GM 파파야 개발 및 생물안전성 평가 연구 동향 (Research status of the development of genetically modified papaya (Carica papaya L.) and its biosafety assessment)

  • 파파야는 열대와 아열대 지역에서 광범위하게 재배되고 있는 주요 작물 중의 하나이다. 파파야 열매는 칼로리가 낮고 비타민 A와 C, 미네랄이 풍부하며, 미숙과에는 단백질 분해 효소인 파파인이 풍부하여 의약품, 화장품, 식품 가공 산업 등에 널리 활용되고 있다. 전세계 파파야 산업에서 가장 중요한 제한 요인 중의 하나가 potyvirus에 속하는 papaya ringspot virus (PRSV)에 의해 야기되는 식물병이다. 1992년에 미국 연구자들에 의해 PRSV의 coat protein (cp) 유전자를 발현하는 최초의 PRSV-저항성 GM 파파야 이벤트($R_0$ '55-1')가 만들어졌으며, 1997년에는 이로부터 유래한 GM 품종('SunUp', 'Rainbow')에 대해 미국 정부가 상업적 재배를 승인하였다. 현재까지 GM 파파야 개발은 해충 저항성, 병 저항성(곰팡이, 바이러스), 수확 후 저장성 증대, 알루미늄과 제초제 저항성 등의 형질에 초점을 맞추어 왔다. 아울러 파파야를 동물단백질(백신 등) 생산을 위한 식물공장으로 활용하기 위한 시도도 이루어졌다. 현재, 미국과 중국을 비롯한 약 17개 국가에서 GM 파파야 개발과 포장 실험 또는 상업적 재배가 이루어지고 있다. GM 파파야의 개발과 더불어 생물안전성 평가 및 GM 판별 기술 개발에 관한 연구도 이루어지고 있다. 생물안전성 평가와 관련하여 주로 인체 위해성과 환경 위해성에 관한 분석이 수행되고 있다. 인체 위해성의 경우, 동물 모델을 대상으로 장기간 식이섭취를 통해 일반 및 유전 독성, 알레르기항원성, 면역 반응, GM 유래 단백질의 안정성에 관한 연구가 수행되었다. 환경 위해성의 경우, GM 재배가 토양 미생물 다양성에 미치는 영향, GM 유래 유전물질의 토양 잔류 및 토양 미생물로의 전이 여부에 관한 연구가 이루어졌다. 우리나라, 유럽 및 일본을 비롯한 많은 나라에서는 상업적 재배를 위한 GM 품종 도입이나, 파파야 가공 식품 제조에 비승인 GM 파파야의 사용을 규제하고 있다. 도입 유전자 특이적 또는 이벤트 특이적인 분자표지를 개발하고, PCR(일반, real-time) 또는 loop-mediated isothermal amplification 방법을 통해 GM 여부를 판별하고 있다. 파파야에 대한 초안 수준의 유전체 정보가 2008년에 해독되었으며, 최근에는 차세대 유전체 분석 기술로 확보된 유전체와 전사체 정보를 활용하여 GM 여부를 판별하는 기술도 확립되었다.

Physicochemical Characteristics and Fatty Acid Composition of Four Papaya Cultivars Grown under Plastic Greenhouse Conditions

  • Some physicochemical characteristics like nutritional values, sugar content, and fatty acid composition (FAs) in fruits of four papaya (Carica papaya L.) cultivars ('Red Lady', 'Sunrise Solo', 'Tainung', and 'BH-65') grown under plastic greenhouse conditions in the Mediterranean region of Turkey were evaluated. The chemical characteristics, except acidity in the fruits, significantly varied among the cultivars. Nitrogen (N) was the most abundant mineral in all papaya cultivars and ranged from $0.80\;g{\cdot}100\;g^{-1}$ in 'Red Lady' to $1.28\;g{\cdot}100\;g^{-1}$ in 'BH-65' in fresh weight. Potassium (K) and calcium (Ca) were found the highest amount in dry weight. Glucose and fructose were identified as the main sugars in all cultivars. Sucrose was in trace amounts in the three cultivars, but not detectable in cultivar 'Red Lady'. Twenty-five FAs were detected in the papaya fruits. Polyunsaturated fatty acids (PUFAs) were found to be in a highest amounts compared to the saturated fatty acid (SFAs) and monounsaturated fatty acids (MUFAs). Palmitic (C16:0), oleic (C18:1), and linoleic (C18:2n6) acids were the major fatty acids detected in all cultivars. The results of this study implied that dietary intake of papayas may supply substantial nutrient components necessary for human health.

