연구배경 : Peroxiredoxin (Prx) 은 최근에 알려진 항산화제로 세포의 증식과 분화, 세포사멸이나 발암과정에 관여하는 것으로 알려져 있다. 하지만, 현재까지 폐암을 비롯한 각종 질병에서의 이들 Prx단백의 역할은 잘 규명되어 있지 않다. 이에 본 연구는 폐암 조직과 정상폐조직에서 Prx 단백의 발현 양상과 분포를 연구하여 이들의 병태 생리학적인 의미를 찾아보고자 하였다. 방 법 : 아주대학교 병원에서 폐암으로 진단된 환자의 폐암조직과, 동일 환자의 정상 폐조직에서, 1 차원전기영동 (reducing 조건과 non-reducing 조건에서의 SDSPAGE) 혹은 2차원전기영동을 시행한 후 Western blot으로 Prx, Trx 및 TR의 발현 양상을 분석 하였으며, 백서의 정상 폐조직과, 환자의 폐암 조직에서 anti-Prx rabbit polyclonal 항체로 하여 면역조직화학염색법을 통해, Prx 단백의 분포를 관찰하였다. 결 과 : 면역조직화학염색법 결과 백서의 정상 폐조직에서는 Prx I, II, III 및 V 유형이, 주로 기관지상피세포, 폐포상피세포 및 폐포대식세포에서 발현되고 있음을 관찰하였다. 인체 폐암조직에서는, 정상 폐조직 부위에 비해서 Prx I 과 Prx III 유형 및 Trx단백의 발현이 선택적으로 증가되어 있고, 특히, 2차원 전기영동을 통한 프로티옴 분석에서 산화된 형태의 Prx I 과 Prx II가 증가 한 것을 비롯하여, 분자량과 등전점(pI)이 약간 변화된 형태의 Prx III가 폐암조직에 존재함을 알 수 있었다. 한편, 폐암 조직의 non-reducing 전기영동 후 Western blot에서는, monomer와 dimer 사이의 중간 크기에 해당하는 약 40 kDa과 200 kDa 이상 크기의, 항 Prx 항체와 반응하는 단백 띠 (reactive bands)가 관찰되었다. 결 론 : 폐암 조직에서 관찰되는 Prx I 과 Prx III 유형 및 Trx의 과발현 양상은, 종양 세포들이 주변의 미세 환경으로부터 겪는 여러 스트레스에 대하여 단백질을 보호하고 세포의 생명력을 유지하는데 있어 주요한 역할을 하는 것으로 사료된다.
단백질-단백질 결합은 수용체 단백질, 효소, 세포 골격 단백질의 세포내 위치 결정 및 기능 조절에 중요한 역할을 한다. Postsynaptic density-95/disks large/zonula occludens-1 (PDZ) 도메인을 가진 단백질들은 시냅스 가소성, 신경세포 성장과 분화뿐만 아니라 많은 질병의 병태생리에 중요하게 관여하는 scaffold 단백질로 작용한다. Multi-PDZ domain protein 1 (MUPP1)은 13개 PDZ 도메인을 가지는 단백질로서 세포막 수용체 군집화, 신호전달 복합체 구성, 세포 골격 조정에 대한 매개 역할을 하는 것으로 알려지고 있지만 MUPP1의 세포 내 기능은 아직 명확히 밝혀지지 않았다. 본 연구에서 MUPP1의 아미노 말단 PDZ 도메인과 결합하는 새로운 단백질을 규명하기 위하여 효모 two-hybrid 방법을 이용하였고 Wdpcp (전에 Fritz로 알려짐)이 MUPP1과 결합하는 것을 확인하였다. Wdpcp는 planar cell polarity (PCP) effector로서 세포 이동과 섬모형성에 관여하는 것으로 알려져 있다. Wdpcp는 MUPP1의 첫 번째 PDZ 도메인과 결합하지만, 다른 PDZ 도메인과는 결합하지 않았다. 또한 MUPP1와 Wdpcp의 결합에서 Wdpcp의 C-말단부위가 결합에 필수적임을 효모 two-hybrid 방법으로 확인하였다. 이러한 단백질간 결합은 glutathione S-transferase (GST) pull-down assay, 공동면역침강, HEK-293T 세포에서의 발현위치를 통하여 추가적으로 확인하였다. 이러한 결과들은, MUPP1과 Wdpcp 결합은 세포내 액틴 다이내믹스(dynamics)와 세포이동 조절에 역할을 할 가능성을 시사한다.
