만성 카드뮴 중독 쥐에서 카드뮴 축적에 미치는 녹차 Catechin의 영향 (Effects of Green Tea Catechin on Cadimium Accumulation in Chronic Cadmium Poisoned Rats)

  • The purpose of this study was to investigate the effects of green tea catechin on the cadmium accumulation in body, cadmium excretion and detoxification functions in chronic cadmium poisoned rats. Sprague-Dawley male rats weighing 100$\pm$10g were randomly assigned to one normal group and three cadmium poisoned groups. Cadmium groups were classified to catechin free diet (Cd-0C group), 0.25% catechin diet(Cd-0.25C group) and 0.5% catechin diet(Cd-0.5C group) according to the levels of catechin supplement. Animals were maintained on 0, 0.25 and 0.5% catechin diets for 20 weeks and simultaneously administered 50ppm Cd(sup)2+ dissolved in the drinking water. Body weight, food intakes and food efficiency ratio in Cd-0C group was lower than the normal group. The accumulation of cadmium in rat liver, kidney, and blood was reduced by catechin supplementation. The excretion of cadmium in urine and feces was increased by catechin supplementation. The metallothionein(MT) contents in liver and kidney were increased in all cadmium groups compared with that of normal group. The ratios of cadmium absorption and retention ratios were significantly decreased in catechin supplementation groups. Accordingly, catechin supplementation resulted to an excretion of cadmium in urine and feces and a lowered accumulation of cadmium in liver and kidney by increasing methallothionein synthesis that led to the significant decrease in cadmium absorption and retention ratios.(Korean J Nutrition 34(4) : 384~392, 2001)

박달나무의 반형매 가계간 Cd Phytoremediation과 축적 특성 (Characteristics of Cd Accumulation and Phytoremediation among Three Half-sib Families of Betula schmidtii)

  • Cd 처리에 대한 박달나무의 반형매 차대간의 Cd 흡수 및 이동 패턴을 알아보고자 수행한 결과, 잎의 주간내 위치에 따른 Cd농도는 지제부에서 높게 나타나고 정단부로 올라 갈수록 농도가 낮아지는 경향을 보였다. 대체로 식물체내에서 Cd의 농도 분포는 뿌리>줄기>잎의 순으로 나타났지만, 가계별, 처리별로 상이한 결과가 나타나 가계간 유전적인 요인이 크게 작용한 것으로 판단된다. 모든 가계의 처리구에서 지상부와 지하부의 Cd 농도 비율이 1.26으로 나타나 박달나무는 Cd을 흡수하여 지상부로 보내는 축적종의 경향을 갖고 있는 것으로 판단된다. 또한 토양 중금속 농도와 식물체내 중금속 농도의 비도 가계간 차이는 있지만, Cd을 능동적으로 흡수하여 지상부로 이동시키는 Cd 정화 수종으로 판단된다. 하지만 박달나무는 Cd에 대한 반응이 가계간 다양하므로 토양 중 Cd정화를 위해 이용할 경우, 정화 능력이 우수한 수종을 선발하는 것뿐만 아니라, 같은 수종 내에서도 내성 능력과 정화능력이 차이가 있으므로 가계 선발이 필요할 것이다.

포도의 부위별 섭취가 흰쥐의 노화 과정 중 Cadmium의 대사 및 중독에 미치는 영향 (Effect of Grape Intake on Cadmium Metabolism of Rats during Aging)

  • This study was carried out to investigate the effect of whole grape, grape pomace and grape juice intake on cadmium (Cd) metabolism during aging in thirteen-month-old Sprague-Dawley male rats. One hundred and twenty rats weighing 548.8 $\pm$ 4.3 g were assigned to eight groups according to body weight and were raised for 3, 5 or 7 months on diets containing 2 % (w/w) dried powders of three different parts of the grape (Campbell Early) and 0.02 % (w/w) CdC12. Food intake tended to decrease with aging, and body weight and epididymal fat pad (EFP) weights of Cd-exposed groups were lower than those of Cd-free groups. Cadmium accumulated in the blood and tissues and Cd concentration was the lowest in the pomace group among Cd-exposed animals. Metallothionein (MT) concentration in the tissues increased through Cd administration. Grape diets, especially grape pomace diets, were effective in decreasing Cd absorption in the tissues by increasing Cd excretion through feces. The intake of grape pomace alleviated the decrease in bone density caused by Cd administration and prevented a decrease in glomerular filtration rates (GFR) with aging. Among the parts of grape, grape pomace, which had highest content of dietary fiber and flavonoids, was the most effective. The results of this study suggest the possibility of using grape pomace as a functional food material, a prospect that previously has been discarded.

