The plastid transformation approach offers a number of unique advantages, including high-level transgene expression, multi-gene engineering, transgene containment, and a lack of gene silencing and position effects. The extension of plastid transformation technology to monocotyledonous cereal crops, including rice, bears great promise for the improvement of agronomic traits, and the efficient production of pharmaceutical or nutritional enhancement. Here, we report a promising step towards stable plastid transformation in rice. We produced fertile transplastomic rice plants and demonstrated transmission of the plastidexpressed green fluorescent protein (GFP) and aminoglycoside 3′-adenylyltransferase genes to the progeny of these plants. Transgenic chloroplasts were determined to have stably expressed the GFP, which was confirmed by both confocal microscopy and Western blot analyses. Although the produced rice plastid transformants were found to be heteroplastomic, and the transformation efficiency requires further improvement, this study has established a variety of parameters for the use of plastid transformation technology in cereal crops.
Near full length cDNA clones encoding the rice seed storage protein, prolamine, were isolated and divided into two homology classes based on cross-hybridization and DNA sequencing analysis. These cDNA clones contain a single open reading frame encoding a putative rice prolamine precursor(M.W.=17,200) possessing atypical 14 amino acid signal peptide. Clones of these two homology classes diverge mainly by insertions/deletions of short nucleotide stretches and point mutations. The deduced primary structures of both types of prolamine polypeptides are devoid of any major tandem repetitive sequences, a feature prevalent in other cereal prolamines. No significant homology teas detected between the rice prolamine and other cereal prolamines, indicating that the rice gene evolved from a different ancestor that gave rise to other cereal prolamine genes. Developing wheat and rice endosperms were examined using ultrathin sections prepared from tissues harvested at various days after flowering. By immunocytochemical localization techniques, wheat prolamines are localized within vesicles from Golgi apparatus and in homogeneous regions of protein bodies. The involvement of the goli apparatus in the packaging of wheat prolamines into protein bodies indicates a pathway which differs from the mode of other cereal prolamines and resembles the mechanism employed for the storage of rice glutelin and legume globulins.
Bacterial blight(BB) caused by Xanthomonas oryzae pv. oryzae(Xoo) is one of the most economically destructive bacterial diseases of rice in worldwide. Utilization of resistant cultivars carrying resistant gene(s) is relatively an effect method to control this disease. About 34 resistant genes for BB resistance have been identified in many countries. Among them, Xa1 and Xa3 genes against bacterial blight have been incorporated into improved korean japonica rice varieties. Now, Ilmi carrying Xa1 gene and severial cultivars carrying Xa3 gene are widely grown in our country. In recent year, xa5, Xa21 and Xa23 genes are using in rice breeding programs for japonica resistant cultivars to bacterial blight. Resistant cultivars incorporated with a diverse single gene and two, three, or the more major gene necessite in the future.

The purpose of this study was to investigate the changes in the content of functional components and the antioxidant activity of cooked rice and porridge composed of selected grains. The results of the study showed that brown rice and oat contained considerable amounts of γ-oryzanol (78.099~238.566 mg/100 g). However, cooked rice showed a decreased content of the same. The highest content of γ-aminobutyric acid (GABA) was observed in brown rice from Samgwang. The contents of biotin in all samples also decreased in cooked rice compared to raw cereal grains. The highest content of total polyphenols and flavonoids were observed in Heukjinmi rice, and the highest radical scavenging activity was also found in this variety. The cooking process led to a decrease in the content of functional components including γ-oryzanol, GABA, biotin, polyphenols, and flavonoids versus the corresponding raw cereal grains. These results may be useful in the development and application of home meal replacements using cereal grains.
This study was carried out to investigate the quality characteristics of commercial breakfast cereals. The results showed that as water absorption index (WAI) increased, water soluble index (WSI) decreased. WSI has an effect on Bowl life, so Bowl life became longer as WSI enhanced. The size, shape and texture of breakfast cereals were also affective factors of their Bowl life. The quality describing terms of breakfast cereals were surveyed and the terms were classified to brittleness, roasted nutty taste, sweetness, hardness and aftertaste, especially brittleness was the most important quality determining terms among these. Significantly, sweetness had positive correlationship with roasted nutty taste. Overall eating quality had positively correlationship with color and roasted nutty flavor, and negatively with pain in mouth, adhesiveness and additive taste.
In order to solve the shortage of roughage supply for dairy farm in Korea, winter cereal forage production after harvesting of rice in the fallow paddy field is studied. This study consist of two parts. One is the model development of the mechanized production which was already reported at the preview paper. This is the 2nd parts of the study. Also, the mechanized production model for cereal forage production in winter was reported in the previous study. In this paper, coverage area and mechanized wrap silge production cost are analyzed and compared to the other available feeds in Korea. Results of the research are summarized as follows; The coverage area for the winter cereal wrap silage production system in Korea is estimated to be 33.7 ha in case of working with a tractor and a set of implements. If two or three tractors are available, the coverage area is estimated to be 68.0 and 101.3 ha, respectively. The break even point (BEP) of the farming size is analyzed as 10 ha and its production(operating) cost is estimated to be 317 to 443 won/TDN-kg at the BEP point. The cost is lowered to 182 won/TDN-kg at 100ha-working, and is much lower compared with prices of imported feeds of 360∼600 won/TDN-kg. Therefore, winter cereal wrap silage model is judged to be feasible and desirable for a large scale production of forage in winter fallow paddy field.
