배추 엽록체의 광계II 활성에 미치는 구리이온의 영향 (Effect of Cupric Ion on the PSII Activity in Isolated Chinese Cabbage Chloroplasts)

  • Copper inhibited PSII-mediated O2 evolution (H2OlongrightarrowDCIP, H2OlongrightarrowSiMo) but not PSImediated O2 uptake(DCIP. Asc.longrightarrowMV) in isolated Chinese cabbage chloroplasts. Copper toxicity on PSII-mediated O2 evolution was higher at alkaline condition than at acidic condition and was inhanced by light illumination after copper treatment. The increased toxicity by light illumination was not recovered by subsequent dark treatment. The inhibitory effect of copper on H2OlongrightarrowDCIP reaction was higher than that on H2OlongrightarrowSiMo reaction. This result suggests that there may be another inhibitory site of copper on PSII other than water oxidizing side of PSII.

Gold Immunolocalization of Rubisco and Rubisco Activase in Pyrenoid of Chlamydomonas reinhardtii

  • The pyrenoid ultrastructure and distribution of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and Rubisco activase in the chloroplasts of Chlamydomonas reinhardtii was studied using the immunogold localization technology with electron microscopy. There were several tubular thylakoids invading in the pyrenoid matrix to form several spokewise channels. The connections between pyrenoid matrix and stroma of chloroplast were the partial of channels. The starch sheath surrounding the pyrenoid was separated into several parts by the connections in transection. Some thylakoids were packed together near the connections in one side of the pyrenoid. Those special structures might be used to transport substance between pyrenoid and stroma of chloroplasts. With the antibody raised against the large subunits of Rubsico from C. protothecoides, the result of the gold immunolocalization of Rubisco in Chlamydomonas reinhardtii showed most of the gold particles heavily labeled the pyrenoid matrix, as well as the starch sheath matrix, and very few in the stroma of chloroplasts. The gold particle density was 880.00 $\pm$ 164.32, 190.00 $\pm$ 152.39 and 9.60 $\pm$ 5.37 ${\mu}m^{-2}$ in pyrenoid matrix, starch sheath and stroma region of chloroplast respectively (background: 5.67 $\pm$ 1.53 ${\mu}m^{-2}$). 99.59% of the total Rubiscos was calculated to be concentrated in the pyrenoid matrix and starch sheath by spatial densities. The gold immunolocalization of Rubisco activase also showed that Rubisco activase was mainly concentrated in the periphery of the pyrenoid and the starch sheath (the density was as high as 229.69 $\pm$ 96.96 ${\mu}m^{-2}$). There were very few gold particles located in the stroma of chloroplasts. These results indicated that pyrenoid surface and starch sheath was the site for Rubisco activation and $CO_2$ fixation, which supported the suggestion that pyrenoids perform photosynthesis function.

  • 피자식물내 엽육조직 미세구조연구의 일환으로 형태해부학적으로 매우 다른 특성을 지닌 국화과 Dubautia속 식물 3종(Dubautia scabra var. leiophylla, D. knudsenii, D. scabra var. leiophylla, D. knudsenii)의 엽육조직이 연구되었다. 부모종 엽육조직의 형태해부학적 특성과 그들의 잡종 엽육조직세포의 분화 정도를 추정하기 위하여 palisade mesophyll내의 chloroplasts, microbodies, mitochondria 및 grana내의 thylakoid membrane system, phytoferritin-like structures, plastoglobuli와 starch grain 등 엽록체 내부의 초미세구조 형질들을 중점적으로 비교 분석하였다. 잡종의 엽육조직세포들은 형태해부학적 엽육형질 및 thylakoidal membrane system 등의 형질에서 부모종 엽육조직의 중간적 특성 (intermediacy)을 나타내어 전형적인 bilateral inheritage를 보여주었으며, thylakoid membrane number/granum, phytoferritin-like structures, starch grain의 분포 등에서는 어느 한 부모종 형질과 더 유사하게 관찰되었다. 미소체 및 미토콘드리아는 대체로 비슷한 양상으로 관찰되었고 phytoferritin-like structures는 국화과 식물 엽록체내에서 처음 보고되는 미세구조이다.

