  • Background: With volatile organic compounds (VOCs) containing aromatic and halogenated hydrocarbons such as benzene, toluene, and xylene that can adversely affect the respiratory and cardiovascular systems when a certain concentration is reached, it is important to accurately evaluate the source and the corresponding health risk effects. Objectives: The purpose of this study is to provide scientific evidence for the city of Seoul's VOC reduction measures by confirming the risk of each VOC emission source. Methods: In 2020, 56 VOCs were measured and analyzed at one-hour intervals using an online flame ionization detector system (GC-FID) at two measuring stations in Seoul (Gangseo: GS, Bukhansan: BHS). The dominant emission source was identified using the Positive Matrix Factorization (PMF) model, and health risk assessment was performed on the main components of VOCs related to the emission source. Results: Gasoline vapor and vehicle combustion gas are the main sources of emissions in GS, a residential area in the city center, and the main sources are solvent usage and aged VOCs in BHS, a greenbelt area. The risk index ranged from 0.01 to 0.02, which is lower than the standard of 1 for both GS and BHS, and was an acceptable level of 5.71×10-7 to 2.58×10-6 for carcinogenic risk. Conclusions: In order to reduce the level of carcinogenic risk to an acceptable safe level, it is necessary to improve and reduce the emission sources of vehicle combustion and solvent usage, and eco-car policies are judged to contribute to the reduction of combustion gas as well as providing a response to climate change.

  • Exposure to hazardous substances occurs through multiple pathways. Aggregated risk assessment, which includes all potential exposure pathways to a single toxicant, is necessary to prevent exposure to harmful substances. We aimed to estimate cadmium and lead exposure through various media, such as food, water, air, smoking, cosmetics, and female hygiene products. This study covered 10,733 subjects from the Seventh Korea National Health and Nutrition Examination Survey(2016, 2017). Dietary exposure was estimated using 24-hour recall data. For water and inhalational exposure, regional variations were considered. Water was classified as tap, bottled, and public water. Inhalational exposure was estimated using the '2014 Time Use Survey' based on daily lifestyle and social status. The frequency and volume of cosmetic usage were randomly approximated by sex and age. Post-menarcheal and premenopausal women were assumed to use feminine hygiene products. Non-carcinogenic aggregated risks were estimated using the Aggregate Risk Index from EPAs and the Total Exposure Hazard Index from Korean government guidelines. For carcinogenic risk assessment, excessive cancer risk was estimated. Ingestion, especially food, was the major route for both cadmium and lead exposure. Smoking was also associated with high cadmium exposure. Exposure to lead from cosmetics was remarkable but not critical. In aggregate risk assessments, median cadmium and lead exposure did not exceed the reference value. Sex, age, smoking status, and income affected exposure levels, unlike to regional variations.

  • Background: Polycyclic aromatic hydrocarbons (PAHs) are generated in petrochemical complexes, can spread to residential areas and affect the health of residents. Although harmful PAHs are mainly present in particle phase, gas phase PAHs can generate stronger toxic substances through photochemical reaction. Therefore, the risk assessment for PAHs around the petrochemical complex should consider both particle and gas phase concentrations. Objectives: This study aimed to investigate the concentration characteristics of particle and gas phase PAHs by season in residential areas around petrochemical complexes, and to assess the risk of PAHs. Methods: Samples were collected for 7 days by seasons in 2014~2015 using a high volume air sampler. Particle and gas phase PAHs were sampled using quartz filter and polyurethane foam, respectively, analyzed by GC-MS. Chronic toxicity and probabilistic risk assessment were performed on 14 PAHs. For chronic toxicity risk assessment, inhalation unit risk was used. Monte-Carlo simulation was performed for probabilistic risk assessment using the mean and standard deviation of measured PAHs. Results: The concentration of particle total PAHs was highest in autumn. The gas phase concentration was highest in autumn. The average gas phase distribution ratio of low molecular weight PAHs composed of 2~3 benzene rings was 85%. The average of the medium molecular weight composed of 4 benzene rings was 53%, and the average of the high molecular weight composed of 5 or more benzene rings was 9%. In the chronic toxicity risk assessment, 7 of the 14 PAHs exceeded the excess carcinogenic risk of 1.00×10-6. In the Monte-Carlo simulation, Benzo[a]pyrene had the highest probability of exceeding 1.00×10-6, which was 100%. Conclusions: The concentration of PAHs in the residential area around the petrochemical complex exceeded the standard, and the excess carcinogenic risk was evaluated to be high. Therefore, it is necessary to manage the air environment around the petrochemical complex.

