가토 수정란의 단기 체외보존에 관한 연구 (Short Term In Vitro Preservation of Embryos in Domestic Rabbit)

  • This experiment was performed to develop simple practical methods for short term preservation of rabbit embryos. A total of 55 cross bred does were superovulated by intramuscular injection of PMSG and HCG. Embryos were recovered at 25~30 hrs, 60~65 hrs and 80~85 hrs after mating and selected by morphological examination. Four cell stage, morulae and blastocyst embryos were stored in PBS enrich with 1, 10, 20 and 40% heat-treated FCS at 4, 20, 30 and 37$^{\circ}C$, respectively. Embryos were examined morphologically at 24, 48 and 72 hrs following storage. The result obtained in this experiment were summarized as follows: The superovulation was induced by PMSG 200 IU and HCG 100 IU. The average number of ovulation points and embryos recovered by collection time were 19.0, 15.6(25~30 hr), 17.3, 13.5(60~65 hr) and 19.2, 14.4(80~85 hr), respectively. And recovery rates of embryos recovered at 25~30 hr, 60~65 hr and 80~85 hr after mating were 62.8%(4 cell), 84.7%(morulae) and 79.6%(blastocyst), respectively. On the other hand, the average number of ovulation points collected by the no, of operations for the repeated collection was 17.3(60~65 hr), 19.2(80~85 hr) in 1st and 9.4(60~65 hr), 10.6(80~85 hr) in 2nd surgery, respectively. There was a significant decrease(P<0.05) in the number of ovulation points the 2nd surgery as compared to the 1st surgery. All of the 4-cell stage embryos stored at 4$^{\circ}C$ for 48 hrs showed the same morphology throught the storage period, on the contrary, 4-cell stage embryos stored at 2$0^{\circ}C$ and 3$0^{\circ}C$ for 48 hrs showed degeneration embryos and stored at 37$^{\circ}C$ for 24 hrs showed degeneration embryos. Morulae and blastodcyst stored at 4, 20, 30 and 37$^{\circ}C$ for 24 hrs showed degeneration embryos. All of the blastocyst stored at 37$^{\circ}C$ for 72 hrs showed degeneration embryos.

효모의 환경내성에 대하여 1 (A Study on Environmental Tolerances of Yeast)

  • Saccharomyces cerevisiae strain M was cultured in a molasses-containing media with repeated transplantations of the yeasts from one culture to another to adapt to molasses. After that only different amounts of phosphorous and nitrogen sources were added to the media. And then some variations during the culture time and the effects of consituents of cell mass on the functional activity and sensitivity of the cell were investigated. The results obtained were summarized as follows : 1. In the same culture condition of yeasts, the carbohydrates and trehaloses contents were more remarkably increased when small amounts of phosphate and nitrogen sources were added, then when alrge amounts were added, but yield percentage on assimilated sugars was lower. 2. The content of trehalose in yeast cells was reduced remarkably at the early stage in the culture, but this increased remarkably at later stage. When small amounts of nitrogen and phosphate were added to the culture medium, the amount of thehalose in the cells increased greasly. 3. The more protein content was present in the yeast cells, the smaller the carbohydrate and trehalose content, but more amino-N, RNA and moisture content were present in the cells. And in this case fermentability of the cells was stronger, but sugar tolerance was lower. 4. During the preservation period of compressed yeast cells at different temperature, the higher the temperature was, the more rapidly the amount of trehalose in the cells decreased. And in the cell where the amount of trehalose in the cells decreased. And in the cell where the amount of trehalose(carbohydrate) was large and the amount of protein was small, the amount of trehalose decreased at a slower rate during the preservation period.

Methodologies for Cryopreservation of Mammalian Germline Cells and Tissues

  • Until today, success in germline cells and tissue cryopreservation is limited mainly due to the poor understanding of the complex physiological processes can lead to cell damage during cryopreservation. Germline cells, from both male and female, have unique ability to differentiate into one or more cell lines and thus it becomes a crucial point to store them in subzero temperature with the minimal damage of their functional properties and maximum recovery of unchanged and viable cells when thawed. In the past three decades, a vast research has been performed using various different animal models which in fact have led to development of new methodologies and optimization of older one. However, successful use of animal model has provided the opportunity in research with human germline cells and tissues preservation, but not in all the cases. Therefore, the use of new cryo-protective chemicals and modified protocols have been often found in different groups of researchers based on the types, physical structures, utility and animal species of the specimens to be cryopreserved. This review discusses about the basics of different types of cryopreservation methodologies and commonly used optimized protocols and cryoprotectants for germline cells and tissues preservation.

