감귤과피로부터 발암 promotion 억제활성성분의 분리 (Isolation of the Anti-tumor Promoters from Citrus Peels)

  • 하귤(Citrus natsudaidai Hayata)의 과피를 cold-press하여 얻어진 정유성분을 column chromatography, HPLC 및 TLC로 분리 정제하여 각 분획에 대하여 EBV활성화에 대한 억제효과를 측정하였다. Column chromatography로 정유성분의 분리를 시도하여 7개 의 peak$(F-I{\sim}VII)$를 얻을 수 있었다. 이것들을 TLC로 분석한 결과, 각각 단일 반점만을 나타냈으며 $R_f$값은 0.31, 0.13, 0.13 0.78, 0.79, 0.69, 0.84이었다. 7개의 peak중에서 F-I, II 및 F-IV 등 3개의 peak에 대하여 HPLC분석을 한 결과 F-I 및 F-II는 단일 peak이며 retention time은 F-I이 3분, F-II는 2.5분이었다. 그러나 F-IV는 2개의 peak가 나타났으며 retention time은 각각 2분과 4.5분이었다. 4.5분에 나타난 peak는 극히 적은 면적으로 보아 극소량의 물질이어서 앞서의 TLC분석에서 나타나지 많았던 것으로 생각된다. $F-I{\sim}F-VII$에 대한 EBV활성화에 대한 억제효과 측정은 Raji cells을 이용하여 간접형광 항체법으로 실시했다. 억제율은 F-VI가 $82.3{\pm}1.3%$으로 가장 높았으며 다음이 F-I$(80.4{\pm}1.6%)$ > F-II$(77.2{\pm}0.9%)$ > F-III$(75.0{\pm}1.2%)$ > F-IV$(74.1{\pm}1.0.%)$ > F-V$(71.0{\pm}1.1%)$ > F-VII$(70.2{\pm}1.2%)$의 순으로 모두 70% 이상의 억제율을 나타냈다. 이상의 결과는 하귤과피에는 발암 promotion 억제활성성분이 존재한다는 것을 시사한 것이다.

제주산 감귤 품종별 carotenoid, 색도, UV 스펙트럼, 유기산 및 유리당 함량 (Carotenoid, Color value, UV Spectrum, Organic Acid and Free Sugar Contents of Citrus Varieties Produced in Cheju)

  • Total carotenoid contents, color values and spectrum characterization in the range of UV of fruit juice and organic acid and free sugar contents determined by HPLC were investigated on 10 varieties of Cheju citrus fruits. Carotenoid contents of juice were 0.47-9.20$\mu\textrm{g}$/ml which showed higher with Dangyooja while showed lowest content with Meiwa Kumquat. Lightness (L) of juice was highest in Meiwa Kumquat, and in the order of Sankyool, Navel orange and Hungjin. Redness(a) and yellowness(b) were in the order of Dangyooja, Natsudaidai, Sambokam and Kinkoji. The maximum absorption wavelengths if citrus Juices were variable at 269.5-285.5nm according to varieties and it was observed that were changed by mixing with Juices of other varieties. Major organic acids of citrus juice were citric and malic acid, and citric acid was 64.4-95.1% of total organic acids. Ascorbic acid was high in Navel orange, Sudachi and Dangyooja, and low in Meiwa Kumquat and Hungjin. Major free sugars were sucrose, glucose and fructose. Sucrose was 49.2-75.2% of total free sugars, and high in Sankyool and Meiwa Kumquat. Glucose and fructose were high in Meiwa Kumquat and Navel Orange.

감귤류 종자 추출물의 향균활성 (Antimicrobial Activity of Extracts from Citrus Seeds)

