Effects of Antihypertensive Diets Mainly Consisting of Buckwheat, Potato, and Perilla Seed on Blood Pressures and Plasma Lipids in Normotensive and Spontaneously Hypertensive Rats (메밀, 감자, 들깨를 이용한 항고혈압 기능성 식이가 정상혈압쥐 및 본태성고혈압쥐에서 혈압 및 혈장지질에 미치는 영향)

  • The study was carried out to investigate a new type of functional foods with hypotensive effect which is critical in the prevention and treatment of hypertension and related circulatory diseases. The experimental diets(A, B, C) were prepared from plant based ingredients such as buckwheat, potato, perilla seed with different ratios formulated as an edible form appropriate for human consumption according to AIN-77 standard. Control group(D) was fed commercial rat chow. Twety-four 15-week-old SDR(Sprague Dawley rats) and twenty-four 20-week-old SHR(spontaneously hypertensive rats), weighing 200g respectively, were assigned to 4 treatments of 6 rats each in a completely randomized design. Blood pressure was measured at 7 day interval by tail-cuff sphygmomanometer using an IITC cuff pump and amplifier. The growth rates of both SHR and SDR were not statistically different in comparions with the control except those of diet B and C in SDR and SHR, respectively(p<0.05). When exprimental diets were fed, systolic blood pressure of SDR and SHR at day 28 had been lowered by 17-20 mmHg(ave. of 19.6mmHg for both strains) compared with the reference pressure at day 0. The levels of HDL-cholesterol were increased, while the levels of LDL-cholesterol consistently decreased in both strains when experimental diets were fed(p<.05). Plasma total cholesterol levels were not different among treatments. Plasma triglyceride levels were higher in control diet(commercial rat chow) due to two times higher fat content of control diet itself(p<.05). Atherogenic indices were lower compared to those of control when the experimental diets were fed in both strains (p,.05). The results suggest that the antihypertensive diets mainly consisting of plant-based ingredients may possibly effective in relieving hypertension as well as circulatory diseases.

ROS-, RNS-Scavenging and Anti-inflammatory Activities of Mori Fructus (상심자 추출물의 ROS, RNS 및 염증 촉진 인자 제어 효과)

  • Objectives : Peroxynitrite $(ONOO^-)$, superoxide anion radical $({\cdot}O_2^-)$ and nitric oxide (NO) are cytotoxic because they can oxidize several cellular components such as proteins, lipids and DNA. They have been implicated in the aging processes, and age-related diseases such as Alzheimer's disease, rheumatoid arthritis, cancer and atherosclerosis. The aim of this study was to investigate the $ONOO^-$, NO, and $({\cdot}O_2^-)$ scavenging and anti-inflammatory activities of Mori Fructus in ob/ob mice. Methods : Mice were grouped and treated for 5 weeks as follows. Both the normal lean (C57/BL6J black mice) and control obese (ob/ob mice) groups received the standard chow. The experimental groups were fed a diet of chow supplemented with 7.5, 15 and 30 mg Mori Fructus per 1 kg of body weight for 14 days. For this study, the fluorescent probes, namely 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4,5-diaminofluorescein (DAF-2) and dihydrorhodamine 123 (DHR 123) were used. Western blotting was performed using anti-phospho $I{\kappa}B-\alpha$, anti-IKK-$\alpha$, anti-NF-${\kappa}B$ (p50, p65), anti-COX-2 and anti-iNOS respectively. Results : Mori Fructus inhibited the generation of $ONOO^-$, NO and $({\cdot}O_2^-)$ in the lipopolysaccharide (LPS)-treated mouse kidney postmitochondria in vitro. The generation of $ONOO^-$, NO and $({\cdot}O2^-)$ were inhibited in the Mori Fructus-administered ob/ob mice groups. The GSH/GSSG ratio decreased in the ob/ob mice, whereas they improved in the Mori Fructus-administered groups. Mori Fructus inhibited the expression of phospho $I{\kappa}B-\alpha$, IKK-$\alpha$, COX-2, iNOS genes, and thereby the activation of NF-$I{\kappa}B$. Conclusions : These results suggest that Mori Fructus is an effective $ONOO^-$, $({\cdot}O_2^-)$ and NO scavenger, and therefore it might be a potential therapeutic drug against the inflammation process and inflammation-related diseases.

