Preparation of the Proteus vulgaris Bacterial Electrodes for the Determination of Urea and Their Application (요소 정량을 위한 Proteus vulgaris 박테리아 전극의 개발과 그 응용)

  • The bacteria containing urease convert each molecule of urea into two molecules of ammonia and one molecule of carbon dioxide gas. Bacterial electrodes have been constructed by immobilizing the Proteus vulgaris on an ammonia and a carbon dioxide gas-sensors, and were investigated for the effects of pH, temperature, buffer solution, bacterial amounts and interferences, and life time. NH3-bacterial electrode based on ammonia gas-sensor had linearity in the range of $7.0{\times}10^{-4}\;-\;3.0{\times}10^{-2}$M urea in pH 7.4, 0.05M phosphate buffer at $25^{\circ}C$ with a slope of 116.7 mV/decade. While $CO_{2-}$bacterial electrode based on carbon dioxide gas-sensor bad linearity in the range of $7.0{\times}10^{-4}\;-\;5. 0{\times}10^{-2}$M urea in pH 7.0, 0.1M phosphate buffer at $30^{\circ}C$with a slope of $45.4{\times}45.7mV/decade$. As the clinical application, urea in urine was determined by these devices and this result was compared with spectrophotometric method. Consequently, these electrodes could be used for the analysis of many samples because of simplicity, rapidity and convenience of the experimental procedure.

  • It was intended in this study to seek for the measures to utilize the USN technique, which has high usability due to low price and low power consumption, in air quality monitoring. As a method, the sensors of temperature, humidity, particulate matters (PM10), and carbon dioxide ($CO_2$) were installed in the self-manufactured sensor nodes; the nodes were installed in the waiting rooms and platforms of a subway station and the measurements were collected at real time with use of a computer which micro gateway was built in. Collected data was to be processed by the statistics program installed in the computer; the collected data is to be used in managing the air quality of stations after transmission to the ventilation system of ventilation chambers.

Assessment of soil moisture-vegetation-carbon flux relationship for agricultural drought using optical multispectral sensor (다중분광광학센서를 활용한 농업가뭄의 토양수분-식생-이산화탄소 플럭스 관계 분석)

  • Agricultural drought is triggered by a depletion of moisture content in the soil, which hinders photosynthesis and thus increases carbon dioxide (CO2) concentrations in the atmosphere. The aim of this study is to analyze the relationship between soil moisture (SM) and vegetation activity toward quantifying CO2 concentration in the atmosphere. To this end, the MODerate resolution imaging spectroradiometer (MODIS), an optical multispectral sensor, was used to evaluate two regions in South Korea for validation. Vegetation activity was analyzed through MOD13A1 vegetation indices products, and MODIS gross primary productivity (GPP) product was used to calculate the CO2 flux based on its relationship with respiration. In the case of SM, it was calculated through the method of applying apparent thermal inertia (ATI) in combination with land surface temperature and albedo. To validate the SM and CO2 flux, flux tower data was used which are the observed measurement values for the extreme drought period of 2014 and 2015 in South Korea. These two variables were analyzed for temporal variation on flux tower data as daily time scale, and the relationship with vegetation index (VI) was synthesized and analyzed on a monthly scale. The highest correlation between SM and VI (correlation coefficient (r) = 0.82) was observed at a time lag of one month, and that between VI and CO2 (r = 0.81) at half month. This regional study suggests a potential capability of MODIS-based SM, VI, and CO2 flux, which can be applied to an assessment of the global view of the agricultural drought by using available satellite remote sensing products.

