Biological characteristics of Scuticociliate with a polymorphic life cycle of ciliate and cyst isolated from cultured olive flounder, Paralichthys olivaceus (양식넙치, Paralichthys olivaceus에서 분리된 Ciliate와 Cyst의 생활사를 갖는 스쿠티카섬모충의 생물학적 특성)

  • Scuticociliatosis has badly settled one of most damaging diseases during the seedling production process of olive flounder. Paralichthys olivaceus in Korea. We isolated a new type of Scuticociliate from flounder. The parasite metamorphoses to ciliate and cyst phases with each other by environmental changes and survive for a relatively long span. The ciliate was measured average 41.8 ${\mu}m$ in length and 21.0 ${\mu}m$ in width, and cyst was 17.0 ${\mu}m$ and 13.5 ${\mu}m$, respectively. Nutritional condition was determined as a major parameter of metamorphosing between ciliate and cyst stages. The ciliate transforms to a cyst stage because of food shortage, and the cyst returns to a ciliate stage with a favorite environmental condition and shows active growth and reproduction. The ciliate multiplied at the maximal density of $2.9 {\times} 10^5 {m\ell}^{-1}$cells in vitro cultivation at $15 ^{\circ}C$temperature using MS BHI medium and bacterial food sources. The ciliate could be proliferated at a 2.5 to $30 ^ \circ}C$ temperature range, pH 6 to 9, and 1 to 55 ppt salinity. Particularly, it survived over one week at $0 ^{\circ}C$temperature showing a high resistance against unfavorable environmental conditions. And the cyst survived for 320 days in the condition of $5 ^{\circ}C$with no feeding, but its survival period was markedly shortened in higher temperature conditions. The chemotherapeutants (formalin and hydrogen peroxide) were clarified as effective chemicals against the ciliate during in vitro trials, but the effect of therapeutants differed in proportion, depending upon the density and the bathing time of chemical compounds.

  • A new trichostome ciliate, Balantidium honghuensis n. sp., was isolated from the recta of Rana nigromaculata and R. limnocharis during parasite surveys in Honghu Lake, Hubei Province, central China in summmer of 2010 and 2011. Its detailed morphometric characters based on LM and SEM studies were described herein. The organism is oval in shape and thickly ciliated. The vestibulum is "V" shaped and occupies about 1/3 to 2/5 of the body length. The vestibular and nearby regions possess strong peripheral fibers which form a marked axial fiber about the cytopharynx. More than 10 contractile vacuoles are distributed along the periphery of the latter body. Comparisions were made between this new species and B. sinensis Nie, 1935. They were discriminated from each other in terms of general body forms, body size, and vestibulum shapes. Besides, special attention was paid to its high-speed daughter swarmers which we believed to be the infective stage of B. honghuensis. Possible infection routes of anura amphibian balantidia were discussed.

Occurrence of scutica-like ciliate (SLC) and its control during the mass seed production of the Japanese scallop, Patinopecten yessoensis (참가리비 (Patinopecten yessoensis) 대량 종묘생산시의 Scutica-like ciliate 발생 및 대책)

  • Few studies on parasitic scutica-like ciliates (SLCs) influencing larval growth and survival during the seed production of the Japanese scallop, Patinopecten yessoensis have been performed in spite of relatively high magnitude of their infectious damage to the larvae. In the mass seed production of the scallop, SLCs were never infectious during D-larval stage which lasted 6 to 7 days. Indeed, the infection first occurred in 2 to 4 days after first umbo-staged larvae. A regular selection of active larvae in swimming behavior kept larval SLC infection low throughout the culture compared to unselected control (P<0.05) in which all the larvae alive were contained. Higher infection frequencies of the unselected control resulted in lower larval survivals and wider range of larval size distribution that drove larval attachment rate lower. To the worse, the nursery spats from the higher SLC infection exhibited a retarded growth and elevated occurrence of abnormal scallop.

  • Concerned to the Ichthyophthiriasis of aquacultural fishes, the developmental features of Ichthyophthirius multifiliis were studied in cultured Korean catfish, Silurus asotus, and rainbow trout, Oncorhynchus mykiss. In the morphological observation, the parasite developed on two kinds of parasitic and non-parasitic phases with 6 developmental stages termed phoront, trophont, protomont, tomont, tomite, and therone. The $60{\times}40-100{\times}70{\mu}m$ fusiform or spherical phoront for the invading stage has 34-38 meridional kineties and begins to develope buccal apparatus. The 80-$800{\mu}m$ spherical or amoeboid trophont for the vegetative stage has a horseshoe shape macronucleus, a inconspicuous cytostome and developmental contractile vacuoles. The 200-$800{\mu}m$ spherical protomont for the encysting stage has a inconspicuous macronucleus, abundant contractile vacuoles; and a fine gelatinous exocyst is exuded, the baccal apparatus begins to resorb. The tomont for the encysted dividing stage has a thick cyst wall, and the buccal apparatus is resorbed completely. A small 35-$50{\mu}m$ spherical tomite for each daughter cell has a cytostome end the conspicuous oral apparatus. The $25{\times}20-60{\times}40{\mu}m$ fusiform theront for the infective stage possesses a perforatorium in the anterior end, a cytostome in the mid-point respectively and has 34-38 meridional kineties. In the experiments of the reproductive, the excysted time is related to water temperature. Tomitogenesis takes 10-14 hours at $28^{\circ}C$, 12-15 hours at $26^{\circ}C$, 16-18 hours at $22^{\circ}C$, 24-28 hour at $18^{\circ}C$, 26-51 hours at $14^{\circ}C$, and 129 hours at $9^{\circ}C$ respectively.
