본 연구에서 우리는 오미자 종자 분획물의 항산화 활성과 자외선에 유도된 human HaCaT cell에서의 MMPs의 발현억제에 의한 항노화 효과를 조사하고자 하였다. 전자공여능, ABTS 라디칼 소거분석, 그리고 hydrogen peroxide 소거분석 실험을 통해, 오미자 종자의 분획물 중 에틸아세테이트 분획물(SCEAf)이 가장 우수한 라디칼 소거활성을 가졌고, collagenase 저해 활성 실험에서 SCEAf는 $500{\mu}g/ml$의 농도에서 92.3% 이상의 저해효과를 보였음을 확인하였다. SCEAf의 HaCaT cell에 대한 세포 독성을 확인하기 위해 MTT assay를 실시한 결과, SCEAf는 세포에 독성이 없음을 나타낼 뿐 아니라 UVB에 손상된 세포의 생존율을 증가시킴으로써 세포 활성에 도움을 주는 것을 확인할 수 있었다. SCEAf의 HaCaT cell에서의 항노화 효과를 조사하기 위해 UVB $50mJ/cm^2$에 유도된 HaCaT cell에 SCEAf를 처리한 후 MMP-1과 -3의 발현을 Western blotting과 reverse-transcription polymerase chain reaction으로 분석하였다. 그 결과 SCEAf가 MMP-1과 -3의 단백질과 mRNAs의 발현을 농도의존적으로 억제시켰다. 이러한 결과는 오미자 종자 에틸아세테이트 분획물이 자외선의 손상을 받은 피부 각질형성세포에서의 collageanse 활성을 억제하여 노화를 예방하고 증상을 완화시킬 것으로 기대할 수 있다. 이러한 결과를 바탕으로 우리는 오미자 종자가 화장품과 식품 산업에서 항노화 효과가 있는 기능성 원료로서 사용하기 위한 잠재성을 가질 것으로 기대한다.
이 연구는 인간진피섬유아(HDF)세포에서 병풀 및 아시아티코사이드가 H2O2 유래 세포주기 정지기, 미토콘드리아 활성 및 염증성 사이토카인에 미치는 영향을 조사하였다. 병풀 80% 메탄올 추출물, 에틸아세티이트 분획물 및 병풀의 대표물질인 아시아티코사이드를 사용하였다. 병풀 추추물, 에틸아세테이트 분획 및 아시아티코사이드로 처리한 세포는 낮은 수준의 TNF-α 및 IL-6을 분비하였고, 아시아티코사이드의 항산화 효과는 병풀 추출물 및 에틸아세테이트 분획물보다 높았다. 아시아티코사이드 처리는 미토콘드리아의 막포텐셜을 증가시키고, 미토콘드리아를 정상으로 되돌렸다. 스트레스 유도 후 에틸아세테이트 분획물 및 아시아티코사이드에 의해 미토콘드리아 산소 소비율이 증가하였고, TLR4-MyD88-TRAF6-p65 경로가 재감소하였다. 이러한 결과는 병풀 추출물, 에틸 아세테이트 분획 및 아시아티코사이드가 HDF 세포의 미토콘드리아 활성을 조절할 뿐 아니라 항산화 및 항염증에 효과 있음을 시사한다.
S-allylcysteine (SAC)은 숙성된 마늘로부터 유래된 수용성 유기황화합물로서, 여러 유형의 암세포에 대한 항암효과를 갖는 것으로 제시되어왔다. 본 논문은 in vitro 및 in vivo 연구결과에 기초하여 SAC가 세포증식, 세포사멸, 세포주기 및 전이에 미치는 세포신호전달경로와 분자적 메커니즘을 정리하였다. SAC는 Bax와 caspase-3을 포함하는 세포사멸촉진 단백질을 활성화하고 Bcl-2 세포사멸억제 단백질군을 억제하여 미토콘드리아-매개 내인성 경로를 통한 세포사멸을 초래 한다. SAC는 또한 PI3K/Akt/mTOR 및 MAPK/ERK 신호전달경로를 억제하여 NF-κB, cyclins, Cdks, PCNA 및 c-Jun의 발현과 활성을 감소시키고, 세포주기 억제단백질인 p16 및 p21의 발현을 증가시킴으로써 세포주기 억제를 유도하여 세포증식을 억제한다. 뿐만 아니라, SAC는 glutathione-s-transferase (GST)와 같은 항산화효소의 활성을 유도하여 독성물질에 의해 유도된 발암작용을 방지한다. 그리고, SAC는 MAPK/ERK 및 PI3K/Akt/mTOR/NF-κB 신호경로의 억제를 통한 전사억제조절인자 Id-1 및 SLUG의 발현억제를 통하여 초래된 COX-2의 발현감소와 E-cadherin의 발현증가에 의해 신생혈관생성과 MET의 억제를 유도함으로써 암세포의 침투와 전이를 억제한다. 따라서, SAC는 암의 예방과 치료를 위한 하나의 잠재적 화학요법제로 간주될 수 있다.
