An alkalophilic mi.croorganism (strain YB380), which produces yeast cell wall hydrolase extracellulary, was isolated from Korean soil. The rod-shaped cells were 0.3~0.4 by 2~4${\mu}{\textrm}{m}$ long, motile, aerobic, gram-positive, and spore-forming. The color of the colony was light yellow. The temperature range for growth at pH 9.0 was 25 to $45{\circ}C, with optimum growth at $35{\circ}C. The pH range for growth at $35{\circ}C was 8 to 11 with an optimum pH of 9.0. Therefore, the strain YB380 is an obligate alkalophile. The 16S rRNA of strain YB380 has a 99% sequence similarity with that of Bacillus alcalophilus. On the basis of physiological properties, cell wall fatty acid composition, and phylogenetic analysis, we propose that the isolated strain is Bacillus alcalophilus. The yeast cell wall hydrolase from Bacillus alcalophilus subsp. YB380 has been purified and partially characterized. The molecular weight was estimated to be 27,000 daltons with an optimum temperature and pH of $60{\circ}C and 9.0, respectively. The N-terminal amino acid sequence of the enzyme was analyzed as Gln- Thr- Val- Pro- Trp- Gly- Ile- Asn- Arg- Val.
GaAs polycrystalline thin films with good performance were prepared on conducting glass by hot wall epitaxy (HWE), which were used for solar cell. Electron probe micro-analyzer (EPMA) was applied for the composition, morphology of surface and cross-section of grown films, and X-ray diffraction (XRD) for their phase structure; Raman scattering spectum (RSS) and photoluminescence (PL) were used for evaluating their optical characteristics. The results show that, there is textured structure on the surface of grown GaAs polycrystalline films, which is greatly promised to be suitable for the candidate of solar cell with low cost and high efficiency. It is concluded that the source and substrate at temperature of 900 ~ 930 $\^{C}$ and 500 $\^{C}$ respectively would be beneficial for such films.

This study was developed to evaluate the growth inhibition effects of cell wall components of Enterococcus faecalis 2B4-1 obtained from feces of neonates against tumor cell lines. Polysaccharide fraction (PS) shown sensitive growth inhibition effect in the cell wall components was isolated and characterized. In growth inhibition effects, residue fractin of whole cell was shown sensitive level of percent survival about 30% when administrated at ehe concentration of 100${\mu}$g/ml, and that was more effective than that of supernatant fraction against the tumor cell lines, SNU-1, 3LL, FARROW and HEC-1-B. Sensitive growth inhibition effects against SNU-1, FARROW and HEC-1-B were performed by whole cell (WC) fraction from Ent. faecalis 2B4-1. Cytoplasm fractin (CP) of WC was shown non-inhibition effect, however, the other part of WC, precipitate of disrupted cell (PD), was sensitive against the tumor cell line mentioned above. Followed by separation to peptidoglycan fraction (PG) and polysaccharide fraction (PS) were all sensitive which the latter was shown more sensitive percent survival than the former. Composed sugars of polysaccharide fraction were determined to D-glucose, L-rhamnose and D-glucosamine, and the rate fo composition was calculated to about 1:1:1 by the data of elemental analysis, IR, TLC and HPLC.
This experiment was conducted to study the effects of lactic acid bacteria (LAB) inoculation either alone or in combination with cell wall degrading enzymes on the fermentation characteristics and chemical compositions of Rhodesgrass silage. Over to 1 kg of fresh Rhodesgrass sample a treatment of inoculant LAB with or without addition of an enzyme of Acremoniumcellulase (A) or Meicelase (M) or a mixture of both enzymes (AM) was applied. The treatments were control untreated, LAB-treated (application rate $1.0{\times}10^5cfu/g$ fresh sample), LAB+A 0.005%, LAB+A 0.01%, LAB+A 0.02%, LAB+M 0.005%, LAB+M 0.01%, LAB+M 0.02 %, LAB+AM 0.005%, LAB+AM 0.01%, and LAB+AM 0.02%. The sample was ensiled into 2-L vinyl bottle silo, with 9 silages of each treatment were made. Three silages of each treatment were incubated at 20, 30 and $40^{\circ}C$ for 2-months of storage period. All silages were well preserved with their fermentation quality has low pH values (3.91-4.26) and high lactic acid concentrations (4.11-9.89 %DM). No differences were found in fermentation quality and chemical composition of the control untreated silage as compared to the LAB-treated silage. Combined treatment of LAB+cellulases improved the fermentation quality of silages measured in terms of lower (p < 0.01) pH values and higher (p < 0.05) lactic concentrations than those of LAB-treated silages. Increasing amount of cellulase addition resulted in decrease (p < 0.05) of pH value and increase (p < 0.05) of lactic acid concentration. LAB + cellulase treatments (all cellulase types) reduced (p < 0.01) NDF, ADF and in vitro dry matter digestibility of silages compared with the control untreated silages. The fermentation quality and the rate of cell wall reduction were higher (p < 0.01) in the silages treated with LAB + cellulase A than in the silages treated with either LAB+cellulase M or LAB + cellulase AM. Incubation temperature of $40^{\circ}C$ was likely to be more appropriate environment for stimulating the fermentation of Rhodesgrass silages than those of 20 and $30^{\circ}C$.

