웅담이 mammalian세포의 생육에 미치는 영향 (Effect of bear's gall on mammalian cell growth)

  • 동물세포에 미치는 웅담의 영향을 조사하기 위해서 원숭이 kidney cell 유래의 COS-7 cell과 hybridoma cell(murine myeloma cell과 rat의 spleen cell을 융합)을 가지고 이들 세포의 생육과 단백질 발현기작에 미치는 영향을 조사했다. COS-7 cell과 hybridoma cell을 웅담이 첨가된 10% FCS DMEM complete medium에서 78시간 배양한 결과 COS-7 cell에는 거의 영향을 미치지 않았으나 myeloma cell과 spleen cell과의 융합세포는 48시간내에 거의 모든 세포가 사멸됐다. 또한 cDNA를 COS-7 cell에 transfection에서 발현되는 단백질양을 조사한바 웅담이 첨가된 배지에서의 단백질 생산량이 첨가되지 않은 배지에서 보다 $30{\sim}40%$ 감소되었다.

Fractionated Aged Black Garlic Extracts Enhance Growth of Anti-My-10 Hybridoma Cells and Production of IgG1 Antibody

  • Aged black garlic (ABG) was extracted with 20% ethanol and water (crude extracts) and fractionated into three categories (>10, 3-10, and <3 kDa). The effect of crude extract supplements on anti-My-10 hybridoma cell growth and IgG1 antibody production was investigated in suspension culture with a chemically defined protein-free medium. We observed that supplementation of ABG to the cell culture medium stimulated anti-My-10 hybridoma cell growth and production of IgG1 antibody, particularly with fractionated ABG of low molecular weight. The stimulation depended upon the concentration and the size of the fractionated ABG. We also found that the growth-promoting activity was not correlated with high antibody production. These results suggest that fractionated ABG is a novel and promising alternative as an animal cell culture supplement.

  • Essential oil of Artemisia lavandulaefolia the chrysanthemum family plant used in the chinese medicine was extracted and antibacterial and antifungal activity with many kinds of the pathogenic bacterium and fungi was experimented by it. Ataphylococcus epidermidis and Streptococcus aureus gram positive bacterium at the concentration of 200ppm and Streptococcus mutans at the concentration of 1,000ppm showed the growth injibition effect of the cell. These showed statistically significant difference(p<0.05) Zymomonas mobilis Entrecoccus faecalis gram negative bacterium at the concentration of 200ppm and Pseudomonas putida at the concentration of 400ppm showedd the growth inhibition effect of the cell)p<0.05) V. Parahaemolyticus at the concentration of 800ppm and Pseudomonas aeruginosa at the concentration of 1,000ppm showed the growth inhibition effect of the cell(p<0.05) Candida albicans and Cryptococcus neoformans yeast-type fungi showed the gorwth inhibition effect of the cell at the concentration of 200ppm(p<0.05) Altenaria mali Aspergillus nidulans and Fusarium oxysporum filamentous fungi took the growth inhibition effect of the cell at the concentration of 600ppm, 400ppm, and 100ppm. respectively.

  • In order to obtain the basic data for the preparation of yogurt containing ginseng component, the effect of ginseng component on cell growth of Lactobacillus casei YIT 9018 and on lactic acid production were investigated. Initial cell growth and acid production were markedly inhibited by the addition of ethanol extracts in the level of 8% into 15% skim milk. Crude saponin did not show any inhibitory effect on cell growth and acid production, but ether layer fraction showed inhibitory effect. It was thought to be more advantageous to add ginseng extracts after the fermentation of milk than before. The addition of ginseng extract at 8% level into liquid yogurt was most suitable in organoleptic test. Cell viability was not affected by the addition of ethanol extracts up to 8% level during storage of liquid yogurt.

  • Mouse malignant T-lymphoma CS21 cells can grow when cocultured with CAl2 lymph node stromal cells, but they undergo apoptotic cell death with DNA fragmentation when separated from CA12 stromal cells. In the course of examining the effects of the soluble factor (s) secreted by CAl2 stromal cells on CS2l cell growth, we found that thiols including cysteine promoted CS2l cell growth. P388 cell growth was also promoted by various thiols. These results suggest that thiols including cysteine participate in CA12 and P388 cell growth.

