The impact of methanol extract of Chaenomeles sinensis (C. sinensis) leaves on acid corrosion of low carbon steel was assessed by gravimetric and electrochemical methods. Phytochemical characterization by total phenolic content (TPC), and total flavonoids content (TFC) of the extract was performed. The TPC and TFC concentrations were identified as 193.50 and 40.55 mg/g. Efficiency increased remarkably in the presence of inhibitor and found as concentration dependent. A maximum inhibition efficiency of 93.19% was achieved using 2000 ppm of the C. sinensis inhibitor. Impedance and surface morphology analysis by SEM and AFM revealed that the anticorrosive activity results from the protective film of phytochemical components of C. sinensis extract adsorbed on the metal surface.
The possible mechanism of the DNA strand breaking activity of the hot-water extract of Fructus Chaenomelis (dried fruit of Chaenomeles sinensis) in a closed circular duplex replica form DNA (RFI DNA) was studied through agarose gel electrophresis under various conditions. Induction of DNA strand scission by the hot-water extract of C. sinensis occurred in dose and time-dependent manners. $Cu^{2+}$ was indispensable for the induction of DNA strand breakage. Exogeneous chelating agents inhibited the DNA breaking activity, conforming the catalytic action of $Cu^{2+}$ on generation of free radicals responsible for oxidative damage. Antioxidant enzymes and some radical scavengers were used to investigate the major radical species triggering the DNA strand scission, demonstrating that a highest inhibitory activity was found in the presence of catalase, while less in the presence of tiron (a scavenger for superoxide radical), 2-aminoethyl-isothiuroniumbromide-HBr, cysteamine (scavengers for hydroxyl radical), and 1,4-diazabicyclo [2,2,2] octane (a scavenger for singlet oxygen) in decreasing order. The findings implied that oxygen radical species generated in presence of transition divalent cation during the oxidation of some compounds contained in the hot-water extract of C. sinensis is mainly responsible for inducing genotoxicity.
Chaenomeles sinensis Koehne (Rosaceae) is a deciduous tree and is distributed in China, Korea and Japan. In previous studies on the fruits of C. sinensis, some triterpenoid compouds such as oleanolic acid, tormentic acid were reported. In an ongoing investigation into biologically active compounds from natural products, the methanol extract of the stems of C. sinensis was investigated. By means of the repeated column chromatography using silica gel, Sephadex LH-20, LiChroprep RP-18, betulin (1), tormentic acid (2), 1-${\beta}$-D-glucopyranosyl-3,4,5-trimethoxybenzene (3), lyoniresinol-2a-O-${\alpha}$-L-rhamnopyranoside (4) were isolated. The chemical structures of compounds 1-4 were determined by the basis of physico-chemical properties and spectroscopic methods such as 1D and 2D NMR. For the isolated compounds (1-4), the inhibitory activity of IL-6 production in TNF-${\alpha}$ stimulated MG-63 cell was examined. Among the isolates, betulin (1), 1-${\beta}$-D-glucopyranosyl-3,4,5-trimethoxybenzene (3), lyoniresinol-2a-O-${\alpha}$-L-rhamnopyranoside (4) showed inhibitory effects on IL-6 production in TNF-${\alpha}$ stimulated MG-63 cell.
Objectives The purpose of this study was to investigate the antiallergic and antioxidative effects of Chaenomeles Sinensis (CS). Methods CS pretreatments inhibited anti-DNP IgE in RBL-2H3 mast cells for an hour. we measured cell viability, $\beta$-hexosaminidase release, IL-4, TNF-$\alpha$ secretion, and IL-4, TNF-$\alpha$ mRNA expression CS pretreatments inhibited DNP-HSA($10\;{\mu}g/ml$) for ten minutes, we measured Dicholrodihydrofluorescein(DCF) and DPPH radical-scavenging activity in 0.5 mM 1,1-diphenyl-2-picrydrazyl(DPPH) radical solution, 0.1ml, 99% ethanol 0.8ml, and CS 0.1 ml mixed solution. Results 0, 1, 2, 3, 4, 5 mg/ml CS treatments were not affect on cell viability and inhibited b-hexosaminidase release, IL-4, TNF-$\alpha$ secretion, CS treatments also decreased IL-4, TNF-$\alpha$ mRNA expression in RBL-2H3 cells. CS treatments inhibited reactive oxygen species(ROS) and DPPH radical-scavenging activity. Conclusions These results suggest that CS may be useful for the prevention or treatment of allergic disease.

Two 2H-1-benzopyran derivatives, methyl 8-hydroxy-2,2-dimethyl-2H-1-benzopyran-5-carboxylate (1) and methyl 8-hydroxy-2,2-dimethyl-2H-1-benzopyran-6-carboxylate (2), including a new compound (1) were isolated from the twigs of Chaenomeles sinensis. Their chemical structures were characterized based on analysis of NMR data including 1H and 13C, COSY, HSQC, and HMBC and HRMS data. The isolated compounds (1 and 2) were assessed for their anti-neuroinflammatory activity by measuring inhibition levels of nitric oxide (NO) production in lipopolysaccharide (LPS)-activated BV-2 cells and for their neurotrophic activity by the secretion of nerve growth factor (NGF) in C6 cells. Compounds 1 and 2 exhibited powerful anti-neuroinflammatory effects with IC50 values of 17.14 and 19.30 μM, respectively, without cell toxicity, and also showed moderate effects on the stimulation of NGF secretion levels with 113.15 ± 3.54 and 130.20 ± 8.03%, respectively. The biosynthetic pathway of 1 and 2 was proposed that they would be derived from a protocatechuic acid and an isoprenyl unit.

