Objective: The inhibitory effects of dietary antioxidants, diallyl disulfide (DADS) and quercetin, in marinade were investigated on the formation of carcinogenic polycyclic aromatic hydrocarbons (EPA priority 16 PAHs) in grilled pork. Methods: The formation of PAHs in grilled sirloin pork with different marinades after charcoal-grilling for 2 min/side were evaluated using high performance liquid chromatography with a photodiode array detector (HPLC-DAD). Results: Compared with the control marinade treatment (without antioxidant), the addition of DADS (500 mg/kg meat sample) in marinade significantly decreased benzo[a]pyrene (BaP) (100%) and heavy PAHs (84%) in charcoal-grilled pork, while the addition of quercetin at the same concentration could reduce 23% and 55% of BaP and heavy PAHs, respectively. Conclusion: The results of this study suggested that the addition of DADS in the marinade could be important in decreasing the levels of PAHs in grilled meat.

The research centers on the concentration profile and risk assessment of toxic metals for ambient air in Taejon industrial complex. Airborne concentrations of each toxic metal for risk assessment were obtained from 2-year sampling by high volume air sampler and analysis by ICP-MS and ICP-AES in the complex. The long-term arithmetic mean of human carcinogen, arsenic, hexavalent chromium and nickel subsulfide was 5.53, 2.16 and $3.46ng/m^3$ while the mean of probable human carcinogen, beryllium, cadmium and lead was 0.08, 2.35, $293.29ng/m^3$, respectively. And the long-term arithmetic mean concentration of non-carcinogenic metal, manganese was $55.91ng/m^3$. The point risk estimate for the inhalation of carcinogenic metals was $3.6{\times}10^{-5}$, which was higher than a risk standard of $10^{-5}$. About 75% of the cancer risk was to the inhalation of human carcinogen, arsenic. Thus, it is necessary to properly manage arsenic risk in Taejon industrial complex. The point hazard index by the inhalation of manganese was 1.1. Therefore, an investigation into Taejon industrial complex is needed to obtain more fine long-term concentration data for airborne non-carcinogenic metals including manganese.

This study was intended to resolve problems caused by different classification criteria and management methods of carcinogenicity, which have made industrial safety & health institutions and business employers difficult to execute projects or to carry out occupational safety and health related works, and have affected how civic groups perceive carcinogens. The content of this study contained the comparison of management and categorization standards for carcinogens between Korea and other countries as well as the current carcinogenicity-related information supply status of each professional institution. Furthermore, this research examined the current state of supplying information on carcinogenicity among major institutional information supply according to the categorization standard for carcinogens by UN GHS, Ministry of Employment and Labor in Korea(KMoEL), and GHS MSDS provided by Korea Occupational Safety & Health Agency(KOSHA). Now, professional agency provide 927 kinds of IARC, 237 kinds of NTP, 351 kinds of ACGIH and 1,006 kinds of EU ECHA information on carcinogenic agents. KMoEL provides carcinogenicity-related information of 58 chemical agents in accordance with the category of carcinogens guided by ACGIH. KOSHA offers 13,232 kinds of GHS MSDS information including 2,484 carcinogenic substances. Therefore, carcinogenicity-related information of chemical substances, which are not available on the existing GHS MSDS DB, should be updated for the future reference.
As currently conducted, standard rodent bioassays do not provide sufficient information to assess carcinogenic risk to humans at doses thousands of times below the maximum tolerated dose. Recent analyses indicate that measures of carcinogenic potency from these tests are restricted to a narrow range about the maximum tolerated dose and that information on shape of the dose-response is limited in experiments with only two doses and a control. Extrapolation from high to low doses should be based on an understanding of the mechanisms of carcinogenesis. We have postulated that administration of the maximum tolerated dose can increase mitogenesis which, in turn. increases rates of mutagenesis and, thus, carcinogenesis. The animal data are consistent with this mechanism, because about half of all chemicals tested are indeed rodent carcinogens, and about 40% of the positives are not detectably mutagenic. Thus, at low doses where cell killing does not occur, the hazards to humans of rodent carcinogens may be much lower than commonly assumed. In contrast, for high-dose exposures in the workplace, assessment of hazard requires comparatively little extrapolation. Nevertheless. permitted workplace exposures are sometimes close to the tumorigenic dose-rate in animal tests. Regulatory policy to prevent human cancer has primarily addressed synthetic chemicals, yet similar proportions of natural chemicals and synthetic chemicals test positive in rodent studies as expected from an understanding of toxicological defenses, and the vast proportion of human exposures are to natural chemicals. Thus, human exposures to rodent carcinogens are common. The natural chemicals are the control to evaluate regulatory strategies, and the possible hazards from synthetic chemicals should be compared to the possible hazards from natural chemicals. Qualitative extrapolation of the carcinogenic response between species has been investigated by comparing two closely related species: rats and mice. Overall predictive values provide moderate confidence in interspecies extrapolation; however, knowing that a chemical is positive at any site in one species gives only about a 50% chance that it will be positive at the same site in the other species.
