Effects of Cirsium setidens nakai on In Vitro Growth and Osteogenic Differentiation of Human Bone-Derived Mesenchymal Stem Cells

  • Cirsium setidens nakai belonging to cirsium has been reported to have various physiological activities including anticancer activity because it contains polyphenols, dietary fiber, minerals and vitamins. Despite these positive positive efficacies, however, no studies have studied cirsium setidens nakai products as biomaterials such as cellular metabolism and bone formation. Thus, the aim of this study was evaluate of osteogenesis differentiation a natural material extracted from cirsium setidens nakai. The natural materials in this studys in this studywere created by 40% ethanol extraction process and then dried. FabricatedFabricatedpowders were added to a medium at various concentrations (0.01, 0.05, 0.1, 0.2, and $0.25{\mu}g/mL$), and pure medium was used as a control. The natural material caused positive increases in cell metabolic activity and mineralized bone formation without cytotoxicity. In addition, we observed higher expression of genes such as ALP, BSP, Runx2 and COL1 in cirsium setidens nakai treatment cells. As a result, this study produced and investigated cirsium setidens nakai extracts and the natural materials showed potential biomaterials. In this research indicated that the cirsium setidens nakai extracts might have promising applications in areas of agricultural, biological and food engineering as a biomaterial.

곤드레 첨가 설기떡의 품질특성 (Quality Characteristics of Sulgidduk added with Cirsium setidens Nakai)

  • 본 연구는 생리활성 기능을 나타내는 곤드레 분말을 설기떡에 첨가하여 품질특성을 조사한 결과, 수분함량은 무첨가구(40.67%)에 비해 곤드레 분말 첨가군(36.51~38.43%)이 낮게 나타났고, 색도는 곤드레 분말 첨가량이 증가함에 따라 L값은 유의적으로 감소하였으며, a값과 b값은 유의적으로 증가하는 경향을 나타냈다(p<0.05). 곤드레 분말 첨가량이 증가할수록 경도, 탄력성, 씹힘성은 증가하나, 응집성은 감소하는 경향을 나타내었다(p<0.05). 관능검사 결과, 전반적인 기호도에서는 곤드레 분말 5% 첨가 설기떡 > 1% 첨가 설기떡 > 3% 첨가 설기떡 > 0% 첨가 설기떡 > 7% 첨가 설기떡 순으로 높게 평가되었다. 또한, 곤드레 분말 첨가량을 달리한 설기떡을 $18^{\circ}C$에서 3일간 저장하는 동안의 총균수 변화는 제조 직후 모든 첨가군의 총균수 함량은 2(logCFU/g) 이하이었고, 3일 저장하는 동안 0%(4.02), 1%(4.94), 3%(5.94), 5%(6.28), 7%(6.9.8)로 곤드레 첨가량이 증가할수록 총균수가 다소 증가하였으나, 곤드레 설기떡은 저장하는 동안 미생물의 증식을 억제하지 못하는 것으로 사료된다. 곤드레 분말 첨가 설기떡 제조시 다른 첨가군에 비해 가장 높게 평가된 곤드레 분말 5% 첨가구가 설기떡 제조의 가장 적합한 것으로 사료되며, 향후 다양한 제품에 곤드레 분말을 이용하고자 한다.

자생 엉겅퀴의 부위별 기능성 성분 및 항산화 효과 (Functional Composition and Antioxidative Activity from Different Organs of Native Cirsium and Carduus Genera)

  • This study was conducted to investigate the functional composition and antioxidant activity of Cirsium and Carduus genera based on different parts. Leaves of Cirsium setidens Nakai contained 23.66% protein and seeds of Carduus crispus L contained 25.30% lipid. Extraction yields of Cirsium and Carduus genera were higher in leaves than in any other parts of the plants. Total phenolics and total flavonoid content were abundant in extracts of leaves, steam and root of C. japonicum var. ussuriense, and the flower extract of C. setidens Nakai. Silymarin was not found in extracts of Cirsium and Carduus genera. Acacetin was identified in leaf or flower extracts of C. setidens Nakai, or in leaf and steam extracts of C. pendulum Fisch ex DC. Apigenin was identified in the flower extracts of Cirsium and Carduus genera and constituted 7.16 mg/g in C. japonicum var. ussuriense. Cynarin was present at 5.55 mg/g in the seed extract of C. setidens Nakai, and narirutin represented 19.56 mg/g and, 4.18 mg/g of the seed extracts of C. pendulum Fisch ex DC and, Carduus crispus L, respectively. 2,2-Diphenyl-1-2-picrylhydrazyl (DPPH) radical scavenger activity was higher in flower extracts of C. setidens Nakai than in the other genera. Photochemiluminescence activity was 2.3 nmol in leaves and flower extracts of C. setidens Nakai and flower extract of Carduus crispus L. Flowers and seeds of C. setidens Nakai shoe potential as new functional materials.

