Isolation and Characterization of Pigment-deficient Mutants from Azomonas agilis PY101

  • To investigate the mechanism of cadmium tolerance in a cadmium-resistant Azomonas agilis PY101 that produces a specific fluorescent pigment promoted by cadmium, we carried out Tn5 mutagenesis and isolated four pigment-deficient mutants. In these mutants, Ppg1, Ppg2, and Ppg3 remarkably reduced the pigment production to 15.3%, 11.2%, and 13.9%, respectively. Especially, Ppg4 mutant did not produce the pigment at all. None of the mutants grew in the presence of 1500 ppm of CdCl2 in growth medium, and they exhibited differential sensitivities to cadmium. Ppg1, Ppg2, Ppg3, and Ppg4 mutants were sensitive to 900 ppm, 1100 ppm, 1000 ppm, and 800 ppm of CdCl2, respectively. These mutants also showed noticeable increase, from 8.8-fold to 13.2-fold, in the size of growth inhibition zone compared with that of the will type after treatment with cadmium. Therefore, the pigment production of A. agilis PY101 was found to decrease the toxic effects of cadmium to the bacterium.

Phosphate Associated Cadmium Immobilization Mechanism Depending on the Original Concentration of Cd in Soil

  • Adsorption and precipitation of cadmium (Cd) could be dependent on rate of P addition and Cd level in soil. Therefore, the objective of this study was to examine how addition rate of P affect mechanisms of Cd immobilization such as adsorption and precipitation in different levels of Cd in soil. Arable soils were spiked with inorganic Cd ($CdCl_2$) to give a total Cd concentration of 10, 100, and $1,000mg\;Cd\;kg^{-1}$. Monopotassium phosphate ($KH_2PO_4$, MPP) was selected as phosphate material and mixed with the pretreated arable soil at the rates of 0, 800, 1,600 and $3,200mg\;P\;kg^{-1}$. The mixture soils were incubated at $25^{\circ}C$ for 8 weeks in dark condition. Soil pH decreased with increasing MPP addition rate in all levels of Cd but negative charge of soil increased, thereby reducing 1 M $NH_4OAc$ extractable Cd. Soil solutions were undersaturated with respect to $CdCO_3$ and $Cd_3(PO_4)_2$ with all P addition rate in soil with low Cd level (${\leq}100mg\;Cd\;kg^{-1}$) but supersaturated in soil with high Cd level ($1,000mg\;Cd\;kg^{-1}$). From the above results, Cd solubility was controlled by precipitation of Cd minerals such as $CdCO_3$ and $Cd_3(PO_4)_2$ in soil with high Cd level but by Cd adsorption induced by increase in negative charge of soil with low level of Cd.

Inbreeding and Genetic Diversity in Three Imported Swine Breeds in China Using Pedigree Data

  • The accumulation of inbreeding and the loss of genetic diversity is a potential problem in the modern swine breeds in China. Therefore, the purpose of this study was to analyze the pedigrees of Chinese Duroc (CD), Landrace (CL) and Yorkshire (CY) swine to estimate the past and current rates of inbreeding, and to identify the main causes of genetic diversity loss. Pedigree files from CD, CL and CY containing, 4529, 16,776 and 22,600 records, respectively, were analyzed. Pedigree completeness indexes of the three breeds, accounting for one generation back, were 83.72, 93.93 and 93.59%, respectively. The estimated average annual inbreeding rates for CD, CL and CY in recent three years were 0.21, 0.19 and 0.13%, respectively. The estimated average percentage of genetic diversity loss within each breed in recent three years was about 8.92, 2.19, and 3.36%, respectively. The average relative proportion of genetic diversity loss due to unequal contributions of founders in CD, CL and CY was 69.09, 57.95 and 60.57%, and due to random genetic drift was 30.91, 42.05 and 39.43%, respectively. The estimated current effective population size for CD, CL and CY was 76, 117 and 202, respectively. Therefore, CD has been found to have lost considerable genetic diversity, demanding priority for optimizing the selection and mating to control future coancestry and inbreeding. Unequal contribution of founders was a major cause of genetic diversity loss in Chinese swine breeds and random genetic drift also showed substantial impact on the loss of diversity.

