치수에서 $Ca^{++}$-ATPase와 $Mg^{++}$-ATPase의 활성도에 관한 실험적 연구 (A BIOCHEMICAL STUDY OF $Ca^{++}$-ATPASE AND $Mg^{++}$-ATPASE ACTIVITY IN HUMAN DENTAL PULP)

  • This study was undertaken to evaluate the physiological roles & mechanism of $Ca^{++}$-ATPase & $Mg^{++}$-ATPase in human dental pulp. Each specimen of dental pulp was obtained from the freshly extracted, freeze-dried 242 teeth. $Ca^{++}$-ATPase & $Mg^{++}$-ATPase activity were measured by the release of inorganic phosphate & protein with Spectrophotometer. The results were as follows; 1. The $Ca^{++}$-ATPase & $Mg^{++}$-ATPase activity were significantly increased in developing teeth. 2. The $Ca^{++}$-ATPase & $Mg^{++}$-ATPase activity were significantly decreased in nonvital teeth. 3. The $Ca^{++}$-ATPase & $Mg^{++}$-ATPase activity were significant decreased in deciduous teeth. 4. The $Ca^{++}$-ATPase & $Mg^{++}$-ATPase activity didn't have relation with dental caries. 5. The $Ca^{++}$-ATPase & $Mg^{++}$-ATPase were activated by either $Ca^{++}$ alone or $Mg^{++}$ alone.

Effect of Repeated Oral Health Education on the Oral Health of Preschool Children

  • This study aimed to investigate the effect of repeated oral health education on the oral health of preschool children. This study classified the control group of children of 5~6 years of age where the oral health education was conducted one time, and the experiment group where the oral health education was conducted 4 times. The oral health conditions of each group and their risk of developing dental caries were also assessed. No significant difference was observed in the participants in terms of general characteristics, oral health conditions, risk of developing dental caries, and oral health behavior after oral health education. The two groups did not show a significant difference (p<0.001) in dental plaque index before and after oral health education, and showed a significant difference (p<0.001) according to the number of education sessions. Before oral health education, the participants in both groups were brushing their teeth incorrectly. However, after the third session, 56.7% of the participants in the experimental group and 9.4% in the control group used the correct method of tooth brushing. A significant difference was observed between the two groups (p<0.05). In addition, when the tooth-brushing time was compared, the two groups showed a significant difference (p<0.001), and a significant difference was observed in terms of the number of education sessions (p<0.05). In summary, repeated oral health education had significant influences on the dental plaque index, tooth-brushing method, brushing occlusal surfaces, and tooth-brushing time. Therefore, when oral health education is carried out, repeated oral health education in children is more effective than one-time oral health education as reflected in the changes in dental plaque index and oral health behavior. Moreover, oral health education is considered effective in changing the oral health behavior of children when conducted at least 3 times.

우식치아와 정상치아의 교합면에서 분리한 Streptococcus mutans의 비교 (THE COMPARISON OF STREPTOCOCCUS MUTANS ISOLATED FROM OCCLUSAL SURFACES OF CARIES AND NON-CARIES TEETH)

  • 우식치아의 교합면과 정상치아의 교합면에서 균을 채취한 결과, 우식치아의 교합면에서는 $3.43\times10^5$ CFU, 정상치아의 교합면에서는$3.43\times10^3$ CFU가 MSB 배지상에서 검출되었다. API test를 이용하여 당발효 및 생화학적 성상을 관찰한 결과, 우식치면에서 분리된 세균은 20종 모두 S. mutans였으나, 건강한 치면에서는 2개의 세균만 S. mutans로 동정되었다. 우식치면과 정상치면에서 분리된 균주 S. mutans SM1과 S. mutans SM2는 $\alpha-galactosidase$ 활성을 제외하곤 당발효 및 생화학적 성상이 모두 일치하였다. 증식에 있어서, 두 균주 모두 pH 5.5에서 증식이 가장 활발하였고, 자당의 농도는 SM1은 20%일 때 SM2는 5%일 때 최대 증식을 보였다. SM1은 배지의 $CaCl_2$농도가 16mM, KCl농도가 160mM, $MgCl_2$농도가 6.4mM 이었을 때 증식이 가장 활발하였고, SM2는 $CaCl_2$ 농도가 16mM, KCl 농도가 40mM, $MgCl_2$ 농도가 6.4mM 이었을때 증식이 가장 활발하였다. Sodium bicarbonate 완충액과 Sodium phosphate 완충액의 경우, SM1과 SM2 모두 1mM에서 증식이 활발하였다. Tris 완충액의 경우, SM1은 1mM에서, SM2는 10mM에서 증식이 활발하였다. Potassium phosphate 완충액의 경우, SM2는 농도가 증가함에 따라 증식이 억제된 반면, SM1은 100mM 까지는 농도가 증가할수록 증식이 활발하였다. SM1과 SM2의 염색체를 추출한 후 Primer gtfB-F961과 gtfC-R5574를 사용하여 gtf 유전자를 PCR 한결과, 4.6kb의 단일 band를 얻었고 이 band를 분리하여 제한효소로 처리한 결과, EcoR I 처치시 S. mutans GS-5, SM1과 SM2모두 약 0.8kb와 3.8kb의 DNA절편을 보여 같은 양상을 나타냈고, Hind III 처치시 GS-5와 SM1은 잘리지 않았고, SM2의 경우 2.4kb, 1.8kb, 400bp의 3조각의 절편으로 나뉘어 SM1과 SM2의 gtf 유전자의 상사성이 관찰되었다. BamH I처치시 SM1과 SM2는 4조각의 절편으로 절단되어 같은 양상을 보였고, GS톤의 경우는 3조각의 절편으로 절단되었다. Kpn I, Sma I, Xho I 그리고 Pst I에는 절단되지 않았다.

