A New Approach for Thermodynamic Study on the Binding of Human Serum Albumin with Cerium Chloride

  • Thermodynamics of the interaction between Cerium (III) chloride, $Ce^{3+}$, with Human Serum Albumin, HSA, was investigated at pH 7.0 and $27\;{^{\circ}C}$ in phosphate buffer by isothermal titration calorimetry. Our recently solvation model was used to reproduce the enthalpies of HSA interaction by $Ce^{3+}$. The solvation parameters recovered from our new model, attributed to the structural change of HSA and its biological activity. The interaction of HSA with $Ce^{3+}$ showed a set of two binding sites with negative cooperativity. $Ce^{3+}$ interacts with multiple sites on HSA affecting its biochemical and biophysical properties.

  • The degradation of reinforced concrete (RC) structures due to physical and chemical attacks has been a major issue in construction engineering. Deterioration of RC structures due to chloride attack followed by reinforcement corrosion is one of the serious problems. The objective of this study is to develop a form of mathematical model of chloride ingress into concrete. In order to overcome some limits of the previous approaches, a mathematical model of chloride ingress into concrete consisting of chloride solution intrusion through the capillary pore and chloride ion diffusion through the pore water was proposed. Moreover, the variability of diffusivity of chloride ion due to degree of hydration of concrete, relative humidity in pore, exposure condition, and variation of chloride binding was considered in the chloride ingress model.

CaAl2O4-CaAl4O7 혼입 포틀랜드 시멘트 결합재의 염소이온 고정 특성 (Chloride Binding Properties of Portland Cement Binder Incorporating CaAl2O4-CaAl4O7)

  • 본 연구는 철근콘크리트 구조물의 염해 내구성 향상의 일환으로써, 보통 포틀랜드 시멘트에 실험체별 다른 비율의 칼슘 알루미네이트 시멘트와 합성 CA2를 혼입하여 혼입 비율에 따른 염소이온 고정능력을 평가하였다. 침지 후 실험체의 물리·화학적 특성을 압축강도, 공극 구조, 수화물 분석으로 염소이온침투깊이를 EPMA를 통하여 고찰하였다. 클링커 조성에 CA가 34%미만일 경우 실험체의 조밀성이나 강도 발현 양상이 구조재료로서 사용 제한이 없을 것이라고 판단되었으며 CAC와 CA2를 5:5비율로 혼입하여 실험체에 치환한 실험체가 CAC 혹은 CA2만 치환한 실험체보다 염소이온 침투억제능력 과 AFm상 및 프리델 염 생성이 높은 것으로 확인할 수 있었다. 결과적으로, 시멘트 페이스트 대비 칼슘 알루미네이트 클링커 치환율이 증가함에 따라 일반적으로 염소이온 고정능력이 향상하였고, CA-CA2비율에 따라 염소이온 고정 능력 및 염소이온 침투억제 능력에 차이가 있음을 확인할 수 있었다. 실험 범위 내에서 CA-CA2비율이 39:60이며 시멘트 페이스트 대비 10% 치환한 실험체인 M 10 가 가장 우수한 것으로 나타났다.

The Binding of Human CLIC1 with SEDL and Its Characterization in vitro

  • Full-length chloride intracellular channel protein 1 (CLIC1) is a member of the family of proteins related to bovine chloride intracellular channel p64. Mutations in the SEDL gene cause spondyloepiphyseal dysplasia tarda (SEDT), a rare X-linked chondrodysplasia. The link between the intracellular chloride channels and SEDL is an important step toward understanding their functional interplay. In the present study, CLIC1 protein was subcloned into the pGEX-KG vector and overexpressed in XL-1 blue cells. We developed a large-scale expression system composed of glutathione S-transferase (GST) fused with a 240-amino-acid CLIC1 protein in Escherichia coli. The soluble CLIC1 protein was successfully purified to homogeneity, and its purity, identity, activity and conformation were determined using SDS-PAGE, MALDI-MS, biophotometer and circular dichroism spectroscopic studies. The binding of both CLIC1 and SEDL proteins in vitro was detected by BIAcore biosensor and fluorescence measurements.

The Effects of Sodium Chloride on the Physiological Characteristics of Listeria monocytogenes

  • Sodium chloride is used to improve various properties of processed meat products, e.g., taste, preservation, water binding capacity, texture, meat batter viscosity, safety, and flavor; however, many studies have shown that sodium chloride increases the resistance of many foodborne pathogens to heat and acid. Listeria monocytogenes has been isolated from various readyto- eat (RTE) meat and dairy products formulated with sodium chloride; therefore, the objective of this paper was to review the effects of sodium chloride on the physiological characteristics of L. monocytogenes. The exposure of L. monocytogenes to sodium chloride may increase biofilm formation on foods or food contact surfaces, virulence gene transcription, invasion of Caco-2 cells, and bacteriocin production, depending on L. monocytogenes strain and serotype as well as sodium chloride concentration. When L. monocytogenes cells were exposed to sodium chloride, their resistance to UV-C irradiation and freezing temperatures increased, but sodium chloride had no effect on their resistance to gamma irradiation. The morphological properties of L. monocytogenes, especially cell elongation and filament formation, also change in response to sodium chloride. These findings indicate that sodium chloride affects various physiological responses of L. monocytogenes and thus, the effect of sodium chloride on L. monocytogenes in RTE meat and dairy products needs to be considered with respect to food safety. Moreover, further studies of microbial risk assessment should be conducted to suggest an appropriate sodium chloride concentration in animal origin foods.

