To investigate the antioxidative effects of an etbylacetate fraction extracted from the flowers of Chrysanthemum indicum L. (Chrysanthemi Flos) on the antioxidative system and lipid profiles of rats with ethanol induced hepatotoxicity. Sprague-Dawley rats weighing $100\~150$ g were divided into 5 groups: normal group (NOR), Chrysanthemi Flos EtOAC fraction (200 mg/kg) treated group (S1), $35\%$ etbanol (10 mL/kg) treated group (S2), Chrysanthemi Flos EtOAC fraction (200 mg/kg) and ethanol concomitantly treated group (S3) and Chrysanthemi Flos EtOAC fraction (400 mg/kg) and ethanol concomitantly treated group (S4), respectively. The antioxidative activity of each fraction was decreased in order of EtOAC, n-hexane, n-BuOH, water and chloroform. The growth rates and feed efficiency ratios were decreased by ethanol treatment, but were gradually restored to similar levels as in the NOR group by administering Chrysanthemi Flos EtOAC fraction. The whole blood concentrations of total cholesterol and LDL-cholesterol, and the activities of ALT and AST that were elevated by ethanol were significantly decreased in the Chrysanthemi Flos EtOAC fraction treated groups. It was also observed that the activities of SOD, catalase, xanthine oxidase and GSH-Px elevated by ethanol in rat liver were markedly decreased in the Chrysanthemi Flos EtOAC fraction treated group as compared to S2. These results suggest that Chrysanthemi Flos EtOAC fraction has possible protective effects against ethanol induced hepatotoxicity in rat liver.

Methanol extract of dried persimmon leaves was fractionated to hexane, chloroform, ethyl acetate, butanol, and aqueous tractions. Hexane, butanol, and aqueous fractions had high yields of extracts. Hexane fraction among these fractions showed the highest inhibition rate on the mutagenicities of aflatoxin (AFB$_1$), dimethyl-amino-bi-phenyl (DMAB), N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), and 4-nitroquinoline-1-oxide (4-NQO) in Salmonella typhimurium TA100. Hexane fraction was further fractionated into eight fractions by silica gel column c-hromatography and thin layer chromatography (TLC). The fraction 5 on TLC exhibited the highest antimutagenic activity on AFB$_1$, DMAB, and MNNC. 1'-oxocannabinol, 3B-acetoxy-17-methyl-5a-18 (13-17) abeoardrost-13-one, 4-methoxy-2'6'-dinitro-3, 5-di-t-butylbiphenyl, 8, 9-dihydro-5, 6-dimethoxy-dibenz [c, h]isoquino [2, 1, 8-1 ma]carbazole-11, 16-dione were tentatively identified from this antimutagenic fraction by GC-MS.

Antimicrobial activity of dandelion(Taraxacum platycarpum D.) was investigated. Methanol extract of dried dandelion was fractionated to hexane, chloroform, ethylacetate, butanol and aqueous fraction. Ethylacetate fraction among these fractions showed the highest inhibitory effect on the microorganisms such as B. subtilis, L. monocytogenes, S. aureus, E. coli and V. parahaemolyticus at $500\mu\textrm{g}/disc$. Ethylacetate fraction was further fractionated into 13 fractions by silica gel column and thin layer chromatography(TLC). The results showed that ethylacetate fractions No. 4, 5 and 6 had high antimicrobial activity. These were mixed again, re separated and five fractions were obtained. Among them, No. 2 fraction had the highest inhibitory effect on the microorganisms, which was then separated into six fractions. In the 3rd fractionation, No. 3 fraction was identified as benzoic acid by HPLC, $^{1}H-NMR$ and GC MS.