Antioxidant enzymes are scavenger reactive-oxygen intermediates and are involved in many cellular defense systems. We previously reported that a crude extract of Garnoderma lucidum, a medicinally potent mushroom, profoundly increased the catalase gene expression and enzyme activities in mouse livers (Park et al., J. Biochem. Mol. Biol. 34. 144-149, 2001). In this study, we elucidated the detailed mechanism whereby G. lucidum stimulates the catalase activity and expression. The major active fraction was isolated from G. lucidum and methyl linoleate was considered the most major component of the fraction. In order to determine whether methyl linoleate increases mRNA and protein synthesis of catalase, Northern and Western blot analyses were performed in vivo with methyl linoleate-treated mouse liver homogenate after feeding methyl linoleate to the mice. Northern and Western blot analyses of the crude liver homogenates in the mice that were administered methyl linoleate revealed that the expression catalase was significantly increased when compared to the untreated controls. In addition, the catalase protein levels and enzymatic activities increased in the mouse liver homogenates. These results suggest that methyl linoleate that is produced by G. lucidum stimulates the catalase expression at the transcription level.

Background: The induction of cellular defensive genes such as phase II detoxifying and antioxidant enzymes is a highly effective strategy for protection against carcinogenesis as well as slowing cancer development. Transcription factor Nrf2 (nuclear factor E2-related factor 2) is responsible for activation of phase II enzymes induced by natural chemopreventive compounds. Methods: Red ginseng oil (RGO) was extracted using a supercritical $CO_2$ extraction system and chemical profile of RGO was investigated by GC/MS. Effects of RGO on regulation of the Nrf2/antioxidant response element (ARE) pathway were determined by ARE-luciferase assay, western blotting, and confocal microscopy. Results: The predominant components of RGO were 9,12-octadecadienoic acid (31.48%), bicyclo[10.1.0] tridec-1-ene (22.54%), and 22,23-dihydrostigmasterol (16.90%). RGO treatment significantly increased nuclear translocation of Nrf2 as well as ARE reporter gene activity, leading to upregulation of heme oxygenase-1 and NAD(P)H:quinone oxidoreductase 1. Phosphorylation of the upstream kinases such as apoptosis signal-regulating kinase (ASK)1, mitogen-activated protein kinase (MAPK) kinase (MKK)4/7, c-Jun N-terminal kinase (JNK), and p38 MAPK were enhanced by treatment with RGO. In addition, RGO-mediated Nrf2 expression and nuclear translocation was attenuated by JNK inhibitor SP600125 and p38 MAPK inhibitor SB202190. Conclusion: RGO could be used as a potential chemopreventive agent, possibly by induction of Nrf2/ARE-mediated phase II enzymes via ASK1-MKK4/7-JNK and p38 MAPK signaling pathways.
Scutellaria baicalensis Georgi. (SBGE) is known to be antioxidant effect. In addition of the effects, we further investigated the SBGE on the antioxidant effect on a renal cortical slices cell and kidney protecting effects. The results were as follows. 1 When renal cortical slices separated from a rabbit's kidney were treated with 1mM tert-Butylhydroperoxide (t-BHP) in the presence of SBGE. SBGE significant prevented t-BHP induced increase in lactate dehydrogenase (LDH) release and lipid peroxidation. 2. When renal cortical slices separated from a rabbit's kidney were treated with oxidant $300{\mu}M$ cisplatin in the presence of SBGE. SBGE significant prevented cisplatin-induced increase in LDH release and lipid peroxidation. 3. Pretreatment with 0.1g/kg SBGE for seven day and treatment with 5 mg/kg cisplatin by the intraperitoneal injection The results were that the pretreatment group with SBGE showed a significant decrease in lipid peroxidation, increase in clearance rate of blood urea nitrogen (BUN) and creatinine in the kidney than the administering single agent group of cisplatin. and pretreatment group with SBGE showed intact microvillus of proximal tubule and no contraction of rumen, it was a similar result with normal group. With the results SBGE showed to be highly effective on antioxidant effect and cellular protection activity against cisplatin that was a toxic agent on a kidney. Therefore, SBGE is considered to have protective effective on a disordered kidney or kidney diseases such as nephritis or renal failure that cause tissue damages in a kidney.

