무즙을 이용한 Candida utilis ATCC 42416균체 생산 (Chinese Radish Juice as a Growth Substrate for the Production of Candida utilis ATCC 42416 Biomass)

  • 단세포 단백질 자원으로 이용 가능한 균주로 알려진 Candida utilis ATCC 42416 균체를 생산하기 위하여 무를 파쇄하여 얻은 무즙을 배지로 균체배양 실험을 하였다. 무즙의 수용성 고형물과 총 당량은 각각 5.0~8.8 Brix와 3.5~6.5%였다. C. utilis ATCC 42416 균주를 무즙에 접종하여 $30^{\circ}C$에서 200rpm으로 진탕 배양하였을 때 24시간 이내에 증식이 끝났으며, 단위 소모당에 대한 최대 균체 생산량은 당농도 1% 회석 무즙에서 L당($5\;\times\;DCRI$) 21.5g의 건조균체를 생산하였다. 5배 회석무즙에 약간의 영양소 glucose(2.0%), yeast extract(0.2%), peptone(0.2%), ammonium sulfate(0.2%) 및 $KH_2PO_4(0.2%)$ 등을 각각 보강하여도 증식 속도, 균체생산 및 세포 단백질 함량 등에 차이가 거의 없어서 무즙자체가 C. utilis ATCC 42416 균체 생산에 좋은 기질이었음을 알 수 있다.

  • Using fossil fuels in existing industrial systems causes a variety of social problems. Recently, many studies have been conducted on bio-refineries, which aim to actively utilize biomass to reduce the use of fossil fuels and solve various social problems. Among them, research using microalgae as a third-generation biomass has attracted considerable attention. Microalgae use inorganic matter to produce organic matter, and cell destruction is necessary to extract useful organic materials from microalgae. The extracted organic materials are currently used in various industrial fields. Numerous cell-destruction methods exist. We have investigated cell disruption by sonication, especially its efficiency. Ultrasound is a sound wave with frequencies above 20 kHz, and destroys cells by sending high energy through a cavitation that occurs, according to the characteristics of the sound wave. The Dunaliella salina microalgae used in this study was cultured in a flat-type photobioreactor. Experiments were performed using a batch low-frequency processing device. Logistic model was applied to analyze the results of cell-destruction experiments using ultrasound. The proper conditions for the most efficient cell destruction were OD 1.4(microalgae concentration)), 54watt(output power) and 200mL(microalgae capacity).

  • A novel and simple purification method for microbial $poly-{\beta}-hydroxybutyrate$ (PHS) was developed. Sodium hydroxide was found to be efficient for digesting cell materials. Initial biomass concentration, NaOH concentation, digestion time, and incubation temperature were optimized. When 40 g/l of biomass was incubated in 0.1 N NaOH at $30^{\circ}C$ for 1 h, PHB purity of 88.4% with a weight average molecular weight ($M_w$) of 770,000 and a polydispersity index (PI) of 2.4 was recovered with a yield of 90.8% from the biomass which initially contained PHB of a $M_w$ of 780,000 and a PI of 2.3.

도광판의 간격이 Chlorella vulgaris 증식에 미치는 영향 (Effect of Optical Panal Distances on the Growth Rate of Chlorella vulgaris in a Photobioreactor)