흰쥐 전뇌 기저부에서 콜린성 신경세포는 내측중격핵, 수직 및 수평 대각 Broca대, 거대세포 시교차앞핵, 복부담창구, Meynert기저핵 그리고 신선조체 등에 분포하는 것으로 알려져 있다. 저자 등은 흰쥐 전뇌 기저부 Meynert 기저핵에서 ChAT항체를 이용한 면역조직 및 세포화학적 방법으로 신경세포들의 발생, 분포, 형태, 형별 출현율 및 세포체의 부피, 조직학적 특성 등을 확인하였다. 또한 전자현미경을 이용하여 ChAT항체에 면역 반응을 하는 신경세포의 미세구조적 특징을 관찰하여 출생 후 발생단계에 따른 Meynert 기저핵에서 ChAT항체 면역반응 신경세포들의 분화를 관찰하고 다음과 같은 결과를 얻었다. ChAT항체에 반응하는 신경세포의 분포는 조직마다 부분적으로 상이하지만, 대부분 담창구가 내포와 접하는 부위, 또는 담창구와 무명질의 연접지역에 군집을 이루거나 혹은 단독으로 관찰되었으며 ,무명질에는 시교차앞핵의 면역반응 세포와 혼재하는 것으로 확인되었다. ChAT면역반응 신경세포들은 세포의 모양과 세포체의 장 단축의 비에 따라 6가지형으로 분류되었다. 원형의 신경세포 출현율은 출생 후 7일에서 가장 높았고, 성체로 되면서 점차적으로 감소하였으며, 난형 신경세포는 출생 후 14일에 가장 높게 나타났고, 원형의 신경세포와 같이 성체로 되면서 점차 감소하였다. 또한, 세장형 신경세포의 출현 율은 출생 후 21일에 가장 높게 나타났다. 삼각형, 방추형 그리고 다각형 신경세포는 성체에서 출현율이 높았다. ChAT면역반응 신경세포체의 부피는 출생 후 7일에 $1,268{\mu}m^3$로 제일 작았으나, 출생 후 21일에서는 $4,453{\mu}m$로 최대치를 보였다. 그 후 성체로 되면서 점차 감소되었다. 전자현미경 관찰에서 성체의 ChAT항체 반응 신경세포의 핵외막, rER의 막, 자유리보즘 그리고 polysorne등이 관찰되었으며, 대칭 및 비대칭 신경연접이 관찰되었다. 이상의 결과들로 미루어 흰쥐 전뇌 기저부 Meynert기저핵에서의 ChAT면역반응 신경세포들은 출생 후 발생과정에서 세포소기관과 신경돌기들의 분화에 따른 세포체 부피의 증가, 분화된 세포형들의 출현 율의 증가 및 세포의 손실이 없는 상태에서 세포 응축 등의 과정을 통하여 세포들이 분화한다고 생각된다.

