평택호 상류 지역 참붕어에 있어서 간흡충 피낭유충의 조사성적 (A Survey on Metacercariae Infection of Clonorchis sinensis on the Fish Host, Pseudorasbora parva from the Upper Stream of Phoung-tack Stream, Kyungki Province)

  • A survey on the incidence of metacercariae infection of Clonorchis sinensis in the fresh water fish, Pseudorasbora parva which is well known as the second intermediate host for this fluke was carried out in the upper stream of Phoung-tack stream, Kyungki province during the period of October to November 1997. Forty fish in P. parva were collected by netting, or using the bait in transparent bowls from the two different districts located in Osung-myun, Phoungtack-gun and transported to the laboratory. The body length and weight were measured. They were from 5.0 to 7.6 cm in length and 1.0 to 5.0 g in weight. A total of 40 P. parva were divided 3 groups by the size (70-79, 60-69 and 50-59 mm) and were digested by artificial digestion with pepsin-Hcl solution in a 36$\circ$C incubator and then examined the infection density of metacercariae of the fresh water fish under a binocular dissecting microscope. The incidence of metacercariae infection in P. parva was 406 of 40 examined and the metacercariae detection rate showed 77.5%. The average number of metacercariae detected in P. parva was 10.2. The results of the survey indicate that Clonorchis sinensis infection may occur by consuming raw fresh water fish caught from the upper stream of Phoungtackho, Kyungki province.

  • An enzyme analysis of the liver fluke, Clonorchis sinensis from Kimhae, Korea and from Shenyang, China was conducted using a horizontal. starch gel electrophoresis in order to elucidate their genetic relationships. A total of eight enzymes was employed from two different kinds of buffer systems. Two loci from each enzyme of aconitase and esterase (${\alpha}-Na{\;}and{\;}{\beta}-Na$) : and only one locus each from six enzymes, gluucose-6-phosphate dehydrogenase (G6PD), ${\alpha}-glycerophosphate$ dehydrogenase (GPD), 3-hydroxybutyrate dehydrogenase (HBDH), malate dehydrogenase (MDH), phosphoglucose isomerase (PGI), and phosphoglucomutase (PGM) were detected. Most of loci in two populations of C. sinensis showed homozygous monomorphic banding patterns and one of them, GPD was specific as genetic markers between two different populations. However, esterase (${\alpha}-Na$), GPD, HBDH and PGI loci showed polymorphic banding patterns. Two populations of C. sinensis were more closely clustered within the range of genetic identity value of 0.998-1.0. In summarizing the above results, two populations of C. sinensis employed in this study showed mostly monomorphic enzyme protein banding patterns, and genetic differences specific between two populations.

  • The WD40-repeat proteins serve as a platform coordinating partner proteins and are involved in a range of regulatory cellular functions. A WD40-repeat protein (CsWD1) of Clonorchis sinensis previously cloned is expressed stage-specifically in the tegumental syncytium of C. sinensis metacercariae. In the present study, interact-ing proteins with the CsWD1 protein was purified by immunoprecipitation and 2 dimension gel electrophoresis from the C. sinensis metacercaria soluble extract, and tryptic peptides were analyzed by LC/ESI-MS. Putative partner proteins were annotated to be actin-2, glyceraldehyde-3-phosphate dehydrogenase, and hypothetical and unmanned proteins. The CsWD1 protein was predicted to contain 3 conserved actin-interacting residues on its functional surface. With these results, the CsWD1 protein is suggested to be an actin-interacting protein of C. sinensis.