The IARC Monographs Programme on the Evaluation of Carcinogenic Risks to Humans has reviewed, summarized and evaluated 869 environmental agents and exposures as oj June 2000. This large collection includes all relevant published epidemiological data on cancer in exposed humans and results of bioassays for carcinogenicity in experimental animals. Since 1986. cancer data have been systematically supplemented by summaries of other toxicological data that are relevant to assessments of carcinogenic hazard. These include summaries qf genetic and related effects of chemicals. which have been prepared as Genetic Activity Profiles (GAP) by the U.S. EPA in collaboration with IARC. As the Mono-graphs have proved increasingly valuable and influential worldwide. they have evolved into an encyclopedia on environmental carcinogenic risks to humans. However. the Monographs have historically been prepared only as printed books with limited distribution. and the Monographs Programme has needed to adjust to expectations oj wider availability. Since 1998 the evaluations and summaries have been globally accessible by Internet from IARC (http://www.iarc.fr) and the GAP profiles by Internet from EPA (http://www.epa.gov/gapdb/). with the two web sites linked. Improved EPN/ARC GAP database and software. GAP2000. now link GAP profiles directly to the appropriate IARC web pages for summaries of evaluations of a given compound and its overall IARC classification. During the year 2000. by means of optical character recognition (OCR) technology the entire series of IARC Monographs is being converted to an electronic version. In order to assess the long-term airborne concentrations of 6 metals such as Cr, Cd, Pb, Zn, K, and Na and the associated health resk, a great number of PM-10 samples were collected and analyzed in Kyung Hee University-Suwon Campus for three years from 1991 to 1993. The 3-year average concentration of Pb in respirable particulate matters was 142.6 ng/m$^{3}$ while that of Zn was 1,210.5ng/m$^{3}$. The corresponding hazard index from Pb Zn for the ingalation route was estimated to be 9.5.times.10$^{-2}$ and 3.5.times.10$^{-2}$ , respectively. Therefore, it can be said that there was no adverse chronic health effects by airborne Pb and Zn. However, the 2-year average concentration of Cr(6) was estimated to be 1.3ng/m$^{3}$ shile the 3-year average concentration of Cd was 3.1ng/m$^{3}$. The total cancer risk by these two metals for the inhalation route was estimated to be about 7.2.times.10$^{-6}$ . This order-of-magnitude risk estimate suggests that the ingalation risk in the study area from all carcinogenic metals including As and Ni (subsulfide) might exceed the acceptable risk criteria of 10$^{-5}$ -10$^{-6}$ by U.S.EPA.For a better risk assessment in the future, alveolar deposition of PM-10 in the study area were also discussed and an assessment was done.
Commonly eaten fish, meat and other protein-containing foods show some level of mutagenic activity following normal cooking such as broiling, frying, grilling, roasting etc. The main food mutagens found in cooked products are“heterocyclic aromatic amines”. Several of them have been shown to be carcinogenic in rodent and suggested to be relevant for human cancer etiology. This review summarizes the chemistry, formation, occurrence and toxicity of food-borne heterocyclic aromatic amines. Factors that influence the formation of them are also discussed with special emphasis on dietary factors. From a health safety point of view, it is desirable to estimate the intake of heterocyclic amines via foods, and reduce or prevent the formation of food mutagens.