In order to evaluate the nutritive value of the forage plants in South Sulawesi, Indonesia, 266 samples (61 grasses and 65 legumes grown in the dry season, 60 grasses and 80 legumes grown in the rainy season) were collected from the highland and lowland in 1998 to 2000, and were subjected to the determination of chemical composition and digestibility. The least-squares analysis of variance demonstrated that the in vitro dry matter digestibility (IVDMD) of grasses was not significantly affected by season or altitude. On the other hand, the some proximate components and cell wall components were significantly affected by season and altitude including the season${\times}$altitude interaction. For the legumes, the in vitro neutral detergent fiber digestibility (IVNDFD) and cellulose content were significantly affected by season. On the other hand, the ether extract (EE) content was significantly affected by season and altitude. The interaction of the season${\times}$altitude for IVDMD, of the year${\times}$season for some proximate components and of the year${\times}$season and the season${\times}$altitude for some cell wall components were significant. These results indicate that the forages grown at highland in dry season have a relatively high quality. The means of the total digestible nutrient (TDN) content estimated from IVDMD in grasses and in legumes were 50.3% and 57.4%, respectively, and the crude protein contents were 7.7% and 17.6%, respectively. The correlation coefficients between IVDMD and the contents of crude fiber, neutral detergent fiber and acid detergent fiber were relatively high in all of forage plants, suggesting that these components would provide an accurate prediction of digestibility or TDN content. A close relationship between IVNDFD and lignin content indicates that the lignin would be the most accurate predictor of cell wall digestibility.
To prevent weakly flocculating characters of bottom brewing yeast during first fermentation, various technical investigations were carried out using strain of Saccharomyces cerevisiae. It appeared that the propagation at $10^{\circ}C$ promoted the molecular structure and biochemical composition of cell wall in favor of flocculation. The yeast grown at $20^{\circ}C$ by addition of zinc ion also had a stimulating effect on flocculation behavior during first fermentation cycle. The zinc ion did not influence directly on the changes of cell wall in favor of stronger flocculence. The increased fermentation activity of yeast due to addition zinc ion was rather responsible for the intensified flocculation capacity. It was concluded that the weakly flocculating characters of bottom brewing yeast during first fermentation can be solved by using yeast propagated at $10^{\circ}C$ or by means of yeast by addition of zinc ion during propagation.
The changes of non-cellulosic neutral sugar composition of strawberry during maturation were investigated. Arabinose, xylose, galactose and glucose were the main non-cellulosic neutral sugar of cell wall and increased until ripe stage. The main non-cellulosic neutral sugar of alkali soluble hemicellulose were arabinose, xylose, mammose, galactose. The contents of non-cellulosic neutral sugar of alkali soluble hemicellulose were increased during maturation.

Since the role of lignin in the wood cell wall is to keep integrity and structure rigidity of lignocellulosic substrate, lignin of the cell wall has to be destroyed before enzymatic hydrolysis of wood polysaccharides. The aspen wood meals were delignified with ozone in acidic condition. The chemical characteristics of wood meal were investigated. The 60% of lignin and almost zero % of polysaccharides in aspen wood meal was degraded with 10min. ozone treatment. The phenolic hydroxyl groups of lignin in ozonated wood meal were increased with ozonation time. The sugar composition of ozonated wood meal showed that the hemicellulose was more susceptible to ozonation than cellulose. The yield of aldehyde was increased in some degree with 10min. ozone treatment and decreased with longer ozone treatment.
Two wood decay ascomycetes fungi identified as Hypoxylon mammatum and H. truncatum were isolated from backyard of Korea Research Institute of Chemical Technology (KRICT) in Korea. Hypoxylon truncatum is newly recorded as a wood degrader in Korea. Unusual germination mechanisms of ascospores in H. mammatum and H. truncatum are described and illustrated. The differences between two species were noticed on the process of germ tube formation. In the process of germ tube formation, the fast movement to pigmented ascospores activated from their perispores was termed as spore eclosion that was only found in H. mammatum. This sophisticated recognition mechanism indicated the existence of specific eclosion and germ tube formation due to the composition of cell wall layers and their preferable host derive, based on examined two species under a genus. The observation on present study postulates different composition of wall layers of ascospore and different nutrient composition for germination.
The results that the effect of 6 detergents on the structural changes and biochemical composition of bacterial membrane of Escherichia coli and Bacillus cereus are as follows ; 1. Population growth of the bacteria was increased in case of the treatment with palmitoyl carnitine and sodium deoxy cholate but was increased in case of the treatment with palmitoyl carnitine and sodium deoxy cholate but was decreased by sodium dodecyl sulfate and palmitoyl choline, in E.coli and was decreased by palmitoyl carnitine and palmitoyl choline at the low concentration, in B. cereus. 2. The electron micrograph showed that cell wall lysis or cell collapse were observed in the treatment of sodium dodecyl sulfate and palmitoyl choline, and also cell wall was condensed by triton X-100 and sodium deoxy cholate, in E.coli. And in B. cereus, endospore formation of the bacteria was stimulated by palmitoyl choline, and cell lysis or structural changes of the membrane were observed in the treatment of sodium dodecyl sulfate, sodium cholate, and triton X-100, respectively. 3. As to the effect of detergent on the biochemical composition of biomembrane, the content of carnitine, in E.coli, and B.cereus, the content of structural protein and phospholipid were decreased by treatment of sodium dodecyl sulfate and structural protein was denatured by palmitoyl choline. 4. The profile of membrane protein revealed that the bacterial membrane were composed of various proteins. By dint of this result, some of membrane proteins were solubilized or changed to small molecules by the treatment of sodium dodecyl sulfate and palmitoyl choline, in E.coli and membrane protein of the biomembrane by treatment of sodium dodecyl sulfate, sodium deoxy cholate, palmitoyl choline, and palmitoyl carnitine were confirmed to be different profile as compared with those of the control, in B. cereus. Therefore, it is suggested that sodium dfodecyl sulfate and palmitoyl choline soulbilized biomembranes or inhibited membrane transport and that palmitoyl carnitine and sodium deoxy cholate were used as an energy source or stimulating the membrane transport, in E.coli. And, it is suggested that all of detergents were inhibited biomembrane synthesis, expet saponin, in B.cereus.
