본 논문에서는 청각 모델을 기초로 잡음에 강한 음성 특징 추출을 연구하였다. 청각모델은 basilar membrane 모델, 섬모세포(hair cell) 모델과 스펙트럼 출력단으로 구성하였다. Basilar membrane 모델은 음파의 진동에 따른 전달 특성을 묘사한 것으로 대역 통과 필터의 열로 나타난다. 섬모 세포 모델은 basilar membrane의 진동에 의한 신경 물질로의 변환을 나타낸다. 이것은 입력의 상대적인 값에 크게 반응하는 adaptation 기능을 이용하게 되며, 잡음 제거에 중요한 역할을 하게 된다. 스펙트럼 출력 단은 각 채널의 평균 firing rate를 이용하여 mean rate spectrum을 형성한다. 그리고 mean rate spectrum을 이용하여 특징 벡터를 추출하였다. 실험 결과는 청각 구조에 기초한 특징 추출이 다른 특징 추출 방법에 비해 잡음에서 더 향상된 성능을 가짐을 보였다.
석유기반 플라스틱의 대체제인 폴리하드록시부틸레이트(polyhydroxybutyrate, PHB)의 기존 추출방법은 분자량 감소 및 물성 변형을 일으킨다. 본 연구에서는 기능화 된 탄소나노튜브(carbon nanotube, CNT)를 부착한 돌기형 탄소나노튜브 분리막의 여과를 통해 물리적 파쇄를 발생시켜 미생물 내 축적된 PHB를 추출하고자 하였다. 돌기형 탄소나노튜브 분리막의 물리적 파쇄를 확인하기 위해 대장균 용액으로 여과 실험을 수행하여 불활성화를 관찰하였다. 또한 PHB를 축적한 미생물 용액의 여과를 수행하여 PHB가 추출되었는지 확인하였더니 가장 대표적인 추출방법인 chloroform과 비교하여도 여과로 인한 추출이 4% 높은 성능을 가진 것을 관찰하였다. 본 결과를 통해 친환경적 바이오 플라스틱 회수를 위한 돌기형 탄소나노튜브 분리막의 적용 가능성을 확인하였다.
셀 카운팅은 세포의 성장을 분석하는 방법으로써 생물학연구에서 가장 많이 사용된다. 최근까지도 다양한 자동 셀 카운팅 기법이 제안되고 있지만 암세포와 같이 분열 속도가 빠르고 군집하려는 성질을 갖는 세포들은 분리 및 검출이 쉽지 않아 세포 이미지 분석을 통하여 셀 카운팅의 신뢰도를 높이기가 어렵다. 본 논문에서는 암 연구의 연구재료로 매우 보편적으로 사용되는 HeLa 세포 이미지 분석을 이용한 자동 셀 카운팅 방법을 제시한다. 세포막 추출 기반의 세포 분할 알고리즘을 통하여 세포의 형태적 상황을 구분하고, 세포 간 경계가 희미한 세포군집 내의 세포 분할을 위하여 역추적 알고리즘을 사용함으로써 셀 카운팅 정확도를 높인다. 실험을 통하여 제안하는 세포 분할 알고리즘이 기존의 세포 분할 알고리즘에 비해 정확함을 입증하였고, 결과적으로 매우 높은 자동 셀 카운팅 정확도를 얻을 수 있음을 확인하였다.
본 논문에서는 한시적 세포포집 구조물을 이용한 다세포 스페로이드의 형성 및 추출칩을 제안하였다. 종래의 웰 플레이트와 플라스크는 작은 스페로이드 형성이 어렵고, 기존 마이크로칩은 고정된 세포포집 구조물을 이용하여 스페로이드 추출이 어려운 단점이 있다. 반면, 제안된 칩은 한시적 세포포집 구조물을 이용하여 스페로이드의 형성과 추출이 모두 용이한 장점이 있다. 50kPa 의 박막압력으로 형성된 세포포집 구조물에 145~155Pa 의 세포입력압력으로 유입되는 세포를 포집한 후, 24 시간 배양하여 스페로이드를 형성하였다. 또한, 박막압력 제거 후 5kPa 의 세포입력압력으로 추출된 스페로이드의 지름과 활성도는 각각 $197{\pm}11.7Bm$, $80.3{\pm}7.7%$로 측정되었다. 제안된 칩은 스페로이드의 균일한 형성과 안정적 추출이 용이하여 스페로이드의 후처리에 적용될 수 있다.
