A comparison of photosynthesis measurements by O2 evolution, 14C assimilation, and variable chlorophyll fluorescence during light acclimatization of the diatom Coscinodiscus granii

  • Photosynthetic rates of the large centric diatom Coscinodiscus granii were measured by means of multicolor variable chlorophyll fluorescence imaging, single cell $^{14}C$ assimilation, and optical $O_2$ sensor measurements during light acclimatization of cultures grown at five different irradiances: 50, 150, 235, 332, and $450{\mu}mol$ photons $m^{-2}\;s^{-1}$. Photo-acclimatization was evident from changes of cellular chlorophyll a content, growth rates, and light response curves. Each of the applied methods evaluates different parts and reactions in the photosynthetic apparatus, which makes a direct quantitative comparison of rates difficult, although a different degree of correlation were found between all three methods. However, when used in combination, they provide information about the internal relationship of photosynthetic pathways as well as the variation in photosynthetic capacity between individual cells within a single algal culture.

CHILLING SENSITIVITY OF CUCUMBER PLANTS MONITORED IN TERMS OF CHLOROPHYLL FLUORESCENCE

  • For three cultivars of chilling-sensitive cucumber plants, chilling sensitivity was evaluated in terms of photosynthetic activity using Chl fluorescence techniques. Low-temperature treatment caused a decrease in photosynthetic activities of cucumber leaves, measured as CO$_2$ exchange, as well as the decrease in the stomatal conductance. FR of the three cultivars decreased after chilling for 24 h in light and the extent of decline of F$_R$ was the greatest in 'Chosaeng' cultivar. When these plants were recovered from light-chilling, 'Chosaeng' and 'Samchuk' cultivars did not fully restore the original value of F$_R$ after 24 h of recovery, in contrast to 'Ilmi' cultivar which showed a rather efficient recovery. The results of FR study showed that 'Chosaeng' was most susceptible, whereas Ilmi was most resistant, to chilling among the three cultivars of cucumber plants. When quenching coefficients for chlorophyll fluorescence was analyzed after chilling the cucumber plants for 24 h in light, 'Chosaeng' elicited more rapid declines in the coefficients for photochemical quenching (qQ), non-photochemical quenching (qNP) and energy-dependent quenching (qE) than 'Ilmi' and 'Samchuk'. The implications of these observations are discussed in relation to the growth habits of the respective cultivars in the field. The results showed that measurement of chlorophyll fluorescence was an effective means of screening chilling tolerance of cucumber plants. Furthermore, the study on the chlorophyll fluorescence induction and fluorescence quenching charactersitics showed that low temperature could accelerate inhibition of photosynthesis in chilling-sensitive plants, by limiting Calvin cycle activity and disrupting, in part, the energy dissipation mechanims of the photosystem II.

Expression of Antioxidant Isoenzyme Genes in Rice under Salt Stress and Effects of Jasmonic Acid and ${\gamma}$-Radiation

  • Analysis of chlorophyll (Chl) fluorescence implicated treatment of 40 mM NaCl decreased maximal photochemical efficiency of photosystem II (PSII) (Fv/Fm), actual quantum yield of PSII (${\Phi}_{PSII}$), and photochemical quenching (qP) in rice, but increased non-photochemical quenching (NPQ). Decreases in Fv/Fm, ${\Phi}_{PSII}$, and qP were significantly alleviated by $30\;{\mu}M$ jasmonic acid (JA), while NPQ increase was enhanced. Transcription levels of antioxidant isoenzyme genes were differentially modulated by NaCl treatment. Expression of cCuZn-SOD2 gene increased, while those of cAPXb, CATb, and CATc genes decreased. JA prevented salt-induced decrease of pCuZn-SOD gene expression, but caused greater decrease in mRNA levels of cAPXa and Chl_tAPX genes. Investigation of vacuolar $Na^+/H^+$ exchanger (NHX2) and 1-pyrroline-5-carboxylate synthetase (P5CS) gene expressions revealed transcription level of NHX2 gene was increased by JA, regardless of NaCl presence, while that of P5CS gene slightly increased only in co-presence of JA and NaCl. Unlike JA, ${\gamma}$-radiation rarely affected expressions of antioxidant isoenzyme, NHX2, and P5CS genes, except for increase in mRNA level of Chl_tAPX and decrease in that of pCuZn-SOD. These results demonstrate enhanced salt-tolerance in JA-treated rice seedlings may be partly due to high transcription levels of pCuZn-SOD, NHX2, and P5CS genes under salt stress.