The experimental exposure of animals to sources of ultraviolet radiation (UVR) which emit their energy primarily in the UVB region (280-320nm) is known to result in a number of well-described changes in the recipient's immune competence. Two such changes include a depressed capacity to effectively respond immunologically to transplants of syngeneic UVR tumors and a markedly reduced responsiveness to known inducers of delayedtype (DTH) and contact hypersensitivity (CH) reactions. The results of experiments that were designed to elucidate the mechanisms responsible for UVR-induced immunomodulation have implicated: 1) an altered pattern of lymphocyte recirculation, 2) suppressor T cells(Ts), 3) deviations in systemic antigen presenting cell (APC) potential. 4) changes in the production of interleukin-1-like molecules, and 5) the functional inactivation of epidermal Langerhans cells in this process. The exposure of skin to UVR, therefore, causes a number of both local and systemic alterations to the normal host immune system. In spite of this seeming complexity and diversity of responses, our recent studies have established that each of the UVR-mediated changes is probably of equal importance to creating the UVR-induced immunocompromised state. Normal animals were exposed to low dose UVR radiation on their dorsal surfaces under conditions where a $3.0\;cm^2$ area of skin was physically protected from the light energy. Contact sensitization of these animals with DNFB, to either the irradiated or protected back skin, resulted in markedly reduced CH responses. This was observed in spite of a normal responsiveness following the skin sensitization to ventral surfaces of the UVR-exposed animals. Systemic treatment of the low dose UVR recipients with the drug indomethacin (1-3 micrograms/day) during the UVR exposures resulted in a complete reversal of the depressions observed following DNFB sensitization to "protected" dorsal skin while the altered responsiveness found in the group exposed to the skin reactive chemical through directly UVR-exposed sites was maintained. These studies implicate the importance of EC as effective APC in the skin and also suggest that some of the systemic influences caused by UVR exposure involve the production of prostaglandins. This concept was further supported by finding that indomethacin treatment was also capable of totally reversing the systemic depressions in CH responsiveness caused by high dose UVR exposure (30K joules/$m^2$) of mice. Attempts to analyze the cellular mechanisms responsible established that the spleens of all animals which demonstrated altered CH responses, regardless of whether sensitization was through a normal or an irradiated skin site, contained suppressor cells. Interestingly, we also found normal levels of T effector cells in the peripheral lymph nodes of the UVR-exposed mice that were contact sensitized through normal skin. No effector cells were found when skin sensitization took place through irradiated skin sites. In spite of such an apparent paradox, insight into the probable mechanisms responsible for these observations was provided by establishing that UVR exposure of skin results in a striking and dose-dependent blockade of the efferent lymphatic vessels in all peripheral lymph nodes. Therefore, the afferent phases of immune responses can apparently take place normally in UVR exposed animals when antigen is applied to normal skin. The final effector responses, however, appear to be inhibited in the UVR-exposed animals by an apparent block of effector cell mobility. This contrasts with findings in the normal animals. Following contact sensitization, normal animals were also found to simultaneously contain both antigen specific suppressor T cells and lymph node effector cells. However, these normal animals were fully capable of mobilizing their effector cells into the systemic circulation, thereby allowing a localization of these cells to peripheral sites of antigen challenge. Our results suggest that UVR is probably not a significant inducer of suppressor T-cell activity to topically applied antigens. Rather, UVR exposure appears to modify the normal relationship which exists between effector and regulatory immune responses in vivo. It does so by either causing a direct reduction in the skin's APC function, a situation which results in an absence of effector cell generation to antigens applied to UVR-exposed skin sites, inhibiting the capacity of effector cells to gain access to skin sites of antigen challenge or by sequestering the lymphocytes with effector cell potential into the draining peripheral lymph nodes. Each of these situations result in a similar effect on the UVR-exposed host, that being a reduced capacity to elicit a CH response. We hypothesize that altered DTH responses, altered alloresponses, and altered graft-versus-host responses, all of which have been observed in UVR exposed animals, may result from similar mechanisms.