성숙중 딸기의 세포벽 비섬유성 중성당 변화 (Changes in Non-cellulosic Neutral Sugar of Strawberry during Maturation)

  • The changes of non-cellulosic neutral sugar composition of strawberry during maturation were investigated. Arabinose, xylose, galactose and glucose were the main non-cellulosic neutral sugar of cell wall and increased until ripe stage. The main non-cellulosic neutral sugar of alkali soluble hemicellulose were arabinose, xylose, mammose, galactose. The contents of non-cellulosic neutral sugar of alkali soluble hemicellulose were increased during maturation.

발포성 포도주의 생산에 고정화 효모의 이용 (Using of Immobilized Yeast Cells for the Production of Sparkling Wine)

  • In order to investigate the possible application of immobilized yeast cells in sparkling wine production instead of riddling puns by the traditional method, fermentation characteristics were tested during the sparkling wine fermentation in the bottle using immobilized yeast cells with alginate. The rates of sugar consumption and alcohol production were faster with free cells than those with immobilized cells during the fermentation. The higher concentration of yeast cells and the lower concentration of alginate in the cell immobilization resulted in the faster sugar consumption and alcohol production. It also resulted in the increase of yeast cell concentration released from immobilized beads during the fermentation. However, no differences were shown in the contents of alcohol, residual sugar and CO2 pressure after fermentation. In case concentration of yeast cells released from immobilized beads during bottle fermentation, the higher concentration of alginate had and the lower had.

진달래꽃 추출물의 항산화 효과 및 인체 KB cell에 대한 세포독성 (Antioxidative Activity and Cytotoxicity on Human KB cell of Extracts from Rhododendron mucronulatum Turcz. Flower)

  • 진달래꽃 추출물의 항산화 효과와 인체 비인두암 세포인 KB cell에 대한 세포독성을 조사하였다. 또한 총 폴리페놀과 총 플라보노이드 함량을 측정하고 항산화성과의 상관성을 검토하였다. 진달래꽃 메탄올 추출물을 용매 분획하여 항산화 활성을 측정한 결과, 에칠아세테이트 분획의 활성이 가장 높게 나타났다. 에칠아세테이트 분획을 silica gel column chromatography로 부분 정제하여 항산화 실험을 한결과 fraction 2와 3은 40 ppm 농도에서 90% 이상의 활성을 나타내었다. 인체 비인두암 세포주인 KB cell의 성장 저해율은 $100{\mu}g/mL$ 농도에서 클로로포름 분획이 81.2%, 헥산분획이 74.6% 였다. 총 폴리페놀과 총 플라보노이드 함량을 측정한 결과, 에칠아세테이트 분획에는 총 폴리페놀 32.70%, 총 플라보노이드 20.30% 함유하고 있었다. 총 폴리페놀 및 총 플라보노이드 함량과 항산화성과의 상관관계는 총 폴리페놀에 대하여는 $r^2=0.8869$, 총 플라보노이드에 대하여는 $r^2=0.8469$로 상관성이 인정되었다.

냉면육수의 보존중 겨자의 첨가효과 (Effects of Mustard Seed (Brassica juncea) during Preservation of Soup for Naengmyon)

  • 겨자의 항균성 및 식품보존제로의 이용가능성을 조사하기 위하여 겨자의 첨가에 따른 냉면육수의 보존 중 그 효과를 조사하고, 대장균의 형태변화를 현미경으로 관찰한 결과는 다음과 같다. 겨자를 첨가시킨 냉면육수의 적정산도는 처음에 겨자 무첨가 육수에 비하여 높았으나 24시간 보존 후에는 겨자 무첨가 육수보다 낮게 나타났으며, 겨자 첨가 냉면육수의 생균수 및 대장균수는 처음에 겨자 무첨가시보다 감소하였으며, 겨자 첨가농도가 높을수록 감소하는 폭이 크게 나타났고, VBN 함량은 보존기간이 지남에 따라 증가하였고, 겨자가 첨가된 냉면육수가 겨자 무첨가보다 낮게 나타났으며, 겨자첨가 농도가 높을수록 더욱 낮게 나타났다. 겨자 물추출물로 처리한 대장균을 전자현미경으로 관찰한 결과 균체표면이 수축되고 표층구조가 허물어져 심한 형태학적 변화를 나타내었다.
