Quantitative Approaches to Assess Key Carcinogenic Events of Genotoxic Carcinogens

  • Chemical carcinogenesis is a multistep process. Genotoxic carcinogens, which are DNA-reactive, induce DNA adduct formation and genetic alterations in target cells, thereby generating mutated cells (initiation). Subsequently, preneoplastic lesions appear through clonal proliferation of the mutated cells and transform into tumors (promotion and progression). Many factors may influence these processes in a dose-dependent manner. Therefore, quantitative analysis plays an important role in studies on the carcinogenic threshold of genotoxic carcinogens. Herein, we present data on the relationship between key carcinogenic events and their deriving point of departure (PoD). Their PoDs were also compared to those of the carcinogenesis pathway. In an experiment, the liver of rats exposed to 2-amino-3,8-dimethylimidazo-(4,5-f)quinoxaline (MeIQx) was examined to determine the formation of MeIQx-DNA adducts, generation of mutations at LacI transgene, and induction of preneoplastic glutathione S-transferase placental form (GST-P)-positive foci and tumors (benign and malignant). The PoDs of the above key events in the carcinogenicity of MeIQx were increased as the carcinogenesis advanced; however, these PoDs were lower than those of tumor induction. Thus, the order of key events during tumor induction in the liver was as follows: formation of DNA adducts ${\ll}$ Mutations ${\ll}$ GST-positive foci (preneoplasia) ${\ll}$ Tumor (adenoma and carcinoma). We also obtained similar data on the genotoxic and carcinogenic PoDs of other hepatocarcinogens, such as 2-amino-3,8-dimethylimidazo(4,5-f)quinoline. These results contribute to elucidating the existence of a genotoxic and carcinogenic threshold.

Ecotoxicological effects of ballast water effluent teated by an electrolytic method on marine environment

  • Ballast water effluent treated by an electrolytic method contains reactive chlorine species and disinfection by-products (DBPs). In this study, we conducted whole effluent toxicity (WET) testing and ecological risk assessment (ERA) to investigate its ecotoxicological effects on marine environment. WET testing was carried out for three marine pelagic organisms, i.e., diatom Skeletonema costatum, rotifer Brachionus plicatilis and fish Paralichthys olivaceus. The biological toxicity test revealed that S. costatum was the only organism that showed apparent toxicity to the effluent; it showed no observed effect concentration (NOEC), lowest observable effect concentration (LOEC) and effect concentration of 50% (EC50) values of 12.5%, 25.0% and 83.3%, respectively, at brackish water condition. In contrast, it showed insignificant toxicity at seawater condition. B. plicatilis and P. olivaceus also showed no toxicities to the effluent at the both salinity conditions. Meanwhile, chemical analysis revealed that the ballast water effluent contained total residual oxidants (TROs) below $0.03{\mu}g/L$ and a total of 20 DBPs including bromate, volatile halogenated organic compounds (VOCs), halogenated acetonitriles (HANs), halogenated acetic acids (HAAs) and chloropicrin. Based on ERA, the 20 DBPs were not considered to have persistency, bioaccumulation and toxicity (PBT) properties. Except monobromoacetic acid, the ratio of predicted environmental concentration (PEC) to predicted no effect concentration (PNEC) of the other 19 DBPs did not exceed 1. Thus, our results of WET testing and ERA indicated that the ballast water effluent treated by electrolysis and subsequently neutralization was considered to have no adverse impacts on marine environment.

The importation of genetically modified crops and its environmental impacts in Korea

  • The global cultivation area of genetically modified crops (GM crops) has been increasing every year. Cultivation of GM crops is not only beneficial to the economy but also has positive effects on the environment in decreasing the use of agrochemicals, chemical fertilizers, and agricultural machinery. However, there have been controversies about the admixture of GM crops and non-GM crops and the unintentional release of GM crops to the environment. Especially in Korea, where consumption of agricultural products is import-dependent, the economic importance of GM crops has been a significant issue. The Act on import and distribution of GM crops was established in 2001 to start the management of GM crops in Korea. Recently, the imported amount of GM crops to Korea has reached over 10 million tons and is increasing very rapidly; consequently, the potential environmental impact of GM crops is becoming a big issue in Korea. In Japan, the discovery of imported GM canola plants around ports in 2005 raised awareness of the unintentional release of GM crops. In Korea, GM maize plants were also found in port and feed factory surroundings from 2005 to 2007. It is now necessary to monitor imported GM crops by tracing distribution, transport process for practical environmental risk assessment. Possible gene transfer from GM crops to non-GM crops should also be investigated in the cultivation area and the surroundings as well.