Effects of Ojung-hwan on Blood Glucose, Hyperlipidemia, Polyol Pathway and Antioxidative Mechanism in Ob/ob Mouse (Ob/ob mouse에서 오정환(五精丸)이 혈당, 고지혈증, Polyol Pathway 및 항산화작용에 미치는 영향)

  • Objectives : Diabetes is a disease in which the body does not produce or properly use insulin. Etiological studies of diabetes and its complications have shown that oxidative stress might play a major role. Therefore, many methods have been tried to regulate free oxygen radicals for treating diabetes and its complications. Ojung-hwan, composed of five crude herbs, has been considered effective for treating symptoms of aging. In male ob/ob mouse of severe obesity, hyperinsulinemia and hyperlipidemia, which are features of NIDDM, the hyperglycemic activities and mechanisms of Ojung-hwan were examined. Methods : Mice were grouped and treated for 5 weeks as follows. Both the lean (C57/BL6J black mice) and diabetic (ob/ob mice) control groups received standard chow. The experimental groups were fed a diet of chow supplemented with 30 and 90 mg Ojung-hwan per 1 kg of body weight for 14 days. The effects of Ojung-hwan extract on the ob/ob mice were observed by measuring the serum levels of glucose, insulin, lipid components, and the kidney levels of superoxide anion radical (${\cdot}\;O{_2}{^-}$), MDA+HAE, GSH/GSSG ratio, and also the enzyme activities involved in polyol pathway. Results : Ojung-hwan lowered the levels of serum glucose and insulin in a dose-dependent manner. Total cholesterol, triglyceride and free fatty acid levels decreased, while the HDL-cholesterol level increased, in Ojung-hwan treated groups. Renal aldose reductase and sorbitol dehydrogenase activities increased in the ob/ob mice, whereas they were inhibited in the Ojung-hwan treated groups. Ojung-hwan inhibited the generation of ${\cdot}\;O{_2}{^-}$ in the kidney. Finally, MDA+HAE levels increased and GSH/GSSG ratio decreased in the ob/ob mice, whereas they improved in the Ojung-hwan treated groups. Conclusions : Ojung-hwan showed antidiabetic and antihyperlipidemic activities by regulating theactivities of polyol pathway enzymes, scavenging reactive oxygen species and reducing the MDA+HAE levels in the ob/ob mice.

Insulin Resistance of Skeletal Muscle was Recovered by Leptin Injection in vivo, but not in vitro, in High-fat Diet Fed Rats

  • We examined the effect of leptin on the insulin resistance in skeletal muscles by measuring the glucose transport. Male Wistar rats were fed with chow or high-fat diets for 30 days. Three days before sacrifice, high-fat fed rats were subcutaneously injected with leptin (1 mg/kg body weight) for 3 days. The glucose transports in the epitrochlearis and soleus muscle were not different among the experimental groups under basal state, however these were decreased significantly in the high fat-diet rats under insulin-stimulation (p<0.01). Leptin treatment recovered the decreased glucose transport in the epitrochlearis (p<0.05) and soleus (p=0.08). Triglyceride concentration in the soleus muscle was increased significantly in the high fat-fed rats, compared to chow diet rats (p<0.01), and it was decreased significantly by leptin treatment (p<0.01). The glucose transport was measured under basal and $60{\mu}u/ml$ of insulin with or without 50 ng/ml of leptin. Leptin had no direct stimulatory effect on glucose transport under both basal and insulin-stimulated conditions in vitro. These results demonstrate that leptin injection to high fat diet fed rats recovered impaired insulin responsiveness of the skeletal muscles and muscle triglyceride concentration. However, there was no direct stimulatory effect of leptin on insulin sensitivity of the skeletal muscle in vitro.

