The chromosome of Fusarium species during the vegetatve nuclear divisions in hyphae were observed by use of HCl-Giemsa technique on light microscope. The haploid chromosome number of Fusarium anthophilum 7472 was n=7, n=6 in F. anthophilum 7481 and n=6 in F. oxysporum 7500. The haploid chromosome number was 7 in F. napiforme 6129 and F. napiforme 6144. Those of F. caucasicum F. caucasicum ATCC 18791 and F. aquaeductuum ATCC 15612 were n=5. F. coeruleum ATCC 20088 was n=6, n=8 in F. camptoceras ATCC 16065 and n=7 in F. sambucinum NRRL 13451. From these results and previous papers, it may be concluded that the basic haploid chromosome number of the genus Fusarium is n=4.
Suspension cell lines have been newly established from the calli derived from the immuature embryo culture of hexapolid (Triticum aestivum var. sicco), tetrapolid (T. durum) and diploid (T. tauchii or Aegilops squarrosa) wheat species. The chromosomal variation in suspension cultured cell lines was examined and old cell line, C82d, established from T. aestivum var. copain was also used. New method using 1-bromonaphthalene for metaphase rapping of suspension cells was developed. Variation in chromosome number was observed among all the suspension lines. Cells with doubled chromosome number and deleted chromosome were also observed. Extensive structural changes in chromosome were found in C82d line. Chromosome aberrations showed loss of chromosome arms and chromosome segment. The mean chromosome number in suspension cells of T. aestivum var. sicco was 40, in C82d line 33, in T. durum 28 and in T. tauchii 14. The stability of chromosome in suspension cells of diploid and tetrapolid wheats was higher than that of hexaploid wheat.
A study on chromosome of leucocytes in blood cultures derived from 6 normal Korean was performed . Exact chromosome counts were carried out on 205 cells in male, 211 in female , of which 86.05% revealed a chormosome mordal number of 46. On the basis of relative chromosome lengths and position of centromeres, the Karyotype that the human chromosomes were classified into 7 groups with 22 airs of autosome and one pair of sex chromosome was determined accoridng to the method of denver report. The chromosome number on metaphase was observed in short term cultures of leucocytes from the peripheral blood of 2 patients with chronic granulocytic leukemia and 1 patient with acute granulocitic leukemia . and the chromosome morpholoogy was also investigated in one acute leukemic patient. In all leukemic cases the leucocytes showed the constant value of 46 in the stem -line of chormosome number. But the frequency of cells with 46 chromosomes appeared in the 3 cases was 67.30% in average with a slightly higher range in hypo-andhyper-diploid chromosome numbers than in normal human, The idiogram analysis did not show any abnormality of chromosome in acute leukemic cells.
This study is concerned with alterations in chromosomes (numbers and morphology) when the culture of Chinese hamster cells (FAF-28 strain) was treated by steroids, testosterone and DOC. 1. In 200 cells of normal untreated cells as control population the chromosome of stemline was decided as which was contained in 158 cells ; that is , in 79 percent of the population. The average chromosome number in above 20 cells observed was calculated as 23.95 with minimum limit at 20 and maximum limit at 70. 2. Many different chromosome numbers, ranging from 19 to 352 were observed in the 200 cells treated by testosterone. The diploid number of 22 showed the peak of variation curve was counted in 71 cells (35.5%) and an average chromosome number of stemline was 22 which was counted in 74 cells (37%). The twenty cells with 44 chromosomes were selected at random from each cell population treated with testosterone and DOC , so that chromosome idiogram and morphology could be studies. In the twenty cells of the testosterone treated population the average ratio of above four groups, LM ; MM;Ss:SM, was found to be 8.6 : 10.8:13.5:10.7. On the other hand, the average ratio in the same number of cells of the DOC treated one was 7.7 :11.4:12.5:12.7. 6. The five types of the altered chromosomes morphologically in the hundred cells selected at random from each cell population treated by testosterone and DOC were observed (Type I-V). The thirty-one altered chromosomes were found to be in the testosterone treated cell population and the sixteen in DOC treated.
The chromosome number of myoga ginger (Zingiber mioga (Thunb.) Roscoe: Zingiberaceae) has been reported as 2n = 22 for Chinese plants and 2n = 55 for Japanese plants. We checked the chromosome number of Z. mioga in plants collected in Jeollabuk-do and Jeollanam-do, Korea, and counted 2n = 44, the first report of this number for the species. As the basic chromosome number of Z. mioga is thought to be x = 11, Z. mioga plants in China, Korea, and Japan appear to be diploids, tetraploids, and pentaploids, respectively. In finding the tetraploid race of Z. mioga in Korea, we can hypothesize that the pentaploid race in Japan is derived through the fertilization of reduced gametes of the diploid race and unreduced gametes of the tetraploid race.