This study was conducted to investigate the antimicrobial activity of Ginkgo biloba L. leaves, seed and outer seedcoat against bacteria. Antimicrobial effects of Ginkgo biloba L. leaves (GBL), seed (GBS) and outer seedcoat (GBO) were examined by paper disc method and optical density method to determine minimum inhibition concentration (MIC), and observed by scanning electron microscope (SEM) to figure out the morphological change on the surface when Ginkgo biloba leaves extract was treated. The extracts of GBL, GBS and GBO were extracted by solvents such as methanol, ethanol, water. The methanol extract of GBL and GBO showed the highest antimicrobial activity against Bacillus cereus, Bacillus subtilis, Klebsiella pneumoniae, Listeria monocytogenes, Salmonella Typhimurium, Staphylococcus aureus, Yersinia enterocolitica except Escherichia coli and thus was further fractionated. The MICs of the chloroform fraction of GBL methanol extract were $125{\mu}g$/mL against B. subtilis, and L. monocytogenes; GBO methanol extract were $62.5{\mu}g$/mL against B. cereus and $125{\mu}g$/mL against B. subtilis, and L. monocytogenes. The microorganisms were treated with chloroform extracts ($2000{\mu}g$/mL) of GBL and GBO methanol extracts. It was observed by scanning electron microscope (SEM). The cells were expanded and a part of cell wall was completely destructed by GBL and GBO. Thus Ginkgo biloba L. leaves and outer seedcoat could be further developed into a natural antimicrobial agent.
For natural antioxidant test, the Rhus javanica $Linn\'{e}$ was extracted by 75% ethyl alcohol and fractionated it by chloroform and ethyl acetate using separatory funnel and column chromatography. In the Rhus javanica L. extract, the effective material was disolved easily in ethyl acetate and chloroform. When 600 ppm of ethyl acetate fraction of the extract was added to palm oil and lard, AI(antioxidant index) of each oil was 1.60 and 3.90 respectively. The $CHCl_3$ and EtOAc fractions were more effective than whole extract of Rhus javanica L. The palm oil, lard and soybean oil containing different levels of the Rhus javanica L. extract were stored at $60^{\circ}C$ to compare their antioxidative activity. Peroxide value, acid value and TBA value of each oil were monitored. The Rhus javanica L. extract was very effective to retard oxidation of palm oil and lard. This result coincided with Ranimat test.
Objectives : The root and rhizome of "Gangwhal" have been used as a traditional medicine for the treatment of cold, fever, headache, swelling, arthritis, rhinitis, and cardiovascular diseases in Korea and china. In china, Gangwhal is well known as a useful oriental medicinal plant that treats cardiovascular diseases such as stroke, headache and hypertension, but little research exists about the effect of O. koreanum on cardiovascular disease. Therefore we investigated the vasorelaxant effects of O. koreanum and compared the vasorelaxant effects of four species of Gangwhal. Methods : The vasorelaxant effects of the ethanol extracts of Ostericum koreanum (NK and BK), Notopterygium incisium (NI), and N. forbesii (NF) on phenyleprine ($1{\mu}M$) or KCl (60mM) pre-contracted rat thoracic aorta rings were compared. The vasorelaxant effects of the water extract and ethanol extract of NK on phenyleprine ($1{\mu}M$) or KCl (60mM) pre-contracted rat thoracic aorta rings were compared. And the vasorelaxant effects of chloroform, ethylacetate and water fraction of ethanol extract of NK on phenyleprine ($1{\mu}M$) or KCl (60mM) pre-contracted rat thoracic aorta rings were compared. Results : Ethanol extracts of NK, BK, NI and NF relaxed rat thoracic aorta rings with a concentration-dependent manner, and NK showed the greatest vasorelaxant effect. And ethanol extract of NK was much more effective than water extract of NK. Finally, chloroform, ethylacetate and water fraction of ethanol extract of NK also relaxed rat thoracic aorta rings, and chloroform fraction showed the greatest vasorelaxant effect.
In order to search for the way to utilize rice hull as a renewable resource, the inhibition on ${\alpha}-glucosidase$ and the fractionation of rice hull extract was investigated. An ethanol extract of rice hull from Japonica-type rice seeds exhibited 30% inhibitory activity on rat intestinal brush border ${\alpha}-glucosidase$ (1.4 mU/mL) in vitro at the concentration of 0.8 mg/mL using 6 mM p-nitrophenyl ${\alpha}-D-glucopyranoside$ as a substrate $(IC_{50}\;162\;mg/mL)$. Among the fractions obtained by partitioning the ethanol extract successively with solvents, the ethyl acetate fraction at the concentration of 0.8 mg/mL was found to exhibit the most potent inhibitory activity i.e. 65% inhibition of ${\alpha}-glucosidase\;(IC_{50}\;0.14\;mg/mL)$. Silica gel column chromatography of the ethyl acetate fraction exhibited slightly higher (90%) inhibitory activity, and its subsequent fractionation by Sephadex LH-20 column chromatography did not improve inhibitory activity. Considering the inhibitory activity and yield, the ethyl acetate fraction obtained by the solvent-partitioning process would be a candidate for the hypoglycemic food if it has in vivo effectiveness.