  • Studies were made to investigate effects of the scale-up of stirred tank bioreactors on cell growth and alkaloids production for suspension cultures of Eschscholtzia californica. In the 1.5 L STR, cell lysis was observed at 110 rpm or higher agitation speed. The agitation speed of 30 L STR was 43.7 rpm to maintain the same shear stress developed in 1.5 L STR of 100 rpm. As a result of scale-up from 1.5 L to 30 L STR, the specific growth rate was decreased from 0.12 to 0.07 day-1. The alkaloids productivity was also decreased from 0.24 to 0.14 mg/L-day. Changes of mixing performance and oxygen transfer were studied to explain the decrease of cell growth and alkaloids production. Decreased oxygen transfer rate coefficient(KLa) and increased mixing time by the scale-up was observed at various aeration rates.

  • Ginkgo biloba의 세포배양을 통하여 세포성장과 flavonol glycosides의 생성에 영향을 미치는 여러 조건들을 연구하였다. 다양한 지역으로부터 유도된 세포주들의 세포성장과 flavonol glycosides의 생합성 능력은 서로 차이가 있었다. 이들의 캘러스 및 현탁 세포는 배지조건과 배양조건에 따라 성장 및 f1avonol glycosides 생합성에 많은 영향을 받았었다. 특히 배양조건 중 빛의 영향은 현저하여 명조건에서의 f1avonol glycosides의 생성은 암조건보다 10배 이상 증가함을 알 수 있었다.

  • The growth characteristics of Candida tropicalis KIST 351 on gas oil substrate under different culture conditions were investigated and the preliminary animal feeding experiments using this yeast as a partial substitute of fish meal was also conducted. The yeast assimilates effectively n-paraffins in gas oil ranging from $C_{16}$ to $C_{16}$ with its maximum cell growth at $33^{\circ}C$ and pH 5.5 with aeration of 3 vvn and agitation of 900 rpm. The optimal concentrations of nitrogen sources, $HK_2PO_4$ and $Na_2HPO$ were 4, 2 and 0.5g/1, respectively. Ferrous sulfate, manganese sulfate and zinc sulfate showed positive effect to cell growth with the optimal range of 5-10 ppm. In the feeding experiment with 3 and 5% incorporation of the gas oil grown yeast, neither adverse effects on growth of chicks nor toxic effect were observed. Protein content of the dried cell was 58.8% and its amino acid composition compared well with other single-cell protein products and FAO reference protein.

  • The biological fixation of carbon dioxide using microalgae have many advantages over chemicals and remove carbon dioxide simultaneously. A ketocarotenoid astaxanthin is hyper-accumulated in the green freshwater microalga, Haematococcus pluvialis. In the present study, the combine effects of carbon dioxide concentration and light intensity on the growth of H. pluvilais were investigated. The carbon dioxide concentration above 10% caused a severe inhibition and around 5% is optimal for growth. Adaptation to high concentration of carbon dioxide enhanced the $CO_2$ tolerance. Specific growth rate calculated differently based upon cell number or dry weight because of the distinctive life cycle patterns of H. pluvialis : small-sized motile green cell and thick cell walled red cyst cell. Based on the light dependence of H. pluvialis, internally illuminated air-lift photobioreactor was designed and operated. Gradual increase of light supply gave more active growth and more effective productivity of astaxanthin than constant light supply.

암모니아산화세균 Brevundimonas diminuta의 분리 및 암모니아 산화 특성 (Isolation and Characteristics of Novel Ammonia Oxidizing Bacteria Brevundimonas diminuta)

  • The microorganism for ammonia gas removal was isolated from composting product. This was identified as Brevundimonas diminuta by morphological, biochemical characteristics study and 16S rDNA sequence analysis. Optimal incubation temperature for cell growth and oxidizing ability of $NH_4-N$ was $30^{\circ}C$ and optimal initial pH was 7. Glucose affected the growth of cell and the removal of $NH4^+$. The growth rate of the isolates were increased when grown in the presence of 0.05-1%(w/v) glucose in the selective medium and lurker increases in glucose concentration to 2% caused significant decreases in the cell growth and oxidizing ability of $NH4^+$.