카드뮴 급성폭로에 의한 Metallothionein 생성과 독성작용 (Induction of Metallothionein and Toxicity in Acute Cadmium Intoxicated Rat)

  • Thirty five male Sprague-Dawley rats were treated with cadmium chloride solution ranging from 0.2 to 3.2mg $CdCl_2/kg$ by intravenous single injection. At 48 hours after administration of cadmium, total cadmium, MT bound cadmium and histopathologic finding in liver, kidney, lung, heart, testis, metallothionein in liver, kidney and total cadmium in bleed were examined. Tissue cadmium concentration was highest in liver, followed by in kidney, heart, lung and testis. Cadmium bound to rnetallothionein (MT-Cd) and ratio of MT-Cd to total cadmium were increased in liver and kidney dependently of cadmium exposure dose, but not significantly changed in other organs. On histopathologic finding, the most susceptible organ was heart in considering cadmium exposed dose, but testis in considering cadmium concentration. Blood cadmium concentration was increased with dose-dependent pattern, and significantly correlated with tissue cadmium concentration, so that we may estimate tissue cadmium concentration by measurement of blood cadmium concentration. Metallothionein in liver and kidney was increased with dose-dependent pattern, higher in liver than in kidney, and was significantly correlated with tissue cadmium concentration. However, metallothionein induction efficiency of tissue cadmium(${\mu}g\;MT/{\mu}g\;Cd$) was eater in liver than in kidney, and reverse to tissue concentration or exposed dose of cadmium.

Effects of Vitamin E on the Changes of Mineral Contents in Chronic Cadmium-Poisoned Rats

  • This study was carried out to investigate the effects of vitamin E on the cadmium contents of bone and on the calcium and phosphorous contents of the blood, urine and feces. Male Sprague-Dawley rats weighing 100$\pm$10g were randomly assigned to one normal group and three cadmium poisoned groups. The cadmium poisoned groups consisted of a vitamin E free diet (Cd-0E) group; a 40 mg vitamin E /kg diet (Cd-40E) group; and a 400 mg/kg diet (Cd-400E) group. Experimental animals were maintained on their respective diets for 20 weeks and were simultaneously administered 50 ppm $Cd^{2+}$ dissolved in the drinking water. At the end of the trial, the average hematocrit value in the Cd-0E group was 28.13% lower than in the normal group. However, the average hematocrit value in the Cd-400E group was significantly higher than in the Cd-0E and Cd-40E groups. WBC levels in the cadmium-poisoned groups were lower than in the normal group, but Cd-400E group levels were significantly higher than in the Cd-0E and Cd-40E groups. The contents of calcium of tibia has no significant difference between normal group and cadmium exposed group at $10^{th}$ week After 20 weeks, the calcium contents of the tibia in the Cd-0E and Cd-40E groups were lower than in the normal group by 25.5% and 22.1 %, respectively, although the calcium contents of the tibia in the Cd-400E group were higher than in the normal group. After 10 weeks, the calcium contents of the femur in the Cd-0E and Cd-40E groups were 19.25% and 15.45% lower than in the normal group, respectively, but the calcium contents of the femur in the Cd-400E group were at the same levels as in the normal group. The levels of calcium in the femur after 20 weeks were similar to the 10-week levels. Calcium levels of the urine in the Cd-0E and Cd-40E groups were 3.92 fold and 2.92 fold higher, respectively, than in the normal group, but levels in the Cd-400E group were significantly lower than in either the Cd-0E group or the Cd-40E group. Calcium levels of the feces in cadmium-poisoned groups were significantly higher than in the normal group, although levels in the Cd-400E group were significantly lower than in the Cd-0E and Cd-40E groups. Phosphorous levels of the blood in the Cd-0E group were 17% lower than in the normal group, although levels in the Cd-400E group were significantly higher than in the Cd-0E group. Phosphorous levels of the urine in the Cd-0E and Cd-40E groups were significantly higher than in the normal group, while Cd-400E group levels were found to be at the same level as in the normal group. Cadmium contents of the tibia in the Cd-40E and Cd-400E groups were 13% and 17% lower, respectively, than in the Cd-0E group. Regarding cadmium levels in the femur, only the Cd-400E group achieved lower levels (10% lower) than the Cd-0E group. In conclusion, vitamin E supplementation resulted in a suppression of the release of calcium from bone, and a reduction in the excretion of calcium via the urine and feces, thus having a normalizing effect on calcium metabolism in rats with chronic cadmium poisoning.

