  • The antifungal activity of silver, copper, and titania nanoparticles against fungal spores was investigated. Cladosporium cladosporioides spores were aerosolized and sampled on a solid agar plate using an Anderson impactor. The solid agar plate contained different concentration of nanoparticles ranging from 0 to $500{\mu}g/mL$. Silver and copper nanoparticles were shown to be an effective antifungal agent, while titania nanoparticles were not. Antifungal activity of these effective nanoparticles appeared at $300{\mu}g/mL$ concentration.

  • Cladosporium cladosporioides H1 was found to be the most abundant microbe in Janggyeong Panjeon. C. cladosporioides H1 produced a 20 kDa xylanase, which was generally stable below $60^{\circ}C$ and had specialized activity in an acidic condition. Our results may lead to the development of a strategy for preservation of organic cultural heritage environments.

Sooty Mold of Persimmon (Diospyros kaki) Caused by Cladosporium cladosporioides

  • In 2002, sooty mold was observed on persimmon (Diopyros kald) from the Jinju Agricultural Products Wholesale Market in Gyeongnam, Korea. The lesion of the sooty mold usually appeared as scars or wounds formed on the surface of the fruits. The symptom started with water soaking lesion, then the fruit softened rapidly. Colony of the causal pathogenic fungus was dark green in color on potato dextrose agar. The diameter of growing hyphae was 1-2 $\mu\textrm{m}$. The mostly one-celled conidia were ovoid, lemon or cylinder in shape, blastophores, and sized 46-132 $\mu\textrm{m}$. Erected conidiophores were long-branched chains and their lengths varied from 24 to 346 $\mu\textrm{m}$ with 2-5 $\mu\textrm{m}$ width. The optimum temperature for mycelial growth was about 2$0^{\circ}C$. Based on their mycological characteristics, the fungus was identified as Cladosporium cladosporioides. This is the first report in Korea of the sooty mold of persimmon caused by the said fungus.

Effect of Vibration on Dispersal of Cladosporium cladosporioides Bioaerosols

  • The vibration of fungal cultures was evaluated to determine its potential effect on the dispersal of airborne fungal microorganisms suspected of being pathogens. An artificial vibration system, which simulates the actual environmental vibration of fungal structures, was designed and constructed for this purpose. Experiments featured the use of low-frequency vibrations similar to those induced by earthquakes. Within the range of conditions tested, the vibration of fungal cultures was found to affect the airflow-driven generation of bioaerosols.

Cladosporium cladosporioides and C. tenuissimum Cause Blossom Blight in Strawberry in Korea

  • Blossom blight in strawberry was first observed in a green house in Nonsan, Damyang, and Geochang areas of Korea, between early January to April of 2012. Disease symptoms started as a grey fungus formed on the stigma, which led to the blossom blight and eventually to black rot and necrosis of the entire flower. We isolated the fungi purely from the infected pistils and maintained them on potato dextrose agar (PDA) slants. To test Koch's postulates, we inoculated the fungi and found that all of the isolates caused disease symptoms in the flower of strawberry cultivars (Seolhyang, Maehyang, and Kumhyang). The isolates on PDA had a velvet-like appearance, and their color ranged between olivaceous-brown and smoky-grey to olive and almost black. The intercalary conidia of the isolates were elliptical to limoniform, with sizes ranging from $5.0{\sim}10.5{\times}2.5{\sim}3.0{\mu}m$ to $4.0{\sim}7.5{\times}2.0{\sim}3.0{\mu}m$, respectively. The secondary ramoconidia of these isolates were 0- or 1-septate, with sizes ranging betweem $10.0{\sim}15.0{\times}2.5{\sim}3.7{\mu}m$ and $8.7{\sim}11.2{\times}2.5{\sim}3.2{\mu}m$, respectively. A combined sequence analysis of the internal transcribed spacer regions, partial actin (ACT), and translation elongation factor 1-alpha (TEF) genes revealed that the strawberry isolates belonged to two groups of authentic strains, Cladosporium cladosporioides and C. tenuissimum. Based on these results, we identified the pathogens causing blossom blight in strawberries in Korea as being C. cladosporioides and C. tenuissimum.

