Inclusion of Cephalotaxus in Taxaceae: Evidence from morphology and anatomy

  • The inconsistent relationship between the monogeneric family Cephalotaxaceae and Taxaceae was discussed and the possibility of merging Cephalotaxus within Taxaceae was also reviewed. Our previous reports (cladistics analysis, leaf anatomy and wood anatomy of Taxaceae s.l.) did not find a feasible reason to create a distinction between Cephalotaxus and other Taxad genera (Taxus, Pseudotaxus, Amentotaxus, Torreya and, Austrotaxus) and thus argued for a broader concept of Taxaceae with Cephalotaxus. The monophyly of Taxaceae including Cephalotaxus is described in various contemporary molecular studies, and some of them are in support of the single large family Taxaceae with six genera. Although additional comprehensive studies in the future may perhaps weaken the precise association between Cephalotaxaceae and other Taxad genera, on the basis of recent corroborations, at this moment Taxaceae should be redefined with broad circumscriptions, including Cephalotaxus.

  • Four biflavonoids were isolated from the leaves of Cephalotaxus koreana. Based on spectroscopic data, the compounds were identified as amentoflavone-7',7",4',4"'-tetramethyl ether, sciadopitysin, ginkgetin and bilobetin, respectively.

  • Cephalotaxus koreana Nakai is endemic species of Korea. Since old days, the fruits have been used as a parasiticide and leaves have been used for a pierced wound by insects. The essential oil components from its aerial parts and stem bark by steam distillation were investigated. Several compounds were characterized by GC-Mass spectra. It was found that the leaves, stembark and flowers contain the compounds of essential oils, 1-octene-3-ol, hexadecanoic acid; $\alpha$-pinene, $\Delta^3$-carene mainly, linalylacetate, $\beta$-cubebene, 3,4-octadine-7-methyl , ferruginol(stem bark) and $\alpha$-pinene mainly, $\beta$-pinene , cyclopropane-1,1-dimethyl-2-(3-methyl-1,3-butadienyl), etc. This Cephalotaxus spp. contains the first components of ferruginol(M.W.286.03, $C_{20}H_{30}O)$ which belong to diterpene.

Analysis of Alkaloid Contents in Korean Plumyew [Cephalotaxus Koreana]: Variation with Location and Season (한국산 개비자 나무 [Cephalotaxus Koreana]의 알칼로이드 함량분석: 지역 및 계절에 따른 변화)

  • The contents and composition of alkaloids homoharringtonine, harringtonine, and cephalotaxine were determined in Korean native plumyew tree (Cephalotaxus koreana) collected from 17 different sites in Korea. The alkaloid contents of C. koreana varied with locations and plant populations from 11.8 mg/g to 195.2 mg/g of dry weight while C. koreana in southern area had higher alkaloid contents. The alkaloid compositions in C. koreana were highest with homoharringtonine (40%) and followed harringtonine (32%) and cephalotaxine (28%). Total alkaloids and homoharringtonine contents in needles were higher than those of stem by 2 fold. Alkaloid contents of C. koreana were the highest in winter, and the lowest in summer. Comparison of alkaloid contents and composition with other species of Cephalotaxus revealed that C. koreana was different to C. harringtonia, and contents of homoharringtonine were higher than other species. C. koreana could be potential source for homoharringtonine and related alkaloids.

Identification and Characterization of Homoharringtonine from Cephalotaxus koreana

  • A novel purification method was developed for producing homoharringtonine from Cephalotaxus koreana, to guarantee high purity and yield. Our simple, efficient procedure for isolating and purifying homoharringtonine from C. koreanabiomass consisted of solvent extraction, synthetic adsorbent treatment, low-pressure chromatography, followed by high performance liquid chromatography (HPLC). The use of active clay treatment and silica gel low-pressure chromatography in the pre-purification process allowed for the rapid, efficient separation of homoharringtonine from interfering compounds and dramatically increased the yield and purity of crude homoharringtonine for high-performance liquid chromatography (HPLC) purification steps compared with alternative processes. Homoharringtonine could be obtained simply with high yield and purity from biomass using this purification method, while minimizing solvent use and the scale and complexity of HPLC operations for homoharringtonine purification. Purified homoharringtonine was identified and characterized.

Modifier Effects on Supercritical $CO_2$ Extraction Efficiency of Cephalotaxine from Cephalotaxus wilsoniana Leaves

  • The effects of modifiers such as methanol, water diethylamine in methanol (10 v/v %), and diethylamine in water (10 v/v %) were investigated at three different concentrations (1, 5, and 10 v/v %) of the modifiers in supercritical $CO_2$ (SC-$CO_2$) in order to enhance the supercritical fluid extraction (SFE) efficiency of cephalotaxine from Cephalotaxus wilsoniana Leaves. Among the modifiers employed, methanol basified with diethylamine was found to greatly enhance the extraction efficiency relative to any other modifiers employed. The results suggest that cephalotaxine in plant matrices may be readily changed from SC-$CO_2$-insoluble salt to SC-$CO_2$-soluble free base by basified modifiers. In addition, SC-$CO_2$modified with basified methanol could enhance the extraction efficiency of cephalotaxine more than 30% when compared to the conventional organic solvent extraction.

  • An effective purification method was developed for producing Homoharringtonine (HHT), to guarantee high purity and yield from Cephalotaxus koreana. This process was a simple and efficient procedure, for the isolation and purification of HHT form the biomass of Cephalotaxus koreana, consisting of extract, adsorbent treatment, precipitation and followed by a chromatography. The extraction, adsorbent treatment and precipitation in pre-purification process allows for rapid and efficient separation of HHT from many compound and dramatically increases the yield and purity of crude HHT for HPLC purification steps compared to alternative processes. This purification processes serves to minimize solvent usage, size, and complexity of the operations for HHT purification.

  • A new isolation and purification method was developed aiming at increasing yield and purity for homoharringtonine. This method was a simple andefficient procedures, for the isolation and purification of homoharringtonine from Cephalotaxus koreana, consisting of solvent extraction, adsorbent treatment, low-pressure chromatography, and high performance liquid chromatography (HPLC). The crude homoharringtonine was efficiently pre-purified adequately to perform HPLC through a combination withadsorbent treatment and low-pressure chromatogaphy. The homoharringtonine can be simply obtained with high purity and yield from crude homoharringtonine by HPLC. Purified homoharringtonine was characterized.

  • Cephalotaxus koreana Nakai is an endemic species in Korea. The EtOH extract of leaf and branch from the plant showed potent antitumor activity in Teruhiro's method. The tumor volume inhibition ratio value is 25.2% with 20mg/kg in the BDF1 mouse injected LLC cell. We isolated one flavone, sciadopitysin (1), two flavone O-glycosides, quercetin 3-O-${\beta}$-D-glucuronide 6"-ethyl ester (2), apigenin 7-neohesperidoside (3) in comparison with literatures data. Compounds 1-3 showed stronger antitumor activity than Taxol used as positive control. The inhibition ratio values of compounds 1-3 is 34.9, 31.6, 34.0%, respectively, and Taxol is 27.0 % compared with control group.