  • Background: When particles are absorbed into the human body, they penetrate deep into the lungs and interact with the tissues of the body. Heavy metals in PM2.5 can cause various diseases. The main source of PM2.5 emissions in South Korea's atmosphere has been surveyed to be places of business. Objectives: The concentration of heavy metals in PM2.5 near the Ulsan Industrial Complex was measured and a health risk assessment was performed for residents near the industrial complex for exposure to heavy metals in PM2.5. Methods: Concentrations of heavy metals in PM2.5 were measured at four measurement sites (Ulsan, Mipo, Onsan, Maegok) near the industrial complexes. Heavy metals were analyzed according to the Air Pollution Monitoring Network Installation and Operation Guidelines presented by the National Institute of Environmental Research. Among them, only five substances (Mn, Ni, As, Cd, Cr6+) were targeted. The risk assessment was conducted on inhalation exposure for five age groups, and the excess cancer risk and hazard quotient were calculated. Results: In the risk assessment of exposure to heavy metals in PM2.5, As, Cd, and Cr6+ exceeded the risk tolerance standard of 10-6 for carcinogenic hazards. The highest hazard levels were observed in Onsan and Mipo industrial complexes. In the case of non-carcinogenic hazards, Mn was identified as exceeding the hazard tolerance of 1, and it showed the highest hazard in the Ulsan Industrial Complex. Conclusions: This study presented a detailed health risk from exposure to heavy metals in PM2.5 by industrial complexes located in Ulsan among five age groups. It is expected to be utilized as the basis for preparing damage control and industrial emission reduction measures against PM2.5 exposure at the Ulsan Industrial Complex.

  • Backgrounds/Aims: Mechanisms for the development of hepatocellular carcinoma (HCC) in hepatitis B virus (HBV)-infected patients remain unclear. The aim of the present study was to identify genes and pathways involved in the development of HBV-associated HCC. Methods: The GSE121248 gene dataset, which included 70 HCCs and 37 adjacent liver tissues, was downloaded from the Gene Expression Omnibus database. Differentially expressed genes (DEGs) in HCCs and adjacent liver tissues were identified. Gene ontology and Kyoto Encyclopedia of Genes and Genome pathway enrichment analyses were then performed. Results: Of 134 DEGs identified, 34 were up-regulated and 100 were down-regulated in HCCs. The 34 up-regulated DEGs were mainly involved in nuclear division, organelle fission, spindle and midbody formation, histone kinase activity, and p53 signaling pathway, whereas the 100 down-regulated DEGs were involved in steroid and hormone metabolism, collagen-coated extracellular matrix, oxidoreductase activity, and activity on paired donors, including incorporation or reduction of molecular oxygen, monooxygenase activity, and retinol metabolism. Analyses of protein-protein interaction networks with a high degree of connectivity identified significant modules containing 14 hub genes, including ANLN, ASPM, BUB1B, CCNB1, CDK1, CDKN3, ECT2, HMMR, NEK2, PBK, PRC1, RACGAP1, RRM2, and TOP2A, which were mainly associated with nuclear division, organelle fission, spindle formation, protein serine/threonine kinase activity, p53 signaling pathway, and cell cycle. Conclusions: This study identified key genes and carcinogenic pathways that play essential roles in the development of HBV-associated HCC. This may provide important information for the development of diagnostic and therapeutic targets for HCC.

  • The United States Environmental Protection Agency (EPA) characterized the cancer hazard of di(2-ethylhexyl)-phthalate (DEHP) as a B2 group (probable human carcinogen) and proposed "Guide-lines for Carcinogen Risk Assessment". This guidelines proposed alternative methods for analyzing carcinogen dose-response data and for extrapolating the effects of observed at high dose to predict that might occur at lower doses relevant to human exposure. This proposed guidelines state that "If in a particular case, the evidence indicated a threshold, as in the case of carcinogenicity being secondary to another toxicity that has a threshold, the margin of exposure analysis for toxicity is the same as is done for a non-cancer endpoint". DEHP is excellent candidate for reconideration under the new guidelines for carcinogen risk assessment (John Doull et al., 1998). This study is conducted about risk assessment for infant exposure on DEHP in powdered milk wing methodology in EPA's new guideline on carcinogenic risk assessment. Estimated cancer risk of DEHP in powdered milk and cow milk is 2.83$\times$$10^5$ (using cancer potency: 1.4$\times$$10^2$/ (mg/kg/day)) as mean and MOE is 12075 (using selected NOEL 20 mg/kg/day) as mean. mg/kg/day) as mean.