브라디키닌 수용체에 관한 연구

  • 브라디키닌은 체내에서 강력한 혈관 확장 작용을 일으키는 autacoid(local hormone)로서 혈압의 항상성 유지, 모세혈관 투과성 증진, 염증 및 통증 반응 등에 관여하고 여러 장관 평활근을 수축시킨다. 또한 septic 혹은 endotoxic shock의 여러 원인 물질로도 생각되어진다. 최근 rhinovirus로 인한 감기의 제증상 원인 물질로도 브라디키닌이 주목을 받고있다. 이와같이 브라디키닌온 다양한 질병에 있어 중요한 원인 물질로 여겨지므로 브라디키닌 길항제들은 한두 질병의 치료제로 개발될 가능성이 높음이 강력히 제시되고 있는 실정이다. 이의 개발을 위해서 브라디키닌 수용체에 대한 연구는 필수적이라고 할 수 있다. 본 연구는 두 부분으로 나누어 진행중인데 첫째, 이제까지 보고된 브라디키닌 길항 물질들은 대부분이 브라디키닌의 특정 아미노산 잔기를 치환시킨 펩타이드 유도체로서 이들을 경구 투여시 peptidase어 의하여 쉽게 분해되고 또한 부분적인 효능제 활성을 갖는 불리한 점을 감안하여, 비펩타이드성 브라디키닌 길항제를 개발할 목적으로 한방 및 민간에서 자주 사용되어온 생약중에 브라디키닌 작용에 선택적 길항효과가 있는 물질을 검색한 바 활성을 보인 황금으로부터 작용 성분을 추적중에 있다. 둘째, 브라디키닌 수용체를 순수하게 분리 정제하기 위한 첫 단계로서 이 수용체의 결합시험(binding assay) 방법을 확립하고 더불어 여러 조직내(흰쥐의 여러 기관, 토끼 및 사람의 신장)의 브라디키닌 수용체의 분포를 파악하는 일이다. 횐쥐 조직의 실험 결과로부터 신장에 브라디키닌 수용체가 많이 분포함을 확인되었고 향후 토끼 신장으로부터 동수용체를 분리하고자 한다. 또한 토끼 신장의 근위세뇨관일차배양세포을 이용하여 브라디키닌의 신장에서의 작용기전도 살펴보고 있다.+}$, $Na^{+}$, NH$_{4}$$^{+}$ 및 H$^{+}$) 수용액 메디움에서의 Cd(II), Mg(II) 및 Zn(II)의 Dowex 1-X8, Cl$^{-}$ 수지에 대한 흡착은 역시 어떤 메디움에서도 Cd(II) 흡착이 제일 크며, 다음이 Zn(II) 이고 착이온을 형성않는 Mg(II)이 제일 작았다. 한편 메디움 종류별 D값의 크기순위는 H$^{+}$>K$^{+}$> $Na^{+}$>NH$_{4}$$^{+}$이였다. 메디움의 종류에 따라 D값의 차이가 나는 것은 금속이온의 착이온 형성과 금속이온의 용액내에서의 이온종의 상태와 관련이 있다고 생각된다. 마. MCl(M:K$^{+}$, $Na^{+}$, NH$_{4}$$^{+}$ 및 H$^{+}$)과 MNO$_{3}$ 용리액에 의한 Cd(II), Mg(II) 및 Zn(II)의 용리는 예상한 바와 같이 MCl에서 작은 Dv 값을 갖는데, 이것은 CdCl$_{4}$$^{2-}$ 착이온을 형성하거나 ZnCl$_{4}$$^{2-}$ , ZnCl$_{3}$$^{-}$같은 이온과 MgCl$^{+}$, MgCl$_{2}$같은 이온종을 형성하기 때문인것 같다. 한편 어떠한 용리액에서던지 NH$_{4}$$^{+}$의 경우