  • To develope natural food preservatives antimicrobial effect of the natural products against food-related bacteria and yeast The purpose of this study was evaluate antimicrobial effect of the citrus seeds. antimicrobial activities of methanol extracts from the citrus seeds investigate against Escherichia coli O26, Staphylococcus aureus 6358, Saccharomyces cerevisiae IBM 4274, Bacillus licheuiformis 9945a and Alcoligenes faecalis. Citrus seeds is containing to moisture 4∼6.5%, curd protein 11∼15%, curd lipid 32∼46%, curd carbohydrate 22∼45 % and ash 2∼4 %, that is containing to flavornoid 12∼48mg% and phenolic compound 22∼53mg%. Solidity content of the methanol extract from the citrus seeds was 0.8∼1.2%. Almost all of the methanol extracts from citrus seeds exhibited growth inhibiting activities for most of microorganisms tested. The methanol extracts from Citrus grandis, C. sunki, C. sulcata showed the growth inhibitory effects against Escherichia coli O26. The methanol extracts from C. obovoidea, C. sulcata, C. aurantium showed the growth inhibitory effects against Staphylococcus aureus 6358. The methanol extracts from C. obovoidea, C. sulcata, C. tangerina showed the growth inhibitory effects against Saccharomyces cerevisiae IBM 4274. The methanol extracts from C. obovoidea, C sunki, C. sulcata, C. tangerinan, C. natsudaidai, C. iyo, C. aurantium showed the growth inhibitory effects against Bacillus licheuiformis 9945a. The methanol extracts from C. obovoidea, C sunki, C. sulcata, C. aurantium showed the growth inhibitory effects against Alcoligenes faecalis. Among this especially, Showed growth inhibiting activity of the methanol extracts from Citrus sulcata that about microorganisms investigated. If apply searching suitable application method about such the citrus seeds antimicrobial activity, role as good antimicorbial material in storage or cooking of food, processing is expected.

감귤 품종별 이화학적 성분 비교 (Comparison of physico-chemical components on citrus varieties)

  • Physico-chemical components which are closely related to processed products were investigated on 10 varieties of Cheju citrus fruits. Juice ratio of Hungjin was the highest, 49.2%, while Sankyool was the lowest, 4.2%, which showed great differences among varieties. Peel ratio of Dangyooja was the highest, 46.1%, while both Navel orange and Hungjin were relatively low. Soluble solid($^{\circ}$Brix) was relatively high In Sankyool, Meiwa Kumquat, Sambokam and Iyo. Acid content was the highest, 4.86% in sudachi and relatively high in Sankyool, Natsudaidai and Dangyooja. The $^{\circ}$Brix to acid content ratio was 13.9 in Meiwa Kumquat and more than 10 in Navel orange and Hungjin. Total sugar contents of Juice were 2.78∼10.94%, while reduced sugar contents were 1.63∼6.38% which showed higher in Meiwa Kumquat, Iyo and Navel Orange. Hesperidin and naringin, the sources of biiter taste and cloudness were low in Hungjin and Iyo. Soluble solid($^{\circ}$Brix) of citrus juice showed highest statistical relationship(r=0.907) with total sugar, and was highly significant at 1% level.

감귤류 종자 추출물의 N-nitrosodimethylamine 생성 억제에 관한 연구 (Inhibition of N-nitrosodimethylamine Formation of Extracts from Citrus Seeds)

  • 감귤 6종류의 종자를 메탄올로 추출하여 이들 추출물에 의한 N-nitrosodimethylamine(NDMA) 생성억제효과를 검정하였다. 감귤류 종자의 수분함량은 4∼6%, 조단백질 함량 11∼15%, 조지방 32∼46%, 탄수화물 함량은 22∼45%이었으며, 회분함량은 2∼4%, 플라보노이드는 12∼24mg%, 폴리페놀함량은 22∼53mg%이었다. 감귤종자 메탄올 추출물의 추출수율은 0.7∼1.1%, 고형분 함량은 0.8∼1.2%이었다. 감귤종자 메탄올추출물에 의한 아질산염의 소거효과는 추출물의 첨가량이 증가할수록 그 소거율이 증가였다. 또한 아질산염 소거능은 pH 의존성이 커서, pH 1.2(42.7∼96.9%)에서 높은 가장 높은 소거효과를 보였으며 pH 6.0(19.9∼62.6%)에서 가장 낮은 효과를 보였다. 게다가 반응시간에 따른 아질산염 소거효과는 3시간 반응시켰을 때 가장 높았다. NDMA 생성 억제효과는 감귤 종자 추출물의 농도가 증가할수록 컸다. 즉 감귤종자 추출물을 각각 1, 3 및 5$m\ell$ 첨가하였을 때 NDMA 생성 억제율은 각각 1.2∼39.8%, 21.3∼60.1% 및 47.4∼94.0%이었다.

