국내에서 유행하는 CPV-2a 분리주에 대한 CPV-2와 CPV-2b 백신의 방어효능 평가 (Evaluation for Protective Effect of CPV-2 and CPV-2b Vaccines against a Korean CPV-2a Isolate in Pups)

  • 본 연구는 상용화되어 판매되고 있는 CPV-2 백신과 CPV-2b 백신이 국내에서 주로 유행하고 있는 CPV-2a 분리주에 대해 방어능력을 가지는 지를 평가하기 위하여, CPV에 대한 예방접종을 실시하지 않은 생후 9주령의 잡종 강아지 20두를 예방접종 미실시군(대조군) 4두와 예방접종군 16두로 편성하였다. 예방접종 군의 강아지 8두씩은 3주 간격으로 3회에 걸쳐 CPV-2 또는 CPV-2b 백신을 각각 예방접종하였다. 3차 예방접종 2주 후에 모든 강아지에 $1{\times}10^6\;TCID_{50}$의 CPV-2a (VR00174 strain) 바이러스를 경구접종하였으며 강아지들의 임상증상, CPV의 변 내 배출, CPV에 대한 혈청학적 반응에 대하여 2주 동안 관찰하였다. 국내 분리 CPV-2a를 공격접종한 후에, 예방접종을 실시한 모든 강아지에서는 어떠한 임상증상도 나타나지 않았지만 예방접종을 실시하지 않은 강아지들은 식욕부진, 우둔, 구토, 점액 또는 혈액성 설사를 나타내었으며 2두는 공격접종 6일째에 폐사하였다. 예방접종을 실시하지 않은 강아지들에서는 공격접종 4일 후부터 변에서 CPV가 검출되었지만 예방접종을 실시한 강아지에서는 변에서 CPV가 검출되지 않았다. 그리고 예방접종 실시한 강아지들은 1차 예방접종에 의하여 방어수준 이상으로 CPV에 대한 항체를 형성하였다. 본 연구는 현재 국내에서 시판 중인 CPV-2와 CPV-2b 백신이 최근 국내에서 주로 분리되는 CPV-2a에 대하여 교차 방어력을 나타내는 것으로 확인되었다.

  • Shigellosis is the most common bacterial gastroenteritis both in developing and developed countries, but bacteremia due to Shigella spp. is very rare. In developed countries recent shigellosis is mostly caused by S. sonnei, but S. flexeri infection is rare. We had rare cases of S. flexeri infections in a family in the Jeonbuk Province: an 8-year-old boy with bacteremic shigellosis and 10- and 12-year-old brothers with diarrhea. The isolates had identical biochemical characteristics, and were resistant to ampicillin, chloramphenicol, and co-trimoxazole. PFGE pattern of Not I-restricted genomic DNA suggested that the isolate from blood was closely related to the two strains isolated from stool which had an identical PFGE pattern.

국내 대학병원에서 분리된 Metallo-β-Lactamase (MBL) 생성 Acinetobacter spp. 분리주의 높은 출현율과 유전형 특징 (High Prevalence and Genotypic Characterization of Metallo-β-Lactamase (MBL)-Producing Acinetobacter spp. Isolates Disseminated in a Korean Hospital)

  • 주요 획득성 metallo-β-lactamase (MBL) 유전자에 의해 매개되는 carbapenem 내성, 특히 Acinetobacter spp. 균종의 임상 분리주에 대한 보고가 증가하고 있다. 본 연구에서 임상에서 비 중복으로 분리된 carbapenem 비감수성 Acinetobacter spp. 191주 중 125 (65.4%)주가 imipenem 혹은 meropenem-Hodge 변법시험에 양성이었고, 49 (25.7%)주가 imipenem-EDTA+SMA double disk synergy (DDS) 시험에 양성이었다. blaVIM-2 allele와 blaIMP-2 allele 검출을 위한 중합효소연쇄반응과 염기서열분석을 시행한 결과, A. baumannii와 A. calcoaceticus에서 각각 29주와 1주가 blaVIM-2를 갖고 있었고, A. baumannii 16주와 A. calcoaceticus 2주가 blaIMP-1을 갖고 있었다. A. genomospecies 3는 blaVIM-2와 blaAIM-1을 동시에 갖고 있었다. 이들 MBL 유전자는 모두 class 1 integron에 있었다. blaVIM-2 혹은 blaIMP-6를 갖는 class 1 integron의 크기는 A. baumannii 분리주에서는 2.8 kb에서 3.2 kb이었고, A. genomospecies 3 분리주에서는 3.2 kb에서 3.5 kb이었다. blaVIM-2는 대부분 class 1 integron에 첫번째 혹은 두번째에 위치하였고, aacA4를 흔히 가지고 있었다. 다양한 내성 유전자를 가질 수 있는 MBL 생성 Acinetobacter spp.뿐 아니라 다양한 내성 유전자를 가질 수 있는 integron의 전파로 imipenem이나 meropenem과 같은 carbapenem 내성을 포함하여 다제 내성 그람음성 세균의 증가가 예상된다. 또한, 위중한 Acinetobacter spp. 감염증 치료를 위한 새로운 항균제 개발이 필요하다.

