Effects of Excess Vitamin E and Cholesterol Supplements on Levels of Vitamin E and Cholesterol in Rat Serum and Liver (과량의 비타민 E 및 Cholesterol 첨가식이가 흰쥐의 혈청과 간의 비타민 E 및 Cholesterol 농도에 미치는 영향)

  • The present study was undertaken to investigate the effects of excess vitamin E supple-mentation (0.5%) and cholesterol (0.5%) on levels of serum and liver vitamin E and cholesterol in two categories of rats, group A and group B. Rats in group A(45-65g) were fed experimental diets for 3 weeks ( I-C, II-E, III-Ch, IV-ECh). On the other hand rats in group B(45g-65g) were first fed control diet for 3 weeks and then fed experimental diets when they were 100-l20g for the subsequent 3 weeks ( I'-C, II'-E, III'-Ch, & IV'-ECh). The levels of serum vitamin E were higher in vitamin supplemented groups as expected. Dietary cholesterol showed a tendency to lower serum vitamin E levels of vitamin E supplemented groups. Serum cholesterol levels tended to stay in a narrow range showing resistancy to dietary cholesterol and were not affected by vitamin E status. Whereas vitamin E supplementation seemed to lower the levels of hepatic cholesterol in both groups A & B( I-C, I'-C vs II-E, II'-E ), simultaneous supplementation of vitamin E with 0.5% cholesterol appeared to increase further the hepatic cholesterol levels which were already increased by cholesterol feeding ( IV-ECh, IV'-ECh, vs III- Ch, III'-Ch). Hepatic vitamin A levels decreased as rats grew older during the experimental period. Even though vitamin A levels did not differ from each other significantly, excess amount of vitamin E supplementation in group B seemed to show a tendency to decrease the vitamin A storage in liver.

  • In this study, cholesterol sensors consisting of a mixture of cholesterol oxidase (ChOx) and zinc oxide (ZnO) nanoparticles (NPs) are constructed on plastic substrates and their sensing characteristics are examined in air. The current of the ChOx-ZnO NP film decreases in magnitude as cholesterol molecules are adsorbed on the film, due to the resulting increase in the number of electrons generated by the reaction between the cholesterol and the ChOx. The cholesterol sensor shows a high sensitivity of $1.08{\mu}A/mM$ and a wide detection range from 10 nM to 1 mM.

Effects of Protein Hydrolysates on Blood and Liver Lipids in Rats fed Fat-enriched Diet (고지방식을 섭취한 흰쥐의 체내지질함량에 대한 단백질 가수분해물의 섭취 효과)

  • The experiments were performed to investigate the effects of protein and protein hydrolysates on lipid metabolism in the hyperlipidemic/hypercholesterolemic rat model induced by feeding fat-enriched diet. In Except 1 male rats were fed four semi-purified high fat and cholesterol diets that contained different nitrogen source, casein(C), casein hydrolysate(CH), corn gluten(G) and corn gluten hydrolysate(GH), for 6 weeks. In Expt. 2 rats were fed high fat and cholesterol diet for 4 weeks to induce hyperlipidemia and hypercholesterolemia. Then the rats were divided into 4 groups and were fed the four kinds of above experimental diets for 4 weeks consecutively. The contents of total lipid , cholesterol and triglyceride in blood, liver and feces were determined. Serum lipid concentrations of CH, G and GH were significantly lower than that of C. Serum cholesterol concentrations of hydrolysate groups(CH and GH) were significantly lower than those of intact protein groups(C and G). Serum HDL -cholesterol concentration tended to increase by hydrolysate intake. The total lipid, cholesterol contents in liver showed similarity results as above. Fecal lipid excretions of CH, G, and GH groups were significantly higher than that of C group. These results indicate that hypolipidemic and /or hypocholesterolemic effect of corn gluten or protein hydrolysates were detected in the process of inducing hyperlipidemia by high-fat and cholesterol diet or after inducing hyperlipidemia.