근교계 마우스에서 간흡충 기생기간과 산란력의 변동 (Egg Production of Clonorchis sinensis in Different Strains of Inbred Mice)

  • 여섯 계통의 근교계 마우스떠에 상흡충의 기생기간과 경시적 산란력을 근거로 하여 숙주 기생충 관계의 계통별 차이를 구명하기 위해 마우스찰 20개의 간흡충 피낭유충을 경구적으로 감염시켰다. 감염 후 간흡충의 충란이 처음 검출될 때까지의 기간은 DDY 마우스에서 평균 21.2일로 가장 짧았고, GPC 21.48, BALB/c 및 DS 23.2 5, ICR 및 nude 23.4일의 순이 었다. 총산란기간 역시 DDY 마우스에서 164일로 가장 길었으며, GPC 132R, BALB/c 97일, nude 37일, DS 32일 및 ICR 28일의 순이었다. DDY 및 GPC 마우스에서는 간흡충의 산란수가 비교적 높고 안정적이었으나 나머지 4계통의 마우스에서는 관찰기간 동안 불규칙하였다. 이상의 성적으로 미루어 보아 마우스의 계통에 따라 간흡충의 숙주 기생충 관계의 차리를 확인하였고 DDY 마우스가 6계통중 가장 호적숙주임을 알았다.

Codon Usage Patterns of Tyrosinase Genes in Clonorchis sinensis

  • Codon usage bias (CUB) is a unique property of genomes and has contributed to the better understanding of the molecular features and the evolution processes of particular gene. In this study, genetic indices associated with CUB, including relative synonymous codon usage and effective numbers of codons, as well as the nucleotide composition, were investigated in the Clonorchis sinensis tyrosinase genes and their platyhelminth orthologs, which play an important role in the eggshell formation. The relative synonymous codon usage patterns substantially differed among tyrosinase genes examined. In a neutrality analysis, the correlation between $GC_{12}$ and $GC_3$ was statistically significant, and the regression line had a relatively gradual slope (0.218). NC-plot, i.e., $GC_3$ vs effective number of codons (ENC), showed that most of the tyrosinase genes were below the expected curve. The codon adaptation index (CAI) values of the platyhelminth tyrosinases had a narrow distribution between 0.685/0.714 and 0.797/0.837, and were negatively correlated with their ENC. Taken together, these results suggested that CUB in the tyrosinase genes seemed to be basically governed by selection pressures rather than mutational bias, although the latter factor provided an additional force in shaping CUB of the C. sinensis and Opisthorchis viverrini genes. It was also apparent that the equilibrium point between selection pressure and mutational bias is much more inclined to selection pressure in highly expressed C. sinensis genes, than in poorly expressed genes.

경남 산청군 생비량면 가계리 주민의 간흡충과 장내 윤충류 충란 양성율 (Egg Positive rates of Clonorchis sinensis and intestinal helminths among residents in Kagye-ri, Saengbiryang-myon, Sanchong-gun, Kyongsangnam-do)

  • 경남 산청군 생비량면 가계리 주민의 대변검사를 modified Kato's cellophane thick smear와 formalin-ether concentration technique로 1987년 11월과 1993년 4월에 실시하였다. 1987년 주민 76명 중 간흡충 감염자는 61명(80.3%)이었으며 EPG 평균은 27.781이었다. 간흡충 충란 양성자들에게 praziquantel 60 mg/kg을 1회 투여하였다. 요꼬가와흡충 감염자는 17명(22.4%)이었으며 간흡충에 동시감염되어 있었다 1993년, 간흡충 감염율은 48.4%이었으며 평균 EPG는 5.929 이었다. 추적조사된 주민중 55.2%가 5년 5개월 동안에 간흡충에 재감염되어 있었다. 요꼬가와흡 충감염자는 3.2%이었다. 이 부락은 간흡충의 재감염이 높게 일어나고 있는 유행지이었다.