Composition and Antimicrobial Activities of Essential Oils in the Peel of Citrus Fruits

  • Volatile components of the essential oils of Satsuma mandarin (C. unshiu), Dangyuza (C. grandis), Yuza (C. junos), Byungkyul (C. playtymamma), Jinkyul (C. sunki), and Hakyul (C. natsudaidai) grown in Jeju Island were isolated from the fruit peels by hydro distillation and determined by GC-MS. GC-MS analysis identified 58 compounds, with main components being d-limonene $(64.01{\sim}79.34%),\;{\beta}-myrcene\;(3.01{\sim}26.53%),\;{\gamma}-terpinene\;(0.11{\sim}12.88%),\;{\beta}-pinene\;(0.78{\sim}4.74%),\;and\;{\alpha}-pinene\;(1.01{\sim}2.55%)$. Differences in compositions and contents of the essential oils were observed among citrus varieties. Effects of citrus oils on growth inhibitions of Escherchia coli, Staphyllococcus epidermidis, and Candida albicans were investigated using disc diffusion assay and minimal inhibitory concentration (MIC) assay. The essential oils inhibited growths of the test organisms, exhibiting higher levels of activity against Gram-positive S. epidermidis (MIC values $0.04{\sim}0.17mg/mL$), whereas Gram-negative E. coli was moderately resistant (MIC values $1.66{\sim}20.30mg/mL$). MIC of citrus essential oils ranged from $0.82{\sim}23.69mg/mL$ against C. albicans. The essential oils obtained from C. sunki, C. grandis, and C. playtymamma showed the highest antimicrobial activities against S. epidermidis and C. albicans, indicating their potential as natural antimicrobial agents.

  • 제주도에서 재배되어 온 재래귤 5종(당유자, 유자, 청견, 하귤, 팔삭) 부위별 에탄올 추출물의 총 폴리페놀 함량 측정, 총 플라보노이드 함량 측정, DPPH radical 소거 활성 실험을 실시하였다. 총 폴리페놀 함량의 경우, 5종 재래귤(당유자, 유자, 청견, 하귤, 팔삭) 부위별 에탄올 추출물 가운데 당유자의 외피(244.84mg/g)와 중피(200.70mg/g) 처리구에서 폴리페놀 함량이 가장 높았다. 총 플라보노이드 함량의 경우, 총 폴리페놀의 함량 측정에서와 마찬가지로 5종 재래귤(당유자, 유자, 청견, 하귤, 팔삭) 부위별 에탄올 추출물 가운데 당유자의 외피에서 102.12mg/g으로 가장 높았으며, 청견과 하귤, 팔삭의 플라보노이드 함량은 폴리페놀 처리구와 같이 당유자 처리구보다 적게 나타났다. DPPH radical 소거 활성의 경우, 총 폴리페놀 함량과 총 플라보노이드 함량 측정의 결과에 같은 경향으로 5종 재래귤(당유자, 유자, 청견, 하귤, 팔삭) 부위별 에탄올 추출물 가운데 당유자의 외피(79.39%)와 중피(70.73%)에서 가장 양호한 항산화 활성을 나타내었다. 이러한 결과들로부터 당유자의 두꺼운 과피 구조인 외피와 중피에 에센셜 성분인 limonene, myrcene, β-ocimenes 등과 같은 향기성분들의 영향으로 생각되어 앞으로 이에 대한 연구가 더 필요하리라 생각된다.

온주밀감 신품종 '하례조생' 육성 (A New Early Maturing Satsuma Mandarin Cultivar, 'Haryejosaeng')

  • A new early maturing satsuma mandarin (Citrus unshiu Marc.) 'Haryejosaeng' was developed as a nucellar seedling selection of 'Tachima Wase' (C. unshiu Marc.) that was crossed with C. natsudaidai Hayata at the National Institute of Subtropical Agriculture in Jeju island in 1992. The 'Jegam ga No. 2', a first selection from the seedlings fruited in 2001 was finally named "Haryejosaeng" through field evaluation trials at three locations in Jeju island from 2003 to 2004. 'Haryejosaeng' produces seedless fruit maturing in early November, and has higher soluble solids and lower acidity than 'Miyagawa Wase', the leading early-maturing satsuma mandarin cultivar in Jeju island. Fruit weight is moderate at about 80~90 g and its shape is compressed-oblate globose with a light orange color. The rind thickness of about 2 mm provides easy peeling. The flesh shows light orange colored and contains 10 to 11 Brix and 1 to 1.1% acidity when mature. Tree shows vigorous growth and spreading thornless twigs. Alternate bearing is similar to 'Miyagawa Wase'. 'Haryejosaeng' is susceptible to citrus scab disease and melanose, but resistant to citrus canker.

