Effects of Mixture of Lycii Radicis Cortex and Gardeniae Fructus on the Change of HPA-Axis and Catecholamine System in the Forced Swimming Test (지골피(地骨皮)와 치자(梔子) 합제(合劑)가 강제수영부하실험에서 HPA-Axis와 Catecholamine system에 미치는 영향)

  • Objectives : In this study the antidepressant effects of mixture of Lycii Radicis Cortex and Gardeniae Fructus on the change of HPA-Axis and Catecholamine system was investigated Methods : The forced swimming test was performed. The expression of CRF, c-Fos in the PVN, and TH in the VTA and LC were measured with immunohistochemical method. And the experimental groups were divided into the Extraction after Mix group(A) and Mix after Extraction group(B). The effects of both group were compared. Results : The duration of immobility in the FST was decreased significantly in three groups except B100(P<0.01). B100 was shown the significant increase(P<0.05). The expression of CRF in PVN was decreased significantly in all group(P<0.05~P<0.01). The expression of c-Fos in PVN was decreased in A100, A400 and was increased significantly in B100, B400(P<0.05). The expression of TH in VTA and LC were decreased significantly in all group(P<0.001). Conclusions : Effects of Extraction after Mix group(A) and Mix after Extraction group(B) on the HPA-Axis system and Catecholamine system were validated.

  • The word of stress was wsed in the field of physics as a external force from 17th century, but the meaning of this stress had chafed to the internal and exteral demand fort the human body in medical area. All the stumulants which make stress was called as j stressor. When animals get stress blucose excreted from liver to adapt for the emergent state ant some related hormone secrete convert protein and lipid to glucose for the purpose of energy supply to muscle. As a results heart rate, blood pressure, respiration rate are increased and musclse are strengthed. These physiolgical reactions ate controlled by autonomic nerve system under the control of hyothalamus in brain. Autonomic nerve system and endocrinary system are react harmoiously to stress reaction. According to the stress reaction, adrenomedullary system are stimulated, and epinephrine, morepinephrine are exceted. Author experimented the effect of the immobilizational stress to rat by analyzing the variaition of catecholamine secrction, the Na concentration and the effect of the antistresf effect by Panax Ginseng which is a traditional Korean herb medicine. The concentration of the norepinephrine, epinephrin, and dopamin in normal rat are 1 578 ng/ml, 0.365 ng/ml, and 0.731 ng/ml respectively, but in the immobilyzed stress groIn the concentration were increased to 1.915 ng.ml, 0.854 ng/ml, and 2,361 ng/ml which she the high show the higher concentration of catecholamine to stressor.

Regulatory Role of Adrenal Medulla and Renin-Angiotensin System in Sympathetic Neurotransmission in Spontaneously Hypertensive and Normotensive Rats (선천성 고혈압 흰쥐와 정상혈압 흰쥐의 교감신경성 신경전달에 미치는 부신수질 및 Renin-Angiotensin계의 역할)

  • To assess the role of adrenal medulla and renin-angiotensin system in the regulation of sympathetic neurotransmission, the pressor response to PNS was evaluated in pithed SHR and normotensive WKY or SDR with or without adrenal demedullation and/or enalapril pretreatment. Three weeks after adrenal demedullation, MAP and the heart rate of demedullated rats were similar to their corresponding sham-operated groups. The pressor response to PNS was frequency-dependent, and blocked by prazosin. In contrast to the normotensive rats, in SHR, the pressor response to PNS was attenuated in demedullated rats as compared with sham-operated rats. However, the attenuation of PNS-induced pressor responses in demedullated SHR was not observed in enalapril-treated SHR. The adrenal demedullation in SHR did not affect the plasma and aortic catecholamine contents in spite of the decreased catecholamine contents of adrenal gland, nor ACE activity in aortic strips. But, in WKY rats, the aortic catecholamines, especially epinephrine, contents as well as ACE activity were increased by adrenal demedullation. These results suggest that the facilitatory role of adrenal medulla in sympathetic neurotransmission depends upon the activation of renin-angiotensin system, and that the compensatory regulation of renin-angiotensin system takes place in normotensive rats but not in SHR.