Effect of Supplement of Korean Pinenut Oil on Plasma Total Fatty Acid Composition in Cholesterol-fed Rabbits (한국산 잣기름이 콜레스테롤 첨가식이로 사육한 토끼의 혈장 총지방산 조정에 미치는 영향)

  • The present study was carried out in atheroscleorotic New Zealend white(NZW) rabbits. to evaluate the effect of dietary supplementation with Korean pinenut oil, on plasma total fatty acid composition. In study I, NZW rabbits were fed 10 weeks on a commercial chow diet supplemented with 5% of energy as fats(soybean oil or pinenut oil) or 10% of energy as fats(soybean oil or pinenut oil) with the addition of 1% cholesterol to the diet. Nineteen fatty acids ranged from myristic acid (14:0) to cervonic acid (22:6 ${\omega}3$) were identified in all the samples. The c5, c9, $c12{\sim}18$ : 3 acid was not reported in the fatty acid methyl ester profiles of each group because it was included in the linoleic acid peak. The major constitutent fatty acids in the chow diet group were linoleic acid, oleic acid, palmitic acid and ${\alpha}$-linolenic acid. In the cholesterol group, oleic acid, linoleic acid and palmitic acid were the major fatty acids. In plasma of cholesterol-fed animals, the levels of 16:1 ${\omega}$ 7 and 18:1 1 ${\omega}$ 9 were increased. Linoleic acid was the major fatty acid in soybean oil/cholesterol and pinenut oil/cholesterol groups. Plasma linoleic acid levels were significantly incresed from 4 to 6% by the supplementation of 5% soybean or 5% pinenut oil in the cholesterol diet for 5 weeks, compared to cholesterol group. Plasma 16 : 1 ${\omega}$ 7 levels in animals fed with 5 or 10% pinenut oils were significantly lower than in those fed cholesterol for 5 weeks. After 10 weeks on the soybean oil and pinenut oil diet there were no significant differences in the fatty acid composition. In study II, the fatty acid composition was not affected by the types or levels of oils supplemented for 5 weeks. After 10 weeks on the oil diets 16:1 ${\omega}$ 7 and 18:1 ${\omega}$ 9 were decreased in 10% soybean in oil/cholesterol and 10% pinenut oil/cholesterol groups, compared to cholesterol group.

A 3D-printing Bone Model for Surgical Planning of Total Hip Replacement after Failed Triple Pelvic Osteotomy

  • A 3-year-old, 26 kg, castrated male Chow Chow was presented for assessment of weight-bearing lameness of the left hind limb. The patient had a history of triple pelvic osteotomy on the left side to correct hip dysplasia 2 years prior to his presentation of clinical signs and underwent total hip replacement on the right coxofemoral joint 1 year later. Upon physical examination, pain and crepitus were noted on the left hip joint during extension. Radiological examination revealed coxofemoral joint subluxation and moderate degenerative bone changes on the left hip joint and pelvic axis, which relates to acetabular angles that were changed after triple pelvic osteotomy (TPO). Preoperative computed tomography was used for 3-dimensional printing to establish an accurate surgical plan. The changed angles of the acetabulum after TPO were evaluated, and rehearsal surgery was performed using a 3-demensional printing bone model. Three months after the THR surgery, the function of the affected limb had improved, with no lameness. Complications, such as luxation and implant failure, were not observed until 6 months after the operation. Accurate evaluation of acetabulum angles and rehearsal surgery using a 3D-printed bone model is effective for total hip replacement after unsuccessful TPO.