The effects of cereal (rice, brown rice, barley, glutinous rice) ethanol extracts on the proliferation, nitric oxide (NO) production, and level of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in B and T lymphocytes, isolated from human blood cells of sasang constitution, were examined in vitro. The rice, brown rice, and glutinous rice ethanol extract, which are known as suitable for eumin (taeumin and soeumin) constitutions, showed the highest activities for proliferation, NO production, and TNF-${\alpha}$ concentration, respectively. However, the barley ethanol extract, which is known tobe suitable for yangin (taeyangin and soyangin) constitutions, showed the highest overall activities for proliferation, NO production, and TNF-${\alpha}$ concentration. The different immunological activities of the cereal extracts in sasang constitution lymphocytes might be due to their individual components. Thus, determination of components from the cereal extracts suitable for sasang constitution could be useful in new food developments.
Fusarium head blight (FHB; scab) caused mainly by Fusarium graminearum is a devastating disease of wheat and barley around the world. FHB causes yield reductions and contamination of grain with trichothecene mycotoxins such as deoxynivalenol (DON) which are a major health concern for humans and animals. The objective of this research was to develop an easy seed or seedling inoculation assay, and to compare these assays with whole plant resistance of twenty-nine Korean winter wheat cultivars to FHB. The clip-dipping assay consists of cutting off the coleoptiles apex, dipping the coleoptiles apex in conidial suspension, covering in plastic bag for 3 days, and measuring the lengths of lesions 7 days after inoculation. There were significant cultivar differences after inoculation with F. graminearum in seedling relative to the controls. Correlation coefficients between the lesion lengths of clip-dipping inoculation and FHB Type II resistance from adult plants were significant (r=0.45; P<0.05). Results from two other seedling inoculation methods, spraying and pin-point inoculation, were not correlated with adult FHB resistance. Single linear correlation was not significant between seed germination assays (soaking and soak-dry) and FHB resistance (Type I and Type II), respectively. These results showed that clip-dipping inoculation method using F. graminearum may offer a real possibility of simple, rapid, and reliable for the early screening of FHB resistance in wheat.
Kjeldahl method used in many materials from various plant parts to determine protein contents, is laborious and time-consuming and utilizes hazardous chemicals. Near-infrared (NIR) reflectance spectroscopy, a rapid and environmentally benign technique, was investigated as a potential method for the prediction of protein content. Near-infrared reflectance spectra(1100-2400 nm) of coarse cereal grains(n=100 for each germplasm) were obtained using a dispersive spectrometer as both of grain itself and flour ground, and total protein contents determined according to Kjeldahl method. Using multivariate analysis, a modified partial least-squares model was developed for prediction of protein contents. The model had a multiple coefficient of determination of 0.99, 0.99, 0.99, 0.96 and 0.99 for foxtail millet, sorghum, millet, adzuki bean and mung bean germplasm, respectively. The model was tested with independent validation samples (n=10 for each germplasm). All samples were predicted with the coefficient of determination of 0.99, 0.99, 0.99, 0.91 and 0.99 for foxtail millet, sorghum, millet, adzuki bean and mung bean germplasm, respectively. The results indicate that NIR reflectance spectroscopy is an accurate and efficient tool for determining protein content of diverse coarse cereal germplasm for nutrition labeling of nutritional value. On the other hands appropriate condition of cereal material to predict protein using NIR was flour condition of grains.
This study evaluated the weight reduction effect of Angook cereal mixture. The cereal mixture was prepared with barley, unpolished rice, corn, soybean, Garcinia cambogia, guar-gum, maltodextrin, glucomannan and a vitamin mixture. Eighteen female college students participated in this 8 weeks weight control program. All subjects were randomly assigned to the treatment and placebo groups. Mean energy intake of the treatment group was $1,356.4{\pm}79.9\;kcal$ (carbohydrate: 67.1%, protein: 18.7%, fat: 14.2%) and placebo group consumed $1,367.6{\pm}71.8\;kcal$ (carbohydrate: 64.2%, protein: 19.7%, fat: 16.1%) during program. The placebo group lost $3.9{\pm}0.8\;kg$ of body weight and the treatment group lost $5.9{\pm}0.7\;kg$ of body weight. There were significant differences in the decrease of total body weight, absolute fat mass, waist circumference (WC) and hip circumference (HC) between the two groups (p<0.05), however, the lean body mass was not significantly decreased in the treatment group compared to the placebo group. There were no differences in the changes in blood glucose, total-cholesterol, HDL-choloesterol, LDL-cholesterol and triglycerides between groups. These findings suggest that the intake of Angook cereal mixture may be beneficial for the reduction of the body weight, absolute fat mass, WC and HC.