Stevia rebaudiana (Asteraceae), a perennial plant, has been used as a low-calorie sweetener and is being developed as a therapeutic agent for diabetes, hypertension, myocardial diseases, and microbial infections. Despite the common use of its leaves and stem, the bioavailability of the components present in S. rebaudiana flowers, when used as ingredients of cosmetics, has not been well investigated. Herein, we investigated the antioxidative and antimelanogenic effects of an aqueous extract of S. rebaudiana flowers (Stevia-F). Total flavonoid and phenolic content in Stevia-F were determined to be $8.64{\pm}0.23mg$ of quercetin equivalents/100 g and $631.5{\pm}2.01mg$ of gallic acid equivalents/100 g, respectively. The $IC_{50}$ values of Stevia-F for reducing power, and 2,2-diphenyl-1-picryl-hydrazyl-hydrate radical, hydrogen peroxide, and nitric oxide scavenging activities were 5541.96, 131.39, 466.34, and $10.44{\mu}g/mL$, respectively. Stevia-F showed inhibitory effects on the tyrosinase ($IC_{50}=134.74{\mu}g/mL$) and ${\alpha}$-glucosidase ($IC_{50}=114.81{\mu}g/mL$) activities. No significant cytotoxicity of Stevia-F was observed in B16F10 cells, treated with up to $100{\mu}g/mL$ of the extract for 24 and 48 h (p > 0.05). Stevia-F ($1-100{\mu}g/mL$) suppressed ${\alpha}$-melanocyte stimulating hormone-induced melanin production in B16F10 cells (p < 0.05) and also inhibited the cellular tyrosinase activity (p < 0.05). Overall, our results show that Stevia-F possesses potential for inhibiting tyrosinase and ${\alpha}$-glucosidase activities and has significant antioxidant capacity. The antimelanogenic potential of Stevia-F should extend the usage of S. rebaudiana flowers in the development of skin-whitening products.
본 연구에서는 잣나무 잎 추출물의 항균, 항산화 및 성분 분석에 관한 연구를 수행하였다. 피부 상재균에 대한 항균활성 측정결과, Propionibacterium acnes (P. acnes), Staphylococcus aureus (S. aureus), Pityrosporum ovale (P.ovale) 및 Escherichia coli (E. coli)에 대한 ethyl acetate 분획의 MIC는 각각 0.06 %, 0.25 %, 0.13 %, 0.50 %로 나타났으며, P. acnes, P. ovale 및 S. aureus에서 큰 항균활성을 나타내었다. 잣나무 잎 추출물의 free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거활성($FSC_{50}$)은 aglycone 분획에서 $22.93\;{\mu}g/mL$으로 나타났다. Luminol-의존성 화학발광법을 이용한 $Fe^{3+}$-EDTA/$H_2O_2$계에서 생성된 활성산소종(reactive oxygen species, ROS)에 대한 잣나무 잎 추출물의 총 항산화능($OSC_{50}$)은 50 % ethanol extract 분획에서 $0.70\;{\mu}g/mL$, ethyl acetate 분획 및 aglycone 분획이 각각 1.04 및 $1.43\;{\mu}g/mL$으로 매우 큰 항산화능을 보였다. Rose-bengal로 증감된 사람 적혈구의 광용혈 실험에서 추출물의 세포보호 효과를 측정하였다. 잣나무 잎 추출물 분획들은 농도 의존적($5{\sim}50\;{\mu}g/mL$)으로 세포보호 효과를 나타내었다. 잣나무 잎 추출물의 가수분해로 얻은 ethyl acetate 분획은 TLC (PK-4, PK-6)와 HPLC (peak 1, peak 2)에서 2개의 주성분으로 나타났다. 이 성분들은 LC/ESI-MS/MS를 통해서 PK-6는 kaempferol-3-O-glucoside (astragalin)로, PK-4는 kaempferol-3-O-arabinoside (juglanin)로 확인되었다. 이상의 결과들은 잣나무 잎 추출물이 ROS를 소광시키거나 소거함으로써, 그리고 ROS에 대항하여 세포막을 보호함으로써 생체계, 특히 태양 자외선에 노출된 피부에서 항산화제로서 작용할 수 있음을 가리키며, 잣나무 잎 성분 분석, 그리고 피부 상재균에 대한 항균작용으로부터 항산화, 항노화 및 항균성 화장품 소재로서의 응용 가능성이 있음을 확인하였다.
