Photoelectrochemical Behavior of Chlorophyll a Langmuir-Blodgett Films

  • The highly efficient photoelectric conversion of chlorophyll a (Chl a) monolayers and multilayers was investigated. Using the Langmuir-Blodgett (LB) technique, Chl a monolayers and multilayers were fabricated onto optically transparent electrode, such as ITO glass. The photocurrent could be observed according to the light illumination. The action spectrum of the Chl a LB films was well consistent with its absorption spectrum. The possible application of the proposed system as a constituent of the artificial color recognition device was suggested.

Prediction of Chlorophyll-a Changes due to Weir Constructions in the Nakdong River Using EFDC-WASP Modelling

  • To evaluate the effect of the 4 major rivers restoration project in the Nakdong River to water quality of the river, the Environmental Fluid Dynamics Code (EFDC) and Water Quality Analysis Simulation Program (WASP), are applied in series. Results showed overall decrease in biochemical oxygen demand ($BOD_5$) concentrations and increase in chlorophyll-a concentrations, while total nitrogen and total phosphorous concentrations did not show significant changes, relatively. Decrease in $BOD_5$ concentrations seems to be influenced by an increased hydraulic residence time, which may allow more time for the degradation of organic material. Changes in Chlorophyll-a (Chl-a) concentration, due to the project were more significant for the upper stream areas that show relatively low Chl-a concentration ranges (less than 20 g/L). After the introduction of the Geumho River in the middle part of the Nakdong River, rapid growth of phytoplankton was observed. However, in this middle part of the Nakdong River, the ratio of Chl-a concentration change are less significant, compared to the upper stream areas, due to the project. In the lower stream area, Chl-a concentration decreased after the project. This seems to be resulted from the decreased light availability, due to increased depth, while the nutrient concentrations have been high enough to support phytoplankton growth.

Winter Algal Bloom and Spatial Characteristics of Water Quality in the Lower Taewha River, Ulsan, Korea (태화강 하류에서 겨울철 조류 발생과 수질의 공간적 특성)

  • This study was carried out to assess the spatial and tidal effects on the water quality in the lower reaches of Taewha River, Ulsan, Korea and to understand the environmental factors affecting winter algal bloom in the river. From May, 2010 to January, 2011, water samples were collected at five locations (New Samho Bridge, Old Samho Bridge, Mungjung Stream, Taewha Bridge, and Mungchon Bridge) along the river at high and low tides of spring tide. We measured environmental parameters including salinity, dissolved oxygen (DO), biological oxygen demand (BOD), chemical oxygen demand (COD), chlorophyll a (Chl a) and various nutrient concentrations. Salinity increased towards the downstream direction. Average values of Chl a concentrations ranged $10-26mg/m^3$ at high tide and $11-53mg/m^3$ at low tide depending on sampling locations. It was noteworthy that there were strong increases in Chl a concentrations during the November 21 to December 22 sampling period especially at the Taewha Bridge. At the location, Chl a concentrations were measured as $138-296mg/m^3$ for the period; Rhodomonas lacustris of class Cryptophyceae was the dominant algal species. Chl a concentrations at the Taewha Bridge were positively correlated with such parameters as salinity, BOD, DO, COD, pH, and T-N, and negatively correlated with temperature and $NO_3{^-}$-N. On the other hand, at the Mungchon Bridge the highest concentration of Chl a was $55mg/m^3$ on August 25, and Chl a concentrations were positively correlated with $NH_3$-N, T-N, $PO_4{^{3-}}$-P, T-P, and heterotrophic plate counts. The results suggested that water quality in the lower Taewha River fluctuated a lot with the sampling locations and the patterns of algal blooms were different between Taewha Bridge and Mungchon Bridge sampling locations.

Suggestion for Trophic State Classification of Korean Lakes (우리나라 호소의 영양상태 분류에 관한 제언)

  • Most of the lakes in Korea are artificial, and their limnological characteristics are significantly different from those of natural lakes in other countries. In this study, the relationship between trophic state parameters was investigated, based on summer average data of the upper layer, in 81 lakes in Korea, 2013-2017. Compared with trends of foreign natural lakes, chlorophyll a (Chl.a) concentration was slightly lower at the same total phosphorus (TP) concentration, and transparency (Secchi depth, SD) was noticeably lower at the same Chl.a concentration. This is because of excessive allochthonous loading of non-algal material during the monsoon period, and the reduction in phosphorus availability to algal growth, by light limitation and short hydraulic residence time. Considering these characteristics, we suggested site-specific thresholds of trophic state classification for Chl.a, TP and SD, based on annual average data at the upper layer of lakes ($3-10{\mu}g\;L^{-1}$ of Chl.a measured by UNESCO method; $13-33{\mu}g\;L^{-1}$ of TP; 1.6-3.2 m of SD for mesotrophic state class, respectively). The threshold value of TP for each trophic state class, corresponded to the upper value of previously reported range, and that of SD was out of the range. We suggested applying only TP and Chl.a in assessment of trophic state of lakes in Korea, excluding SD.