  • Effluent treated by an NaDCC injection method in Ballast water management system (BWMS) contains reactive chlorine species and disinfection by-products (DBPs). WET testing was carried out for four marine pelagic and freshwater organisms, i.e., diatom Skeletonema costatum, Navicula pellicuosa, chlorophyta Dunaliella tertiolecta, Pseudokirchneriella subcapitata, rotifer Brachionus plicatilis, Brachionus calyciflorus and fish Cyprinodon variegatus, Pimephales promelas. The biological toxicity test revealed that algae was the only biota that showed apparent toxicity to the effluent; it showed no observed effect concentration (NOEC), lowest observable effect concentration (LOEC) and effect concentration of 50% (EC50) values of 25-50%, 50-100% and >100%, respectively, at three water condition, but did not show any significant toxicities on other biota. Meanwhile, chemical analysis revealed that the BWMS effluent contained total residual oxidants (TROs) below $0.03{\mu}g/L$ and a total of 25 DBPs such as bromate, volatile halogenated organic compounds (VOCs), halogenated acetonitriles (HANs), halogenated acetic acids (HAAs), chloropicrin and Isocyanuric acid. Based on ERA, the 25 DBPs were not considered to have persistency, bioaccumulation and toxicity (PBT) properties. The ratio of predicted environmental concentration (PEC) to predicted no effect concentration (PNEC) of the other DBPs did not exceed 1 for General harbor environment. However, four substances (Isocyanuric acid, Tribromomethane, Chloropicrin and Monochloroacetic acid) were exceed 1 for Nearship environment. But observed toxicity in the test water on algal growth inhibition would be mitigated by normal dilution factor of 5 applied for nearship exposure. Thus, our results of WET testing and ERA showed that the BWMS effluent treated by NaDCC injection method would have no adverse impacts on marine environment.

작업자 무영향도출수준(worker-DNEL)의 사업장 적용을 위한 평가 연구 (Evaluation of the Application of worker-DNELs under REACH Guidance as Provisional Occupational Exposure Limits in the Workplace)

  • Objectives: The purpose of this study is to calculate the worker-DNEL (Derived No Effect Level) values using the REACH guidance and compare the calculated DNELs with existing Korea occupational exposure limits (KOELs) for evaluation of the applicability of the worker-DNELs as provisional occupational exposure limits for chemicals that are not established KOELs in the workplace. Methods: The worker-DNELs for 46 chemicals among 113 hazardous substance requiring management were calculated using the REACH guidance, and a paired t-test was performed to see if there is any statistical difference between two lists (worker-DNELs vs KOELs). The ratios of KOELs over worker-DNELs were also calculated to compare the overall levels of two lists using the geometric means method. Results: The calculated worker-DNELs for 46 chemicals ranged from 0.001 to $329mg/m^3$ (GM= 6.9, GSD = 10.8), and appeared to be a significant difference between the worker-DNELs and the KOELs (p < 0.01). In addition, the ratios of KOELs over worker-DNELs ranged from 0.3 to 394 times (GM = 10.2, GSD = 3.9), indicating that the worker-DNELs were, on average, 27 times lower than the KOELs. Conclusions: Therefore, the study results show that the calculated worker-DNELs can be applied and used as provisional occupational exposure limits in the workplace in order to reduce worker exposures to chemicals and health risks, and manage potential worker exposures based on the precautionary principle through comprehensive chemical risk assessment.

보육시설, 산후조리원의 실내공기질 농도 및 위해성평가 (Concentration and risk assessment of indoor air quality in day care centers and postnatal care centers)

  • In this study, we measured the concentration of Particulate Matter($PM_{10}$), Formaldehyde(HCHO), and Total Bacteria Count (TBC) at two facilities: day care centers, and postnatal care centers located in the cities of Gyeonggi, Gangwon, Jeolla and Gyeongsang from January 1, 2012 to December 31, 2015. $PM_{10}$ concentration was similar to the day care centers and postnatal care centers. HCHO concentration was the highest in the postnatal care centers. TBC concentration was the highest in the day care centers. Comparing the different cities, $PM_{10}$ concentration was the highest in Gyeonggi, HCHO concentration was the highest in Gyeonggi, and TBC concentration was the highest in Gyeonggi. As a result of HCHO's risk assessment, it was found that adults exceeded the carcinogenicity tolerance of $10^{-6}$ specified by the US EPA. This study is expected to be helpful in preventing damage to health from the contaminated indoor air at sensitive facilities, and can be used as basic data for indoor air quality management.

