TNBS로 유발된 마우스 대장염모델에서 설파살라진, 육계와 시호 복합 추출물의 항염증 및 세포 자멸 개선효과 (Anti-inflammatory and apoptosis improving effects of sulfasalazine and Cinnamomi cortex and Bupleuri radix mixture in TNBS-induced colitis mouse model)

  • 염증성 장 질환에 일반적으로 사용되는 설파살라진은 고용량 및 장기 섭취 후 다양한 부작용이 있다. 본 연구에서는 TNBS로 유발된 마우스 대장염 모델에서 설파살라진, 육계와 시호 복합 추출물의 항염증 및 세포 자멸 개선효과를 확인하고자 하였다. 실험은 정상군, TNBS 대조군, Sulfasalazine (30 mg/kg)군, Sulfasalazine (60 mg/kg)군, Sulfasalazine (30 mg/kg)+육계 및 시호 혼합 (30 mg/kg)군, 총 5개의 군으로 나누었으며, 7일간 경구투여 하였다. 염증 및 세포 자멸 단백질은 western blot을 통해 발현량을 확인하였다. SCB 투여는 염증 단백질 및 세포 자멸과 관련된 단백질의 억제에 유의한 효과를 나타냈다. 이러한 결과로 보아 설파살라진, 육계와 시호 복합 추출물은 염증과 세포 자멸의 억제를 통해 염증성 장 질환을 개선시킬 수 있으며, 염증성 장 질환의 치료 대안 가능 물질로 사료되는 바이다.

  • Objectives: In this study, we investigated the antiobesity and antidiabetic effects of polyherbal extract, DM2 consisting of Atractylodis Rhizoma, Anemarrhenae Rhizoma, Cinnamomi Cortex, and Moutan Radicles Cortex in high fat diet-induced obesity mice. Methods: DM2 extract was prepared with a hot water. Six-week-old male C57BL/6N mice were fed a high-fat diet (HFD) for 8 weeks and then administrated with DM2 extract (500 mg/kg, p.o.) for 4 weeks. The changes of physiological markers, body weight (BW), food and water intakes, and the levels of fasting blood glucose (FBG) were measured once a week for 4 weeks in mice. The the serum levels of glucose, insulin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), total cholesterol (T-CHO), triglyceride, and low density lipoprotein cholesterol in sera were measured in mice using autometic chemical analyzer and enzyme linked immunosorbant assay. We also observed the histological changes of liver and pancreatic tissues with Hematoxylin & Eosin staining. Results: In physiological change, the increases of BW, calorie intake, and FBG in HFD-induced obese mice were significantly decreased after administration of DM2 extract for 4 weeks. The decrease of water intake was significantly increased in DM2 extract-administrated mice. In serological change, the administration of DM2 extract in obesity mice was significantly decreased the serum levels of glucose, insulin, T-CHO, AST, and ALT levels. We also found that DM2 extract inhibited the increase of lipid droplets in liver and the structural destruction of pancreatic tissues in obesity mice. Conclusion: Our study demonstrated that DM2 extract has antiobesity antidiabetic effects with body weight loss, decrease of glucose and insulin levels, and lipid accumulation on liver tissue.

사상체질 약물의 항당뇨효능에 관한 실험적 연구 (Experimental Study of Inhibitory Activity of Sasang Medicines on Diabetes)

  • The purpose of this research was to investigate inhibitory activity of Sasang medicines on diabetes. Reduce 36 kind of medicines to powder, abstract add 100 times methanol. Examine inhibitory activity on α-amylase and α-glucosidase in vitro. Thirty six oriental herbs were screened for inhibitory activities against carbohydrate digesting enzymes in vitro. Methanol extract of Pleuropterus multiflorus Turez, Ephedra sinica, Moutan cortex Radicis, Magnolia obovata, Alpinia officinarum, Amomi Semen, Rubus coreanus, and Cinnamomi cortex inhibited yeast α-glucosidase activity and extract of Moutan cortex Radicis and Rubus coreanus inhibited porcine α-amylase activity by more 50%. Rubus coreanus showed the strongest inhibition against both α-glucosidase and α-amylase. Therefore, Rubus coreanus was the most effective medicice of 36 kind of medicines.