회분식 발효를 실시하여 균주의 최적 발효조건을 구하여 세포 재순환식 연속 발효 공정에 적용한 결과, CSL의 최적 함량은 2.5%로 나타났으며, 수율과 생산성은 각각 0.80g total acid/g glucose, 0.26g total acid/L/h이였다. 연속 발효 결과, 최대 생산물 생산성은 희석률 0.2/h에서 3.32g organic acid/L/h을 얻었다. 최적 추출시스템은 30%w/w TOA/MIBK로 나타났으며, Membrane Contactor을 이용한 추출에서는 수용액상의 200ml/min, 추출상은 100ml/min의 유량을 갖는 Counter-current가 최적 조건임을 확인했고, 또한 수용액상과 추출상의 접촉 부피비는 2:1이 여러 가지 요인을 고려했을 때 최적 조건임을 확인했다. 실제 발효를 통해 생성된 유기산의 추출은 1:1의 부피비로 counter-current로 접촉시켰을 때 Acetic acid 가 56% Propionic acid가 72.41%의 추출력을 얻을 수 있었다.
A novel process was developed to simultaneously produce invertase and yeast extract from baker's yeast using ultrafiltration (UF) and microfiltration (MF) membrane processing. After the extraction of invertase under the optimal condition obtained in this study, invertase was separated from yeast cells using a hollow fiber membrane with a pore size of 0.1 $\mu\textrm{m}$. The resulting permeate containing invertase was concentrated using a hollow fiber membrane with a nominal molecular weight cut-off of 30 kDa. The yeast cell and permeate solutions, which were obtained after MF and UF membrane processing, respectively, were mixed together, and the autolysis was performed at 50$^{\circ}C$ in the presence of 5% (w/v) ethanol and 1% (w/v) NaCl. As a result, the yeast extract and invertase could be simultaneously produced from baker's yeast by this novel process.
Voacanga globosa (Blanco), a plant endemic to the Philippines, is traditionally used especially by indigenous people of Bataan in the treatment of ulcers, wounds and tumorous growths. This study aimed to provide scientific evidence to therapeutic properties by determining cytotoxic and pro-apoptotic activity of HPLC fractions from leaves on HCT116 human colon carcinoma and A549 human lung carcinoma cell lines. Ethanolic extraction was performed on V globosa leaves followed by hexane and ethyl acetate partitioning. Silica gel column chromatography and high performance liquid chromatography (HPLC) produced MP1, MP2 and MP3 fractions. Cytotoxic activity of the fractions was determined through MTT assay against the cancer cell lines HCT116 and A549 and the non-cancer AA8 Chinese hamster ovarian cell line. Pro-apoptotic activities of the most active fractions were further assessed through DAPI staining, TUNEL assay and JC-1 mitochondrial membrane potential assay with HCT116 cells. While the MPI fraction exerted no significant activity against all cell lines tested, MP2 and MP3 fractions demonstrated high toxicity against HCT116 and A549 cells. The MP3 fraction induced formation of apoptotic bodies, condensed DNA and other morphological changes consistent with apoptosis of HCT116 cells and TUNEL assay showed significant increase in DNA fragmentation over time. In these cells, the MP3 fraction also induced mitochondrial membrane destabilization, which is generally associated with the beginning of apoptosis. Phytochemical analysis demonstrated the presence only of saponins and terpenoids in the MP3 fraction. The results indicate that the MP3 fraction exerts cytotoxic activity on HCT116 cells via induction of apoptosis triggered by loss of mitochondrial membrane potential crucial for cell survival.