A Sensitivity Analysis of Model Parameters involved in Clark Method on the Magnitude of Design Flood for urban Watersheds (CLARK 유역추적법에 의한 계획홍수량 산정에 미치는 매개변수의 민감도 분석)

  • A Sensitivity analysis on the model parameters involved in the Clark watershed routing method is made to demonstrate the effect of each parameter on the magnitude of 50-year design flood for small urban streams. As for the rainfall parameter the time distribution pattern of design storm was selected. For short duration storms Huff, Yen & Chow and Japanese Central type distributions were selected and the Mononobe distribution of 24-hour design storm was also selected and tested for Clark method application. The effect of SCS runoff curve number for effective rainfall and the methods of subbasin division for time-area curve were also tested. The routing parameter, i.e. the storage constant(K), was found to be the dominating parameter once design storm is selected. A multiple regression formula for K correlated with the drainage area and main channel slope of the basin is proposed for the use in urban stream practice for the determination of design flood by Clark method.

PMP Estimation and Its Application for the Design Flood Determination in River Basin (하천유역의 설계 홍수량 결정을 위한 P.M.P의 산정 및 적용)

  • This study aims at analysis and application of PMP(Probable Maximum Precipitation) for the determination of design of major hydraulic structures. PMP was estimated by hydro-meteorolgical method and envelope curve method. PMF(Probable Maximum Flood) was then estimated from this PMP by synthetic unit hydrograph method and chow method. From the comparison of three methods for PMP estimation of magnitude of PMP in order of statistical, hydro-meteorlogical, envelope curve method. Amon PMP results estimated by each method it is believed that the hydro-meteorological method gave the best proper value in comparison with historical maximum rainfall because of this method reflected upon all meteorological factor. From the comparison of PMP with probable rainfall and flood, it was shown that estimated value by statistical method and hydro-metelogical method were nearly equivalent to the value of return period 200 to 500 year. It was found that PMF estimated from would be more safe for the design of major hydraulic structures in the consinderation.

Effects of Two Traditional Chinese Cooking Oils, Canola and Pork, on pH and Cholic Acid Content of Faeces and Colon Tumorigenesis in Kunming Mice

  • Faecal pH and cholate are two important factors that can affect colon tumorigenesis, and can be modified by diet. In this study, the effects of two Chinese traditional cooking oils (pork oil and canola/rapeseed oil) on the pH and the cholic acid content in feces, in addition to colon tumorigenesis, were studied in mice. Kunming mice were randomized into various groups; negative control group (NCG), azoxymethane control group (ACG), pork oil group (POG), and canola oil Ggroup (COG). Mice in the ACG were fed a basic rodent chow; mice in POG and COG were given 10% cooking oil rodent chow with the respective oil type. All mice were given four weekly AOM (azoxymethane) i.p. injections (10mg/kg). The pH and cholic acid of the feces were examined every two weeks. Colon tumors, aberrant crypt foci and organ weights were examined 32 weeks following the final AOM injection. The results showed that canola oil significantly decreased faecal pH in female mice (P<0.05), but had no influence on feces pH in male mice (P>0.05). Pork oil significantly increased the feces pH in both male and female mice (P<0.05). No significant change was found in feces cholic acid content when mice were fed 10% pork oil or canola oil compared with the ACG. Although Kunming mice were not susceptible to AOM-induced tumorigenesis in terms of colon tumor incidence, pork oil significantly increased the ACF number in male mice. Canola oil showed no influence on ACF in either male or female mice. Our results indicate that cooking oil effects faecal pH, but does not affect the faecal cholic acid content and thus AOM-induced colon neoplastic ACF is modified by dietary fat.

Effects of Cheonghyul-san on Blood Glucose, Hyperlipidemia, Polyol Pathway and Reactive Oxygen Species in ob/ob Mice (청혈산(淸血散)이 ob/ob mouse의 혈당, 고지혈증, Polyol Pathway 및 Reactive Oxygen Species에 미치는 영향)

Finally, MDA+HAE level was increased and the GSH level was decreased in the ob/ob mice, whereas those were improved in the Cheonghyul-san-administered groups. The results suggested that Cheonghyul-san exerted the antidiabetic and antihyperlipidemic activities by regulating the activities of polyol pathway enzymes, scavenging ROS, regulating the MDA+HAE and GSH levels in the ob/ob mice.