Preparation of the Citrobacter freundii Bio-Sensor for the Determination of Glucose and Its Applications (Glucose 정량을 위한 Citrobacter freundii Bio-Sensor의 개발과 그 응용)

  • A bio-sensor for the determination of glucose has been constructed by immobilizing the Citrobacter freundii or its organelle on carbon dioxide gas-sensor. The bacterial sensor was better than organelle in response, but the latter showed a shorter response time. The bacterial sensor gave linearity between 7.0 ${\times}\;10^{-4}$ and 1.0 ${\times}\;10^{-2}$ M glucose with a slope of 42.2 mV/decade in pH 7.0, 0.2 M tris-HCl buffer at 30$^{\circ}C$. The selectivity of this sensor was very high for glucose. Employing for the determination of glucose in serum, the sensor showed a good agreement with a routine analyzer.

Highly Sensitive and Fast-Responding Ethanol Sensor using Au Doped-In2O3 Hollow Spheres

  • Pure and 0.3 wt% Au-doped In2O3 hollow spheres were synthesized via ultrasonic spray pyrolysis of droplets containing an In-source and sucrose in air and their gas sensing characteristics to 1 ppm ethanol, 1 ppm toluene, 1 ppm xylene, 2 ppm nitrogen dioxide (NO2), and 30 ppm carbon monoxide (CO) were measured at 400 - 450℃. The pure In2O3 hollow spheres exhibited relatively low gas responses and sluggish recovery kinetics. In contrast, the doping of Au into In2O3 hollow spheres significantly increased the gas response (S= resistance ratio) to 1 ppm ethanol (S= 20.6) at 400℃ with low cross-responses (S = 1.3-8.8) to other interference gases. Furthermore, the hollow spherical morphology of In2O3 provides a large surface area and facilitates rapid gas diffusion, resulting in fast response and recovery times. The sensor exhibited excellent performance with a low detection limit of 1.6 ppb. These findings indicate that the Au-In2O3 hollow spheres are promising candidates for advanced ethanol-sensing applications, particularly in breath-alcohol monitoring for ignition interlock devices.

Optical waveguide structure design of Non-dispersive Infrared (NDIR) CO2 gas sensor for high-sensitivity (이산화탄소 검출을 위한 고감도 비분산 적외선 가스센서의 광도파관 구조 설계)

  • The Non-dispersive Infrared (NDIR) gas sensor has high selectivity, measurement reliability, and long lifespan. Thus, even though the NDIR gas sensor is expensive, it is still widely used for carbon dioxide (CO2) detection. In this study, to reduce the cost of the NDIR CO2 gas sensor, we proposed the new optical waveguide structure design based on ready-made gas pipes that can improve the sensitivity by increasing the initial light intensity. The new optical waveguide design is a structure in which a part of the optical waveguide filter is inclined to increase the transmittance of the filter, and a parabolic mirror is installed at the rear end of the filter to focus the infrared rays passing through the filter to the detector. In order to examine the output characteristics of the new optical waveguide structure design, optical simulation was performed for two types of IR-source. As a result, the new optical waveguide structure can improve the sensitivity of the NDIR CO2 gas sensor by making the infrared rays perpendicular to the filter, increasing the filter transmittance.

Development of a Greenhouse Environment Monitoring System using Low-cost Microcontroller and Open-source Software (저비용 개방형 Microcontroller를 사용한 온실 환경 측정 시스템 개발)

  • Continuous monitoring of environmental parameters provides farmers with useful information, which can improve the quality and productivity of crops grown in greenhouses. The objective of this study was to develop a greenhouse environment measurement system using a low-cost microcontroller with open-source software. Greenhouse environment parameters measured were air temperature, relative humidity, and carbon dioxide ($CO_2$) concentration. The ranges of the temperature, relative humidity, and $CO_2$ concentration were -40 to $120^{\circ}C$, 0 to 100%, and 0 to 10,000 ppm, respectively. A $128{\times}64$ graphic LCD display was used for real-time monitoring of the greenhouse environments. An Arduino Uno R3 consisted of a USB interface for communicating with a computer, 6 analog inputs, and 14 digital input/output pins. A temperature/relative humidity sensor was connected to digital pins 2 and 3. A $CO_2$ sensor was connected to digital pins 12 and 13. The LCD was connected to digital pin 1 (TX). The sketches were programmed with the Arduino Software (IDE). A measurement system including the Arduino board, sensors, and accessories was developed (totaling $244). Data for the environmental parameters in a venlo-type greenhouse were obtained using this system without any problems. We expect that the low-cost microcontroller using open-source software can be used for monitoring the environments of plastic greenhouses in Korea.

