Studies on Biological Activity of Wood Extractives (XI) - Compounds from Heartwood of Taxus cuspidata and Their Antioxdative Activities - (수목추출물의 생리활성에 관한 연구(XI) - 주목(Taxus cuspidata) 심재 추출성분 및 항산화활성 -)

  • Antimicrobial and antioxidative activities of heartwood extractives of domestic species were investigated to develop a natural fungicide or preservative. Four lignan derivatives and one taxane were isolated from heartwood of Taxus cuspidata which has been selected due to its high antioxidative activity among the tested species. The chemical structures were identified as : taxusin, isolariciresinol (4, 4', 9, 9'-tetrahydroxy-3', 5-dimethoxy-2, 7'-cyclolignan), lariciresinol (4, 4', 9-trihydroxy-3, 3'-dimethoxy-7, 9'-epoxylignan), taxiresinol (3, 4, 4', 9-tetrahydroxy-3'-methoxy-7, 9'-epoxylignan) and isotaxiresinol (3', 4, 4', 9, 9'-pentahydroxy- 5-methoxy-2, 7'-cyclolignan) on the basis of spectroscopic data and their chemical correlations. According to the results of free radical scavenging activity, isolariciresinol, lariciresinol and isotaxiresinol showed higher radical scavenging activity than those of 𝛼-tocopherol and butylated hydroxytoluene (BHT), the strongest natural and synthetic antioxidants. However, taxusin did not show any free radical scavenging activity. In this regard, it could inferred that high antioxidative activity of extractives of T. cuspidata was derived from isolariciresinol, lariciresinol and isotaxiresinol.

Evaluation of phlorofucofuroeckol-A isolated from Ecklonia cava (Phaeophyta) on anti-lipid peroxidation in vitro and in vivo

  • Lipid peroxidation means the oxidative degradation of lipids. The process from the cell membrane lipids in an organism is generated by free radicals, and result in cell damage. Phlorotannins, well-known marine brown algal polyphenols, have been utilized in functional food supplements as well as in medicine supplements to serve a variety of purposes. In this study, we assessed the potential anti-lipid peroxidation activity of phlorofucofuroeckol-A (PFF-A), one of the phlorotannins, isolated from Ecklonia cava by centrifugal partition chromatography in 2,2-azobis (2-amidinopropane) dihydrochloride (AAPH)-stimulated Vero cells and zebrafish system. PFF-A showed the strongest scavenging activity against alkyl radicals of all other reactive oxygen species (ROS) and exhibited a strong protective effect against ROS and a significantly strong inhibited of malondialdehyde in AAPH-stimulated Vero cells. The apoptotic bodies and pro-apoptotic proteins Bax and caspase-3, which were induced by AAPH, were strongly inhibited by PFF-A in a dose-dependent manner and expression of Bcl-xL, an anti-apoptotic protein, was induced. In the AAPH-stimulated zebrafish model, additionally PFF-A significantly inhibited ROS and cell death, as well as exhibited a strong protective effect against lipid peroxidation. Therefore, these results suggest that PFF-A has excellent protective effects against ROS and lipid peroxidation induced by AAPH in both an in vitro Vero cell model and an in vivo zebrafish model.

Isolation and Purification of Antibacterial Components in Cortex Phellodendri (황백나무로부터 항균성분의 분리 및 정제)

  • Cortex Phellodendri (CP) is derived from the dried bark of Phellodendron amurense. It has been widely used as a drug in traditional Korea medicine for treating diarrhea, jaundice, swelling pains in the knees and feet, urinary tract infections and infections of the body surface. In this study, preparative centrifugal partition chromatography (CPC) was successfully carried out to separate antibacterial compounds from a CP methanol extract. The optimum two-phase CPC solvent system was composed of n-butanol: acetic acid: water (4:1:5 v/v/v). The flow rate of the mobile phase was 3 $m{\ell}/min$ in ascending mode with rotation at 1,000 rpm. The CPC-separated fraction and purification procedures were carried out by preparatory HPLC. Palmatine weas identified by $^1H$, $^{13}C$-nuclear magnetic resonance and electrospray ionization-mass spectroscopy spectral data analysis.