  • The anti-allergy activity of Cinnamomum cassia cortex extract and its fractions were investigated in mice. The extract exhibited potent anaphylactic shock and Arthus reaction. Among the fractions obtained in the successive fractionation with n-hexane, butanol and acetone, the butanol insoluble portion was shown to have the activities.

천연산물로부터 항산화물질의 탐색

  • 30 kinds of natural products were considered for developing natural antioxidants by improved D.O. analyzing method including simple calculation of Area Under Curve. Several natural products such as Cimicifuge Rhizoma, Epimedii Herba, Atractylodis Rhizoma Alba, Acori Graminei Rhizoma, Mori Cortex Radicis, Aurantii Nobilis Pericarpium were bore nearly same antioxidant effects compared to synthetic powerful antioxidant BHT and also expressed powerful antioxidant effect than ${\beta}-carotene$ such as Eucommiae Cortex, Cinnamomi Cortex, Angelicae Gigantis Radix, Lycii Furctus, Acanthopanacis Cortex, Sophorae Radix, Paeoniae Radix, Geranii Herba. Another method of DPPH was performed for searching natural antioxidant from natural product. Sophorae Radix, Puerariae Radix, Aurantii Nobilis Pericarpium, Atractylodis Rhizoma Alba, Acori Graminei Rhizoma, Corni Fructus, Angelicae Gigantis Radix, Paeoniae Radix were carried higher antioxidant capacity than ${\beta}-carotene$ by DPPH method.

계지탕(桂枝湯)의 기미(氣味) 표준화 연구(I) (In order to standarization of Kye-Ji Tang of ${\ulcorner}$Treatise on Febrile Diseases${\lrcorner}$)

  • This study was conducted to standardize the 'Ki-Mi' of Kye-Ji Tang in the basis of ${\ulcorner}$Treatise on Febrile Diseases${\lrcorner}$ and the results were as follows 1. The medicinal plants used are processed with hot water right after picking. 2. One seung(升) of water equals to 10cc. 3. One yang(兩) equals to 6 grams. 4. Medicinal plants are shattered to the size of beans by the process called Boo-Jeor. 5. Kye-Ji Tang, which is composed with 5 medicinal plant(Cortex Cinnamomi : 18g; Radix Paeoniae : 18g; Radix Glycyrrhizae : 12g; Rhizoma Zingiberis : 18g; Fructus Ziziphi Jujubae : 18g), is prepared by adding 490cc water to above 5 medicinal plants and boiled to 210cc. After boiling, the residue is discarded and the extract is taken orally 3 times a day as one dosage of 70cc. It is recommended that 20 minitues after taking of Kye-Ji Tang, the additional uptake of rice porridge might enhance the pharmacological effect of Kye-Ji Tang.

다류원료 식물류 물 추출물의 항산화 효과 (The Antioxidative Effects of the Water-Soluble Extracts of Plants Used as Tea Materials)

  • 본 연구는 다류원료 식물류의 물 추출물에 대해 항산화성을 검토하고 그 항산화 물질(페놀성 화합물, 비타민 C, 셀레늄)을 분석하였다. 대두유에 식물류의 물 추출물을 0.02%(추출고형분 함량, w/w) 첨가하여 $55^{\circ}C$ 공기순환식 인큐베이터에서 36일 저장기간 동안 과산화물가 측정과 $110^{\circ}C$에서 Rancimat 방법에 의해 항산화성을 검토하였으며 첨가제가 들어있지 않은 대두유를 대조구에 사용했고 0.02% BHT 첨가한 대두유는 비교구로 사용하였다. 대조구와 BHT 첨가구에 비해 산화 유도기간이 $55^{\circ}C$에서는 구기자, 우롱차, 귤껍질(진피), 유자, 매실 물 추출물 등이 길었으며 $110^{\circ}C$에서는 우롱차, 인스탄트커피, 유자, 구기자, 오미자, 매실 물 추출물 등이 길었다. 추출고형분 g당 총페놀성 물질 함량은 우롱차, 녹차, 홍차, 커피, 계피, 결명자 물 추출물 등에서 높았고 비타민 C 함량은 커피, 두충, 홍차, 영지, 계피, 감잎, 치커리, 녹차, 우롱차 물 추출물 등에서 높았으며 셀레늄은 모든 물 추출물에서 검출되지 않았다. 이들의 항산화성은 단일 항산화성 물질의 효과에 기인하기 보다는 페놀성 화합물, 비타민 C 등 복합적 효과에 의한 것으로 사료된다.