Analysis of algal spatial distribution characteristics using hyperspectral images and machine learning in upstream reach of Baekje weir (초분광영상과 머신러닝을 이용한 백제보 상류구간 조류 공간분포 특성분석)

  • 부영양화된 호수나 유속이 느린 하천에서 발생하는 녹조의 과도한 발생은 하천 생태계 훼손, 동식물의 건강, 담수의 오염 등 환경 사회 경제적으로 큰 피해를 준다. 현재 수질 측정망은 정해진 지점에서 Chlorophyll-a(Chl-a), Phycocyanin(PC)을 대표농도로 산정하고 조류경보에 활용하고 있으나, 일주일에 한번씩 샘플링을 통해 Chl-a 및 PC를 측정하여 시공간적인 신뢰성의 문제가 제기될 수 있다. 본 연구에서는 기존 점단위 조류 모니터링의 한계점을 개선하기 위해 초분광영상 자료를 머신러닝 기법에 적용하여 Chl-a 및 PC 산정 알고리즘을 개발하였다. 이를 위해 Chl-a와 PC의 최대 흡수, 반사 파장대, 주요 물 흡수 파장대 자료를 조합하여 9개의 파장비를 구축하였으며, 기존 연구에서 활용한 머신러닝 기법인 Partial Least Square, Random Forest, Gradient Boosting, Support Vector Machine, K-Nearest Neighbor, Artificial Neural Network를 검토하여 최적 모델을 선정하였다. 학습된 머신러닝의 성능을 R2, NSE, RMSE 목적함수를 이용해 평가하였으며, 그 결과 ANN이 각각 PC 0.801, 0.755, 11.774 mg/m3, Chl-a 0.733, 0.622, 8.736 mg/m3로 가장 우수한 성능을 보였다. 최적화 된 ANN 모델을 백제보 상류 2016-2017년 항공 초분광영상에 적용하여 시공간에 따른 조류 분포변화를 평가하고자 한다.

Phytoplankton Community in Adjacent Waters of Ulchin Nuclear Power Plant

  • To understand the phytoplankton community in adjacent waters of Ulchin nuclear power plant (UNPP), abundance and the size fractionated $chl-\alpha$ concentrations were evaluated through seasonal interval sampling from April 2003 to February 2004. A total of 211 different phytoplankton species was observed and mean abundance of phytoplankton in each study period ranged from 244,286 to 1,221,779 cells $L^{-1}$. The contributions of microplankton $(>20\mu{m})$ to total phytoplankton abundance ranged from 42.5 to 83.6% (average 66.1%) and those of nanoplankton $(>20\mu{m})$ ranged from 16.4 to 57.5% (average 33.9%). Total chl-$\alpha$ concentrations of phytoplankton ranged from 0.52 to $2.26\mu{g}\;L^{-1}$. The contribution of chl-$\alpha$ concentrations of microplankton was higher than that of nano- and picoplankton through the study period with exception of July 2008. The results of abundances and $chl-\alpha$ concentrations suggest that microplankton has an important role in adjacent waters of UNPP. The diminution of abundances and $chl-\alpha$ concentrations of phytoplankton was observed after passage through the cooling water system, but it was gradually recovered by mixing with the ambient waters. Our results suggested that the influence of thermal discharges on phytoplankton should be restricted within narrow limits around outlet area of thermal effluents.

Effect of Temporary Loading of Nonylphenol on a Summer Planktonic Community in a Eutrophic Pond

  • Recent studies reveal one of the representative endocrine disrupters of nonylphenol affects on the composition of a planktonic community. Since nonylphenol is sometimes discharged into eutrophic waters, we monitored planktonic community composition of a eutrophic pond after receiving nonylphenol when cyanobacterium Microcystis aeruginosa mainly dominated. The experiment was carried out two times using small-scale microcosms in a laboratory. In both two experiments, ciliate abundances significantly decreased when nonylphenol was added. On the seventh day, the ciliate abundances in $10{\mu}g\;L^{-1}$ added treatments decreased by 36.9% in the first experiment and 33.6% in the second, when compared to the control. The response of other planktonic groups was less obvious to nonylphenol addition. In particular, in the first experiment, Chl. b/Chl. $\alpha$ and Chl. c/Chl. $\alpha$ significantly increased with the addition of nonylphenol, while total Chl. $\alpha$ concentration did not change. Indeed, bacillariophyceae and chlorophyceae abundances tended to increase with nonylphenol dosing. From these results, we tentatively hypothesized that nonylphenolloading positively affects on abundances of edible phytoplankton such as Scenedesmus spp. and diatoms by releasing from grazing pressure due to decrease in ciliate abundances. The present study emphasizes that the indirect effect of endocrine disrupters should be paid more attention when freshwater resources are polluted by them.

