were represented by a linear model. Lightness decreased and redness increased with increasing amounts of leek powder (p<0.001). In addition, hardness displayed significant differences (p<0.001) with increasing amounts of leek powder. The sensory evaluation results showed significant differences for color, flavor, texture, and overall quality (p<0.05). Based on the numerical and graphical methods, the optimal formulation was determined as 8.30 g of leek power, 88.37 g of sugar, and 81.70 g of butter.

Adipose-derived stem cells decolonize skin Staphylococcus aureus by enhancing phagocytic activity of peripheral blood mononuclear cells in the atopic rats

  • Staphylococcus aureus (S. aureus) is known to induce apoptosis of host immune cells and impair phagocytic clearance, thereby being pivotal in the pathogenesis of atopic dermatitis (AD). Adipose-derived stem cells (ASCs) exert therapeutic effects against inflammatory and immune diseases. In the present study, we investigated whether systemic administration of ASCs restores the phagocytic activity of peripheral blood mononuclear cells (PBMCs) and decolonizes cutaneous S. aureus under AD conditions. AD was induced by injecting capsaicin into neonatal rat pups. ASCs were extracted from the subcutaneous adipose tissues of naïve rats and administered to AD rats once a week for a month. Systemic administration of ASCs ameliorated AD-like symptoms, such as dermatitis scores, serum IgE, IFN-γ+/IL-4+ cell ratio, and skin colonization by S. aureus in AD rats. Increased FasL mRNA and annexin V+/7-AAD+ cells in the PBMCs obtained from AD rats were drastically reversed when co-cultured with ASCs. In contrast, both PBMCs and CD163+ cells bearing fluorescent zymosan particles significantly increased in AD rats treated with ASCs. Additionally, the administration of ASCs led to an increase in the mRNA levels of antimicrobial peptides, such as cathelicidin and β-defensin, in the skin of AD rats. Our results demonstrate that systemic administration of ASCs led to decolonization of S. aureus by attenuating apoptosis of immune cells in addition to restoring phagocytic activity. This contributes to the improvement of skin conditions in AD rats. Therefore, administration of ASCs may be helpful in the treatment of patients with intractable AD.

  • A new process has been developed for nitrate and other salts removals from polluted waters. Alumina cement and calcium oxide served as precipitating agents to remove nitrate with stirring at basic pH. Low content of alumina in the commercialized alumina cements resulted in a increasing in nitrate removal yield. It is found that the compositions of aluminium and calcium are the most important factors in successful nitrate insolubilization. In order to remove high concentration of nitrate in polluted water, multi-stage precipitation was found to be very effective. Sulfate, chloride, and phosphate ions as well as nitrate were also removed by the precipitated reaction. After precipitation, post-treatments including Na2CO3 addition and neutralization with acid alleviated the level of aluminium and calcium in the treated water.

  • The expression, purification, and characterization of cold-adapted lipase from the psychrophile, Janthinobacterium sp. were investigated. The gene encoding lipase from Janthinobacterium sp. PAMC 25641 was cloned into a pET28a(+) vector and heterologously expressed in Escherichia coli BL21 (DE3). The amino acid sequence deduced from the nucleotide sequence (930 bp) corresponded to a protein having 309 amino acid residues with a molecular weight of 32.7 kDa and a pI of 5.55. Recombinant E. coli harboring the Janthinobacterium lipase gene were induced by addition of isopropyl-${\beta}$-D-thiogalactopyranoside. $Ni^{2+}$-NTA affinity chromatography was used to purify the lipase, which had a specific activity of 107.9 U/mg protein. The effect of temperature and pH on the activity of lipase was measured using p-nitrophenyl octanoate as a substrate. The stability of the lipase at low temperatures indicated it is a cold-adapted enzyme. The lipase activity was increased by $Na^{2+}$, $Mg^{2+}$, and $Mn^{2+}$, and decreased by $Zn^{2+}$ and $Co^{2+}$. Analysis of the lipase activity using various p-nitrophenyl esters showed a strong preference toward short acyl chains of the esters, indicating the ability of the cold-adapted lipase to hydrolyze short-chain esters.
