The effects of total ginseng saponin. extracts of Panax ginseng C.A. Meyer, on the differentiation of F9 teratocarcinoma stem cells were studied. F9 stem cells cultured in the presence of ginseng saponin together with dibutyric cAMP became parietal endoderm - like cells. Moreover, the expressions of differentiation marker genes. laminin. type IV collagen. and retinoic acid $receptor-{\beta}(RAR{\beta})$ were increased after treatment of ginseng saponin. Among various ginsenosides purified from crude ginseng saponin, $Rh_1\;and\;Rh_2$ caused the differentiation of F9 cells most effectively. Since ginsenosides and steroid hormone show resemblance in chemical structure. we studied the possibility of the involvement of a steroid receptor in the differentiation process induced by ginsenosides. According to Southwestern blot analysis, a 94 kDa protein regarding as a steroid receptor was detected in F9 cells cultured in the medium containing ginseng saponin. Based on these data, we suggest that ginseng saponin, especially ginsenosides $Rh_1\;and\;Rh_2$ cause the differentiation of F9 cells and the effects of ginsenosides might be exerted via binding with a steroid receptor or its analogous nuclear receptor.

Shade-dried flowers of Sarcostemma brevistigma Wight Syn (SBF) belonging to Asclepiadaceae yielded a rare flavonol glycoside viz., Quercetin 5a prenyl, 3-O-glucosyl, 7-O-(4c-p-coumaroyl) neohesperidoside which were characterized based on chemical and spectral (including 2D NMR) studies. The ethyl acetate fraction of alcoholic concentrate (test sample) of flowers of this plant (SBF) was evaluated for its hepatoprotective and inhibition of lipid peroxidation activities to investigate the scientific basis of the traditional uses. The oral administration in varying doses viz., 125 and 250 mg/kg of aqueous suspension of SBF to rats for 7 days produced significant (P < 0.01) hepatoprotective effect comparable to that of standard drug silymarin. The SBF afforded good hepatoprotection against $CCl_4$ induced elevation levels of serum marker enzymes, serum bilirubin and liver weight. The free radical scavenging effects of SBF and flavonol glycoside (SA) were assigned by $Fe^{2+}$/ascorbate induction method (in vitro), which revealed the inhibition of lipid peroxidation. The SBF and SA showed prominent anti-lipid peroxidation activity ($IC_{50}$ about $\sim$180 mg/ml and 11.0 mg/ml respectively), which was comparable to standard drug curcumin ($IC_{50}$ about $\sim$8.25 mg/ml). Thus the hepatoprotective activity of SB could be correlated to the free radical scavenging property of the flavonol glycoside.
Epidemiologic studies have demonstrated an association between short-term exposure to particulate air pollutants and increased mortality. However the biological mechanism underlying these associations have not been fully established and also the chemical and physical characteristics of the pollutant particles are not well understood. The metal constituents of air pollutant particles and their bioavailability are considered to Play an important role as possible mediators of Particle-induced airway injury and inflammation. Sprague-Dawley rat alveolar macrophage cells (NR8383) were exposed to airborne and acid-leached particulate matter (PM). Titanium oxide and nickel subsulfide were used as negative and positive controls. Particle-induced reactive oxygen species formation in cells was detected using the fluorescent probe 2',7'-dichlorofluorescin diacetate. Expression of TNF-$\alpha$ and IL-6 were measured by enzyme-linked immunosorbent assay, and PM-induced DNA double-strand breaks were determined with $\lambda$DNA/Hind III marker. Metals associated with air pollutant particles mediated intracellular oxidant production in alveolar macrophages, and the cytotoxicity and proinflammatory cytokine production induced by PM were associated with oxidative stress. The oxidants produced by air pollutant particles also are likely to induce DNA double-strand breaks. Our findings in alveolar macrophage cells exposed to PM and acid-leached PM support the hypothesis that metal components in urban air pollutants and their bioavailabilities might play an Important role in the induction of the adverse health effects.

In order to facilitate the quality control of the fruits of Crataegus pinnatifida, a simple, accurate and reliable HPLC method was developed for the simultaneous determination of the three bioactive compounds: chlorogenic acid (1), rutin (2), and hyperin (3), which were selected as the chemical markers of C. pinnatifida. Separation was achieved on an Agilent Eclipse XDB-C18 column with a gradient solvent system of 0.1% trifluoroacetic acid aqueous-acetonitrile at a flow-rate of 1.0 mL/min and detected at 254nm. All three calibration curves showed good linearity ($R^2$ > 0.998). The recoveries of three marker compounds were in the range of 94.87~111.52 %. The contents of chlorogenic acid (1), rutin (2), and hyperin (3) of the fruits of C. pinnatifida collected from 23 district markets in Korea, Japan, and China were 0.16~0.65 mg/g, 0.07~1.24 mg/g, and 0.03~0.62 mg/g, respectively. The results demonstrated that this method is simple and reliable for the quality control of the fruits of C. pinnatifida.