Rapid PCR Method for Detecting Candida albicans Using Primers Derived from the Integrin-like Protein Gene $\alpha$INT1 of Candida albicans

  • Oligonucleotide primers amplifying a 344 bp fragment on the integrin-like protein alpha-INT1p gene (${\alpha}$INT1) of Candida albicans were synthesized for screenign of C. albicans from clinicalsamples by the polymerase chain reaction (PCR). The PCR specifically amplified DNA from C. albicans and none from any other Candida, fungal, or human DNA in standard used here. The PCR assay showed that the primers (LH1 and LH2) were specific for 26 isolates of C. albicans from clinical smaples, whereas the positive fragment, 344 bp, was not amplified from 15 clinical isolates including 14 other medically important Candida species and an isolate of Saccharomyces cerevisiae. PCR was conducted on the urine samples of 20 patients and 4 samples were C. albicans positive. The detection limit of the PCR assay for C. albicans was shown to be approximately 10 cells/ml saline. The PCR system using 344 bp ${\alpha}$INT1 as a target is more specific and rapid than the conventional culture method, and the sensitive detection method is applicable to clinical diagnosis of C. albicans infections.

Usefulness of PCR to Mycobacterium Tuberculous and Nontuberculous Mycobacteria from Paraffin-embedded Tissues

  • The purpose of this study was to evaluate the clinical utility of TB/NTM PCR by comparing the results of TB PCR to detect Mycobacterium tuberculous (MTB) and nontuberculous mycobacteria (NTM) from paraffin-embedded tissue specimens. A total of 60 cases were tested using TB PCR and TB/NTM PCR. The MTB and NTM rate of TB/NTM PCR was 84.2% (16/19), 10.5% (2/19) in TB positive of TB PCR. The NTM rate of TB/NTM PCR was 29.3% (12/41) in TB negative of TB PCR. Fourteen different species of NTM were identified, the common isolate was M. gordonae (21.4%), M. avium (14.3%), M. ulcerans (7.1%), M. interjectum (7.1%), M. gilvum (7.1%), M. fortuitum (7.1%), M. mucogenicum (7.1%). The rare species identified were M. farcinogenes (7.1%), M. tokaiense (7.1%). Therefore, TB/NTM PCR is useful to differentiate MTB and NTM from paraffin-embedded tissue specimens and it is more effective in detecting NTM with TB PCR.

Relationship between AdeABC Efflux Pump Genes and Carbapenem in Multidrug-resistant Acinetobacter baumannii

  • Multidrug-resistant strain of Acinetobacter baumannii (MDRAB) is an emerging pathogen in health care facilities, preventing MDRAB is a public health concern. We conducted this experiment on a clinical isolate of A. baumannii with two main goals: the role of the efflux pump system in the stress provision of carbapenem and the response to the transcription level of the efflux pump gene. A total of 34 strains of A. baumannii was isolated from the Yangsan Hospital of Pusan National University. First, when we compared and observed the expression of the efflux pump gene and antibacterial resistance to carbapenem, a strong correlation was observed between carbapenem resistance and overexpression of adeB (P=0.0056). Second, a correlation between the efflux pump and concentration gradient and tolerance to carbapenem stress at the AdeABC efflux pump genes transcription level was confirmed. Our results revealed that the expression of the AdeABC efflux pump is an important resistance determinant in obtaining antibiotic resistance of the carbapenem group in A. baumannii.

임상검체에서의 Haemophilus aphrophilus분리 5예 (Isolation of Haemophilus aphrophilus from Five Clinical Specimens)

  • Haemophilus aphrophilus is a fastidious gram-negative bacillus found in mouth of normal individuals. Though H. aphrophilus infection is quite rate, it includes such serious ones as endocarditis and brain abscess. The authors isolated H. aphrophilus from five patients with the diagnosis of lung abscess, conjunctivitis, brain abscess and facial masticator space abscess. Two of the patients died. Three of the patients also yielded other species of bacteria from the same specimens. One of the isolate was intermediately susceptible to amikacin and resistant to tobramycin, indicating the necessity of a routine susceptibility test in order to select the proper antimicrobial agents. Since H. aphrophilus can be differentiated from other similar organisms by morphological and biochemical characteristics, one should determie the possibility of this organism when fastidious gram-negative bacilli are isolated from blood or from sites adjacent to upper respiratory tract.