Inhibitory Effects of Resina Pini on the Growth and Glucosyltransferase activity of Streptococcus mutans

  • The purpose of this study is to evaluate the inhibitory effects of Resina Pini against Streptococcus mutans (S. mutans) that is one of the major causes of dental caries and oral diseases. Topically applied Resina Pini (RP) would be incorporated in saliva and thus the factor associated with water solubility should be considered. In this paper, therefore, effects of various treatment for RP and activities of water extracts from unprocessed and processed RP were compared. The crude RP (RP1) and the recrystallized RP (RP2) in ethanol solution showed strong antimicrobial activities (d.>15mm) against S. mutans. All RP samples exhibited considerable inhibitory effect against glucosyltransferase produced by S. mutans $(IC_{50}=91.2\;to\;276.2\;{\mu}g/ml)$. The very considerable increase in cellular permeability of S. mutans was observed with RP1, RP2 and their water extracts. These results suggest that RP1 and RP2 may be a potential source for pharmaceutical products used for prevention and/or treatment of dental caries and periodontal disease.

콩 추출물의 구강미생물에 대한 항균효과 (Anti-microbial Activity of Soybean Extract Against Oral Microbes)

  • This study was performed to investigate anti-microbial activity of soybean extract against oral microbes, and to determine the minimum inhibition concentration (MIC) for microbes causing oral diseases. The soybean extract was prepared using ethyl acetate and it was treated with 16 types of oral microbes at a concentration of 5.00 mg/ml (0.5%). The MIC of soybean extract for three major microbes causing oral diseases was determined. The anti-microbial activity and MIC were measured using broth dilution method. Significant reduction of microbial activities of 9 types oral microbes when the soybean extract was added to the broth compared to the control (p<0.01). The extract showed higher anti-microbial activity against some anaerobic strains (P. gingivalis and P. intermieia). S. mutans, which causes dental caries, showed MIC at a concentration of 40 mg/ml for the soybean extract. P. gingivalis, which causes adult periodontal disease, showed MIC at a concentration of 20 mg/ml for the extract. C. albicans, which causes denture stomatitis and angular stomatitis, showed MIC at a concentration of 20 mg/ml for the extract. These results indicate that soybean extract showed anti-microbial effort against 9 types of oral microbes, and the anti-microbial effect of the extract against oral microbes was stronger against fungi than against bacteria. The anti-microbial mechanism of soybean extract against oral microbes should be investigated, and more research for clinical application is required at a level of actual intake.

Streptococcus sobrinus에 의한 일부 시판음료의 pH 변화 (pH changes by Streptococcus sobrinus on some commercial drinks)

  • Objectives: The purpose of this study was to investigate the dental caries activity of some commercial drinks. Methods : Acidity levels were recorded as pH values of original drinks and amount of 50%-sodium hydroxide(NaOH) required to neutralize by titration 50.00ml of the drink(It was called titratable acid). Acidity of the drinks with S. sobrinus was measured at an interval of one hour after incubating ($37^{\circ}C$) the test drinks and control solution which was composed 20.00ml of neutralized drinks as pH 7.0 with 0.2ml of a pooled suspension of S. sobrinus. Results : The drink with the highest pH value was the Cow's milk(pH $6.70{\pm}0.01$), and the most acidic was the Carbonated drink(pH $2.40{\pm}0.02$). The drink with the highest buffer effect was the Fruit juice($139.78{\pm}0.76$), and the lowest was the Glucose solution(control, $4.42{\pm}0.20$). The drinks with S. sobrinus during incubation at $37^{\circ}C$ for 24h were decreased to below critical pH 5.5. Spending times from pH 7.0 to pH 5.5~5.0 were Glucose solution(less than 1h). Carbonated drink(3h), Fruit juice(5h), Cow's milk(8h) and Mixed drink(21h). Conclusions: These results show that some commercial drinks with S. sorbrinus have the possibility of tooth decalcification. Thus, it is suggested that the people who have the low salivary secretion rate and the children who sleep without toothbrushing after drinking need the instruction for diet control and oral health education.