  • The term of stress was used by many scientists, but there were several difinitions about stress, and this term are using in societies frequently. Generally stress is defined as equilibrium, harmony, and adaptation to external stimulations, in 1930 s Hans Selye was the first scientist who used the term of stress and he pointed out the stress mean the function of force in body. When we get stress the stimulation go to the sympathetic-adrenomedulla system and epinephrine and norepinnephrine are secreted from adrenal medulla. Author tried to investigate the change of catecholamine secretion from rat urine which were exposed to 80 dB noise environment. The control rat urine released 0.03 $\pm$ 0.01 ng/ml of epinephrine and 0.18 $\pm$ 0.04 ng/ml, but in noise exposed group the epinephrine was 0.42 $\pm$ 0.07 ng/ml and norepinephrine was 2.16 $\pm$ 0.48 ng/ml.

  • Noise is not only affecting the ear and the auditory cortex locally, but its influence is widely spread throughout the brain structures, e. g., the reticular formation, the brain stem nuclei or the subcortical forebrain area. Hence, any of the organism's activities can be hindered or stimulated by noise. High noise is a stressor and the catecholamine level can be used both as a stress marker and as an indicator of modified sympathetic nervous system activity. Several recent studies have found that the urinary excretion of catecholamines is increased due to high noise intensity, especially unexpectedly high and long lasting noise. The present study was conducted in order to examine the effects of noise stress on urinary excretion of ctecholamines in rats and humans. Rats were exposed to 90 dB noise for 10, 30, and 60 minutes, 3 and 12 hours. 24 hour . urinary samples were collected and the catecholamones were extracted by alumina and analyzed by HPLC-ECD. Catecholamine levels increased with time of exposure up to 60 minutes : norepinephrine concentration at 60 min of noise=1.038 ng/ml, epinephrine=0.636 ng/ml. Urine catecholamines of blue collar workers exposed to 90 dB of noise at the work place were collected between 2 and 4 p.m. and compared to that of white collar workers exposed to 70 dB. Mean norepinephrine level of the blue collar workers was 0.89 ng/ml (${\pm}0.25$), epinephrine 0.24ng/m1 (${\pm}0.09$), and that of the white collar workers 0.48 ng/ml (${\pm}0.12$), epinephrine 0.19 ng/ml(${\pm}0.05$). It was concluded that noise acts as a stressor and increases the catecholamine levels in both rats and humans.

Effect of the bee venom aqua-acupuncture on the neuronal activities of catecholaminergic system in brainstem (봉독약침자극이 Catecholamine성 신경세포의 활성변화에 미치는 영향)

  • This study was designed to evaluate the effect of the bee venom(BV) aqua-acupuncture on the neuronal activities of catecholaminergic(tyrosine hydroxylase : TH, dopamine ${\beta}$ hydroxylase : D${\beta}$H) system in the brainstem. After the BV aqua-acupuncture was applied on Chok-Samni(ST36) and the gluteal part(Blank locus) in rats. Also, the number of colocalization between catecholamine containing neurons and Fos immunoreactive neurons were analyzed by using the double immunohistochemical technique. The results of the experiments were summarized as follows : 1. In DR and LC, Chok-Samni group and the Blank locus group showed more significant increase in the number of colocalization between TH containing neurons and Fos immunoreactive neurons than the control group. Furthermore, Chok-Samni group showed more significant increase than the Blank locus group. Also, in Arc, Chok-Samni group showed more significant increase than the Blank locus group and the control group. 2. In LC, Chok-Samni group showed more significant increase in the number of colocalization between D${\beta}$H containing neurons and Fos immunoreactive neurons than the Blank locus group and the control group. Also, in A5, Chok-Samni group and the Blank locus group showed more significant increase than the control group. Chok-Samni group showed more significant increase than the Blank locus group. However, there was no significant change in A7. Consequently, the BV aqua-acupuncture increased more potent the number of Fos immunoreactive neurons and the activity of catecholaminergic neurons. Furthermore, the BV aqua-acupuncture was more effective on Chok-Samni than Blank locus group. These results indicate that the BV aqua-acupuncture is very effective therapy to control pain. The therapeutic effect of BV aqua-acupunture may associated with the endogenous modulatory system such as catecholamine. Those data from the study can be applied to establish the effective treatment of the BV for pain control in the clinical field.

  • Capillary zone electrophoresis (CZE) which is highly efficient separation technique has been domestically established having optimum detection sensitivity. By applying 20∼35 kV of electric potential to the narrow (50 ${\mu}m$ i.d.) capillary tubing filled with running buffer, this technique can quickly (< 20 min) separate the small quantities of sample with high separation efficiency (number of theoretical plates : 200,000∼500,000). Factors affecting the separation efficiency and resolution in CZE were examined by analyzing adenine and catecholamine derivatives.