해인사 장경판전으로부터 분리한 곰팡이의 Xylanase 특성 (Characterization of Xylanase of Fungi Isolated from Janggyeong Panjeon in Haeinsa Temple)

  • 팔만대장경판과 이를 보존하고 있는 건물인 장경판전으로부터 미생물조사를 실시하였고 조사를 통해 분리된 미생물들이 생산하는 xylanase의 특성을 조사하였다. 조사 당시에 장경판전의 내부 온도는 $20^{\circ}C$ 안팎으로 적절하게 유지되고 있었지만 상대습도는 최대 80%에 이르는 곳도 있어 미생물과 같은 생물적 열화원의 발생으로 문화재의 손상 가능성이 있을 것으로 생각된다. 분리된 5종류의 미생물인 Cladosporium cladosporioides H1, Penicillium citreonigrum H3, Penicillium toxicarium H4, Aspergillus versicolor H6, Acremonium alternatum H7은 모두 재질의 표면을 오염시키고 유기질 (cellulose, xylan, lignin)을 분해시킬 수 있는 곰팡이였다. 이들은 $20^{\circ}C$의 저온에서도 성장이 활발하여 xylanase의 생산도 활발히 이루어졌다. 이들에 의해 생산되는 xylanase는 균주의 종류에 따라 생산 속도의 차이가 있었으며 대부분 산성인 조건에서 xylan을 효과적으로 분해하는 것으로 조사되었다. 또한, Cladosporium cladosporioides H1와 Penicillium toxicarium H4의 xylanase는 산성 조건에서 열에 안정한 것으로 나타났지만 그 외 균주들이 생산한 xylanase는 열에 대한 안정성은 높지 않은 것으로 조사되었다. 이러한 조사 결과는 문화재의 보존 환경의 조절이나 보존제 개발을 하는데 기초자료로 활용될 수 있을 것으로 보이며 생물학적 열화를 일으키는 미생물을 제어할 수 있는 대안을 마련하는데 도움을 줄 것으로 기대된다.

  • As the first step to develop efficient means to control fungal spore bioaerosols, we designed, manufactured, and evaluated a fungal spore aerosol generator. We studied the physical and biological properties of the fungal spore bioaerosols on two common fungal species. The results demonstrated that the fungal spore bioaerosol generator effectively produces fungal spore bioaerosols.

  • Fungal particles have been known to aggravate indoor air quality. To develop fungal particle cleaning devices requires a well-controlled generator of fungal aerosol particles. In this study, a novel fungal aerosol generator was designed and tested for anti-fungal experiment. Cladosporium cladosporioides was selected as test fungal particle. After aerosolization, the number concentration and the size of particles were measured by aerodynamic particle sizer. The number concentration depended on the vibration strength and vibration period of the designed fungal aerosol generator. For the vibration strength of 10volt and the period of 10 sec (5 sec on and 5 sec off), the stable particle generation with concentration of 10#/cm3 was maintained during 35 minutes.

The Origin of Meju Fungi - Fungal Diversity of Soybean, Rice Straw and Air for Meju Fermentation

  • Meju is a brick of dried fermented soybeans and is the core material for Jang such as Doenjang and Ganjang. Jang is produced by addition of salty water to Meju and is considered the essential sauces of authentic Korean cuisine. Meju is fermented by diverse microorganisms such as bacteria, fungi and yeasts. It is known that fungi play an important role in the Meju fermentation and they degrade macromolecules of the soybeans into small nutrient molecules. In previous study, 26 genera and 0 species were reported as Meju fungi. However, it is not comprehensively examined where the fungi present on the Meju are originated. In order to elucidate the origin of the fungi present on the Meju, the mycobiota of 500 samples soybean kernels, 296 rice straw pieces and air samples of Jang factories was determined in 0, 2 and 7 Jang factories respectively. Forty-one genera covering 86 species were isolated from the soybeans and 33 species were identical with the species from Meju. From sodium hypochlorite untreated soybeans, Eurotium herbariorum, Eurotium repens, Cladosporium tenuissimum, Fusarium fujikuroi, Aspergillus oryzae/flavus and Penicillium steckii were the predominant species. In case of sodium hypochlorite-treated soybeans, Eurotium herbariorum, E. repens and Cladosporium tenuissimum were the predominant species. Of the 4 genera and 86 species isolated from soybeans, 3 genera and 33 species were also found in Meju. Thirty-nine genera and 92 species were isolated from the rice straws and 40 species were identical with the species from Meju. Fusarium asiaticum, Cladosporium cladosporioides, Aspergillus tubingensis, A. oryzae, E. repens and Eurotium chevalieri were frequently isolated from the rice straw obtained from many factories. Twelve genera and 40 species of fungi that were isolated in the rice straw in this study, were also isolated from Meju. Especially, A. oryzae, C. cladosporioides, E. chevalieri, E. repens, F. asiaticum and Penicillium polonicum that are abundant species in Meju, were also isolated frequently from rice straw. C. cladosporioides, F. asiaticum and P. polonicum that are abundant in low temperature fermentation process of Meju fermentation, were frequently isolated from rice straw incubated at $5^{\circ}C$ and $25^{\circ}C$, while A. oryzae, E. repens and E. chevalieri that are abundant in high temperature fermentation process of Meju fermentation, were frequently isolated from rice straw incubated at $25^{\circ}C$ and $35^{\circ}C$. This suggests that the mycobiota of rice straw have a large influence in mycobiota of Meju. Thirty-nine genera and 92 species were isolated from the air of Jang factories and 34 species were identical with the species from Meju. In outside air of the fermentation room, Cladosporium sp. and Cladosporium cladosporioides were the dominant species, followed by Cladosporium tenuissimum, Eurotium sp., Phoma sp. Sistotrema brinkmannii, Alternaria sp., Aspergillus fumigatus, Schizophyllum commune, and Penicillium glabrum. In inside air of the fermentation room, Cladosporium sp., Aspergillus oryzae, Penicillium chrysogenum, A. nidulans, Aspergillus sp., C. cladosporioides, Eurotium sp., Penicillium sp., C. tenuissimum, A. niger, E. herbariorum, A. sydowii, and E. repens were collected with high frequency. The concentrations of the genus Aspergillus, Eurotium and Penicillium were significantly higher in inside air than outside air. From this results, the origin of fungi present on Meju was inferred. Of the dominant fungal species present on Meju, Lichtheimia ramosa, Mucor circinelloides, Mucor racemosus, and Scopulariopsis brevicaulis are thought to be originated from outside air, because these species are not or are rarely isolated from rice straw and soybean; however, they were detected outside air of fermentation room and are species commonly found in indoor environments. However, A. oryzae, P. polonicum, E. repens, P. solitum, and E. chevalieri, which are frequently found on Meju, are common in rice straw and could be transferred from rice straw to Meju. The fungi grow and produce abundant spores during Meju fermentation, and after the spores accumulate in the air of fermentation room, they could influence mycobiota of Meju fermentation in the following year. This could explain why concentrations of the genus Aspergillus, Eurotium, and Penicillium are much higher inside than outside of the fermentation rooms.