세포의 대사과정에서 생성되는 활성산소종(reactive oxygen species : ROS)은 세포의 성숙과 발생 과정을 저해하며, 인간의 생식 수관에서 불임의 원인이 된다. 많은 세포생물학적 연구를 통해 ROS에 대한 세포 내의 보호 기작이 밝혀지고 있다. Activating transcription factor 4(ATF4)는 세포 내에서 산화적 스트레스를 비롯한 여러 스트레스 요인으로부터 세포를 보호하는 기작에 관여하는 중요한 인자로서, 스트레스에 의한 세포 사멸을 유도하는 유전자의 활성화와 관련이 있다고 알려져 있다. 본 연구에서는 착상 전 초기 배아의 발생 단계에서 ROS에 의한 산화적 스트레스가 배아의 발생에 영향을 준다는 보고와 관련하여 생쥐 초기배아에 산화적 스트레스 요인인 $H_2O_2$(hydrogen peroxide)를 처리한 후 ATF4 유전자의 발현 변화를 추적하였으며, ROS 방어에 관여하는 SOD1 유전자와 apoptosis 유전자인 Bax의 발현 양상을 함께 비교하였다. 또한 면역형광염색법을 이용하여 착상전 초기배아의 ATF4 단백질 발현 부위를 조사하였다. $H_2O_2$를 0.1 mM 농도로 처리한 2-세포기 배아에서는 처리 8시간 후인 4-세포기 단계부터 발생율이 감소하기 시작하였으며, 0.5 mM과 1.0 mM 농도에서는 배아의 발생이 진행되지 않았다. RT-PCR결과 SOD1 유전자의 발현은 $H_2O_2$를 처리한 모든 그룹에서 처리 1시간째인 2-세포기 배아단계에서 대조군보다 증가하였으며, ATF4 유전자 역시 2-세포기 배아단계에서 발현이 증가하였다. Bax 유전자도 통일한 시기에 발현이 증가하였다. ATF4 단백질의 배아 세포 내 발현부위는 스트레스 방어 기작이 주로 일어나는 세포질에서 많이 발현이 되었으며 포배기 배아에서는 내세포괴(inner cell mass)부위 보다는 영양외배엽(trophectoderm)에서 발현됨을 확인하였다. 2-세포기 배아에서 ATF4 immunoreactivity는 모든 $H_2O_2$농도 처리군에서 대조군보다 증가하였다. 이상의 결과에서, 착상 전 초기 배아에서 ROS에 의해 ATF4 발현이 유도됨을 확인하였다. 따라서 산화적 스트레스에 대해 배아를 보호하기 위한 방어 기작에 ATF4가 관여하는 것으로 사료되며, 세포 사멸 유전자의 발현과도 밀접한 관련이 있는 것으로 사료된다.
본 연구에서는 H2AX의 생리학적인 기능 및 분자세포 생물학적 기전 해석에 대한 보다 명확한 정보를 제시하고자, H2AX 관련 단백질들을 literature review 및 생물정보학적인 기술을 이용하여 최적의 결합 단백질체를 40개를 예측하곤 이들 가운데 상호작용 가능성이 높은 BRCA1와 BARD1 단백질을 선별하여 in vitro 결합실험을 통해 이를 증명하였다. 이들 두 가지의 유전자를 발굴하여, 클로닝하였다. 클로닝된 유전자를 이용하여 두 가지 단백질을 발현 및 정제하였으며, 단백질들의 자체적인 구조에 의한 결합능력을 판단하기 위해 in vitro binding assay법을 실시하였다. 단백질의 구조적 안정과 비특이적 결합을 억제하는 detergent만이 포함된 상태에서, 구조학적 및 물리학적 상호 결합의 유무를 판정할 수 있게 하였으며, BRCA1과 BARD1은 모두 H2AX에 결합함을 확인하였다. 이런 실험결과를 바탕으로 각각의 단백질에 대해 H2AX와의 최적 결합 부위를 알아내기 위해 각 유전자의 domain을 생물정보학적으로 분석하였다. 이에 RING domain, NES, NLS 및 BRCT domain에 해당하는 유전자 부분을 새로 클로닝하여, 다시 in vitro 결합실험 및 실험결과에 대한 literature review를 통한 분석을 실시한 결과, H2AX는 BRCA1의 NLS, BARD1의 BRCT domain 부분과 결합하는 것을 확인하였다. H2AX에 대한 BRCA1과 BARD1과의 결합은 DNA repair에 있어 BRCA1의 NLS와 BARD1의 BRCT domain을 통해 H2AX foci의 관련 세포 신호전달 기전에 중요한 역할을 하여 전체적으로 genomic stability에 영향을 미칠 가능성이 농후할 것으로 사료된다.