Infection rates of Enterobius vermicularis and Clonorchis sinensis of primary school children in Hamyang-gun, Gyeongsangnam-do (Province), Korea

  • The egg Positive rate of Enterobius vermicularis and Clonorchis sinensis of school children in the rural area was studied in Hamyang-gun, Gyeongsangnam-do in Korea. Cellotape anal swab and formalin ether concentration methods were performed one time to 720 Primary school children. The total egg Positive rate of E. vermicularis was 12.6% in two schools (Baekjeon and Wiseong). In the Baekjeon and Wiseong primary school, the egg Positive rate of E. vermicularis was 4.6% and 13.4%, respectively Pinworm egg Positive rate was 17.6% in the lower grades (1st, 2nd and 3rd), and 7.7% in higher grades (4th, 5th and 6th). The total egg Positive rate of male and female was 12.6% and 12.7%, respectively. The egg Positive rate of C. sinensis of Baekjeon and Wiseong Primary school was 1.5% and 0.46%, respectively The total e99 Positive rate of C. sinesis was 0.56%. This survey showed that continuous education and chemotherapy is necessary to treat and prevent reinfection of E. vermicularis. In the case of C. sinensis, health education for school children is recommended to prevent potential infection of adolescents.

Susceptibility of experimental animals to reinfection with Clonorchis sinensis

  • The present study observed the resistance to reinfection with Clonorchis sinensis in various experimental animals including mice, guinea pigs, rabbits, and dogs, as well as rats and hamsters. The resistance rates to reinfection in rats, mice, hamsters, guinea pigs, rabbits, and dogs were 79.7%, 58.0%, -12.6%, 54.8%, 62.6%, and 6.0%, respectively. Worms recovered from reinfected rats and mice were immature, and significantly smaller than those from the primarily infected (P < 0.01), whereas those from other animals were fully matured to adults. These findings indicate that the protective response against reinfection with C. sinensis is prominent in rats and mice, and that they may be a good animal model to investigate the mechanism of resistance to reinfection with C. sinensis.

Toll-Like Receptor-Mediated Free Radical Generation in Clonorchis sinensis Excretory-Secretory Product-Treated Cholangiocarcinoma Cells

  • Clonorchiasis, caused by direct contact with Clonorchis sinensis worms and their excretory-secretory products (ESPs), is associated with chronic inflammation, malignant changes in bile ducts, and even cholangiocarcinogenesis. Our previous report revealed that intracellular free radicals enzymatically generated by C. sinensis ESPs cause NF-${\kappa}B$-mediated inflammation in human cholangiocarcinoma cells (HuCCT1). Therefore, the present study was conducted to examine the role of upstream Toll-like receptors (TLRs) on the initial host innate immune responses to infection. We found that treatment of HuCCT1 cells with native ESPs induced changes in TLR mRNA levels in a time-dependent manner, concomitant with the generation of free radicals. ESP-mediated free radical generation was markedly attenuated by preincubation of the cells with TLR1-4-neutralizing antibodies, indicating that at least TLR1 through 4 participate in stimulation of the host innate immune responses. These findings indicate that free radicals triggered by ESPs are critically involved in TLR signal transduction. Continuous signaling by this pathway may function in initiating C. sinensis infection-associated inflammation cascades, a detrimental event leading to progression to more severe hepatobiliary diseases.

Functional characterization of Clonorchis sinensis choline transporter

  • Clonorchis sinensis is commonly found in East Asian countries. Clonorchiasis is prevalent in these countries and can lead to various clinical symptoms. In this study, we used overlap extension polymerase chain reaction (PCR) and the Xenopus laevis oocyte expression system to isolate a cDNA encoding the choline transporter of C. sinensis (CsChT). We subsequently characterized recombinant CsChT. Expression of CsChT in X. laevis oocytes enabled efficient transport of radiolabeled choline, with no detectable uptake of arginine, α-ketoglutarate, p-aminohippurate, taurocholate, and estrone sulfate. Influx and efflux experiments showed that CsChT-mediated choline uptake was time- and sodium-dependent, with no exchange properties. Concentration-dependent analyses of revealed saturable kinetics consistent with the Michaelis-Menten equation, while nonlinear regression analyses revealed a Km value of 8.3 µM and a Vmax of 61.0 pmol/oocyte/h. These findings contribute to widen our understanding of CsChT transport properties and the cascade of choline metabolisms within C. sinensis.