  • 본 연구는 도광판의 간격이 Chlorella vulgaris (C. vulgaris)의 증식률에 미치는 영향을 알아보고자 하였다. C. vulgaris (FC-16) ($3-8{\mu}m$)는 Jaworski's Medium 에 $22^{\circ}C$에서 15일 증식시킨 뒤 사용하였다. 실험을 위하여 네 개의 샘플을(Run 1: 225 mm 간격, Run 2: 150 mm 간격, Run 3: 112.5 mm 간격, Run 4: 90 mm 간격) 준비하여 도광판의 간격이 C. vulgaris의 증식률에 미치는 영향을 비교 실험하였다. 광효율은 Run 1이 86%, Run 2가 90%, Run 3가 92% 그리고 Run 4가 94%의 광효율을 나타내어 도광판의 간격이 짧을수록 광효율은 높았으며, 도광판의 간격에 따라 Run 1은 7일째, Run 2는 5일째, Run 3는 3일째 그리고 Run 4는 2일째에 최대 바이오매스 증식률을 나타내었다. 또한, 최대 증식률은 Run 4가 Run 1에 비해 3배 높았다. 그러나, 증식속도, 셀 당 클로로필 함량 및 단위면적 당 Cell volume 그리고 Doubling time은 Run 1과 Run 2에 비해 Run 3와 Run 4가 높았지만 Run 3과 Run 4는 크게 차이가 없었다. 따라서 경제적인 부분을 고려한다면 바이오매스의 대량생산을 위하여 Run 3의 사용이 가장 효율적이라 생각된다. measured, and the results for the toxic effect was not observed.

  • Rhodotorula marina IFO 0879의 균체생산과 균체 내 유지생산을 위한 최적배양조건에 관하여 조사하였다. 균체량과 유지생산량은 $30^{\circ}C$, pH $6.0{\sim}7.5$에서 최고량을 나타내었다. $30^{\circ}C$에서 8일간 진탕배양하는 동안 질소성분이 어느정도 소모되어 균체가 증식한 후 지방질의 생성이 시작되었으며, 이때 최대균체량은 9.82g/l이고 세포내 지방질은 효모균체에 대하여 최고 35.4%까지 축적하였다. 탄소원으로서 유당(乳糖)과 포도당을, 질소원으로서 ammonium sulfate를 사용하였을 때 균체량과 지방질 함량이 가장 많았다. 배양액의 질소농도가 높을수록 균체 지방함량은 감소하고 균체량이 증가하였으며, 농도가 낮을수록 반대로 되었다. 배양액에 주기적으로 당을 첨가했을 때 초기에는 균체량과 지방질생성량이 대조구보다 적었으나 후기에는 더 많았다. 생산된 지방질의 주지방산은 palmitic acid(20.3%), oleic acid(46.6%) 및 linoleic acid(16.2%)이었다.

Influence of $CO_2$ on Growth and Hydrocarbon Production in Botryococcus braunii

  • Botryococcus braunii is a green colonial fresh water microalga and it is recognized as one of the renewable resources for production of liquid hydrocarbons. CFTRI-Bb-l and CFTRI-Bb-2 have been reported for the first time and their performance with regard to growth and biochemical profile is presented here. The present study focused on effect of carbon dioxide $(CO_2)$ on biomass, hydrocarbon, carbohydrate production, fatty acid profile, and carotenoid content in various species of B. braunii (LB-572, SAG 30.81, MCRC-Bb, N-836, CFTRI-Bb-l, and CFTRI-Bb-2) at 0.5, 1.0, and 2.0% (v/v) levels using a two-tier flask. $CO_2$ at 2.0% (v/v) level enhanced growth of the organism, and a two-fold increase in biomass and carotenoid contents was observed in all the B. braunii strains studied compared with control culture (without $CO_2$ supplementation). At 1 % and 2% (v/v) $CO_2$ concentrations, palmitic acid and oleic acid levels increased by 2.5 to 3 folds in one of the strains of B. braunii (LB-572). Hydrocarbon content was found to be above 20% at 2% $CO_2$ level in the B. braunii LB-572, CFTRI-Bb-2, CFTRI-Bb-l, and N-836 strains, whereas it was less than 20% in the SAG 30.81 and MCRC-Bb strains compared with control culture. This culture methodology will provide information on $CO_2$ requirement for growth of algae and metabolite production. B. braunii spp. can be grown at the tested levels of $CO_2$ concentration without much influence on culture pH.