Chlorella ellipsoidea 엽록체의 인지질 생합성 및 지방산 조성에 미치는 항생제의 효과 (Effect of Antibiotics on Phospholipid Biosynthesis and Fatty Acid Composition in Chlorella ellipsoidea Chloroplasts)

  • Chlorella 엽록체에서 인지질 생합성과 그의 지방산 조성에 미치는 amphotericin B($150\;\mu\textrm{g}/ml$)와 cycloheximide($10\;\mu\textrm{g}/ml$)의 영향을 분석하였다. Total lipid와 인지질 함량(PC, PE, PI)은 항생제 처리구가 대조구에 비하여 낮았다. Whole cell system에서의 주요 지방산은 대조구에서는 palmitic acid(31.96%)와 linoleic acid(16.96%)로 나타났으나 amphotericin B 처리구에서는 palmitic acid(36.15%)와 linolenic acid(16.71%)로 분석되었다. 또한 cycloheximide 처리구에서는 palmitic acid(31.90%)와 stearic acid(15.32%)가 인지질 형성에 이용되었다. 분리한 엽록체의 대조구(33.75%, 18.90)와 amphotericin B 처리(36.75%, 9.46%)에서 분리한 엽록체에서 인지질 형성에 이용되는 주요 지방산은 palmitic acid와 linolenic aicd였으나 cycloheximide 처리구에서는 palmitic acid(28.01%)와 oleic acid(19.27%)가 인지질생합성에 도입된 것으로 분석되었다.

보리 엽록체의 전자전달과 광인산화 활성에 미치는 ${Zn}^{2+}$의 영향 (Effects of ${Zn}^{2+}$ on the Activities of Electron Transport and Photophosphorylation of Barley Chloroplasts)

  • ${Zn}^{2+}$이 엽록체의 광합성계에 미치는 영향을 구명코져 보리 엽록체를 사용하여 전자전달, 광인산화반응의 활성측정 및 전기영동에 의한 CP복합체의 안정성을 조사하였다. ${Zn}^{2+}$은 추출된 분리엽록체 조건에서 뿐아니라, 생육시 반응용액 조건에서도 전자전달, 특히, PS II의 활성을 저해하였으며, 이보다 광인산화활성을 더욱 더 크게 저해하였다. 전자전달능의 저해는 ${Mn}^{2+}$에 의하여 회복될 수 있었으며, 광인산화반응에서 ${Mg}^{2+}$${Zn}^{2+}$는 서로 기능상의 경쟁을 나타냈다. 한편 전기영동에서 CP I은 ${Zn}^{2+}$과 mercaptoethanol이 존재하면 쉽게 파괴됨을 볼 수 있었다.

Feeding specificity and photosynthetic activity of Korean sacoglossan mollusks

  • During feeding on algal cytoplasm, some sacoglossans are known to keep the chloroplasts photosynthetically active for days to months in their digestive cells. Korean sacoglossan mollusks containing functional chloroplasts were screened using an in vivo chlorophyll fluorescence measuring system (pulse amplitude modulation, PAM). We collected six sacoglossans feeding on siphonous and siphonocladous green algae (Elysia atroviridis, E. nigrocapitata, E. ornata, Ercolania boodleae, Placida dendritica, Stiliger sp.) and one feeding on ceramiaceaen algae (Stiliger berghi) and performed feeding experiments using 37 algal species. Three species of Elysia showed strong photosynthetic activity for months. However, P. dendritica maintained functional chloroplasts only for several hours after feeding. E. boodleae, S. berghi, and Stiliger sp. showed no photosynthetic activity in any circumstances. Among all species, E. nigrocapitata was capable to tolerate the longest period of starvation for over 4 months. Four 'solar powered' sacoglossans bonded avidly to their specific algal food. Each species attached to and consumed only one algal species when several algae were given together. While they occasionally consumed other algae after prolonged starvation, they always reverted to their specific algae when available.