Synergistic Antibacterial Activity of an Active Compound Derived from Sedum takesimense against Methicillin-Resistant Staphylococcus aureus and Its Clinical Isolates

  • There are a growing number of reports of hospital-acquired infections caused by pathogenic bacteria, especially methicillin-resistant Staphylococcus aureus (MRSA). Many plant products are now being used as a natural means of exploring antimicrobial agents against different types of human pathogenic bacteria. In this research, we sought to isolate and identify an active molecule from Sedum takesimense that has possible antibacterial activity against various clinical isolates of MRSA. NMR analysis revealed that the structure of the HPLC-purified compound was 1,2,4,6-tetra-O-galloyl-glucose. The minimum inhibitory concentration (MIC) of different extract fractions against numerous pathogenic bacteria was determined, and the actively purified compound has potent antibacterial activity against multidrug-resistant pathogenic bacteria, i.e., MRSA and its clinical isolates. In addition, the combination of the active compound and β-lactam antibiotics (e.g., oxacillin) demonstrated synergistic action against MRSA, with a fractional inhibitory concentration (FIC) index of 0.281. The current research revealed an alternative approach to combating pathogenesis caused by multi-drug resistant bacteria using plant materials. Furthermore, using a combination approach in which the active plant-derived compound is combined with antibiotics has proved to be a successful way of destroying pathogens synergistically.

오리의 Pasteurella anatipestifer 감염증 발생 (Pasteurella anatipestifer infection in ducklings in Korea)

  • This study was conducted to investigate the cause of a new duck disease occured in southern part of Korea. A meat type duck farm located in Kangjin, Chonnam Province had experienced outbreaks of septicemic disease at around 20 days of age in nearly every batch of ducklings from early spring to early summer in 1989. Main symptoms of the birds were eye and nasal discharge, depression, inappetence, diarrhea and nervous signs such as tremor and ataxia. Some birds died suddenly without any signs. Mortality reached from 20% to 80% in severe cases. The autopsy findings of the affected ducklings revealed consistantly severe airsacculitis, fibropericarditis, perihepatitis and occasionaly enteritis and distended ureter with urate deposit. A rod shaped gram-negative bacterium was isolated purely from brain and liver of the diseased ducks by culturing the specimens on blood agar for 48 hours in candle jar. The isolate neither produced hemolysis nor grew on MacConKey Agar. It formed colony relatively slowly being recognizable at least 36 hours after culturing, reaching colony size of about 1mm in diameter at 48 hours culture. The colony looked iridescent under oblique light and had muddy odor. The isolate did not ferment carbohydrates tested but produced gelatinase, hippuricase and oxidase which were considered as characteristics of P anatipestifer. The isolate induced similar signs and lesions when infected experimentally into ducks of 3 to 38 days age via intraperitoneum or intratrachea. However it did not produce any clinical signs wen inoculated via intranasal route. It produced only mild signs in chicken just injected with a very large dose. The bacteria did not produce any signas or lesions in mice. It was concluded through biochemical and physiological tests and animal inoculation tests that the new disease was caused by P anatipestifer.

Progesterone 및 Acetyl-L-Carnitine이 정자의 동결-융해에 미치는 영향 (The Effects on Sperm Parameters and Membrane after Treatment with Progesterone and/or Acetyl-L-Carnitine; Cryopreservation-Thawing)

  • Objective : To assess the effects of progesterone and acetyl-L-carnitine used after treated with Isolate�� gradient before semen cryopreservation-thawing on sperm parameters and membrane integrity. Material and Methods : From April 2001 to July 2001, ten normal male partner of couples who were visited in vitro fertilization (IVF) clinics. the semens were treated with $Isolate^{(R)}$ gradient before cryopreservation, spermatozoa was incubated with progesterone (1, 5 and $10{\mu}M$), acetyl-L-carnitine (2.5, 5 and $10{\mu}M$), or both (progesterone, $1{\mu}M$; and acetyl-L-carnitine, $5{\mu}M$) for 30 min. Results: There were no differences in sperm parameters and vital stain among isolate only treated group, progesterone (1, 5 and $10{\mu}M$), acetyl-L-carnitine (2.5, 5 and $10{\mu}M$) and both (progesterone, $1{\mu}M$; and acetyl-L-carnitine, $5{\mu}M$). But, in high concentration of acetyl-L-carnitine ($10{\mu}M$) treated group, sperm parameters and vital stain were decreased. The statistical method was used ANOVA (Kruskal-Wallis test) and p value was <0.01. Conclusions : Neither progesterone nor acetyl-L-carnitine show to be protective effect on the cryodamage assessed by sperm parameters and vital stain (eosin-Y stain) in normal sperm. High concentration of acetyl-L-carnitine ($10{\mu}M$), however, was harmful effect on cryoprevention.