Steady state and Lifetime Measurements of Primary Fluorescence from Phytoplanktons (식물플랑크톤 색소의 형광 특성과 lifetime 측정)

  • The steady state and decay characteristics of primary fluorescenece of phytoplanktons including Cyanophyceae and Cryptophyceae were investigated in vivo. At 580-640 nm region, fluorescence emission spectra were obtained from all algae examined. The observed fluorescence emission maxima were similiar$(\pm3\;nm)$ except Synechocorcus sp. (SYN). Considered $\lambda_{max}$ of emission spectra of phycobiliproteins and the excitation spectra with $\lambda_{max}=540-560nm$, it seems to be originated from biliproteins. Fluorescence lifetimes $(\tau)$ and decay curves were compared with standard solution of candidate organic compounds, b-phycoerythrin. The $\tau$ values obtained for phytoplankton with $\lambda_{max}=580nm$ were different depending upon the species of algae. The observed $\tau$ values were ranged from 1.39 ns to 1.95 ns. These are considerably shorter than $\tau(3.23\;us)$ for standard solution of b-phycoerythrin. The reduction of $\tau$ for phycoerythrin in vivo seems to be originated from effective energy transfer system between Chl. a and phycobiliprotein in intact cell. There are subtantial differences in fluorsecence spectra and lifetimes at the class level. At the species level, differences seems to be much smaller. The result of experiment suggests that measurement of fluorescence lifetimes may be helpful in the rapid characterization of algae. Direct application will likely be found in combination with the measurement of other luminescence parameters.

Decrease of Photochemical Efficiency Induced by Methyl Viologen in Rice(Oryza sativa L.) Leaves is Partly due to the Down-Regulation of PSII

  • In the rice leaves treated with methyl viologen (MV), the photochemical efficiency of PSII (or $F_{v/}$F $m_{m}$) was significantly decreased, and significant portion of the photoinactivation process was reversible during the dark-recovery. The dark-reactivation process was relatively slow, reaching its plateau after 2-2.5 h of dark incubation. The damaged portion of functional PSII was 13%, based on the value of I/ $F_{o}$- I/ $F_{m}$ after this dark-recovery period. The reversible photoinactivation process of PSII function in the MV-treated leaves consisted of a xanthophyll cycle-dependent development of NPQ and a xanthophyll cycle-independent process. The latter process was reversible in the presence of nigericin. As well as the increase in the values of Chl fluorescence parameters, the epoxidation process during the dark-recovery after the MV-induced photooxidation was very slow. These results suggest that the photooxidative effect of MV is partly protected by the down-regulation of PSII before inducing physical damages in core proteins of PSII.I.I.I.I.

Seasonal Investigation of Natural Organic Matters from Yeongsan River Basin by Fluorescence Spectroscopy (영산강 수계 자연유기물질의 계절별 형광특성 연구)

  • This study investigated the characteristics of natural organic matter (NOM) with general water characteristics (pH, DO, electrical conductivity, BOD, COD, TN, TP, Chl-$a$, DOC, $UV_{254}$, SUVA) and the 3D fluorescence excitation-emission matrix (FEEM) in the Yeongsan River basin. FEEM was used to classify protein-like and fulvic & humiclike substances with fluorescence intensity in the matrix of excitation and emission wavelength. The concentration of BOD, COD, TN, electrical conductivity and DOC in the region of Gwangju city (Gwangju sewage treatment plant: GJS, Gwangjucheon: GJC, Gwangju 2: GJ2) was relatively higher than the upper reaches and lower reaches of the Yeongsan River basin. SUVA in most sites was lower than 3 L $mg^{-1}\;m^{-1}$ as the hydrophilic substances, except Damyang (DY) in the upper reaches of Yeongsan river was higher than 3 L $mg^{-1}\;m^{-1}$ as the hydrophobic substances during winter and autumn. In the FEEM investigation the fulvic and humic substances were found in most sites, and in sites regarding Gwangju city (GJS, GJC, GJ2) during winter and GJC in summer, protein-like substances were found. The trend of fluorescence intensities from the upper reaches to the lower reaches in most sites corresponded to that regarding the concentration of water characteristics (BOD, COD, TN, DOC). That is why the region of Gwangju city (GJS, GJC, GJ2) was relatively higher. This results were an equivalent trend to those of fluorescence index (FI) in most sites, and the higher FIs in the sites of Gwangju city indicate more microbial-derived substances due to enormous effluent organic matters (EfOM) from huge Gwangju sewage treatment plants.

The effect on photosynthesis and osmotic regulation in Beta vulgaris L. var. Flavescens DC. by salt stress