  • Sulfamethazine has been widely used in swine for prevention or treatment of infections. Recently, the safety of the drug to consumers has been questioned because of carcinogenic effects. To prevent unwanted drug residues entering the human food chain, both government authorities and industries have established extensive control measures. The demands for reliable, simple, sensitive, rapid and low-cost methods for residue analysis of foods are increasing nowadays. In this study, we established a rapid prediction test for the detection of cattle with violative tissue residues of sulfamethazine. The recommended therapeutic dose of sulfamethazine (withdrawal time, 15 days) was administered to each of 10 cattle. Blood was sampled before drug administration and during the withdrawal period. The concentration of sulfamethazine in plasma, determined by a semi-quantitative ELISA, was compared to that of an internal standard (10 ppb). The absorbance ratio of internal standard to sample (B/Bs) was employed as an index to determine whether drug residues in cattle tissues were negative or positive. That is, a B/Bs ratio less than 1 was considered residue positive and if larger than 1 was considered negative. All 10 plasma samples from non-treated cattle showed negative to sulfamethazine. Sulfamethazine was detected in plasmas of treated cattle until Day 7 of withdrawal period. The present study showed that the semi-quantitative ELISA could be easily adapted in predicting residues of sulfamethazine in live cattle.

  • 17-${\beta}$-hydroxysteroid dehydrogenase type 1 ($17{\beta}$-HSD type 1) mediates the reaction of $17{\beta}$-estradiol (E2) production from estrone (E1). Inhibitory effects of curcuminoids on $17{\beta}$-HSD type 1 activity were investigated to find a lead compound for treating estrogen-dependent diseases including breast cancer. Among curcuminoids, demethoxycurcumin showed potent inhibitory effect ($IC_{50}=2.7{\mu}M$) on mouse $17{\beta}$-HSD type 1. Curcuminoids also displayed their inhibitory effects on the production of $17{\alpha}$-estradiol which is a carcinogenic metabolite produced by the enzyme. Bisdemethoxycurcumin ($IC_{50}=1.3{\mu}M$) showed potent inhibitory effect on the $17{\alpha}$-estradiol production by chicken $17{\beta}$-HSD type 1. Curcuminoids did not inhibit ERE transcriptional activity with and without E2. Taken together, curcuminoids can be used for treating and preventing E2-dependent diseases via inhibition on $17{\beta}$-HSD type 1 activity.

  • The increasing consumption of pseudo-petroleum products (PPP) has been disturbing the national petroleum market. The use of PPP lead to tax evasion, disturbance of sound trading principles, component corrosion of cars, and explosion accident. Also, PPP have emitted hazardous air pollutants (HAP) including the carcinogenic aromatic hydrocarbons, PAHs and aldehydes more than regular-petroleum products. It thus has potentials to cause many environmental and health care problems. In this study, benzene, toluene and xylene emissions from road transport vehicles due to the use of pseudo-gasoline are estimated for the year 2008. The results of our study provide emission estimates of benzene, toluene and xylene for the year as 405, 1,711, 717 tonne/yr, respectively for regular-gasoline. BTX emissions are calculated as 452~515, 1,882~2,264 and 732~752 tonne/yr when the amount of pseudo-gasoline is estimated to account for 6~13% for regular-gasoline consumption. BTX emissions increased as much as 12~27, 10~32, 2~5% by using pseudo-gasoline. It is found that the pseudo-gasoline should be the key component to produce HAP in urban area.

  • Breast cancer is the most commonly diagnosed cancer in women in the world and is one of the leading causes of death due to cancer. Health benefits have been linked to additive and synergistic combinations of phytochemicals in fruits and vegetables. Nigella sativa has been shown to possess anti-carcinogenic activity, inhibiting growth of several cancer cell lines in vitro. However, the molecular mechanisms of the anti-cancer properties of Nigella sativa phytochemical extracts have not been completely understood. Our data showed that Nigella sativa extracts significantly inhibited human breast cancer MDA-MB-231 cell proliferation at doses of $2.5-5{\mu}g/mL$ (P<0.05). Apoptotic induction in MDA-MB-231 cells was observed in a dose-dependent manner after exposure to Nigella sativa extracts for 48 h. Real time PCR and flow cytometry analyses suggested that Nigella sativa extracts possess the ability to suppress the proliferation of human breast cancer cells through induction of apoptosis.