고려엉겅퀴 및 컴프리를 이용한 양조간장의 개발 (Decelopment of Fermented Soysauce Using Cirsium setidens Nakai and Comfrey)

  • In order to develop new soysauce using mountain edible herbs, cirsium setidens Nakai and comfrey were added to the raw material of soysauce as a partially substituting ingredient to wheat composing the total nitrogen. The changes of chemical composition of soysauce by the substitution of 1~5% cirsium setidens after four months of aging were similar to those of the control soysauce. On the other hand, total solid content was decreased, while the pH was increased by more than 7%. The total nitrogen and solid contents in the confrey substituted soysauce were increased with the increment of the amount of comfrey, but showed little changes of the pH. The mineral contents of calcium and potassium in the soysauce after four months of aging at 10% substitution of cirsium setidens were increased by 1.7 and 1.4 times, respectively. A significant increase in calcium(2.3 times as compared to the control) was also observed in 7% substitution of comfrey. The amino acid contents such as methionine, tyrosine and histidine were markedly increased at 3% substitution of cirsium setidens. With 5% substitution of comfrey, the contents of tyrosine, glutamic acid methionine were increased by 4.4, 1.8 and 1.7 times, respectively as compared to the control. The results of sensory evaluation revealed that overall acceptability of soysauce with 1~5% substitution of cirsium setidens and 5% of comfrey exceeded other samples composing different ratios of substituting ingredients.

고려엉겅퀴, 정영엉겅퀴 및 동래엉겅퀴의 분류학적 실체 검토 (Taxonomic Identities of Cirsium setidens, C. chanroenicum and C. toraiense)

  • 엉겅퀴속의 한국 특산종으로 인식되어온 고려엉겅퀴 (Cirsium setidens)와 정영엉겅퀴 (Cirsium chanroenicum) 그리고 동래엉겅퀴 (Cirsium toraiense)를 대상으로 분류군의 한계와 그 실체를 명확히 밝히고자 외부 형태학적 형질을 중심으로 수리분류학적 연구를 수행하였다. 고려엉겅퀴와 정영엉겅퀴는 주요 식별형질인 잎, 화서 및 총포에 관한 29개의 외부 형태학적 형질과 잎에 관한 12개의 주요 식별형질들을 비교 분석한 결과, 거의 구별이 되지 않는 하나의 집단으로 나타났다. 이러한 결과, 한국 특산종인 정영엉겅퀴는 고려엉겅퀴에 포함하는 것이 적절하다고 판단되며, 또한 동래엉겅퀴는 원기재문에 의한 생육지 조사, 기준표본과 표본관 소장 석엽표본 조사 등의 결과, 그 실체를 확인할 수 없었다.

고려엉겅퀴(Cirsium setidens (Dunn) Nakai)의 구성성분 및생리활성에 관한 리뷰 (A review on Phytochemistry and pharmacological Activities of Cirsium setidens (Dunn) Nakai)

  • Objectives : The objective of this study was to investigate the phytochemistry and pharmacological activities of Cirsium setidens. Methods : Domestic and international articles about Cirsium setidens were investigated. A review was perfoemed via DB searching engine such as Sci.Direct, Springer, DBpia, KISS, Google scholar, Kipris, and so on. Total 73 listed literature were classified by compound analysis and pharmacological efficacy. Results : C. setidens contains pectolinarin and its glycoside, pectolinarigenin as index compounds, and linarin, apigenin, diosmetin, scopoletin, acacetin, cirsimarin, cirsimaritin, setidenosides A and B, silymarin, hispidulin, 92 volatile compounds, and 15 fatty acids. The Pharmacological activities of C. setidens has been reported to inhibit of platelet aggregation and fat accumulation in the liver, inhibit to hepatitis, anti-cancer, antibacterial, skin improvement, hair growth, liver protection, anti-diabetic, anti-inflammatory, sedative. Also, It has been reported the effect of cholesterol-lowering and anti-obesity, neuroprotective effects, increasing human stem cell viability, inhibiting osteoclast formation and osteogenic differentiation. Conclusion : This reviews showed that C. setidens which has been traditionally used for the treatment of inflammation and hypertension, has anticancer and river protective effect, as well as hair loss and diet. In order to maximize the efficacy of C. setidens, research has also begun on the effect of processing processes such as fermentation or fine powdering and combining natural plant resources.