Water Quality Simulation and Sensitivity Analisys in Nakdong River Using WASP7.2 Model (WASP7.2를 이용한 낙동강의 수질모의 및 민감도분석)

  • 낙동강 하류지점인 물금은 2003년${\sim}$2005년의 대부분이 부영양화의 기준을 넘고 있다. 본 연구에서는 WASP 모형을 이용하여 낙동강 하류에서 부영양화의 주요원인이 되는 영양물질과 결과적으로 나타나는 Chl-a 농도변화를 모의하였다. Chl-a의 농도모의를 위한 주요 입력자료로서 측정된 동물성 플랑크톤의 개체수가 입력되었다. 선정된 수질조사 지점은 낙동강 본류(왜관, 고령, 적포, 남지, 하남, 낙동강 하구둑)와 지류인 금호강, 회천, 황강, 남강, 밀양강이다. 수질모의 구간은 낙동강 본류(왜관${\sim}$낙동강 하구둑)와 지류인 금호강, 회천, 황강, 남강, 밀양강을 포함한 총 72개의 구간이다. 모형의 모의시간과 출력시간은 각각 1일로 하였다. 2003년의 자료를 이용하여 추정된 수질 매개변수들의 적합성을 확인하기 위해 2004년${\sim}$2005년의 수질자료와 유량자료를 이용하여 검증하였고, 입력된 매개변수와 기상 입력자료(유량, 수온, 일사량, 일조율)의 값을 중심으로 10% 씩 상하로 변화시켰을 때의 민감도를 분석하였다. Chl-a는 민감도분석결과 Chl-a의 성장률과 동물성 플랑크톤의 섭식률에 의해 영향을 많이 받았다. 인은 Chl-a에 영향을 주었지만 질소는 거의 영향이 없었다. 기상자료의 민감도분석결과 유량, 수온, 일조율, 일사량의 순서로 민감하게 모의 되었다. WASP 모형에서 동물성 플랑크톤 개체수의 입력 유 무에 따른 수질모의결과는 동물성 플랑크톤을 입력했을 때가 더 정확히 모의되는 것으로 나타났다.

Inhibition Characteristics of Chlorsulfuron and Imazaquin on Acetolactate Synthase Activity of Corn Plants (Chlorsulfuron 및 Imazaquin에 의한 옥수수 Acetolactate Synthase 활성의 저해특성)

  • The inhibition characteristics of chlorsulfuron [CHL, 2-chloro-N-[{ (4-methoxy-6-methyl-1,3,5-triazin-2-yl)amino}carbonyl]benzenesulfonamide] and imazaquin [IMA, 2-{4,5-dihydro-4-methyl-4-(1-methy-lethyl)-5-oxo-1H-imidazol-2-yl}-3-quinolinecarboxylic acid] on acetolactate synthase(ALS) activity of corn plants were investigated. CHL and IMA rapidly inhibited ALS activity of corn plants in vitro. Their $I_{50}$ values for ALS activity were 100nM and $5{\mu}M$, respectively, indicating that CHL had 50 times more inhibitory effect on ALS activity than IMA. The first applied herbicide had a dominant inhibitory effect on ALS activity when the two herbicides were applied sequentially. Branched-chain amino acids, valine(Val), leucine(Leu), and isoleucine(Ile) showed a feedback inhibition on ALS activity ; Val or Leu had a more inhibitory effect on ALS activity than Ile. Branchedchain amino acids and CHL or IMA exhibited an additive effect on inhibiting ALS activity. This suggests that branched-chain amino acids inhibit ALS activity by a different mechanisms) from that of CHL or IMA. Apparent ALS activity, which was measured on the basis of the conversion of pyruvate to acetolactate, was decreased by the addition of 2-ketobutyrate into the ALS reaction mixture in a concentration-dependent manner. In addition, kinetic studies revealed that CHL acts as a noncompetitive inhibitor, while IMA acts as an uncompetitive inhibitor to ALS with respect to pyruvate.

Alternative Fungicide Spraying for the Control of Powdery Mildew Caused by Sphaerotheca fusca on Greenhouse Watermelon (Citrullus lanatus) (시설 수박 흰가루병 방제를 위한 살균제 교차방제 체계)

  • Alternative spraying was developed using fungicides and potassium phosphonate to control watermelon powdery mildew (WPM) caused by Sphaerotheca fusca under greenhouse condition. In 2010 trial, a total of 9 combinations of 6 fungicides which are legally registered in Korea and currently used on watermelon farms were tested for the control efficacy against WPM. Though the combination of chlorothalonil (Chl)-myclobutanil (My)-fenarimol (Fen)-hexaconazole (Hex) was the most effective (82.6%) in the first-year trial, it had the problem that My, Fen and Hex have the same mode of action. Therefore, in the second-year trial to improve the fungicide combination and to reduce fungicide application, fungicides and potassium phosphonate (P) combinations were tested for WPM control using 500 ppm aqueous potassium phosphonate solution which is both eco-friendly and showing different mode of action from the fungicides tested. The P-My-Fen-Hex applied at 10-day intervals provided relatively high control (75.6%), whereas Chl-My-Fen-P provided 31.8% control. To improve the control, fungicides and P were applied at 7-day intervals in the third-year trial. Though Chl-My-Fen-Hex provided the highest control (89.6%), P-My-Fen-Hex also provided relatively high control (83.5%) compared to 65.9 % control with Chl-My-Fen-P, which suggests that Chl-My-Fen-Hex could be replaced with P-My-Fen-Hex for WPM control.
