Lower Antibody Response in Chickens Homozygous for the Mx Resistant Allele to Avian Influenza

  • The chicken Mx gene has been regarded as a candidate gene for resistance to avian influenza virus (AIV). In this study, three groups of chickens with homozygotes (AA, GG) and heterozygotes (AG) of the resistant (A) and susceptible alleles (G) to AIV of the Mx gene were constructed from a line of dwarf egg-type chickens. These chickens were not examined for their resistant activities to AIV because the differential resistance had only been detected in vitro. The birds of the three groups were vaccinated with inactivated H5N2 AIV vaccine and the level of hemagglutination inhibition (HI) antibody to AIV was detected. The association between disease resistant activity to AIV and antibody response to AIV vaccination in the three groups was analyzed. The chickens with homozygous resistant allele A showed the lowest antibody levels, whereas the heterozygous chickens (AG) presented the highest antibody level after the boosting vaccination, which indicates that the efficiency of artificial selection on the resistant allele of Mx gene will be compromised since the homozygotes of the allele presented the weakest antibody response to the corresponding vaccine.

Prevalence of fowl adenovirus and chicken anemia virus in Jeonbuk, Korea (전북지역 조류아데노바이러스 및 닭전염성빈혈 감염률 조사)

  • Fowl adenovirus (FAdV) and chicken anemia virus (CAV) have gained much importance as an immunosuppressive and economically important emerging pathogen of poultry. This study was carried out to investigate the prevalence of FAdV and CAV infection in chickens. The groups were divided into Korean native chickens, broiler, layer hens and broiler breeder and set up groups according to age. As results, 12.5% of the native chicken, 2.5% of broiler and 6.7% of layer chicken were positive, respectively by PCR for FAdV. Serological test showed that 84.8%, 79.0%, 97.7% and 96.1% of chickens were positive for antibody to FAdV in native chickens, broiler, layer hens and broiler breeder. The prevalence of CAV infection were 20.0%, 7.5%, 16.7% and 10.0%, based on CAV gene detection by PCR. In serological test of CAV, 40.6%, 35.9%, 84.8% and 73.9% of chickens were positive in that groups.

Involvement of GABAergic Mechanism in the PIasticity Phenomenon of Chicken (닭의 Plasticity 현상에서 GABAergic 기작의 관련)

  • In monocular vision, bead and eye optokinetic nystagmus (OKN) display directionnal asymmetry, in lower vertebrates such as chickens, T-N stimulation being more efficient in evoking this visuomotor reflex than N-T stimulation. The N-T component of monocular OKN is significantly weaker in chickens. Coil recordings and observation showed that in adult chickens, prolonged monocular visual deprivation by unilateral eyelid suture provoked significant and progressive increase of the N-T component in chickens. This plasticity phenomenon involved the eye and head OKN in chickens. The administration of THIP, a GABA agonist, abolished reversibly the increase of the N-T component in chickens. This fact suggests that the GABAergic system could be involved in determining this plasticity phenomenon observed in adult lower vertebrates.

  • This experiment was conducted to investigate the effect of feed restriction in early stage of growth on the compensatory growth in male and female broiler chickens. Day-old 480 male and 480 female commercial broiler chickens(Ross strain) were randomly allotted to 32 pens(replicates), 30 birds per pen, to compare the sex difference in growth performance and carcass characteristics. Birds in control group(C) were full fed throughout the study. T1, T2 and T3 were restricted at 4∼11d, 4∼14d and 4∼18d, respectively. Chickens were weighed every week, and one bird was selected from each pen to measure carcass characteristics at the end of experiment. In female chickens, all restricted-fed groups except for T3 weighed same as or heavier than the control group at 7 weeks of age. In male chickens, both T2 and T3 that had longer feed restriction period were significantly different from the control group. Viability was not different significantly. Feed intake in both male and female chickens of T3 group statistically differed from the control group. The female chicks showed larger abdominal fat pad sizes than did male birds up to market age.

Dexamethasone reduces infectious bursal disease mortality in chickens

  • Very virulent infectious bursal disease virus (vvIBDV) causes high mortality in chickens but measures to reduce the mortality have not been explored. Chickens (8-9 weeks) were treated with 3 agents before and during vvIBDV inoculation. Dexamethasone treatment reduced the mortality of infected chickens (40.7% vs. 3.7%; p < 0.001), but treatment with aspirin or vitamin E plus selenium did not affect the mortality. The bursa of Fabricius appeared to have shrunk in both dead and surviving chickens (p < 0.01). The results indicate that dexamethasone can reduce mortality in vvIBDV-infected chickens and may provide therapeutic clues for saving individual birds infected by the virus.