제주산 감귤류의 숙기에 따른 유리당, 유기산, 헤스페리딘, 나린진, 무기물 함량의 변화 (Free sugar, Organic acid, Hesperidin, Naringin and Inorganic elements Changes of Cheju Citrus Fruits According to Harvest Date)

  • 대표적인 제주산 감귤류인 하귤, 당유자, 병귤, 스타치, 지각, 궁천조생, 6품종을 수확시기에 따라 유리당, 유기산, 헤스페리딘, 나린진, 무기물을 각각 분석하였다. 수확시기별 유리당의 함량에는 조금씩 차이를 보였으나 모든 품종에 있어서 과즙내 유리당 중에서 sucrose가 $44.9{\sim}66.0%$로 가장 많이 함유되어 있었고 glucose와 fructose는 $15.7{\sim}25.7%,\;17.5{\sim}30.1%$로 서로 비슷하게 함유되어 있었다. 유리당의 함량은 성숙함에 따라 계속적으로 증가하는 경향을 보였다. 유기산 함량은 감귤이 성숙됨에 따라 점차적으로 감소되며, 이는 산 함량의 변화와 비슷한 경향을 보였다. 감귤과즙에서 검출된 주요한 유기산은 citric acid, malic acid, oxalic acid였으며 전체 유기산 중에 citric acid가 90%이상으로 대부분을 차지하고 있었고, 그 외로 malic acid가 $0.98{\sim}9.45%$ 내외로 함유되어 있었고 oxalic acid가 3.58% 이하로 미량 함유되어 있었다. 품종별 과즙과 과피내의 flavonoid계 성분인 naringin과 hesperidin의 함량을 분석한 결과 이 두 성분은 과피 중에서 월등히 많이 함유되어 있었고 미숙과일 때 그 함량이 높았으나 착색이 진행됨에 따라 점차적으로 감소하는 경향을 보였다. 감귤을 이용한 가공품 제조시에는 완숙과를 사용해야 가공품의 혼탁과 쓴맛이 적을 것으로 예상되었다.

Chemical profile and antioxidant activity of peel of Yellowball, a novel citrus variety

  • Yellowball (Citrus hybrid cv. Yellowball ) is a new citrus hybrid between Haruka (C. tamurana × natsudaidai ) and Kiyomi (C. unshiu × sinensis) and is known to possess strong antioxidant activity. However, detailed information on the antioxidant components of its peel has not yet been reported. This study evaluated the antioxidant activity of the peel and identified the antioxidant components by fractionating a methanolic extract of Yellowball peels using liquid-liquid extraction with n-hexane, ethyl ether (ether), ethyl acetate (EA), butanol, and water. The phenolic contents and antioxidant activities of the n-hexane, ether, and EA fractions were higher than those of the other fractions, and these fractions were further separated by semi-preparative high-performance liquid chromatography (HPLC). Four antioxidant peaks, EA1, EA2, EA3, and He1, were isolated and analyzed using ultra-performance liquid chromatography-quadrupole-time- of-flight mass spectrometry (UPLC-Q-TOF MS). Sinapoyl glucoside and hesperidin were identified in EA2 and EA3, respectively, and a polymethoxylated flavone (PMF) complex (5-hydroxy-3,6,7,8,3',4'-hexamethoxyflavone, natsudaidain, tetrameth- oxyflavone, and tangeretin) was identified in He1. A compound in EA1 with m/z 223.0246 [M-H] could not be identified and was named unknown2. The antioxidant activity of unknown2 (IC50=69.17 ㎍/mL) was similar to that of Trolox, which was noted as a major antioxidant in Yellowball peel. Further studies on the antioxidant capacity of Yellowball peel are required; however, these results provide a foundation for using Yellowball peel as an antioxidant.