Three acidic triterpense were isolated from the ethyl acetate soluble fraction of the fruits of Chenomeles sinensis (Rosaceae) together with previously reported oleanolic acid and ursolic acid by repeated silica gel colunm chromatography. Their structures have been elucidated as $2{\alpha},{\;}3{\betha}-dihydroxyolean-l2-en-28-oic$ acid(l. maslinic acid), $2{\alpha},{\;}3{\alpha},{\;}19{\alpha}-trihydroxy-urs-12-en-28-oic$ acid (2, euscaphic acid), and $2{\alpha},{\;}3{\betha}-dihydroxyolean-l2-en-28-oic$ acid (3,tormentic acid), respectively, on the basis of chemical and spectral evidence.
Matrix metalloproteinase-9 (MMP-9) is considered to be an important component in the progression of inflammation. Monocytes/macrophages are prominent at inflammation sites, and activation of these cells by stimulants such as lipopolysaccharide (LPS) leads to the production of significant amounts of MMP-9. Here, we show that LPS-induced MMP-9 production and activation was inhibited by the water extract from the fruit of Chaenomeles sinensis (CS), the root of Polygonum cuspidatum (PC), but increased by the extract from Boswellia carterii (BC). To investigate the mechanism by which those extracts inhibits MMP-9 activation, we examined the level of MMP-9 mRNA expression. We observed a significant change in the MMP-9 expression between LPS alone and LPS plus Chaenomeles sinensis and Polygonum cuspidatum extracts-treated cells. In addition, LPS significantly up-regulated MMP-9 promoter activity in Raw 264.7 cells, which was attenuated by the CS and PS extracts. However, water extracts from Boswellia carterii increased MMP-9 expression and MMP-9 promoter activity which were induced by LPS treatment in Raw 264.7 cells. These data suggest that water extracts from Chaenomeles sinensis and Polygonum cuspidatum can modulate anti-inflammatory immune response, which may be in part associated with the regulation of MMP-9 production and/or activation through the regulation of MMP-9 expression in mouse macrophage cells.
장미과 식물(에탄을 추출)의 항산화력을 측정하기 위하여 1,1-diphenyl-2-picrylhydrazyl (DPPH) 자유 라디칼 생성 시스템을 이용하여 하였다. 또한 산화 환원 과정을 통하여 피부의 과잉 색소 생성에 관련된 멜라닌 생합성 과정의 조절 가능성을 조사하였다. 장미과 식물중에서 Prunus sargentii, Rubus coreanus, Chaenomeles sinensis, Photinia glabra와 Pyrus pyrifolia은 생합성 과정에서 dopa [3-(3,4-dihydroxyphenyl) alanine]를 도파크롬으로 변환시켜 주는 tyrosinase 저해 효과를 나타내었다. MTT 실험법은 장미과 에탄올 추출물의 사람 섬유아세포에 미치는 독성정도를 실험하기 위해 사용되었다. 장미과 식물중 특히 Prunus sargentii의 껍질, Phorinia glabra의 껍질과 나무 그리고 Chaenomeles sinenis의 잎, 껍질, 나무 모든 부분에서 mushroom tyrosinase에 대해 $100{\;}{\mu}g/mL$에서 50\%$ 이상의 저해 활성을 보였으며, $10{\;}{\mu}g/mL$ 농도에서 강한 라디칼 소거 효과를 나타내었다. 또한 이들 추출물은 사람 섬유아 세포에 대해 높은 생존율을 나타냄으로써 멜라닌 형성 과정을 조절할 수 있을 것으로 기대한다.
Objectives : This study was conducted to compare Chaenomelis Sinensis Fructus with Chaenomelis Lagenariae Fructus, and to examine Fructus chaenomelis having an influence on Intestinal Motility. Methods : We compared Chaenomelis Sinensis Fructus with Chaenomelis Lagenariae Fructus, by observing their effects on Intestinal Motility. Oral administration of water extracts of Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus into albino rats was followed by dealing with carbachol or loperamide, injecting charcoal meal and measuring the moving length in the intestine. Results : By Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus, the Intestinal Motility of normal albino rats did not change significantly. Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus controled the accelerated Intestinal Motility of albino rats. Chaenomelis Sinensis Fructus was not different from the extracting method, Chaenomelis Lagenariae Fructus had effects of control on extracting by methanol significantly. When Chaenomelis Sinensis Fructus was compared with Chaenomelis Lagenariae Fructus, the former was proved to have control effects on Intestinal Motility in decoction extracts and extracts by ethyl ether higher than the latter. By both Chaenomelis Sinensis Fructus and Chaenomelis Lagenariae Fructus, the declined Intestinal Motility was not changed significantly. Conclusion : Chaenomelis Fructus suppressed the exasperated Intestinal Motility by carbachol, but did not influence the dropped Intestinal Motility by loperamide. In addition, Chaenomelis Sinensis Fructus was more excellent than Chaenomelis Lagenariae Fructus in its control effects of Intestinal Motility. inhibitory effects by DKT represent a potential therapeutic approach to the treatment of inflammatory diseases.