Comparative Analysis of Latex Plants by GC-MS using Methanol Extraction

  • Plants are able to produce a large number of diverse bioactive compounds. Solvent extraction is used for isolation of plant metabolites. The extract yield for plant metabolite extraction strongly depends on the nature of solvent. A review showed the methanol can yield more bioactive compounds. Drying of the sample material is also important for the extraction of plant material. The present study was carried out to analyze the phytocomponents of 5 different latex producing plants. The plants like Calotropis gigantea, Carica papaya, Nerium oleander, Ficus benghalensis and Plumeria alba leaves and latex. The GC-MS analysis of the metabolites revealed phytocomponents. Calotropis gigantea leaves showed 14 compounds and latex produced 5 compounds out of this 4,4,6A,6B,8A,11,11,14B-Octamethyl-1,4,4A,5,6,6A,6B,7,8,8A,9,10,11,12,12A,14,14A,14B-Octadeca-hydro-2 and 2R- Acetoxymethyl-1,3,3-trimethyl-4T-(3-Methyl-2-Buten-1-Yl)-1T-Cyclohexanol compound was present in both latex and leaf extraction. Beta. -carotene compound was present in both latex and leaf of Carica papaya. It was observed that Ficus benghalensis contained 2R-Acetoxymethyl-1,3,3-trimethyl-4T-(3-Methyl-2-Buten-1-Yl)-1T-Cyclohexanol was same in latex and leaf extraction.

Hybrid 'Sinta' Papaya Exhibits Unique ACC Synthase 1 cDNA Isoforms

  • Five ripening-related ACC synthase cDNA isoforms were cloned from 80% ripe papaya cv. 'Sinta' by reverse transcription-PCR using gene-specific primers. Clone 2 had the longest transcript and contained all common exons and three alternative exons. Clones 3 and 4 contained common exons and one alternative exon each, while clone 1, the most common transcript, contained only the common exons. Clone 5 could be due to cloning artifacts and might not be a unique cDNA fragment. Thus, there are only four isoforms of ACC synthase mRNA. Southern blot analysis indicates that all five clones came from only one gene existing as a single copy in the 'Sinta' papaya genome. Multiple sequence alignment indicates that the four isoforms arise from a single gene, possibly through alternative splicing mechanisms. All the putative alternative exons were present at the 5'-end of the gene comprising the N-terminal region of the protein. 'Sinta' ACC synthase cDNAs were of the capacs 1 type and are most closely related to a 1.4 kb capacs 1-type DNA(AJ277160) from Eksotika papaya. No capacs 2-type cDNAs were cloned from 'Sinta' by RT-PCR. This is the first report of possible alternative splicing mechanism in ripening-related ACC synthase genes in hybrid papaya, possibly to modulate or fine-tune gene expression relevant to fruit ripening.

Anti-tumor Promoting Activity of Some Malaysian Traditional Vegetables (Ulam)

  • Ethanolic extracts of different parts of 10 local traditional vegetables (ulam) (Amaranthus gangeticus, Jussiaea linifolia, Eugenia polyantha, Trapa incisa, Trichosanthes anquina, Mangifera indica, Pachyrrhirus erosus, Barringtonia mcarostachya, Carica papaya, and Coleus tuberosus) were screened for in vitro antitumor promoting activity using the inhibition test of Epstein-Barr virus (EBV) activation in Raji cells induced by phorbol 12-myristate 13-acetate and sodium-n-butyrate. All the extracts were found to have strong inhibition activity toward EBV-activation, except for leaf extract of T. anquina. The extracts were non-cytotoxic to the Raji cells except for the extracts of A. gangeticus (leaves), B. macrostachya (leaves), E. polyantha (young leaves), and J. linifolia (leaves) where the viability of the cells were decreased significantly.

  • The extracts of Carica papaya (flower), Barringtonia macrostachya (leaves), Coleus tuberosus (tuber), Mangifera indica (fruit skin) and Eugenia polyantha (leaves) showed strong in vitro anti-tumor promoting activity when assayed using Raji cells (Mooi et al., 1999). The antitumor promoting activity of the crude extracts was further analyzed by immunoblotting analysis of Raji cells carving Epstein-Barr virus genome. The expression of early antigens diffuse (EA-D) and early antigens restricted (EA-R) was determined by performing western blotting of treated Raji cells with human sera of nasopharyngeal carcinoma patients. All the plant extracts were shown to be able to suppress both EA-D and EA-R.

