Forskolin의 흰쥐적출관류부신으로 부터 Ach, Excess $K^+$, DMPP, McN-A-343에 의한 Catecholamine 분비효과의 증강작용 (Forskolin-Induced Potentiation of Catecholamine Secretion Evoked By Ach, DMPP, McN-A-343 and Excess $K^+$ From the Rat Adrenal Gland)

  • Adenylate cyclase 효소를 활성화시키는 약물인 Forskolin의 흰쥐 적출관류 부신으로부터 Ach, excess $K^+$, McN-A-343 및 caffein에 의한 catecholamines (CA) 분비작용에 대한 영향을 검색하고, 그 기전을 규명코자 연구를 시행하여 다음과 같은 연구결과를 얻었다. Forskolin (1.0 uM)은 흰쥐 부신적출정맥내로 1분동안 관류시킨 후 Ach(50 ug), excess $K^+$(56 mM), DMPP (100 uM) 및 caffeine (0.3 mM)에 의한 CA 분비작용을 현저히 증강시켰으나 McN-A-343에 의한 CA분비작용에는 영향을 미치지 않았다. Forskolin 자체는 CA분비작용을 일으키지 못하였다. 또한 세포의 calcium을 제거한 상태에서도 위 약물에 의한 CA분비작용에 대하여 유의한 증강작용을 나타내었다. 그러나 McN-A-343의 CA작용에는 영향이 없었으나 위의 약물의 CA분비작용을 유의하게 강화시켰다. Cyclic AMP를 증가시키는 약물로 알려져 있는 dibutyryl cyclic AMP (DBcAMP)는 300 uM농도를 1분간 관류시 Ach, excess $K^+$ 및 DMPP의 CA 분비작용을 뚜렷하게 증강시켰으나 McN-A-343 및 caffeine의 CA분비에는 별다른 영향이 없었다. DBcAMP 자체도 CA분비작용에는 영향을 미치지 못하였으나 또한 DBcAMP는 세포의 칼슘제거시에도 위의 약물에 의한 CA분비작용을 의의있게 증강시켰다. 그렇지만, McN-A-343의 CA분비작용은 증강시키지 못하였다. 이상의 연구결과로 보아 Forskolin는 adenylate cyclase를 활성화 시킴으로써 cyclic AMP 농도를 증가시켜 세포내로 칼슘유입을 증강시키며, 또한 세포내의 칼슘이동에도 관여함으로써 cholinergic nicotinic stimulation 및 depolarization에 의한 CA분비작용을 상승시키는 것으로 사료되어진다.

  • The present study was designed to compare the effects of conotoxin GVIA, a selective blocker of N-type voltage-dependent calcium channels (VDCC) and cilnidipine, a blocker of both L- and N-type VDCC, on the secretion of catecholamines (CA) evoked by cholinergic stimulation and membrane-depolarization in the isolated perfused rat adrenal gland, and also to establish the mechanism of action. 1. The inhibition of the CA secretory response evoked by acetylcholine (5.32 x 10$\^$-3/ ${\mu}$M) was stronger in cilnidipine-treated glands than in conotoxin GVIA-treated glands. (omitted)

  • The present study was designed to investigate the effects of green tea extract (GTE) and epigallocatechin gallate (EGCG) on secretion of catecholamines (CA) in the isolated perfused rat adrenal gland. In the presence of GTE (100 ${\mu}$g/$m\ell$) into an adrenal vein for 60 min. CA secretory responses evoked by ACh (5.32 mM), high K+ (56 mM) and Bay-K-8644 (10 ${\mu}$M for 4 min) from the isolated perfused rat adrenal glands were greatly inhibited in a time-dependent fashion. 흰쥐 적출 부신에서 DMPP 및 McN-A-343의 Catecholamine 분비작용에 관한 연구 (Studies on Secretion of Catecholamines Evoked By DMPP and McN-A-343 in the Rat Adrenal Gland)