Effects of Cecal Ligation and Colostomy on Food and Water Intake and loafer Excretion in Chickens Fed Restrictedly and Freely (닭에 있어서 사료섭취의 자유 및 제한급여시킬 때의 사료섭취량, 음수량 및 수분 배설량에 미치는 맹장결찰 및 인공항문 수술의 효과)

  • It was examined whether the ceca and the back-flow of urine into the lower intestine are involved in water intake and excretion in chickens and food intake affects those. Colostomy significantly increased water intake, total water excretion and the ratio of the water intake to food intake in the ceca-ligated chickens under restrict and ad libitum feeding conditions (P<0.05), but the increases were much larger in chickens fed ad libitum than in those fed restrictedly. Cecal ligation increased water intake, total water excretion and the ratio of water intake to food intake in the colostomised chickens which were fed freely (P<0.05). but not in those fed restrictedly, None of colostomy and cecal ligation affected the resultant water balances in chickens under both feeding conditions. Colostomy increased food intake in the ceca-ligated chickens (P<0.05), but no increase by cecal ligation was observed in colostomised chickens. It is concluded that the lower intestine takes a very important role in water recovery from urine to maintain water balance in chickens.

Effect of Red Pepper [Capsicum annuum L.] on Growth Rate and Resting Metabolic Rate in Chickens (고추 [Capsicum annuum L.]가 병아리의 성장률(成長率)과 휴식시대사(休息時代謝)에 미치는 영향(影響))

  • The effect of administration of red pepper Capsicum annuuml on the growth rate and resting metabolic rate in male chickens was investigated while varying the dose of red pepper. Growth of the chickens fed the rations containing small amount of red pepper(0.1% or 0.5%) was significantly enhanced. In the chickens given the ration containing 1% of red pepper, which showed the result similar to that of control chickens fed the ration without red pepper, retarded growth was seen as compared with the growth of the chickens fed the rations containing small amount of red pepper. Chickens administered the rations containing large amount of red pepper (5%, 10%, 60%) showed significantly retarded growth as compared with the growth of control chickens. There was no effect on the resting metabolic rate of chickens when administered varying amount of red pepper. The oxygen of resting chickens was varied $9.0-10.5ml/kg^{0.75}/min$ and rectal temperature was varied within the normal range ($41-42^{\circ}C$).

Isolation of Salmonella from the layer chickens reacting in pullorum-typhoid agglutination test (추백리 혈청검사 양성 산란계로부터 Salmonella속균 분리)

  • To investigate the specificity of rapid slide agglutination test for pullorum-gallinarum diseases and to obtain a basic data for avian salmonellosis control, salmonella isolation was peformed for the layer chickens positively reacted in pullonlm-typhoid agglutination test. The biochemical, serological and antimicrobial properties of the isolates were examined. The results obtained through this study were summarized as follows; 1. Of 2,384 chickens tested by the agglutination test, 606 chickens (25.4%) were positive reactors. 154 of 606 reactors and 49 of the non-reacting chickens were investigated for salmonella isolation, resulting in isolation of 68 strains of salmonellae from 27 chickens. 2. By organs, the isolation frequency from liver, cecum, spleen, ovary and gall bladder showed 8.9% (18 strains), 8.9% (18 strains), 7.4% (15 strains), 4.4% (9 strains) and 3.9% (8 strains), respectively. 3. By culture medium the combination of selenite broth and MacConkey agar revealed the highest isolation rate and the enrichment culture by delayed secondary enrichment culture method was found the most effective for salmonella isolation. 4. The serotypes of 68 salmonella isolates were identified as 3 strains of S pullorum, 24 strains of S gallinarum, 15 strains of S typhimurium, 8 strains of S enteritidis, 7 strains of S paratyphi A, 5 strains of S typhimurium and 6 strains of the other salmonellae. 5. The serotypes of 8 salmonella strains isolated from 49 chickens non-reacting in pullorum-typhoid agglutination test were identified as 3 strains of S typhimurium and 5 strains of S infantis. 6. When 24 chickens of which 68 strains of salmonellae isolated were examined by microplate agglutination test, the average antibody titer for pullorum antigen was $2^{5.25}$. The chickens at antibody titer between $2^3$ and $2^5$ showed the higher frequency of isolation as compared with the chickens at the other titers. 7. When salmonella isolates were tested the antimicrobial drug sensitivity by disk diffusion method, S paratyphi A were highly sensitive by 100% to ATM and GM, S typhimurium, by 88% to AM, CIP, IMP and TN, S infantis, by 100% to AM, CRO, ENR and PIP, S enteritidis,by 100% to IMP and PIP, S pullorum, by 100% to ATM, CRO, ENR and PIP and S gallinarum, by 92% to CRO, CIP and PIP.