건조 방법에 따른 곤드레 나물의 품질 특성 (Quality Characteristics of Cirsium setidens Nakai by Different Drying Method)

  • 본 연구에서는 고품질의 곤드레 묵나물 제품을 얻기 위한 방법으로 열풍건조와 순환형감압건조기술을 이용하여 품질 특성을 비교 분석하였다. 건조방법에 따른 곤드레의 건조시간별 중량 변화는 0~6시간 구간에서 가장 많은 중량 감소를 나타내었으며, 시간별 중량변화의 폭은 순환형감압건조에 비해 열풍건조에서 더 크게 나타났다. 건조 곤드레의 색도 변화는 L값(명도), a값(적색도), b값(황색도) 모두 순환형감압건조에 의한 곤드레가 높은 값을 나타내었다. 건조 방법에 따른 곤드레의 일반성분 및 무기질을 분석한 결과, 열풍건조 및 순환형감압건조의 건조 방법은 일반성분 및 무기질 함량에는 큰 영향을 미치지 않는 것으로 나타났다. 건조 방법에 따른 곤드레 추출물의 총 폴리페놀 함량은 순환형감압건조된 시료가 열풍건조 시료보다 높은 함량이 측정되었으며, 이는 열에 의해 페놀성분의 일부가 파괴되었음을 나타낸다. 또한, 건조 방법에 따른 곤드레 추출물의 DPPH radical 소거능을 비교한 결과, 순환형감압건조 시료는 생시료와 비슷한 수준의 높은 활성을 유지하고 있는 것으로 나타났다. 곤드레 묵나물을 제조하기 위한 효율적인 건조방법은 열풍건조방법보다는 순환형감압건조를 통해 우수한 품질과 기능성을 갖는 곤드레 묵나물을 제조할 수 있을 것으로 판단된다.

고려엉겅퀴 주정추출물의 안정성 조사 (Stability of Ethanolic Extract from Cirsium setidens Nakai)

  • 본 연구에서는 고려엉겅퀴 추출물에 대한 열안정성, pH 안정성을 조사하고자 고려엉겅퀴 주정추출물은 온도별(4, 25 및 $50^{\circ}C$), pH별(4.0, 7.0, 및 10.0)로 저장하면서 추출물의 pectolinarin 함량, 총 페놀 함량, 2,2-diphenyl-1-picrylhydrazyl (DPPH) 라디칼 소거능 및 FRAP assay를 분석하였다. 고려엉겅퀴 추출물의 pectolinarin 함량은 저장기간이 증가함에 따라 다소 감소하는 경향을 보였으며, 온도에 의한 영향보다 pH에 의한 영향이 큰 것으로 나타났다. 총 페놀 함량도 pectolinarin 함량과 저장기간이 증가함에 따라 같이 감소하는 경향을 보였으며 저온($4^{\circ}C$)에서 보다 높은 온도인 $25^{\circ}C$$50^{\circ}C$에서 큰 감소치를 보였고 산성(pH 4.0)과 중성(pH 7.0)에서는 비교적 변화가 적었다. 항산화 활성은 저장기간이 증가함에 따라 DPPH 라디칼 소거능은 감소하는 경향을 보였으나 FRAP은 온도별, pH별 저장조건에 따른 큰 변화를 보이지 않았다. 이상의 결과를 고려해 볼 때, 고려엉겅퀴 주정 추출물은 산성(pH 4.0)과 중성(pH 7.0)조건에서 저온($4^{\circ}C$) 저장 시 추출물의 안정성을 최적화 할 수 있을 것으로 판단되었다.

사자발쑥과 고려엉겅퀴 추출물의 항산화 및 간암세포 활성 효과 (Antioxidant and Anticancer Effects of Extracts and Components from Artemisia princeps Pampanini and Cirsium setidens Nakai)

  • In the present study, we investigated the compositions, antioxidant activities and anti-tumor effects of Artemisia princeps Pampanini (APD) as well as blanched leaves (CNBD) and dried leaves (CND) from Cirsium setidens Nakai on HepG2 cells. Water and ash contents were increased in CND. Protein and lipid contents were increased in CNBD. K, Ca, Mg, Fe and Cu contents of CND were higher than those of CNDB and APD. P contents was significantly decreased in CND. Yields of CND was reached high levels, but TPC, TFC, acacetin, apigenin, cynarin contents, and antioxidant activity were higher in APD. Viability of HepG2 liver cells was significantly decreased in APD. Therefore, extracts of APD are more effective preventing the liver cancer than extracts of CND and CNBD.

Pectolinarin 고함유 곤드레 미세분말의 항산화 및 항비만 활성 (Antioxidant and Anti-Obesity Properties of Pectolinarin-rich Cirsium setidens Nakai Fine Powder)

  • The aim of this study was to evaluate the basic data of Cirsium setidens Nakai fine powder (FPC), which will then be used in the development of functional fooditems. We measures and evaluated the level of pectolinarin content, phenol content, flavonoids content, antioxidants and anti-obesity properties of FPC. Our results from the study showed that the pectolinarin, phenol, and flavonoids contents of FPC measured at 10.95±0.1 5mg/g, 12.92±0.18 mg gallic acid equivalent (GAE)/g and 26.47±0.77 mg rutin equivalent (RE)/g, respectively. The exhibited antioxidant activity of FPC increased significantly depending on the dosage, and additionally. FPC did not show any cytotoxicity up to the dosage level of 500 μg/mL. During adipocyte differentiation, FPC significantly inhibited ROS production and lipid accumulation, compared with the control substance. These results suggest that FPC could be considered a promising resource of natural antioxidants and could serve a variety of health-improving roles in the production of functional food ingredients.