NC 가공을 위한 곡면 모델링에 관한 연구

  • 자동차, 항공기, 선박등과 같은 산업제품과 전화기, TV브라운관과 같은 가전제품, 심미적 기능을 갖는 제품등과 같은 일상용품등은 많은 부분이 자유곡면(sculptured surface)으로 이루어져 있다. 이러한 해석적으로 정의 하기 어려운 제품 또는 그것을 생산하는데필요한 금형을 가공하는데 있어서 기존에는 석고 모형이나 목형을 이용한 모방 절삭을 하였다. 그러나 근래에는 자유곡면으로이루어진 제품을 설계하고 가공하는데 있어서 CAD/CAM system을 이용하고 있다. 제품의 곡면을 표현하거나 이들을 NC가공하기 위해서는 곡면을 나타내는 형상 정보가 필요 하고 이들 형상정보로 곡면모델링을 하여공구 경로를 구한다. 그러므로 이들 형상정보를 허용한도내에서 적절히 정하여 실제형상에 가까운 곡면을 형성하여야 한다. CAD/CAM 기술의 발달에 따라 다양한 형태의 곡면을 형성하는 기능을 가진 system이 많이 출현되었고 점토, 석고 또는 나무등으로 만든 physical model로 부터 얻어진 형상정보에 의해 surface fitting을 함으로서 자유곡면을 표현하는 방법이 많이 사용되고 있다. 어떠한 곡면을 표현할때는 곡면의 특성을 잘 표현하면서 전체적으로 smooth 한 것이 바람직하다. 그러므로 곡면의 형태를 설계자가 쉽게 이해할 수 있고 적은 수의 patch로도 복잡한 형상을 나타내야 하며 또한 국부적으로 수정이 용이하여야한다. 본 논문에서는 자유곡면을 나타낼 수 있는 수학적 표현 방법에 관하여 논하고, 해석적 곡면으로 부터 형상정보를 얻어 곡면 모델링을 한 후의 Ferguson곡면, UBS와 NUBS의 차이점을 분석하고, 이들 곡면과 해석적 곡면으로부터 얻은 실제곡면과의 오차를 측정하여 실제형상에 가까운 모델링 곡면을 찾고자 하였다.. 라. Optimum Process Condition by Experimental Design Method for the Synthesis of Methylchlorosilanes(MCS) (실험계획법에 의한 메틸염화실란 합성의 최적공정조건)

  • An optimum synthetic condition was studied for the MCS used as a silicone monomer. The contact mixture was made from the four component catalyst system($CuCl/ZnCl_2/Sn/Cd$) and silicon particles. The contact mass was used for a series of experiments with methyl chloride, which were designed and done to explore the optimum condition for MCS synthesis by an experimental design method. The optimum temperature and MeCl flow rate, which were obtained using 50g contact mass at 60rpm and 1 atm, were in the range of $300-305^{\circ}C$ and of 70-80ccm. Also a continuous run was performed to confirm the conditions. The results showed that the average reaction rate and selectivity were 170(g-MCS/hr.kg-Si) and 0.05 respectively at 67% conversion of MeCl and 92% silicon utilization rate. Also the parameters of overall reaction rate equation and a total pressure were estimated on the basis of the results of the continuous run.

Response of Photobacterium phosphoreum to Heavy Metal (Photobacterium phosphoreum의 중금속에 대한 반응성)

  • Photobacterium phosphoreum was used in order to study response to heavy metal including $HgCl_2$, $CdCl_2$, $MnSO_4$ and $ZnSO_4$ in view of developing monitoring system for toxic substances. The concentrations of heavy metal causing 50% reduction($EC_{50}$) in bioluminescence intensity were determined with both free and immobilized P. phosphoreum. The bioluminescence responses were examined at various concentrations of heavy metal after 10, 20 and 30 min of exposure. The linear correlation between Gamma values and concentrations of heavy metal was obtained and $EC_{50}$ was calculated from the linear correlation. The significant inhibitory concentrations for bioluminescence emission were found to be 0.05mg/L for $HgCl_2$, 25mg/L for $CdCl_2$, 50mg/L for $MnSO_4$ and 12.5mg/L for $ZnSO_4$, respectively. The free cell and disc type were shown to be more sensitive to heavy metal than cells mixed with Na-alginate or immobilized on Sr-alginate. However, the linear regression curves were derived from the Sr-alginate immobilized cells indicating the immobilization method is a useful tool for monitoring of heavy metal under more stable condition of bioluminescence.

