Viability of preserved Cryptosporidium baileyi oocysts

  • The present study was undertaken to determine the viability and infectivity of oocysts of Cryptosporidium baileyi that had been stored from 1 to 40 months at $4^{\circ}C$ preserved in 2.5% potassium dichromate solution. Oocysts of C. baileyi were purified from the feces of experimentally infected chickens using discontinuous sucrose gradients. Subsequently, the purified oocysts were suspended in 2.5% potassium dichromate solution at a concentration of $1{\;}{\times}10^7$ organism/ml, and their viabilities were assessed by nucleic acid staining, histologic examination, and infectivity to 2-day-old chickens. All chickens inoculated with oocysts that had been stored for 1-18 months developed patent infections, while chickens infected with older oocysts remained uninfected. Between 5.8% and 82.2% of the oocysts, stored at $4^{\circ}C$ in 2.5% potassium dichromate solution, were found to be viable, as determined by nucleic acid staining. Parasite colonization in the bursa of Fabricius was detected in the microvillus border of bursal epithelium. The finding that C. baileyi oocysts remain infective to chickens for at least 18 months offers important time-saving advantages to investigators who frequently require large numbers of oocysts.

Effects of Siegesbeckiae Herba Extract on the Blood Pressure of Dogs and Chickens(II) (회첨 엑기스의 혈압에 미치는 영향(II))

  • This study was performed in dogs and chickens to observe differences of responses by species of animals to hypotensive action of Siegesbeckiae Herba, using its ethanol extract. Siegesbeckiae Herba e ethanol extract (SGEE), when injected into the vein of dogs or chickens, elicited depressor actions in both a animals. The hypotensive effects of SGEE in dogs and chickens were markedly inhibited by the pretreatment of atropine, but not affected by bethanidine, propranolol and diphenhydramine. Phentolamine significantly weakened the action of SGEE in dogs, but did not alter the action of SGEE in chickens. SGEE inhibited the pressor action by norepinephrine or carotid artery occlusion, whereas it did not change the pressor action b by angiotensin in dogs.

  • The fat deposition is an important factor affecting chicken meat quality, which is closely related to lipid metabolism of chickens. Therefore, it is important to regulate the lipid metabolism of chickens to improve the chicken meat quality. Plant extracts have special regulatory effects on animal's growth and health and have been widely used in chicken breeding. Some plant extracts have been reported to have functions of changing the fatty acid composition, reducing abdominal fat percentage, and enhancing the intramuscular fat content of chickens by improving the antioxidant capacity, regulating the expression of genes, enzymes, and signaling pathways related to lipid metabolism, modulating intestinal microbiota, affecting hormones level, and regulating DNA methylation. This paper reviewed the application and mechanism of plant extracts on regulating lipid metabolism of chickens to provide a reference for the further application of plant extracts in chicken breeding.

Pathological findings of leucocytozoonosis in chickens (닭 leucocytozoonosis의 병리학적 소견)

  • Pathological findings of leucocytozoonosis naturally occurred in Korea are described. Most of the affected chickens showed paleness of comb and wattle, egg drop, approximately 20 to 30% mortality, green diarrhea, anorexia and weakness. On the blood smear merozoits and gametocytes were detected. At necropsy, petechial and ecchymotic hemorrhages in liver, kidney, heart, muscle, and splenomegaly were frequently observed. Microscopically schizonts with fine granular merozoits were often detected in kidney, intestine, brain, lung, myocardium, muscle, liver with cenrilobular fatty change, and spleen.

  • IBD's antibody level and morphological change of immune organ was examined in chicken. The results were as follows ; 1. Seventy percent at 42 day and 40% at 45 day of age chickens were reacted positively by the agar gel immunodiffusion test and 42 day and 45 day of age chickens indicated 1859, 1425 by the ELISA test, respectively. 2. In 2 and 5 day young broiler chickens, the level of maternal antibody was not proper. B/B ratio showed low level, but S/B ratio and BS were normal. 3. In layer, 100% of 8 and 86 day of age chicken and 70% at 40 day of age chicken had antibody aganist IBDV. The level of antibody was high as 2293 and 3336 in 8 and 86 day of age chicken, while was low as 1186 in 40 day of age chicken. B/B ratio showed low level and S/B ratio high level, but BS was normal.