Investigation of the Insulin-like Growth Factor System in Breast Muscle during Embryonic and Postnatal Development in Langshan and Arbor Acres Chickens Subjected to Different Feeding Regimens

  • Nutrient availability may control muscle growth directly and indirectly through its influence on regulatory factors. We analyzed the effects of nutrient availability on the breast muscle insulin-like growth factor system. Real time RT-PCR was used to quantify the level of transcription in breast muscle from Langshan (LS) layer and Arbor Acres (AA) broiler chickens subjected to different feeding regimens during embryonic and postnatal development. The AA chickens were fed AA diet (AA, control group) while the LS chickens were either fed LS diet (LL) or AA diet (LA). According to our results, insulin-like growth factor (IGF)-II (embryonic day 16 (E16) - postnatal day 42 (P42)), IGF-I receptor (IGF-IR, E18-P42), and IGF binding protein (IGFBP)-2 (E18-P42), -5 (E16-P14), -7 (E12-P0), and -3 (E12-P0) were positively correlated with IGF-I, while IGFBP-3 (P0-P28) was negatively correlated with IGF-I. In comparison, IGF-IR (E18-P42), IGFBP-2 (E18-P42), IGFBP-5 (E14-P0), and IGFBP-3 (E16-P0) were positively correlated with IGF-II, while IGF-IR (E10-E16) and IGFBP-3 (P0-P28) were negatively correlated with IGF-II. Moreover, IGFBP-2 (E16-P42), -7 (E10-E16), and -3 (E10-E16) were positively correlated with IGF-IR, while IGFBP-3 (P0-P28) was negatively correlated with IGF-IR. Finally, IGFBP-7 (E12-P0) was positively correlated with IGFBP-3, while IGFBP-2 (P0-P28) and -7 (P0-P42) were negatively correlated with IGFBP-3. Overall, the AA chickens exhibited higher levels of IGF-I, IGF-IR, and IGFBP-2 mRNA expression than the LL chickens, while the opposite was true for IGFBP-7. No strain differences in IGF-I, IGF-IR, and IGFBP-7 mRNA expression were detected between LA and AA chickens; however, a strain difference was observed for IGFBP-2. LA chickens exhibited higher levels of IGFBP-2 than LL chickens, while the opposite was true for IGFBP-7. Our data show the first evidence that certain genes may be correlated during specific developmental periods and that strain differences in the expression of those genes in LS and AA chickens are due to differential responses to the same diet.

Effect of Mixed Rearing of Male and Female Chickens on the Stress Response of Korean Native Chickens (토종 종계의 암수 합사가 개체의 스트레스 반응 정도에 미치는 영향)

  • This study was performed to investigate the effect of mixed rearing of male and female chickens on the stress response in Korean native chickens. To identify the degree of the stress response, heterophil-lymphocyte ratio (H/L ratio), heat shock protein genes (HSPs) expression, and intracellular nuclear DNA damage rate were analyzed before and after the mixed rearing of male and female chickens. The results showed that the H/L ratio of chickens after mixing males and females was more than thrice as higher than before mixing (P<0.001), but the differences between males and females were not significant. HSP-70, HSP90-α, and HSP90-β expression levels were 2.5 to 3.4 times higher after mixing male and female chickens, compared to before mixing (P<0.01). In the mixed rearing of male and female chickens, the increase in HSPs expression in females was higher than in males. Comet indicators in intracellular DNA damage rate analysis showed a significant increase after mixing male and female chickens compared to before mixing (P<0.001). However, there was no significant difference between males and females with respect to DNA damage rate. Taken together, these results suggest that male and female mixed rearing acts as a strong external stressor in both male and female chickens.