We report the meiotic chromosome numbers of four Carex taxa from Korean populations. Three are the first reports made on taxa from Korean populations: Carex appendiculata (Trautv. & C. A. Mey.) $K{\ddot{u}}k$. ($n=27_{II}$), C. fernaldiana H. $L{\acute{e}}v$. & Vaniot ($n=33_{II}$), and C. metallica H.$L{\acute{e}}v$. ($n=15_{II}$). Reports on the other species expand the range of variation in the chromosome number within a taxon, C. miyabei Franch. (n = $43_{II}$, $44_{II}$, $45_{II}$). Carex L. (Cyperaceae) consists of more than 2,000 species worldwide and is the most species-rich genus in Korea. The species diversity in the genus has been hypothesized to be associated with the chromosome variation, but chromosome information pertaining to Korean Carex taxa is not well known. This report updates the chromosome number inventory on Korean Carex to 24 out of 180 taxa.
The chromosome number was identified and fluorescence in situ hybridization(FISH) mapping of 5S and 45S rDNAs were conducted for C. minima and C. lanceolata in the genus Codonopsis from Jeju island. In this study, we have confirmed that the somatic metaphase chromosome number determined as 2n=2x=16 was the same as the findings from the previous studies. While the conventional staining method makes it rather difficult to distinguish satellite chromosomes due to high degree of variability, FISH analysis produced the exact number and location of 5S and 45S rDNAs. Both species in the genus Codonopsis have a pair of 5S rDNA and their gene loci were observed on chromosome 3. Although two pairs of 45S rDNAs (one on chromosome 1 and the other on chromosome 8) were identified in both species, the 45S rDNA signals on chromosome 8 in C. minima were significantly weaker than those on chromosome 1. In addition, the 45S rDNA signals on chromosome 1 in C. lanceolata showed that the chromosome is non-homologus. In this study, we have determined cytogenetic characteristics of C. minima and C. lanceolata according to their gene replication patterns.

This study was carried out to obtain the basic information on morphological and chromosomal charateristics in the three species of Chinese carps (grass carp; Ctenopharyngodon idellua, bighead carp; Aristichthys nobilis, and silver carp; Hypophthalmichthys molitrix) introduced to Korea from China. C. idellua was differ from A. nobilis and H. molitrix by the number of gill rakers, scales, fin rays, body proportion. A. nobilis and H. molitrix were similar in having ventral keel and many scale number, but H. molitrix was differ from A. nobilis by the connected gill rakers and body color pattern. Diploid chromosome and arm number (fundamental number, NF) of the three species were all the same to 2n=48 and NF=84. Diploid chromosome numbers in the three species are consisted of 10 pairs of metacentric chromosome, 8 pairs of submetacentric chromosome and 6 pairs of acro and/ or telocentric chromosome. Morphological and karyological relationship of the three Chinese carps are discussed.
A copy number amplification system for yeast artificial chromosomes (YACs) was combined with simultaneous overexpression of genes integrated into a YAC. The chromosome VII (1,105 kb) was successfully split to 887 kb, 44 kb containing the element for copy number amplification, and a 184-kb split-YAC. The 44-kb split-mini YAC was amplified a maximum of 9-fold, and the activity of the reporter enzymes integrated into the split-mini YAC increased about 5-7-fold. These results demonstrate that the mini-YAC containing a targeted chromosome region can be readily amplified, and the specific genes in the mini-YAC could be overexpressed by increasing the copy number.
The somatic chromosome number of four Korean species of Leontopodium were investigated. The chromosome number of L. leiolepis (2n = 24) is reported here as for the first time. The chromosome number of L. japonicum (2n = 28) is not varied among the 3 populations on the Korean Peninsula, but that condition is different from the previous reports for Korea (2n = 26) and Japan (2n = 21, 26). L. hallaisanense and L. japonicum, both of which are in sect. Nobilia and similar to each other in gross morphology, have the same chromosome number of 2n = 28. On the other hand the chromosome number of Korean L. leontopodioides (2n = 24) is different from that in Russian reports (2n = 26). The chromosome numbers of all Korean species of the genus Leontopodium could be inferred as tetraploid or aneuploid.