계피함유 제제의 Cinnamaldehyde 및 Cinnamic acid의 안정성 (Stability of Cinnamadehyde and Cinnamic acid of Cinnamomi Cortex Preparations)

  • Using HPLC, we examined quantitative change of cinnamaldehyde and cinnamic acid, caused by time-progress and temperature-change for the Cinnamoni Cortex preparations, Gye Ji Ryung Whan(桂枝茯?丸), Pal Mi Ji Whang Whan(八味地黃丸) and Do Haig Sung Ki Tang(桃核承氣湯). The content of cinnamaldehyde significantly decreased by the increase of temperature. It is suggested that cinnamaldehyde and cinnamic acid are considerably stable when kept frozen.

Helicobacter pylori에 대한 항균활성을 가진 천연물의 검색과 항균력 비교 연구 (Study on the antimicrobial activities of herbal extracts against Helicobacter pylori)

  • Infection with Helicobacter (H.) pylori is strongly associated with duodenal and gastric ulcers. Substantial epidemiological data has revealed that high rates of H. pylori infection might be related to high rates of gastric cancer. In this study, a medicinal herbal extracts were examined and screened for anti-H. pylori activity. Seventy percent ethanol was used for herbal extraction. For anti-H. pylori activity study, the inhibitory zone tests with 74 herbal compounds were conducted. As the results, thirteen compounds including Cinnamomi Cortex, Magnoliae Cortex and Meliae Fructus were revealed strong anti-H. pylori activities.

서울지역 유통한약재의 통계적 품질관리 - 회분, 산불용성회분, 건조감량, 유해중금속을 중심으로 - (Statistical Quality Control of Herbal Drugstuffs Distributed in Seoul Area - Centering around Ash, Acid-insoluble Ash, Loss on Drying and Hazardous Heavy Metals -)

  • To survey the status of quality control of major ingredients of 'Ssanghwatang (Oriental medicine used for fatigue)', we analyzed 1,024 samples (9 kinds) of medical herbs to determine amounts of ashes, acid-insoluble ashes, loss on drying that are major elements to ensure basic quality of herbal drugstuffs. After ash analysis, Paeoniae Radix (from China) failed to meet the herbal standard criterion. After yielding Z-score(indicating the probability of exceeding its criterion) Zizyphi Fructus, Zingiberis Rhizoma, Paeoniae Radix, Rehmanniae Radix Preparata became objects of intense quality control (=IQC). Analysis on loss on drying shows Cinnamomi Cortex was unsuitable and was put under the IQC. In case of respective heavy metals, Angelicae Gigantis Radix, Cnidii Rhizoma, Cinnamomi Cortex exceeded the maximum permissible range for Cd. The 3 kinds of sub-standards were put to the IQC. Statistic figures showed a significant correlation (t-test, p<0.01) between country-of-origin and sub-quality rates. Compared to domestic one, imported medicinal herbs were less suitable in regard of acid-insoluble ash, loss on drying and Pb/Hg content. Meanwhile, amounts of Acid-insoluble ash showed positive relation with amounts of Pb (r=0.202) and As (r=0.243) among heavy metals leading to an inference that herbs of which root/rhizome is used for medical purposes attribute its high heavy metal content to the fine earth/sand being stuck to its root stalk.