  • 균체 생산성 실험과 chitinase 생산성 실험을 비교해 볼 때, chitinase만을 생산하는 조건 에서는 배지성분에 chitin을 첨가해 주는 것이 좋으나, 해충 방제용으로 살균력을 증진시키기 위하여 균체량과 chitinase의 생성량 및 산업적, 경제적 사용이 용이한 배지를 고려할 때에는 쌀겨와 밀기울이 첨가된 배지가 좋은 배지임을 알 수 있었다. 또한 이 배지를 이용하였을 경우 균체는 1X$10^8$ cfu/g, chitinase는 370mU/g로 생산되었으며 생물검정결과 53-64%의 탁월한 살충효과를 확인 할 수 있었다.

Effects of Nutrient Levels on Cell Growth and Secondary Carotenoids Formation in the Freshwater Green Alga, Chlorococcum sp.

  • The freshwater green alga Chlorococcum sp. grew on NH_4^{+},{\;}NO_3^{-}$, urea, yeast extract, and peptone as the nitrogen source showing similar pattens of growth and secondary carotenoid (SC) production. However, the most suitable nitrogen source for the induction fo SC was urea. The dffects of nutrient levels (urea, phosphate, sulfate, ferrous iron, and salt) on growth and SC production were stydied by varying the concentration of each nutrient in batch cultures. High biomass production was achieved in cultures containing 20-28 mM urea, 4.8-10 mM phosphate, 1.6 mM sulfate, 70 mM phosphate, 1.6 mM sulfate, 170 mM NACl, and $50{\;}\mu\textrm{M}$ iron. The optimum concentrations of nutrients for biomass and for the SC accumulation in biomass were evaluated and the two media for achieving high biomass production and SC production were thus developed. The extent to which each parameter to stimulate the formation of SC in the alga were varied and the potentially improned SC prodution by manipulating the nutrient levels in the modified media were descussed.

Power Sharing and Cost Optimization of Hybrid Renewable Energy System for Academic Research Building

  • Renewable energy hybrid systems look into the process of choosing the finest arrangement of components and their sizing with suitable operation approach to deliver effective, consistent and cost effective energy source. This paper presents hybrid renewable energy system (HRES) solar photovoltaic, downdraft biomass gasifier, and fuel cell based generation system. HRES electrical power to supply the electrical load demand of academic research building sited in $23^{\circ}12^{\prime}N$ latitude and $77^{\circ}24^{\prime}E$ longitude, India. Fuzzy logic programming discover the most effective capital and replacement value on components of HRES. The cause regarding fuzzy logic rule usage on HOMER pro (Hybrid optimization model for multiple energy resources) software program finds the optimum performance of HRES. HRES is designed as well as simulated to average energy demand 56.52 kWh/day with a peak energy demand 4.4 kW. The results shows the fuel cell and battery bank are the most significant modules of the HRES to meet load demand at late night and early morning hours. The total power generation of HRES is 23,794 kWh/year to the supply of the load demand is 20,631 kWh/year with 0% capacity shortage.

Rhodobactor sphaeroides의 수소생산에 미치는 광세기 및 광합성 배양기 영향 (Influence of light intensity and photo-bioreactor design for photo biological hydrogen production by Rhodobactor sphaeroides)

  • Purple non-sulfur bacteria, Rhodobacter sphaeroides KD131 grew to reach the maximum cell concentration in 45 hrs of incubation in the synthetic media containing (NH4)2SO4, L-aspartic acid and succinic acid as the carbon and nitrogen sources, respectively, at 30oC under 8 klux irradiance using halogen lamp. The strain produced hydrogen from the middle of the logarithmic growth phase and continued until the cell growth leveled out. The strain grew and produced hydrogen under the irradiance of 3-30 klux, but cell growth was inhibited over 100 klux. In addition, anaerobic/light culture condition was better than the aerobic/dark on the hydrogen production. Among various photo-bioreactors examined, the flat-vertical reactor manufactured using clear acrylic plastic material showed the best hydrogen production rate at the given culture condition.