벼 이삭마름병해(病害)에 관여(關與)하는 균류(菌類)의 검출(檢出)과 검출(檢出)된 균류(菌類)가 종자(種子) 변색(變色) 및 수량(收量)에 미치는 영향(影響) (Detection of Fungi Associated with Ear Blighting of Rice Plants and Their Effect on Seed Discoloration and Yield)

  • 1. 벼 종자(種子)의 변색정도(變色程度)에 따른 균류(菌類)의 검출(檢出)을 조사(調査)한 결과(結果) 변색(變色)이 심한 종자(種子)에서 Alternaria tenuis, Cladosporium cladosporioides, Curvularia lunata, Fusarium semitectum, Gerlachia oryzae, Phoma sp. Trichoconiella padwickii 등이 높은 비율(比率)로 검출(檢出)되었고 특히 반점종자(斑點種子)에서는 Helminthosporium 균(菌)이 높은 비율(比率)로 검출(檢出)되었다. 2. 벼 종자상(種子上)에서 진균(眞菌)의 검출(檢出)에 미치는 광선(光線)의 영향(影響)을 조사(調査)한 결과(結果) Alternaria tenuis, Helminthosporium oryaze, Trichoconiella padwickii, Fusarium moniliforme, Fusarium semitectum 등(等)은 암흑구(暗黑區)에서 보다 광선조사구(光線照射區)에서 검출율(檢出率)이 높았고 Phoma sp. Curvularia lunata, Epicoccum purpurascens, Verticillium sp.는 광선조사구(光線照射區)나 암흑구(暗黑區)나 거의 차이가 없었으며 Gerlachia oryzae는 암흑구(暗黑區)에서 오히려 검출율(檢出率)이 높았다. 3. 벼 종자상(種子上)의 배양온도(培養溫度)가 균류(菌類)의 검출(檢出)에 미치는 영향(影響)을 조사(調査)하였던 바 Alternaria tenuis, Cladosporium cladosporioides는 저온구$(15^{\circ}C)$에서 검출율(檢出率)이 높았고 Phoma sp.는 고온구$(30^{\circ}C)$에서 검출율(檢出率)이 높았다. 4. 검출(檢出)된 각(各) 균류(菌類)가 종자(種子) 변색(變色)및 수량(收量)에 미치는 영향(影響)을 조사(調査)하였던 바 공시(供試)한 7게(個)의 균류(菌類) 모두가 종자(種子) 변색(變色)에 관여(關與)하여 인공접종(人工接種)한 이삭에서의 천립중(千粒重)은 대조구의 천립중(千粒重)에 비해 모두 낮았고 특히 공시(供試)한 7개(個)의 균(菌)을 혼합접종(混合接種)한 이삭의 천립중(千粒重)과 등숙비율(登熟比率)은 대조구보다 저하(低下)됨을 알 수 있었다.