In this study, a wide range of volatile organic constituents of environmental tobacco smoke (ETS) were determined using an environmental chamber, where ETS is the sole source of target compounds. ETS was generated in an environmental chamber by a number of different cigarettes, including the Kentucky reference cigarette and eight different commercial brands. More than 30 compounds were measured simultaneously for a total of twelve experimental runs. The target compounds are classified into three major classes, i.e. vapor phase ETS markers including 3-ethnylpyridine (3-EP) and nicotine, carbonyl compounds including formaldehyde, and volatile organic compounds (VOCs). The results from the chamber study were used to generate characterized ratios of selected VOCs to 3-EP, a vapor phase ETS marker. Emission factors for VOCs associated with ETS were also estimated. The characteristic ratios appeared to be generally in good agreement with published data obtained by environmental chamber studies similar to this study. This implies that the ratios may be useful for identifying and quantifying the impact of ETS as a source of target compounds in 'real world' indoor environments, which is affected by a complex mixture of multi-sources. The environmental chamber method described here provides a direct and reliable method to compare the ETS generated by different cigarettes. The method can also be applied to the simultaneous determination of many different ETS components.
Scutellaria baicalensis $G_{EORGI}$ (Scutellariae Radix) has been used to clear heat and to dry dampness in the stomach or intestines, which manifests as diarrhea or dysenteric disorder. In this study, we investigated the effective preparation of active components in Scutellariae Radix using the methods of solvent extraction and absorption fractionation for the development of new functional food or pharmaceuticals. The marker substances, baicalin, baicalein, wogonoside, and wogonin were directly isolated from the Scutellariae Radix. There chemical structures were elucidated by spectroscopic analysis. The Scutellariae Radix was extracted with hot water. To enhance yield of flavonoids in Scutellariae Radix, the hot water extract was dissolved in ethanol with concentration dependent manner. The precipitates were separated using centrifugal techniques at 10,000 rpm. Supernatant liquid was applied to the HPLC for quantification of major compounds. Separately, the hot water extract was absorbed on Diaion HP-20 resin. And then, the absorbed fraction was eluted with methanol for HPLC. The contents of baicalin, baicalein, wogonoside and wogonin in different treatment methods were analyzed by HPLC. Total amount of four major components were 16.9% in 50% ethanol extract, 21.7% in 70% ethanol extract, 20.5% in 90% ethanol extract, and 39.3% in absorbed fraction of Diaion HP-20 resin. In these results, we found that resin absorption method is suitable for the extraction of enriched flavonoids from Scutellariae Radix.
Hydrocarbons have been successfully used as a chemical marker in order to identify irradiated from non-irradiated foods. The method for determining hydrocarbons consists of extraction of fats, followed by separation of hydrocarbons by florisil column chromatography, and then identification of hydrocarbons by GC/MS. Currently, solvent extraction method for fats has certain limitations with regard to extraction time and solvent consumption. Commercial hams and sausage were irradiated at 0 and 5 kGy, and the efficiency of microwave-assisted extraction (MAE) and conventional solvent extraction (CSE) methods on the extraction of radiation-induced hydrocarbons from the meat products was compared. Significant levels of hydrocarbons, mainly composed of 1,7-hexadecadien, 1,7,10-hexadecatriene, and 6,9-heptadecadiene, were detected in the extracts from irradiated hams and sausages by both CSE and MAE methods. Both methods were acceptable in extracting hydrocarbons from samples, but MAE method required apparently reduced amounts of solvent from 150 (CSE) to 50 mL and reduced extraction time from 23 (CSE) to 5 min.
Curcuma caesia Roxb. (Zingiberaceae) is commonly known as 'Black turmeric'. In India it grows in West Bengal, Madhya Pradesh, Orissa, Bihar, North-East and Uttar Pradesh and is widely used by ethnic communities for various ailments. Rhizomes of the plant are used for sprains and bruises and are also employed in cosmetics. In West Bengal it is an important place in traditional system of medicine and is also used as a substitute for turmeric in fresh stage. Present communication deals with the detailed pharmacognostical evaluation of the rhizome sample. Inner part of the rhizome is bluish-black in colour and emits a characteristic sweet smell, due to the presence of essential oil. On steam distillation the rhizome yields an essential oil rich in camphor. A detailed HPTLC studies has been carried out for quantitative evaluation of active marker component. HPTLC, physico-chemical, morphological and histological parameters presented in this paper may be proposed as parameters to establish the authenticity of C. caesia rhizome and may possibly help to differentiate the drug from its other allied species.
The purpose of this study was to develop a cocktail approach for the measurement of the permeability of marker compounds, caffeine and propranolol (high permeability), ofloxacin (intermediate), atenolol (low), and quinidine (P-glycoprotein substrate), simultaneously. Then we compared the permeability in Caco-2 cells with that in rat intestinal segments. The difference between individual measurement and cocktail approach was less than 20 %, and the permeabilities of these compounds were similar to those previously reported, suggesting that the cocktail transport study and simultaneous drug analysis were successfully developed and validated in this study. Additionally, in the application of this cocktail method, the permeability of five drugs in rat jejunum was similar to that in ileum but different from that in colon, which was measured using the Ussing chamber system. Moreover, permeability in jejunum and ileum was similar to that in Caco-2 cells. In conclusion, the permeability in Caco-2 cells was equivalent to the permeability in rat jejunum and ileum determined with the Ussing system. Therefore, this newly developed cocktail assay and its application to the Ussing system can be a useful tool for robust and rapid screening for site-specific permeability in rat intestine, thus accelerating the prediction of absorption of new chemical entities.