In this study, we investigated the antibacterial, antioxidant, and whitening effects of Prunus persica Batsch var. davidiana Max. (Feral Peach) and Prunus mume (white and pink) flowers. The extracts of the three kinds of flowers showed antibacterial activity against Staphylococcus. aureus. The chloroform fractions of the white Prunus mume, Feral Peach, and pink Prunus mume flowers exhibited antibacterial activities of 84, 49, and 30%, respectively, against Staphylococcus. aureus at a concentration of 0.5 mg/mL. The flower extracts of the three species also exhibited antibacterial effects against Pseudomonas. aeruginosa. The chloroform fractions of the Feral Peach and pink Prunus meme flowers exhibited antibacterial activities of 36 and 30%, respectively, at a concentration of 0.5 mg/mL. These extracts did not exhibit any significant antibacterial activity against Staphylococcus. epidermidis and Escherichia. coli. The extracts of the three kinds of flowers did not significantly affect the survival of HaCaT cells. The distilled water fraction of the pink Prunus mume flower extract exhibited antioxidant effects at concentrations of both 20 and 40 ㎍/mL. The ethyl acetate fraction of the pink Prunus mume flower extract exhibited an antioxidant activity superior to glutathione at a concentration of 40 ㎍/mL. The flower extracts did not significantly affect the survival rate of B16F10 cells. The chloroform fraction of the Feral Peach flower exhibited a whitening effect of 18% at a concentration of 40 ㎍/mL. Based on these results, we conclude that the three kinds of flower extracts are raw materials exhibiting antibacterial, antioxidant, and whitening effects.
In order to elucidate the defence mechanism of Cyprinus carpio nudus to Clonorchis sinensis, clonorchicidal substance in the epidermal mucus of this fish was isolated by silica gel column and thin layer chromatography and analyzed for its chemical nature by UV, IR and NMR-spectroscopy. The epidermal mucus of the fish was extracted by ethyl ether and separated into 2 fractions by column chromatography using petroleum ether/chloroform(30/70,v/v) as first solvent and methanol as second solvent. The second fraction with greyish white colour showed clonorchicidal effect. The second was again fractionated into greyish white precipitate and clonorchicidal greenish yellow supernatant fraction, by adding petroleum ether and standing the mixture for 5 days at $5^{\circ}C$. The latter was divided into 7 fractions in column chromatography with acetone/ benzene (10/90, v/v) as carrier. The fraction on equivalent to the spot of Rf. 0.225 value among them disclosed the strongest clonorchicidal effect. By this purification procedure, clonorchicidal substance was purified 154-fold with 0.9% yield from the mucus of the fish, and 10mg of the final fraction killed tne metaceicariae in 22 min. Infra red, nuclear magnetic resonance and ultraviolet spectrometric analysis of the purified substance revealed that the substance is linoleic acid. According to the results of the present studies it seemed that this fish could not be proper intermediate host of Clonorchis sixensis, and that defence mechanism of this fish to the worm seems to be correlated with clonorchicidal substance in epidermal mucus.
Antimicrobial and antioxidative effects of 14 different herbal flower extracts on skin microorganisms and oxidation were tested in this research. Herbal flower extracts were prepared with 70% ethanol. Among the herbal flower extracts, roselle (Hibiscus sabdariffa L.) flower extract showed the highest antimicrobial activity against Staphylococcus epidermidis as determined by a paper disc method. The seventy % ethanol extract of roselle flower was fractionated by sequential hexane, chloroform, ethyl acetate, n-butanol, and water fractionation. The growth of S. epidermidis, Streptomyces collinus, Streptomyces coeruleoprunus, Salmonella enteritidis, Vibrio parahaemolyticus, and Malassezia pachydermatis was most efficiently inhibited by ethyl acetate fraction of roselle flower extract as determined by a paper disc method and growth inhibition curves. In addition, the ethyl acetate fraction, water fraction and butanol fraction showed free radical scavenging and DNA cleavage inhibition activities. These results demonstrate that roselle flowers hold antimicrobial and antioxidative activities against skin microorganisms and oxidants.