Purification and Characterization of PC-Like Cadmium-Binding Peptide from Root of Rumex crispus

  • This research investigated the process of removing cadmium and tested the detoxification mechanism of the cadmium-binding peptide (Cd-BP) from Rumex crispus. Phytochelatin-like cadmium-binding peptide (PC-Cd-BP) of Rumex crispus was purified and identified. Rumex crispus was exposed to 4.3 mg Cd/L for seven days. Heat-treated supernatant fraction taken by root tissues showed traces of PC-Cd-BP An analysis of the material through Gel-filteration chromatography on the Sephadex G-75 column showed two symmetrical Cd-BP peaks. The major peak with the smaller molecular weight was further purified by $C_{18}$ reverse-phase HPLC to produce apparent homogeneity. The amino acid composition of Cd-BP from Rumex crispus included cysteine (22.6%), glutamate and glutamate acid (20%), and glycine (12%). It was similar the amino acid composition of most PC. The molecular weight of the purified peptide was determined at 568-706 Da by MALDI-TOF MS. Therefore, the Cd-BP of Rumex crispus was PC-Cd-BP consisting of isopeptides.

오미자 추출물이 카드뮴을 급여한 흰쥐의 대사와 신장내 카드뮴 함량에 미치는 영향 (Effects of the Omija (Schizandra chinensis Baillon) extract on the metabolism and renal Cadmium contents in Cadmium administered rats)

  • 카드뮴에 중톡린 흰쥐에 대한 오미자 추출물의 해독 효과를 알아 보는 본 연구에서 50 ppm의 카드뮴액과 함께 3% 오미자 추출액을 급여한 흰쥐군(CDOE)의 체중증가량과 사료 섭취 효율은 50 ppm의 카드뮴액만을 급여한 흰쥐군(CD)의 이들 측정값과 비교할 때 유의한 차이는 없었다. 그러나 CDOE군은 CD군과 비교하여 감소되었고 신장 무게는 정상흰쥐와 같은 수준으로 회복하였고 GOT 활성도 역시 정상 흰쥐와 같은 수준으로 감소를 보였다. 이로써 오미자는 카드뮴 중독 흰쥐에서 신장등의 장기내 카드뮴 축적을 감소시켜 카드뮴 중독 작용에 대한 경감 효과를 갖는 것으로 나타났다.

Animals were raised for 20 weeks. Cadmium was supplied in drinking water which contained 50ppm Cd$^{2+}$. Effects of catechin were analyzed on changes of mineral contents in chronic cadmium poisoned rats by determining the calcium accumulation in bones, blood, urine and faces and phosphorus In blood and urine. Cd-poisoning inducted the decrease of red blood cell(RBC), white blood cell(WBC), contents of blood hemoglobin and hematocrit, but the levels of those indices were increased by catechin supplementation. The contents of tibia and femur in Cd-0C group was significantly lower than in normal group, but those of catechin supplemetation group was similar to normal group. The calcium contents of urine and faces were higher in Cd-poisoned groups than in normal group, but they was lowered by catechin supplementation. The phosphorus contents of blood and urine in Cd-0C group was significantly lower than in normal group, but that of catechin supplementation group was similar to normal group. Catechin supplementation improved the calcium metabolism in chronic cadmium poisoned rats by increasing the contents of minerals such as calcium and phosphorus in blood and femur and by lowering the urinary and fecal calcium.m.