Effect of Ovarian Steroid Hormones on Catecholaminergic Nervous System in the Hypothalamus of SHR (선천성 고혈압 쥐에서 시상하부 카테콜아민성 신경계에 미치는 난소 스테로이드 호르몬의 영향)

  • A question whether abnormal responsiveness of hypothalamic catecholaminergic nervous system to ovarian steoid hormones in spontaneously hypertensive rats (SHR) exist was investigated. Four groups of experimental animals were prepared for SHR and normotensive Wistar rats (NW) respectively: 1) intact, 2) ovariectomized (OVX+V), 3) ovariectomized and estrogen treated (OVX+E), 4) ovariectomized and estrogen plus progesterone treated (OVX+E+P) groups. Hypothalami from experimental animals were dissected out and used for determination of .alpha.-adrenergic receptor binding characteristics and catecholamine contents. Norepinephrine(NE) content and B$_{max}$ of $\alpha_1$-adrenergic receptors in hypothalami were greater in intact SHR than in intact NW, but dopamine(DA) content was lower in SHR than in NW. Neither contents of NE and DA nor binding characteristics of $\alpha_1$-adrenergic receptors were different in OVX+V and OVX+E group from intact group of both SHR and NW. Kd and B$_{max}$ of $\alpha_1$-adrenergic receptors in OVX+E+P was lower than that in intact SHR but not in NW. DA content was lower in OVX+E+P than in intact group of SHR and NW. The result of the present study indicates that there is an abnormal responsiveness of hypothalamic catecholaminergic nervous system to ovarian steroid hormones in SHR which may be one of genetically-determined factors probably not responsible for the development of hypertension.

The Effects of Qigong training on the cardiopulmonary functions and catecholamine levels after physical traning stress in untrained college students (‘내경일지선(內徑一指禪)’ 기공수련이 대학생의 운동부하 스트레스 후의 심폐기능 및 Catecholamine 변화에 미치는 영향)

  • This study was performed to investigate the effects of Qigong training after physical training stress in untrained college students For this study, 6 voluntary subjects(male 4, female 2) were chosen in untrained students of K University. they were trained by teachers during 6 weeks and tested just before Qingong training and after 6 weeks. Each subject was performed a treadmill exercise(model Q65, Quinton Co, U.S.A.) to the all-out state. During exercise stress test, electrocardiogram, heart rate were checked by stress test monitor(model Q4500, Quinton Co, U.S.A) and also oxygen uptake, maximal oxygen uptake analyzed continuously by automatic gas analysis(model QMC, Quinton Co, U.S.A). During physical training the serum were collected 3 times, pre-experimental rest time, and serum catecholamine were measured by HPLC.T-test of statistical analysis system was used in every experiment for statistical assessment. The results of T-test on these data were summarized as follow:1.Heart rate change during exercise stress test after Qigong training was shown more decreasing tendency than before training. Especially, heart rate change after Qigong training during resting periods was decreased significantly than before training.2. Oxygen uptake change during exercise stress test after Qigong training was shown more increasing tendency than before training, And also maximal oxygen uptake after Qigong training was shown more increasing tendency than before 6 weeks.3. Epinephrine level of after Qigong training was more decreased significantly than before training in all-out state. And norepinephrine level of after Qigong training was shown more decreasing tendency than before training in all-out state and after 30 minutes rest time. Above results indicate that Qigong training for 6 weeks could be effective to elevate the cardiopulmonary functions and diminish the stress responses of the physical stress.

Physiological Markers in Anxiety and Depression (생리적 지표를 이용한 불안과 우울 측정에 대한 고찰)

  • Purpose: Anxiety and depression are the concepts that commonly used as an emotional approach in the nursing researches and most of researches have been used questionnaires as a marker of anxiety and depression. There were few researches using physiological markers in measuring anxiety and depression. Methods: Journals published between 1950 and 2007 that include depression and anxiety measuring through physiological markers were reviewed. Results: As in the case of the hypothlamus-pituitary-adrenal axis system, it appeared that cortisol, epinephrine, and norepinephrine belonged to the category of hormones which were responsive to anxiety and depression. Plasma norepinephrine was a marker of the anxiety whereas plasma cortisol was a marker of the depression. The anxiety and depression were correlated with immune and taste, but it considered as an outcome variable not a physiological marker. Conclusion: Catecholamine and cortisol reflect anxiety and depression state. Our findings suggest that further researches are needed to distinguish between markers and outcomes of depression and anxiety using physiological markers.