  • 흰쥐 적출 부신에서 DMPP와 McN-A-343의 카테콜아민(CA) 분비작용의 차이와 특성에 대해서 연구한 결과 다음과 같다. DMPP(100 uM)와 McN-A-343(100 uM)은 부신정맥내로 투여시 유의한 카테콜아민 분비작용을 나타내었다. Mol농도로 비교시 McN-A-343의 CA분비작용은 DMPP의 약 1/5정도였다. DMPP나 McN-A-343의 반복투여시 반응 급강현상은 관찰할 수 없었다. DMPP의 CA분비작용은 chlorisondamine이나 desipramine또는 $Ca^{2+}-free$ Krebs + EGTA 관류등의 전처치로 의의있게 억제되었으나, pirenzepine, ouabain 및 physostigmine등 전처치에 의해서는 영향을 받지 않았다. 그러나 atropine 전처치시 DMPP의 분비작용은 오히려 증강되었다. McN-A-343의 CA분비작용은 atropine, pirenzepine, chloriondamine, physostigmine 및 $Ca^{2+}-free$ medium plus EGTA 관류등의 전처치에 의해서현처히 차단되었으나 desipramine등에 의해서는 영향을 받지 않았다. 그러나 ouabain의 전치치시 McN-A-343의 분비효과는 크게 증강되었다. 이상의 실험결과로 보아 DMPP와 McN-A-343은 횐쥐 적출관류 부신에서 현저한 CA분비작용을 일으키며, 이는 $Ca^{2+}$ 의존성 임을 보였으며, DMPP의 분비작용은 부신의 nicotine 수용체의 흥분을 통해서 나타내며, 또한 McN-A-343의 분비작용은 $M_{1}-muscarine$ 수용체의 흥분에 의하여 유발되는 것을 생각된다. DMPP의 분비활성이 McN-A-343보다 훨씬 강력한 것으로 사료된다.

Influence of Cilnidipine on Catecholamine Release in the Perfused Rat Adrenal Medulla

  • The present study was attempted to investigate the effect of cilnidipine (FRC-8635), which is a newly synthesised novel dihydropyridine (DHP) type of organic $Ca^{2+}$ channel blockers, on secretion of catecholamines (CA) evoked by acetylcholine (ACh), high $K^+$, DMPP and McN-A-343 from the isolated perfused rat adrenal gland. Cilnidipine $(1{\sim}10{\mu}M)$ perfused into an adrenal vein for 60 min produced relatively dose- and time-dependent inhibition in CA secretory responses evoked by ACh $(5.32{\times}10^{-3}M),\;DMPP\;(10^{-4}M\;for\;2\;min)$ and McN-A-343 $(10^{-4}M\;for\;2\;min)$. However, lower dose of cilnidipine did not affect CA secretion by high $K^+\;(5.6{\times}10^{-2}\;M)$, higher dose of it reduced greatly CA secretion of high $K^{+}$. Cilnidipine itself did fail to affect basal catecholamine output. In the presence of cilnidipine $(10{\mu}M)$, the CA secretory responses evoked by Bay-K-8644 $(10{\mu}M)$, an activator of L-type $Ca^{2+}$ channels and cyclopiazonic acid $(10{\mu}M)$, an inhibitor of cytoplasmic $Ca^{2+}$-ATPase were also inhibited. Moreover, ${\omega}-conotoxin\;GVIA\;(1{\mu}M)$, a selective blocker of the N-type $Ca^{2+}$ channels, given into the adrenal gland for 60 min, also inhibited time-dependently CA secretory responses evoked by Ach, high $K^+$, DMPP, McN-A-343, Bay-K-8644 and cyclopiazonic acid. Taken together, these results demostrate that cilnidipine inhibits CA secretion evoked by stimulation of cholinergic (both nicotinic and muscarinic) receptors from the isolated perfused rat adrenal gland without affecting the basal release. However, at lower dose, cilnidipine did not affect CA release by membrane depolarization while at larger dose inhibited that. It seems likely that this inhibitory effect of cilnidipine is exerted by blocking both L- and N-type voltage-dependent $Ca^{2+}$ channels (VDCCs) on the rat adrenomedullary chromaffin cells, which is relevant to inhibition of both the $Ca^{2+}$ influx into the adrenal chromaffin cells and intracellular $Ca^{2+}$ release from the cytoplasmic store. It is thought that N-type VDCCs may play an important role in regulation of CA release from the rat adrenal medulla.

파낙스디올의 가토적출부신의 카테콜아민분비 작용에 관한 연구 (Studies on Secretion of Catecholamines Evoked by Panaxadiol in the Isolated Rabbit Adrenal Gland)

  • 본 연구에서 한국산 인삼의 활성성분의 하나인 panaxadiol(PD)의 가토적출분신에서 카테콜아민(CA)의 분비작용과 작용기전을 파악하고자 실험을 시행하여 다음과 같은 결과를 얻었다. PD($400\;{\mu}g$)을 가토적출부신에 투여 하였을 때 카테콜아민의 분비를 유의성 있게 증가시켰다. PD의 이러한 CA분비작용은 atiropine처리로 현저히 억제되었다. Physostigmine 처리시 PD뿐만 아니라 Ach의 CA 분비작용은 뚜렷이 증가 되었다. 그러나 chlorisondamine처리로 PD나 Ach의 분비효과는 억제되었다. 또한 PD($400\;{\mu}g$/30min)을 주입한 후에 Ach의 CA분비작용은 증강되었다. Ouabain에 의해서 PD의 작용은 약화되었으나 Ach의 CA분비효과는 오히려 강화되었다. PD나 Ach의 작용은 adenosine 전처리시 현저히 증강되었다. EDTA(5mM)와 함께 Ca-free Krebs액으로 30분 주입한 경우에 Ach의 분비작용은 거의 전적으로 차단되었으며, PD의 작용도 약화되었다. 이상의 실험결과로 보아, PD는 가토적출부신에서 $Ca^{++}$의존적으로 CA분비를 증가시키며, PD의 이러한 작용은 부신내의 cholinergic muscarinic 및 nicotinic receptor의 흥분작용에 기인되고, chromaffin cell에 대한 일부 직접작용에 의해서 나타나는 것으로 사료된다.