  • Chicken has been one of the most useful animals for life. They have been not only one of the most economical and adundant sources of valuable nutrients, but also the very good experimental animal to develop modern sciences. In Japan, eggs laid by the chickens have long been good sources of cash income for farmers and in many cases even for Samurais. Although nearly 50 millions of chickens were raised and they were of considerable importance in Japanese agricultre before the World War II, the industrialization or specializatin started after the War, Substantiallysince the 1960s. Among other spcies of poultry then chickens, ducks and Japanese quails are of some importane in Japan. Duck meat is often used in various cuisines at higher class restaurants and Japanese quail eggs are widely used. Hower those those species of commpared with chickens.

  • Reciprocal crosses were conducted between three strains of Taiwan Country chickens, developed in the National Chung-Hsing University, and two strains of Single Comb White Leghorns, developed in the Taiwan Livestock Research Institute. Traits studied were growing performances, laying performances, egg quality traits and traits concerning disease resistance, including resistance to Marek's disease virus and immune responses to Newcastle disease virus vaccine and to sheep red blood cell. Results indicated that laying performances of Taiwan country chickens were much inferior to White Leghorns, but they matured earlier, their eggs had better shell strength and larger proportion of yolk, and their general disease resistance was much better than White Leghorns. Heterosis were found in laying performances and egg quality traits. The heterosis in laying traits was so large that the hybrid laid as many eggs and as large eggs as did pure strains of White Leghorns. Strategies on the improvement of native chickens and the utilization of genetic merits of native chickens were also discussed.

  • Three mature layer chickens from a farm in which chickens showed green diarrhea, cyanosis, lethargy, loss of appetite were pathologically examined. Grossly, multiple variable sized caseous nodules were detected in the liver, intestinal serosa and mesentery. In addition, parathypoid nodules in the liver and fibrous serositis on the several peritoneal organs and tissues were noticed. One of spleens had multiple infarction areas. Histologically caseous nodules consisted of central caseous core and peripheral epithelioid cells overlying the fibrous connective tissue. Multinucleated giant cells were scattered between the epithelioid cells and fibrous connective tissue. In these nodules Gram negative cocobacilus bacterial colonies were present, whereas Periodic Schiff reaction and Ziehl-Neelsen stain detected neither fungi nor acid fast bacteria. From these results multiple granulomas might be induced by Escherichia coli. In addition, severe Ascafdiodf and Salmonellosis were coinfected in these chickens.

Immunohistochemistry and RT-PCR for pathogenesis of Newcastle disease in chickens (닭 뉴캣슬병의 발병기전 규명을 위한 RT-PCR 및 면역조직화학적 연구)

  • The present experiment was carried out to study the pathogenesis of Newcastle disease(ND), ND virus (NDV) antigens and genes in various organs from NDV inoculated chickens were detected by immunohistochemistry and RT-PCR. Immunohistochemically, NDV antigens were detected in the spleen, thymus, cecal tonsil, proventriculus, trachea and lungs at 12 hour post-inoculation (hpi). Viral antigens were localized mainly in the cytoplasm of lymphocytes and macrophages. After 48 hpi, clinical findings of the affected chickens were open-mouth breathing, conjunctivitis, watery diarrhea and edema around the eye and neck. After 72 hpi, chickens showed muscular tremor, paralysis of the legs and wings, and coma. Histopathological results consist of multi-focal necrosis with hemorrhages in lymphoid aggregates of the intestinal tracts, necrosis of the lymphoid tissues, neuronal degeneration and necrosis, and perivascular cuffing. Using RT-PCR, virus genes were detected in the spleen and proventriculus at 48 hpi, and in the brain at 60 hpi.

  • This experiment was carried out for 42 days to investigate the effect of caponizing and diethylstilbestrol (DES) on body weight gain, rate of feed conversion, weight of carcass and percentage of dressing in young chickens. The chickens used were 128 male Hamphorn of 30 days old. In the body weight gain of chickens, control group was higher than the other groups (P<0.01); $T_1$ group was the lowest group. Rate of feed conversion of control group was higher than $T_2$ and $T_3$ groups.