Effect of Citrus Pectin Oligosaccharide Prepared by Irradiation on High Cholesterol Diet B6.KOR-ApoE Mice

  • Effect of citrus pectin oligosaccharides produced by irradiation was studied on the ability to improve lipid metabolism and hypercholesterolemia in mice fed high cholesterol diets. A total of 35 mice were divided into 5 groups and fed the following diets for 6 weeks: normal diet (C), 0.5% cholesterol (CH), 0.5% cholesterol+5% non-irradiated pectin (P), 0.5% cholesterol+5% irradiated pectin at 20 kGy (PIR), and 0.5% cholesterol+5% irradiated at 20 kGy and dialyzed (PIR-F). CH group had significantly higher serum triglycerides, total cholesterol, and low density lipoprotein (LDL)-cholesterol contents than pectin oligosaccharide-treated groups (p<0.05). Triglycerides and total cholesterol contents was the lowest in C and PIR-F and followed by PIR and P group, and CH group had significantly higher LDL-cholesterol. Serum high density lipoprotein (HDL)-cholesterol content in C group was not different from that in CH and P groups, but lower than that of PIR and PIR-F groups. These results suggest that pectin oligosaccharides produced by irradiation can reduce the levels of serum triglyceride, total cholesterol, and LDL-cholesterol in the blood of mice fed high-cholesterol diets and therefore, irradiation can be used as a tool to produce functional oligosaccharides from citrus pectin.

Effect of Surface Hydrophobicity of Soybean Peptides on the Concentration of Serum Cholesterol and Fecal Steroid Excretion in Rats (대두 펩타이드의 표면소수도가 흰쥐의 혈청 콜레스테롤 농도 및 분변 스테로이드의 배설량에 미치는 영향)

  • Effect of surface hydrophobicity of soybean peptides on serum cholesterol in rats was investigated. Soybean protein(ISP), casein(CNP), and their peptic hydrolyzates fractionated by acid precipitations (SHT, SH8, SH6, SH4, CHT, CH6, CH5, CH4) were fed to rats and the concentration of serum cholesterol and the fecal steroid excretion were measured. And surface hydrophobicities of the peptide fractions were measured by determining by the ANS flourescence intensity and SDS binding capacity. It was found that the higher the surface hydrophobicity of peptides was, the more the fecal steroids excreted(r=0.801) and the lower the concentration of serum cholesterol became(r=-0.868). However, there was no relationship between SDS surface hydrophobicity and fecal steroids or serum cholesterol. ANS surface hydrophobicity of soybean protein was increased by enzymatic hydrolysis. These results suggest that high surface hydrophobicity of peptides formed during digestion is responsible for the hypocholestrolemic effect of soybean protein through the hydrophobic interaction between the peptides and bile salts in rats.

Effects of Kimchi Solvent Fractions on Accumulation of Lipids in Heart, Kidney and Lung of Rabbit Fed High Cholesterol Diet (김치 용매획분이 고 콜레스테롤 식이를 섭취한 토끼의 심장, 신장 및 폐의 지질 축적에 미치는 영향)

  • The effects of dichloromethane (CH$_2$C1$_2$), ethylacetate (EtOAc) or water ($H_2O$) fraction of Korean cabbage kimhi on accumulation of lipids in the heart, kidney and lung of rabbit fed 1% cholesterol diet for 16 weeks were studied. The amount of kimhi fraction added to the 100 g of diet was 8.3 mg of CHaC12.5.6 W of EtOAc, and 221.9 mg of $H_2O$, which are equivalent to 5% of freeze-dried kimhi added to the diet. Each group had 6 rabbits and rabbit was housed individually. Cholesterol and triglyceride concentrations of three organs were noticeably decreased due to these solvent fractions of kimhi but for phospholipid and total lipids, only CH$_2$C1$_2$ fraction group showed decreasing effects. In the heart, compared to the control, cholesterol concentrations for CE$_2$C1$_2$, EtOAc, and H2O fraction group were decreased by 42 (P<0.05),21 (P<0.05), and 8%, respectively, and triglyceride for these groups were decreased by 29 (p<0.05),4, and 11%, respectively. In kidney, cholesterol concentration for CH$_2$C1$_2$, EtOAc, and H2O fraction group were decreased by 23, 12, and 11%, respectively (p<0.05) and triglyceride concentration for CH$_2$Cl$_2$ and H2O fraction groups were significantly decreased by 51 and 21%, respectively (p<0.05). In lung, cholesterol concentrations for CItCIB, EtOAc, and H2O fraction groups were decreased by 37,20, and 22%, respectively (p<0.05) and triglyceride concentration of these groups were significantly decreased by 39, 28, and 28%, respectively (p<0.05). And phospholipid and total lipid of CH$_2$CI$_2$ fraction group were significantly decreased by 33 and 34% respectively (p<0.05). Among three organs the cholesterol content of lung was the highest followed kidney and heart. For triglyceride, heart, kidney and lung is in the order of showing the highest concentration. The phospholipid concentration was not significantly different among three organs. According to these results, we may conclude that CH$_2$C1$_2$, fraction of kimhi might have the most active component, which decreases cholesterol, triglyceride, phospholipid and total lipids concentrations in heart, kidney, and lung of rabbit fed high cholesterol diet.