  • The main objective of this study is to determine the critical chloride ion concentrations in the pore solutions causing depassivation of steel reinforcement in concrete made with cements of different $C_3A$ contents. Cement pastes with water-ratio of 0.5 were prepared using four cements with $C_3A$ contents of 0.46, 5.97, 9.14, and 9.65 percent. The pastes were allowed to hydrate in sealed containers for 28days and then objected to pore solution expression. The expressed pore fluids were analyzed for chloride and hydroxyl ion concentrations. It was found that the free cholride concentration in the pore solution decreases significantly with an increase in the $C_3A$ content of the cement. With increasing level of chloride addition, although the alsolute amount of bound chloride increase, the ratio of bound to total chlorides decreases.

대기 환경조건을 고려한 콘크리트 교량 바닥판의 염소이온 침투 예측 모델 (Prediction Model of Chloride Penetration in Concrete Bridge Deck Considering Environmental Effects)

  • Recently, the deterioration of reinforced concrete structures, primarily due to corrosion of steel reinforcement, has become a major concern. Chloride-induced deterioration is the most important deterioration phenomenon in reinforced concrete structures in harsh environments. For the realistic prediction of chloride penetration into concrete, a mathematical model was developed in which the effects of diffusion, chloride binding and convection due to water movement can be taken into account. The aim of this research was to reach a better understanding on the physical mechanisms underlying the deterioration process of reinforced concrete associated with chloride-induced corrosion and to propose a reliable method for estimating these effects. Chloride concentrations coming from de-icing salts are significantly influenced by the exposure conditions such as salt usage, ambient temperature and repeated wet-dry cycles.

  • 콘크리트용 천연골재의 공급이 어려워지고 건설폐기물은 발생량이 지속적으로 증가하고 있는 추세이다. 따라서 건설폐기물을 골재로 만든 순환골재 사용의 필요성이 대두되고 있다. 이에 순환골재를 사용한 콘크리트의 특성에 대한 많은 연구가 진행되고 있으나 대부분의 연구들이 부정적인 결과를 나타내고 있다. 하지만 최근 연구들을 중심으로 긍정적인 연구결과들이 보고되고 있으며 그 중 순환굵은골재의 부착 모르타르의 염화물고정효과에 관한 연구도 보고되고 있다. 따라서 본 연구에서는 순환굵은골재에 비해 모르타르 함량이 많아 염화물고정에 유리하다고 예상되는 순환잔골재를 사용한 모르타르의 역학적 특성과 염화물확산을 평가한 결과 순환잔골재의 염화물고정효과를 확인했다.

Chloride diffusion in concrete associated with single, dual and multi cation types

  • Currently, most of the investigations on chloride diffusion were based on the experiments and simulations concerning single cation type chlorides. Chloride diffusion associated with dual or multi cation types was rarely studied. In this paper, several groups of diffusion experiments are conducted using chloride solutions containing single, dual and multi cation types. A multi-ionic model is also proposed to simulate the chloride diffusion behavior in the experimental tests. The MATLAB software is used to numerically solve the nonlinear PDEs in the multi-ionic model. The experimental and simulated results show that the chloride diffusion behavior associated with different cation types is significantly different. When the single cation type chlorides are adopted, it is found that the bound rates of chloride ions combined with divalent cations are greater than those combined with monovalent cations. When the dual/multi cation type chlorides are adopted, the chloride bound rates increase with the $Ca^{2+}/Mg^{2+}$ percentage in the source solutions. This evidence indicates that the divalent cations would markedly enhance the chloride binding capacity and reduce the chloride diffusivity. Moreover, on the basis of the analysis, it is also found that the complicated cation types in source solutions are beneficial to reducing the chloride diffusivity.

환경조건을 고려한 염소이온 침투해석 프로그램 개발 (Development of Chloride Penetration Analysis Program Considering Environmental Conditions)

  • 본 논문에서는 온도 및 습도 등 환경조건의 변화와 염소이온의 확산, 이송 및 구속 현상을 고려하는 콘크리트 내부로의 염소이온 침투해석 프로그램을 개발하였다. 외부 환경조건의 변화를 고려하기 위하여 온도와 습도분포 해석을 동시에 수행할 수 있도록 하고, 확산계수의 온도, 습도 및 재령에 대한 변동성을 반영하였다. 실측데이터와의 비교를 통해 개발된 프로그램이 실측결과와 잘 일치함을 검증하였으며, 건습의 반복 등 외부 환경조건의 변화에 따라 콘크리트 내부 수분량이 변화하는 경우 염소이온이 표면부에 축적되는 현상의 기본 메커니즘과 그에 따른 철근부식 발생의 촉진과정을 예제해석을 통해 규명하였다.