청과용 파파야 무가온 생산시 육묘기간이 생육특성 및 수량에 미치는 영향 (Effect of Nursery Period on the Growth and Yield of Green Papaya (Carica papaya) Production under Non-Heated Greenhouse)

  • 본 시험은 청과용 그린 파파야 무가온 생산시 육묘기간이 생육 및 수량특성에 미치는 영향을 구명하고자 'Red lady' 품종을 이용하여 수행하였다. 육묘기간은 3개월부터 13개월 까지 6처리로 하였으며 2015년 4월 15일 무가온 하우스내에 정식하였다. 육묘기간에 따른 정식 전 파파야 묘의 초장, 마디수, 생체중 등은 육묘기간이 긴 묘일수록 높았으며, 13개월 육묘에서 가장 생육이 좋았다. 첫 수확일수는 육묘기간이 짧아질수록 길어졌으며 13개월 육묘에서 137일(8월 18일)이 소요되었고 3개월 육묘에서 184일(10월 2일)로 가장 길었다. 과일특성은 육묘기간이 짧았던 3개월 육묘에서 과장과 과경이 가장 작았으나 나머지 처리 간에는 차이를 보이지 않았다. 수량은 육묘기간에 따라 영향을 받았으며 상품수량 역시 육묘기간이 길어짐에 따라 증가하였다. 13개월 육묘에서 가장 많았으며(3,172kg), 다음이 11개월(2,247kg) 및 9개월(2,357kg) 이었다. 7개월과 5개월에서 각각 1,942kg 및 1,787kg 이었으며, 3개월 육묘에서 1,443kg으로 가장 적었다. 특히 7개월 육묘 이하에서 상품수량은 1,942kg 이하로 크게 감소되었다. 이상의 결과 청과용 그린 파파야 무가온하우스 재배시 수확기를 앞당기기 위해서는 11개월 육묘가 유리하겠으나, 경영비 등을 고려한다면 9개월 정도 육묘가 바람직할 것으로 생각된다.

파파인 가공한 양모/폴리에스터 혼방직물의 정련 및 염색성 (Dyeing Properties and Scouring of Wool/Polyester Blend Fabrics Using Papain from Carica Papaya)

  • This study provides the optimum papain treatment method and its effect on wool/polyester blend fabrics. The enzymatic treatment condition is optimized depending on its pH level, temperature, concentration of enzyme, treatment time and concentration of activators. The characteristics of samples treated with the papain are measured using weight loss, tensile strength, whiteness, WCA, dyeing property and surface micrographs. The results are described as follows: According to measuring weight loss, tensile strength and whiteness, a pH level of 7.5, $70^{\circ}C$, 10% papain(o.w.f.) and 60minutes of treatment time are optimized for papain treatment. L-cysteine and sodium sulfite are able to activate the papain. The optimum concentrations of them are 10mM and 50mM respectively. The WCA of fabrics is decreased since papain treatment makes wool/polyester blend fabrics more hydrophilic. Scouring with papain treatment improves whiteness and dyeing property of fabrics. The dyeing property of papain-treated fabrics is enhanced simply by a single step dyeing process using a basic dye. The surface of wool treated with papain in the presence of L-cysteine shows to be descaled. The surface of wool fibers in the presence of sodium sulfite, however, shows it is hydrolyzed evenly instead of being descaled. The surface of papain treated polyester fibers shows cracks and voids.

양모.폴리에스터 혼방직물의 효소가공 시 활성제 복합사용의 효과 (Effects of Mixed Activators on Enzymatic Activation for Wool.polyester Blend Fabrics)

  • This study provides effects of mixed activators on enzymatic activation and determines optimum mixture ratio for enzymatic treatment. Wool 80% and polyester 20% blend fabric and papain from carica papaya are used in this experiment. L-cysteine and sodium sulfite are used as activators for papain treatment process. The treatment condition is pH 7.5, $70^{\circ}$, papain concentration 10%(o.w.f), 60 minutes. L-cysteine and sodium sulfite are added in enzyme solution with various concentrations($0{\sim}50mM$). The optimum treatment condition is determined by measuring weight loss, tensile strength, whiteness, water contact angle(WCA), dyeability and surface micrographs. The results are as follow; The optimum mixture ratio of activators is L-cysteine 2mM and sodium sulfite 10mM. Mixed activators assists in improving the activation of papain. WCA of papain treated fabrics is decreased since papain treatment with activator mixture makes wool polyester blend fabrics more hydrophilic. Dyeing property of papain-treated fabrics more improves by the treatment with mixed activators than with single activator. It means that this method can save time and lower cost. After papain treatment in the presence of mixed activator, the surface of fabrics is modified. The surface of wool fiber shows to be descaled and hydrolyzed, and that of polyester fiber shows to be cracked.