  • 본 연구는 공동주택에 도시가스를 공급하기 위해 정압기 대신 설치하는 공용 압력조정기의 안전성 향상을 위한 대안을 마련하기 위한 목적으로 진행되었다. 먼저, 국내 압력조정기의 안전기준 및 설치 실태를 조사하여 문제점을 도출하고 해결방안을 제시하였다. 또한 정량적 위험성평가 솔루션인 DNV Phast-Safeti 프로그램을 이용해 공동주택에 설치되는 압력조정기와 주변 가스설비에 내포된 가스누출 등의 위험성을 개인적 위험도와 사회적 위험도로 구분하여 평가하고, 압력조정기의 설치와 관리 측면에서 위험성 감소를 위한 방안을 도출하였다.

Effect of Dosage Level of Carcinogen and Clonorchis sinensis Infestation on Cholangiocellular Carcinoma Induction in Hamsters

  • The infection of liver flukes, Clonorchis sinensis (CS) and Opisthorchis viverrini (OV), has been known as a risk factor to induce cholangiocellular carcinoma (CCC) in human living in the endemic area, providing promoting effect on the liver initiated by chemical carcinogens. The present study evaluated the relationship between the dosage level of dimethylnitrosamine (DMN) and the infection load of CS in the neoplastic development by histopathological examination of the treated hamsters. To evaluate the effects of DMN, different doses of DMN ranging from 0 to 25 ppm were administered to hamsters with 20 CS metacercariea. For the risk assessment of the infection load, 0, 5, 15, 50 CS metacercariae were respectively infected with 12 ppm DMN. The mortality was closely related to the infection load rather than the concentration of DMN. The infection of CS clearly promoted the induction of CCC even at dose level of 6 ppm DMN. Only five metacercariae were enough to promote CCC induction at the concentration of 12 ppm DMN.

다중판별 장치 스크리닝 기법을 이용한 가스공정의 위험성 평가 (Hazard Evaluation of Gas Processes Using a Multi-distinction Equipment Screening Algorithm)

  • 본 연구에서는 가스공정에서 중요시되는 화재/폭발에 대한 위험성 평가 기법으로, 공정위험 지수법인 Dow's F&EI와 정성적 위험판별기법인 장치스크리닝 기법의 특성에 산업별로 위험시되는 사항에 대한 전문가 의견을 수렴할 수 있도록 전문가 시스템-장치 및 공청의 사고사례 및 전문가의 의견 수렴된 시스템을 도입하여 위험 지수에 전문가적 reward/penalty를 주고 공정 장치의 종합위험도 지수를 계산하여 보다 정밀한 위험장치 검색을 할 수 있는 형태인 다중판별 장치스크리닝 기법(Multi-distinction Equipment Screening Algorithm, MESA)을 제안하였다. 그리고 이에 대한 사례연구로 LPG 지하저장공정의 위험도 순위에 대해 전문가 시스템이 도입되지 않은 경우와 도입된 경우의 위험판단 해상도를 파악하여 제안 알고리즘의 유용성을 입증하였다. 그리고 이와 같은 위험도 순위에 따른 장치의 물리적 특징을 파악함으로써 안전장치 및 mitigation 장치 그리고 비상대응계획에 대한 정보를 제공할 수 있으리라 기대한다.

고엽제 노출에 따른 건강위해의 보건학적 고찰 (Review of Public Health Aspects of Exposure to Agent Orange)

  • Objectives: Controversy regarding the relationship between exposure to Agent Orange and disease has progressed for more than four decades, both at home and abroad. Recently, the allegation by US veteran Steve House of the burial of Agent Orange at the US Army base Camp Carroll located in Waegwan-eup, Korea, has emerged. We reviewed published articles and reports related to Agent Orange. Methods: Articles and reports were collected online using the keywords 'agent orange' and 'health' and then reviewed. Results: A number of epidemiologic studies have reported disease outcomes due to exposure to Agent Orange, while others were unable to establish a link to the injuries of veterans of the Vietnam War. This can be explained by the fact that accurate exposure assessment should be carried out since exposure misclassification in epidemiologic studies can affect estimates of risk. In the case of the burial of Agent Orange at Camp Carroll, an exposure pathway could be through underground water supplies, which differs from the cases of Vietnam and Seveso in Italy. Conclusion: There still remains a dispute among academics regarding the relationship between exposure to Agent Orange and disease, although Agent Orange is a highly toxic chemical. This dispute indicates that accurate exposure pathway and exposure assessment is needed.