This study was undertaken to determine whether Salviae-radix (SVR) extraction prevents the oxidant-induced cell injury and thereby exerts protective effect against oxidant-induced inhibition of tetraethylammonium uptake (TEA) in renal corticaJ sices. SVR (5%) attenuated $H_2O_2-induced$ inhibition of TEA uptake. $H_2O_2$ increased LDH release and lipid peroxidation in a dose-dependent manner. These changes were prevented by SVR extraction. The protective effect of SVR on LDH release was dose-dependent over the concentration range of 0.1-0.5%, and that on lipid peroxidation over the concentration ranges of 0.05-2%. SVR significantly prevented Hg-induced lipid peroxidation. SVR extraction (0.5%) increased cellular GSH content in normal and $H_2O_2-treated$ tissues. When slices were treated with 100 mM $H_2O_2$, catalase activity was decreased, which was prevented by 0.5% SVR extraction. The activity of glutathione peroxidase but not superoxide dismutase was significantly increased by 0.5% SVR extraction in $H_2O_2-treated$ tissuces. These results suggest that SVR has an antioxidant action and thereby exerts benefical effect against oxidant-induced impairment of membrane transport function. This effect of SVR is attributed to an increase in endogenous antioxidants such as GSH, catalase and glutathione peroxidase.
The secretion of the alkaliphilic Bacillus sp. S-1 extracellular pullulanase involves translocation across the cytoplasmic membrane of the Gram-positive bacterial cell envelope. Translocation of the intracellular pullulanase PUL-I, was traced to elucidate the mechanism and pathway of protein secretion from an alkaliphilic Bacillus sp. S-1. Pullulanase could be slowly bue quantitatively released into the medium during growth of the cells in medium contianing proteinase K. The released pullulanase lacked the N-terminal domain. The N-terminus is the sole membrane anchor in the pullulanase protein and was not affected by proteases, confirming that it is not exposed on the cell surface. Processing of a 180,000M$\_$r/ pullulanase to a 140,000M$\_$r/ polypeptide has been demonstrated in cell extracts using antibodies raised against 140,000M$\_$r/ extracellular form. Processing of the 180,000 M$\_$r/ protein occured during the preparation of extracts in an alkaline pH condition. A modified rapid extraction procedure suggested that the processing event also occured in vivo. Processing apparently increased the activity of pullulanase. The western blotting analysis with mouse anti-serum against 140-kDa extracellular pullulanase PUL-E showed that PUL-I is processed into PUL-X via intermediate form of PUL-E. Possible explanationa for the translocation are discussed.

In this study, poly(vinyl-alcohol) and water insoluble ${\beta}$-cyclodextrin polymer (${\beta}$-CDP) cross-linked with citric acid, have been used as macrocyclic carrier in the preparation of polymer inclusion membranes (PIMs) for aniline (as molecule model) extraction from aqueous media. The obtained membranes were firstly characterized by X-ray diffraction, Fourier transform infrared and water swelling test. The transport of aniline was studied in a two-compartment transport cell under various experimental conditions, such as carrier content in the membranes, stirring rate and initial aniline concentration. The kinetic study was performed and the kinetic parameters were calculated as rate constant (k), permeability coefficient (P) and flux (J). These first results demonstrated the utility of such polymeric membranes for environmental decontamination of toxic organic molecules like aniline. Predictive modeling of transport flux through these materials was then studied using design of experiments; the design chosen was a two level full factorial design $2^k$. An empirical correlation between aniline transport flux and independent variables (Poly ${\beta}$-CD membrane content, agitation speed and initial aniline concentration) was successfully obtained. Statistical analysis showed that initial aniline concentration of the solution was the most important parameter in the study domain. The model revealed the existence of a strong interaction between the Poly ${\beta}$-CD membrane content and the stirring speed of the source solution. The good agreement between the model and the experimental transport data confirms the model's validity.