Cadmium Selenide Nanoparticles을 함유하는 액상실리콘 고무의 제조와 형광특성 (Preparation and Photoluminescence Characteristics of Liquid Silicone Rubber Containing Cadmium Selenide Nanoparticles)

  • Poly [(dimethylmethylvinyl)siloxane 공중합체(PMViS)를 phosphorus oxychloride $(POCl_3)$와 반응시켜 poly [(dimethylmethylvinyl) siloxane] phosphine oxides (PMViSPO)를 제조하였다. Cadmium selenide (CdSe)는 cadmium oxide(CdO), tetradecylphosphonic acid(TDPA), trioctylphosphine oxide(TOPO)를 $300^{\circ}C$에서 반응시키고 여기에 Se를 용해시킨 tributylphosphine(TBP)과 trioctylphosphine(TOP)을 가한 다음 $320^{\circ}C$에서 반응시켜 제조하였다. 또한 CdSe 제조 용액에 PMViSPO를 가하여 CdSe-SPO adduct를 제조하였다. 백금 촉매 존재 하에서 $\alpha,\omega-vinyl$ poly(dimethylsiloxane) (VPMS), HPMS, CdSe 또는 CdSe-SPO를 고속 교반기에 취하고 컴파운딩하여 CdSe 함유 액상실리콘 고무 composite (LSRC-1)와 CdSe-SPO 함유 LSR composite (LSRC-2)를 제조하였다. 제조한 LSR composites 내에 함유된 형광 물질인 CdSe nanoparticles의 분산형태를 측정하여 입자 크기가 $30\sim50nm$인 입자가 균일하게 분포되어 있음을 확인하였고 LSRC-2의 분산도가 LSRC-1보다 우수함을 확인하였다. 또한 CdSe 입자의 개수를 측정한 결과 동일한 면적에 대하여 166개와 202개로 보다 많은 개수의 CdSe가 LSRC-2에 함유됨을 알 수 있었다. LSR composites의 열적 특성을 측정한 결과 CdSe-SPO가 함유된 LSRC-2의 열안정성이 높게 나타났다.

Effect of Chlorella vulgaris intake on cadmium detoxification in rats fed cadmium

  • The aim of this study was to investigate if dietary Chlorella vulgaris(chlorella) intake would be effective on cadmium(Cd) detoxification in rats fed dietary Cd. Fourteen-week old male Sprague-Dawley(SD) rats weighing $415.0{\pm}1.6\;g$ were randomly divided into two groups and fed slightly modified American Institute of Nutrition-93 Growing(AIN-93G) diet without(n=10) or with(n=40) dietary Cd(200 ppm) for 8 weeks. To confirm alteration by dietary Cd intake, twenty rats fed AIN-93G diet without(n=10) and with(n=10) dietary Cd were sacrificed and compared. Other thirty rats were randomly blocked into three groups and fed slightly modified AIN-93G diets replacing 0 (n=10), 5 (n=10) or 10% (n=10) chlorella of total kg diet for 4 weeks. Daily food intake, body weight change, body weight gain/calorie intake, organ weight (liver, spleen, and kidney), perirenal fat pad and epididymal fat pad weights were measured. To examine Cd detoxification, urinary Cd excretion and metallothonein (MT) concentrations in kidney and intestine were measured. Food intake, calorie intake, body weight change, body weight gain/calorie intake, organ weight and fat pad weights were decreased by dietary Cd intake. Urinary Cd excretion and MT concentrations in kidney and small intestine were increased by dietary Cd. After given Cd containing diet, food intake, calorie intake, body weight change, body weight gain/calorie intake, organ weights and fat pad weights were not influenced by dietary chlorella intake. Renal MT synthesis tended to be higher in a dose-dependent manner, but not significantly. And chlorella intake did not significantly facilitate renal and intestinal MT synthesis and urinary Cd excretion. These findings suggest that, after stopping cadmium supply, chlorella supplementation, regardless of its percentage, might not improve cadmium detoxification from the body in growing rats.