Chromatic Detection of Cholesterol Using Polydiacetylene Vesicles

  • In this study, a new system to determine the concentration of cholesterol using a color change was developed. The system comprised diacetylene vesicles and cholesterol oxidase (ChOx). 10,12-Pentacosadiynoic acid (PCDA) was used as the diacetylene compound, and PCDA vesicles were formed using sonication. The H2O2 produced during the reaction between cholesterol and ChOx was used to initiate the polymerization of the PCDA in the vesicles. During polymerization, the vesicles changed from colorless to blue. Therefore, the cholesterol concentration was proportional to the intensity of the blue color. The absorption at 665 nm indicated that the blue color was directly proportional to the cholesterol concentration. This indicates that the system can be used for cholesterol detection. The minimum cholesterol concentration detected using this system was 1.0 mM.

Effect of Digestibility of Soybean Peptides on the Concentration of Serum Cholesterol in Rats (대두 펩타이드와 소화율이 흰쥐의 혈청 콜레스테롤 농도에 미치는 영향)

  • In order to investigate the hypocholesteremic effect of soybean perptides, soybean protein(ISP), casein(CNP), their peptic hydrolyzates fractionated by acid precipitation at different pH's(SHT, SH8, SH6, SH4, CHT, CH5, CH4) and amino acid mixtures of the same composition as the proteins(SAA, CAA) were fed to rats and the concentration of serum cholesterol was measured. Then in vitro digestibility and molecular weight distribution of the peptides by pepticpancreatic hydrolysis was measured by FPLC. The lower the in vitro digestibility of peptides is, the lower the concentration of serum cholesterol becomes(r=0.986) and the higher the ratio of macropeptides is, the lower the concentration of serum cholesterol becomes(r=-0.932) in rats. These results suggest that the in vitro digestibility of peptides has close relationship to the concentration of serum cholesterol in rats and non-digestible meacropeptides or polypeptides especially more than 1 kDa, formed through digestion in gut, may lower the serum cholesterol in rats.

  • The extracellular cholesterol oxidase produced from a soil microorganism HSL613 was purified and partially characterized. Through a series of purification procedures including concentration with CH2 concentrator, DEAE-cellulose column chromatography and gel filtration on Superose12, the purified enzyme was shown to have a specific activity of 108 units/mg protein giving 30.8-fold purification and final yield of 66%. The molecular weight of the enzyme was estimated to be 59,500 daltons by SDS-PAGE. The optimum temperature and pH for this enzyme were $50^{\circ}C$ and 6.0, respectively. The activity of the purified cholesterol oxidase was inhibited by $Ag^{2+}$, $Hg^{2+}$ and SDS.

  • Cholesteryl p-[3-(2-chloroethyl_-3-nitrosoureido] phenylthioaccetate (2) was synthesized : an intermediate, p-[3(2-chloroethyl_-3-nitrosoureido] phenylthioacetic acid (1) is a congener of an antitumor chlorambucil which both the -CH$_{2}$CH$_{2}$-linkage and -N(CH$_{2}$CH$_{2}$Cl)$_{2}$ group of chlorambucil molecule is doubly modified into the respective -S-linkage and -NH-CO-NNO-CH$_{2}$CH$_{2}$CL group. The attackment of cholesterol moiety as a carrier group to p-[3-(2-chloroethyl-3nitrosoureido] phenylthioacetic acid was accomplished through the esterification of cholesterol with p-[3-(2-chloroethyl-3-nitrosoureido] phenylthioacetyl chloride which was obtained from the treatment of p-[3-(2-chloroethyl)-3-nitrosourei-do] phenylthioaceticacid with SOCL$_{2}$, p-[3-(2-chloroethyl)ureido]]-phenythioacetic acid was nitrosated with NaNO$_{2}$ IN 98-100% HCOOH to give exclusively p-[3-(2-chloroethyl_-3-nitrosoureido] phenylthioacetic acid.