백질려 추출물이 Streptococcus mutans에 대한 항치아우식에 미치는 영향 (Anticariogenic Properties of the Ethanol Extract of Tribuli fructus against Streptococcus mutans)

  • Streptococcus mutans is considered one of the primary etiologic agents of dental caries. we studied the effect of the ethanol extracts of Tribuli fructus (T. fructus) on the growth, biofilm formation, acid production, adhesion and insoluble glucan synthesis of S. mutans. The ethanol extracts of T. fructus showed concentration dependent inhibitory activity against the growth and acid production of S. mutans, and produced significant inhibition at the concentration of 0.025, 0.05, 0.1, 0.2, 0.3, 0.5 mg/ml compared to the control group. In the biofilm assay, the ethanol extracts of T. fructus inhibited formation of biofilm synthesized by S. mutans at the concentration of 0.05 mg/ml. The extracts markedly inhibited S. mutans adherence to HA treated with saliva, and cell adherence was repressed by more than 50% at the concentration 0.05 mg/ml. On the activity of glucosyltransferase which synthesizes water insoluble glucan form sucrose, ethanol extract of T. fructus showed more than 10% inhibition over the concentration of 0.025 mg/ml. Hence, we conclude that T. fructus might be a candidate of anticaries agent. Further studies are necessary to clarify the active constituents of T. fructus responsible for such biomolecular activities.

Effect of the Antimicrobial Peptide $\small{D}$-Nal-Pac-525 on the Growth of Streptococcus mutans and Its Biofilm Formation

  • Streptococcus mutans is the primary etiological agent of dental caries. The antimicrobial peptide $\small{D}$-Nal-Pac-525 was designed by replacing the tryptophans of the Trp-rich peptide Pac-525 with $\small{D}$-${\beta}$-naphthyalanines. To assess the effect of $\small{D}$-Nal-Pac-525 on cariogenic bacteria, the activity of $\small{D}$-Nal-Pac-525 on the growth of S. mutans and its biofilm formation were examined. $\small{D}$-Nal-Pac-525 showed robust antimicrobial activity against S. mutans (minimum inhibitory concentration of 4 ${\mu}g/ml$). Using scanning electron microscopy and transmission electron microscopy, it was shown that $\small{D}$-Nal-Pac-525 caused morphological changes and damaged the cell membrane of S. mutans. $\small{D}$-Nal-Pac-525 inhibited biofilm formation of S. mutans at 2 ${\mu}g/ml$. The results of this study suggest that $\small{D}$-Nal-Pac-525 has great potential for clinical application as a dental caries-preventing agent.

유치열에서 Chlorhexidine Varnish와 Polyurethane Sealant의 Streptococcus mutans억제 효과에 대한 연구 (THE INHIBITORY EFFECT OF CHLORHEXIDINE VARNISH AND POLYURETHANE SEALANT ON STREPTOCOCCUS MUTANS IN THE PRIMARY DENTITION)

  • Numerous chemical agents have been developed to reduce the activity of cariogenic bacteria. Of these, chlorhexidine is acknowledged as the most effective. Gel and mouthrinse have been the traditional method of its application in the mouth. It has been reported that chlorhexidine varnish has prolonged inhibitory effect on the number of streptococcus mutans in saliva and plaque. Recently, chlorhexidine varnish and polyurethane sealant have been developed to promote prolonged anticariogenic effect of chlorhexidine. Products containing 10% chlorhexidine varnish and polyurethane sealant have been developed to prevent caries by reducing the number of streptococcus mutans in the oral cavity. The purpose of this study is to investigate the inhibitory effect of chlorhexidine varnish and polyurethane sealant on streptococcus mutans in the primary dentition. Children with primary dentition containing no active carious lesion were divided into two groups. To the experimental group (n=11), chlorhexidine varnish and polyurethane sealant ($Chlorzoin^{(R)}$, Knowell Therapeutic Technologies, Inc. Canada.) was applied once a week for four weeks according to the manufacturer's instruction. Only oral prophylaxis was performed on the control group(n=7). Caries activity was measured after using $Cariescreen^{(R)}$SM (Knowell Therapeutic Technologies, Inc. Canada,) to incubate streptococcus mutans before and 5, 12, 24 weeks after initial varnish application. The following results were observed.; 1. There was statistically significant decrease in the number of streptococcus mutans in the experimental group for 5 weeks(P<0.01), 12 weeks(P<0.05) after the initial application. but, by 24 weeks significant difference had disappeared. 2. As the inhibitory effect of chlorhexidine varnish and polyurethane sealant application is not everlasting, reapplication at 12-24 weeks should be needed.