  • This study was to investigate the effect of salt stress on physiological characteristics such as plant growth, photosynthesis, solutes related to osmoregulation of Beta vulgaris. A significant increase of dry weight was observed in 50 mM and 100 mM NaCl. The contents of Chl a, b and carotenoid were lower in NaCl treatments than the control. On 14 day after NaCl treatment, photosynthetic rate (PN), the transpiration rate (E) and stomatal conductance of CO2 (gs) were reduced by NaCl treatment. On 28 day after NaCl treatment, the significant reduction in gs and E was shown in NaCl 200 mM. However, PN and water use efficiency (WUE) in all NaCl treatments showed higher value than that of control. Total ion contents (TIC) and osmolality were higher than the control. On 14 day after treatment, the contents of proline (Pro) increased significantly in 200 mM and 300 mM NaCl concentration compared with control, whereas on 28 day in all treatments it was lower than that of the control. The contents of glycine betaine (GB) increased with the increase of NaCl concentration. The contents of Na+, Cl-, GB, osmolality and TIC increased with the increase of NaCl concentrations. These results suggested that under severe NaCl stress conditions, NaCl treatment did not induce photochemical inhibition on fluorescence in the leaves of B. vulgaris, but the reduction of chlorophyll contents was related in a decrease in leaf production. Furthermore, increased GB as well as Na+ and Cl- contents resulted in a increase of osmolality, which can help to overcome NaCl stress.

Accuracy Evaluation and Alert Level Setting for Real-time Cyanobacteria Measurement Using Receiver Operating Characteristic Curve Analysis (ROC 분석을 이용한 수질자동측정소 실시간 남조류 측정의 정확성 평가 및 경보기준 설정)

  • With the need to evaluate accuracy of real-time measurement of cyanobacterial fluorescence to determine cyanobacterial blooms, this research examined 357 paired data (2013-2016) comprising both microscopic toxic cyanobacterial cell counts and concurrent real-time cyanobacterial concentrations at 2 sites (YS1 and YS2) in Yeongsan river. The increase in real-time cyanobacterial concentration was closely associated with the exceedance of 5,000 cyanobacterial cells/ml (odds ratio [OR] 1.07, 95% confidence interval [CI] 1.03-1.12) and 10,000 cells/ml (OR 1.08, 95% CI 1.04-1.12) at YS2 site. The area under the receiver operating characteristic (ROC) curve for the real-time cyanobacterial measurement at the YS2 site was 0.93, which indicates the measurement provides a high accurate detection of cyanobacterial blooms. On the ROC curve, the early alert levels of real-time cyanobacteria ranging $16-23{\mu}g$ chl-a/L would produce acceptable sensitivity of 79% and specificities greater than 90%. The real-time fluorescence measurement was found to be an accurate indicator of cyanobacteria and can serve as a tool for detecting toxic cyanobacterial bloom events in Youngsan river.

Growth regulation of cow1 rice mutant seedlings by blue light

  • We assessed whether the cow1 mutant defects are associated with growth of Tos17 and T-DNA insertional rice in blue light (BL). Growth of oscow1 mutants which encoded a member of the YUCCA protein family was retarded in BL. Root to shoot ratios of the mutants were reduced about 2 times lower in the absence of NAA and about 2.5 times lower in the presence of NAA; the shoot growth was not significantly changed by NAA addition. Photosynthetic activity of the mutants was however inhibited in high light. Pigment analysis showed significant difference between wild-type (Chl a:b = 3.02) and mutants (3.84). Carotenoid contents of the mutants were also decreased considerably, implying the involvement of cow1 in pigment formation. These findings lead us to suggest that the growth retardation of oscow1 mutant plants by BL results from the difference of photosynthetic activity in part.

PHOSPHATE-DEFICIENCY REDUCES THE ELECTRON TRANSPORT CAPACITIES OF THYLAKOID MEMBRANES THROUGH LIMITING PHOTOSYSTEM II IN LEAVES OF CHINESE CABBAGE

  • Experiments were carried out to investigate whether P, deficiency in detached 25 mM mannose-feeding led to a decline of the photosynthetic electron transport rates through acidification of the thylakoid lumen. With increasing mannose-feeding time, the maximal CO2 exchange rates and the maximal quantum yields of photosynthesis decreased rapidly up to 6 h by 73% then with little decrease up to 12 h. The ATP/ADP ratio declined by 54% 6 h after the treatment and then recovered to the control level at 12 h. However, the NADPH/NADP~ ratio was not significantly altered by mannose treatment. Electron transport rates of thylakoid membranes isolated from 6 h treated leaves did not change, but they decreased by 30% in 12 h treated leaves. The quenching analysis of Chl fluorescence in mannose-treated leaves revealed that both the fraction of reduced plastoquinone and the degree of acidification of thylakoid lumen remained higher than those of the control. The reduction of PSI in mannose fed leaves was inhibited due to acidification of thylakoid lumen (high qE). The reduction of primary quinone acceptor of PSII was inhibited by mannose feeding. Mannose treatment decreased the efficiency of excitation energy capture by PSII. Fo quenching was induced when treated with mannose more than 6 h, and had a reverse linear correlation with (Fv)m/Fm ratio. These results suggest that Pi deficiency in Chinese cabbage leaves reduce photosynthetic electron transport rates by diminishing both PSII function and electron transfer from PSII to PSI through acidification ofthylakoid lumen, which in turn induce the modification of photosynthetic apparatus probably through protein (de)phosphorylation.