생 표고버섯을 5 g씩 나누어 실험용 건조기와 일광을 이용하여 건조한 후 100 mL의 유기용매(acetone, ethanol, methanol)와 $90^{\circ}C$의 물 100 mL로써 추출하였다. 총 페놀함량을 측정하고 농축 또는 동결 건조하여 DMSO에 녹인 후 적당한 농도로 희석하여 DPPH 라디칼 소거능, ORAC value 및 CAC 등의 항산화 활성을 측정하였으며 comet assay를 이용하여 항 유전독성을 조사하였다. 그 결과, 아세톤과 에탄올 추출물에서 일광건조 한 표고버섯의 총 페놀 함량이 생 표고나 오븐건조 한 표고의 함량보다 유의적으로 높았으며 물 추출한 표고버섯 시료의 총 페놀이 유기용매로 추출한 시료보다 최소 2배 이상 월등히 높은 함량을 나타내었다. $IC_{50}$으로 나타낸 표고버섯의 DPPH 라디칼 소거능은 생 표고버섯이 모든 용매에서 유의적으로 가장 낮은 값을 나타내었으며, 특히 물 추출에서는 일광건조 시료가 생 표고와 거의 비슷한 활성을 보였다. AAPH로 유도된 peroxyl radical 소거능은 $50{\mu}g/mL$ 농도에서 오븐건조 한 표고버섯보다 생 표고나 일광건조 한 표고버섯이 더 뛰어남을 알 수 있었다. HepG2 cell을 이용하여 세포 내에서의 peroxyl radical에 대한 항산화 활성 검색에서는 시료의 최종농도를 $100{\mu}g/mL$로 처리했을 때 일광건조 한 표고버섯의 활성이 두드러지게 높은 활성을 보였으며 특히 메탄올과 물 추출 시료에서 각각 107.5%와 108.1%로 AAPH를 처리하지 않은 control과 비슷한 값을 나타내었다. 또한 comet assay에서는 표고버섯 추출물을 $50{\mu}g/mL$의 농도로 백혈구에 처리한 후 $H_2O_2$$200{\mu}M$의 농도로 처리하여 DNA 손상을 유도한 결과 모든 표고 버섯 추출물이 $H_2O_2$만을 처리한 positive control에 비해 DNA 손상에 대한 보호 효과를 나타내었으며 특히 생 표고버섯의 항 유전독성 효과가 건조 표고버섯에 비해 뛰어났다. 총 페놀 함량과 항산화 활성 및 항 유전독성 활성 간에 상관관계는 나타나지 않았다. 이는 페놀 화합물 외에 표고버섯에 함유되어 있는 여러 가지 다른 생리활성 물질이 건조 조건에 따라 각각 다른 용매에 용출되어 작용한 것으로 사료되며 이에 대하여 앞으로 더욱 심도 있는 추가적인 연구가 수행되어야 할 것이다. 결론적으로 표고버섯을 식품으로 섭취 시 생 표고를 이용하며 장기간 보존을 위해 건조를 한다면 일광을 이용하는 것이 높은 페놀 함량과 항산화 활성을 기대할 수 있을 것이다.
본 연구는 Monascus purpureus (Mp), Aspergillus oryzae (Ao), Aspergillus kawachii (Ak) 및 Rhizopus oryzae (Ro) 균주로 Cordycepin-고함유 동충하초(Cordyceps militaris)(CM${\alpha}$)를 발효시켜 수용성 추출물을 얻어 페놀화합물 및 플라보노이드 농도와 항산화 및 티로시나제 저해 활성을 측정한 결과 Ak로 발효시킨 CM${\alpha}$ (AkF-CM${\alpha}$)에서 각각 46 mg/g 및 093 mg/g과 6274% 및 7997%로 가장 우수한 효과를 나타내었다. 이러한 결과로부터 AkF-CM${\alpha}$를 선택하여 멜라닌 세포(B16F0 mouse melanoma cell)에서 미백효과를 검토하였다. 양성 대조구 arbutin 처리 B16F10 melanoma 세포는 92% 이상의 세포 생육과 43%의 멜라닌 생성 억제 효능을 보였고, AkF-CM${\alpha}$ 1, 3 및 5 mg/ml 처리 시 멜라닌 생성은 각각 35, 45 및 53% 억제되었다. 또한 AkF-CM${\alpha}$은 멜라닌 세포 내 tyrosinase 활성과 mushroom tyrosinase 활성 모두를 저해시켰고, 멜라닌 생성 관련 tyrosinase 단백질 발현량도 무첨가군에 비해 처리 농도 의존적으로 억제되었다. 이상의 결과에 따라 Aspergillus kawachii 균주로 발효시킨 Cordycepin-고함유 동충하초(Cordyceps militaris)의 수용성 추출물은 미백 화장품 소재로 개발 가능성이 높은 것으로 사료된다.

① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