Effects of Paraquat on Ascorbic Acid and Malondialdehyde Contents, and Superoxide Dismutase Activity in Spinach Chloroplasts under Light and Dark

  • The amounts of ascorbic acid in chloroplasts treated with light and light+paraquat (PQ) were reduced by 81 and 82% of initial level, respectively at 24 hr at incubation. And those treated with dark and dark+PQ were decreased by W and 55% of the original level, respectively. Malondialdehyde (MDA) contents at 24 hr of dark and dark+PQ treatment were increased by 6 and 31% of the initial level, respectively. When chloroplasts were treated with light and light+PQ, MDA contents after 24 hr were increased by 88 and 146% of the initial level, respectively. SOD activities treated with light and light+PQ were increased by 10 and 20% of the initial level, respectively for 3 hr and thereafter reduced by 46 and 49% of the original level, respectively at 24 hr. However, the SOD activities treated with dark and dark+PQ were decreased by 37 and 30% of the initial level, respectively. It is considered that PQ triggers the oxidation of ascorbic acid, the induction of lipid peroxidation and the inactivation of SOD under light so that PQ has inhibitors effect on the pathway of plant metabolism. VaSpoU1 (SpoU gene) may be involved in organelle rRNA/tRNA modification in Viscum album

  • The SpoU family of proteins catalyzes the methylation of transfer RNAs (tRNAs) and ribosomal RNAs (rRNAs). We characterized a putative tRNA/rRNA methyltransferase, VaSpoU1 of the SpoU family, from Viscum album (mistletoe). VaSpoU1 and other plant SpoU1s exhibit motifs of the SpoU methylase domain that are conserved with bacterial and yeast SpoU methyltransferases. VaSpoU1 transcripts were detected in the leaves and stems of V. album. VaSpoU1-GFP fusion proteins localized to both chloroplasts and mitochondria in Arabidopsis protoplasts. Sequence analysis similarly predicted that the plant SpoU1 proteins would localize to chloroplasts and mitochondria. Interestingly, mitochondrial localization of VaSpoU1 was inhibited by the deletion of a putative N-terminal presequence in Arabidopsis protoplasts. Therefore, VaSpoU1 may be involved in tRNA and/or rRNA methylation in both chloroplasts and mitochondria.

Viola속 식물의 원형질체 및 융합세포의 전자현미경 관찰 (Electron Microscopic Observations of Protoplast and Fusion Cell of Viola Species)

  • To obtain a basic information on the development of Genus Viola, ultrastructure and electrofusion process between the two protoplasts from wild Viola callus cells and pansy mesophyll cells were observed with a scanning electron microscopy(SEM) and transmission electron microscopy(TEM). In the ultrastructural observation of wild viola callus protoplasts and pansy mesophyll protoplasts using SEM, their cell walls were removed completely. A knob-like formation was observed on the enlarge surface of viola callus protoplasts. On the surface of pansy mesophyll protoplasts net-like chloroplasts were observed. In SEM observation of pansy mesophyll protoplasts, chloroplasts devoid of membrane were observed on the surface the protoplasts. Pearl chain was formed by applying AC field of 200 V/cm at 1.0 MHz for 43 sec. The lysis of plasma membranes and fusion process occurred by applying a 1,600 V/cm DC pulse twice for 1 sec. After 1-2 hours of a DC pulse application, it was observed that the two protoplasts were fused completely into one cell. In TEM observation of the fused cell, many small vacuoles were located in the fusion area of the two protoplasts. Indeed, two distinct regions were observed during fusing process; in one region, a nucleus was found, while in the other region, both nucleus and nucleous were found.