  • The influence of glucocorticoids on the secretory responses of catecholamines (CA) evoked by acetylcholine (ACh), DMPP, McN-A-343, excess K$\^$+/ and Bay-K-8644 from the isolated perfused rat adrenal gland and to clarify the mechanism of its action. The perfusion of the synthetic glucocorticoid dexamethasone (10-100 uM) into an adrenal vein for 20min produced relatively a dose-dependent inhibition in CA secretion evoked by ACh (5.32mM), excess K$\^$+/ (56mM), DMPP (a selective nicotinic receptor agonist, 100uM for 2min), McN-A-343 (a muscarinic receptor agonist, 100uM for 4min), Bay-K-8644 (a calcium channel activator, 10 uM for 4min) and cyclopiazonic acid (a releaser of intracellular Ca$\^$2+/, 10uM for 4min).

흰쥐 시상하부의 정중융기에서 카테콜아민 신경세포와 GnRH 신경세포와의 연접에 관한 연구 (Catecholaminergic Innervation of GnRH Neurons in the Rat Median Eminence)

  • The present study was carried out 1) to show the ontogenic development of CA-and GnRH-containing nerve fobres in the median eminence, 2) to simultaneously demonstrate the synaptic contact between these two nenre fibres in the rat median eminence at the ultrastructural level using light and electron microscopic doublelabel immunostainlngs. GnRH-and CA-nenre terminals were detectable in the median eminence at embryonic day 19.5. The CA-newe terminals were obsenred in the entire legion of the extern31 lavers, while GnRH-newe terminals only in the lateral portion. At the 14th postnatal daw, both %ropes of nerve terminals showed a very similar distribution to those of adult one. In the median eminence of adult rats, a substantial overlap existed in the distribution of GnRH fibres with CA-containing nerve fibres. This overlap was most intense throughout the external palisade zone. Furthermore, an electron microscopic double label immunostaining showed that there was a close apposition of CA- and GnRH-nenre fobres. These axo-axonic contacts occurred frequently in the internal and palisade zones, i.e. at the level of the fobre preterminals. These morphological results suggest that the CA-mediated GnRH secretion may occur via sxo-axonic interaction in the median eminence.

Mechanism of Vasoactive Intestinal Polypeptide-Induced Catecholamine Secretion from the Rat Adrenal Medulla

  • The present study was attempted to investigate the effect of vasoactive intestinal polypeptide (VIP) on secretion of catecholamines (CA) and to establish whether there is the existence of a noncholinergic mechanism in adrenomedullary CA secretion from the isolated perfused rat adrenal gland. The perfusion into an adrenal vein of VIP $(3{\times}10^{-6}\;M)$ for 5 min or the injection of acetylcholine (ACh, $5.32{\times}10^{-3}\;M$) resulted in great increases in CA secretion. Tachyphylaxis to releasing effect of CA evoked by VIP was not observed by the repeated perfusion. The net increase in adrenal CA secretion evoked by VIP still remained unaffected in the presence of atropine or chlorisondamine. However, the CA release in response to ACh was greatly inhibited by the pretreatment with atropine or chlorisondamine. The releasing effects of CA evoked by either VIP or ACh were depressed by pretreatment with nicardipine, TMB-8, and the perfusion of $Ca^{2+}$-free medium. Moreover, VIP- as well as ACh-evoked CA secretory responses were markedly inhibited under the presence of $(Lys^1,\;Pro^{2.5},\;Arg^{3.4},\;Tyr^6)-VIP$ or naloxone. CA secretory responses induced by ACh and high $K^+\;(5.6{\times}10^{-2}\;M)$ were potentiated by infusion of VIP $(3{\times}10^{-6}M\;for\;5\;min)$. Taken together, these experimental results indicate that VIP causes CA release in a fashion of calcium ion -dependence, suggesting strongly that there exists a noncholinergic mechanism that may be involved in the regulation of adrenomedullary CA secretion through VIP receptors in the rat adrenal gland, and that VIP may be the noncholinergic excitatory secretagogue present in the chromaffin cells.