Forward-Mode $Na^+-Ca^{2+}$ Exchange during Depolarization in the Rat Ventricular Myocytes with High EGTA

  • During depolarization, extrusion of $Ca^{2+}$ from sarcoplasmic reticulum through forward-mode $Na^+-Ca^{2+}$ exchange was studied in the rat ventricular myocytes patch-clamped in whole-cell configuration. In order to confine the $Ca^{2+}$ responses in a micro-domain by limiting the $Ca^{2+}$ diffusion time, rat ventricular myocytes were dialyzed with high (14 mM) EGTA. $K^+$ current was suppressed by substituting KCl with 105 mM CsCl and 20 mM TEA in the pipette filling solution and by omitting KCl in the external Tyrode solution. $Cl^-$ current was suppressed by adding 0.1 mM DIDS in the external Tyrode solution. During stimulation roughly mimicking action potential, the initial outward current was converted into inward current, $47{\pm}1%$ of which was suppressed by 0.1 mM $CdCl_2.$ 10 mM caffeine increased the remaining inward current after $CdCl_2$ in a cAMP-dependent manner. This caffeine-induced inward current was blocked by $1\;{\mu}M$ ryanodine, $10\;{\mu}M$ thapsigargin, 5 mM $NiCl_2,$ or by $Na^+\;and\;Ca^{2+}$ omission, but not by $0.1\;{\mu}M$ isoproterenol. The $I{\sim}V$ relationship of the caffeine-induced current elicited inward current from -45 mV to +3 mV with the peak at -25 mV. Taken together, it is concluded that, during activation of the rat ventricular myocyte, forward-mode $Na^+-Ca^{2+}$ exchange extrudes a fraction of $Ca^{2+}$ released from sarcoplasmic reticulum mainly by voltage-sensitive release mechanism in a micro-domain in the t-tubule, which is functionally separable from global $Ca^{2+}{_i}$ by EGTA.

Characteristics of Cadmium Biosorption and Desorption by Brown Marine Algae (갈색해조류에 의한 카드뮴의 흡착 및 탈착 특성)

  • The biosorption and desorption of Cd were carried out using brown marine algae, known as the good biosorbent of heavy metals. The content of alginate bound to light metals could be changed by the physical and the chemical pretreatment of Sargassum fluitans biomass. The Cd uptake was independent of the alginate content. In case of protonated biomass, Cd uptake was the lowest because the alginic acid of biomass was dissolved to cadmium solution during the biosorption. The maximum Cd uptake of Sargassum biomass was ranged from 79 mg/g to 139 mg/g. In case of raw biomass, the higher the alginate content of biomass, the higher was the Cd uptake. 100% of Cd and light metals sorbed in the biomass were eluted at 0.1N HCI(pH 1.1). However, the elution efficiency in $CaCl_2$ and $Ca{(NO_3)}_2$solution was varied by the concentration, the solid to liquid ratio and the pH of calcium solution. The distribution coefficient between Cd and protons in the desorption solution at pH ranged from 1.6 to 2.9 was observed on the constant stoichometric coefficient(1.3).

  • This study was designed to investigate the effects of varying concentrations of $CdCl_2$ on the enzyme activities such as LDH and SOD related with the accumulations of cadmium(Cd) in liver and kidney of BALB/c mice. 1. Cd accumulations in liver and kidey were increased in a dose dependent fashion. And also, this pattern was more conspicuous in kidney than that in liver. 2. LDH activities of liver and kidney were increased in a dose dependent fashion except for that of liver in 200 ppm Cd group which was similar to that of 25 ppm Cd group. 3. SOD activities of Cd exposed liver and kidney in Cd-fed mice similar to those of controls except for elevation of SOD activity of liver in 25 ppm Cd group. The results of this study suggest that the activities of various enzymes can be modulated by Cd intoxication.