The method for total organic carbon analysis employing TiO2 photocatalyst (이산화티타늄 광촉매를 이용한 총유기탄소 분석방법)

  • Biochemical oxygen demand (BOD) and chemical oxygen demand (COD) methods are conventional analytical methods to analyze water quality. Both of these methods are technically indirect measurement methods, require complicated preconditions, and are time-consuming. On the other hand, the total organic carbon (TOC) method is a direct and fast measurement method which is more intuitive and accurate than the BOD and COD methods. However, general TOC analysis methods involve complicated processes and high power consumption owing to the process of phase transition from liquid to gas by a high-temperature heater. Furthermore, periodic consumables are also required for the removal of inorganic carbon (IC). Titanium dioxide (TiO2) is one of the most suitable photocatalysts for simple processes. Its usage involves low power consumption because it only reacts with the organic carbon (OC) without the requirement of any other reagents and extra processes. We investigated a TiO2 photocatalyst-based TOC analysis for simple and affordable products. TiO2-coated fiber substrate maintained under carbon included water was exposed to ultraviolet (UV) radiation of wavelength 365 nm. This method is suitable for the real-time monitoring of water pollution because of its fast reaction time. Its linear property is also sufficient to match the real value.

Fabrication and Evaluation of a Total Organic Carbon Analyzer Using Photocatalysis

  • Water quality is crucial for human health and the environment. Accurate measurement of the quantity of organic carbon in water is essential for water quality evaluation, identification of water pollution sources, and appropriate implementation of water treatment measures. Total organic carbon (TOC) analysis is an important tool for this purpose. Although other methods, such as chemical oxygen demand (COD) and biochemical oxygen demand (BOD) are also used to measure organic carbon in water, they have limitations that make TOC analysis a more favorable option in certain situations. For example, COD requires the use of toxic chemicals, and BOD is time-consuming and can produce inconsistent and unreliable results. In contrast, TOC analysis is rapid and reliable, providing accurate measurements of organic carbon content in water. However, common methods for TOC analysis can be complex and energy-intensive because of the use of high-temperature heaters for liquid-to-gas phase transitions and the use of acid, which present safety risks. This study focuses on a TOC analysis method using TiO2 photocatalysis, which has several advantages over conventional TOC analysis methods, including its low cost and easy maintenance. For TiO2, rutile and anatase powders are mixed with an inorganic binder and spray-coated onto a glass fiber substrate. The TiO2 powder and inorganic binder solutions are adjusted to optimize the photocatalytic reaction performance. The TiO2 photocatalysis method is a simple and low-power approach to TOC analysis, making it a promising alternative to commonly used TOC analysis methods. This study aims to contribute to the development of more efficient and cost-effective approaches for water quality analysis and management by exploring the effectiveness and reliability of the developed equipment.

ENVIROMENTAL CONDITION DURING AIR SHIPMENT OF HORTICULTURAL PRODUCTS FROM OKINAWA TO TOKYO

  • Air shipment affords the quickest possible delivery of horticultural products. The price of air shipped horticultural products are relatively high as most of these products are perishable. Usually the temperature in the cargo compartment is not controlled during flight. Thus, special attention should be paid to procooling prior to shipment. The environmental condition during transportation of horticultural products is an essential parameter for maintaining the quality of perishable products. Commonly horticultural products were loaded by ULD(Unit Load Devices) as a container or pallet in the aircraft (except for small aircraft) . Therefore, inside temperature of the container and cargo compartment came into question. Scarce literature on the relationship between environmental condition and quality changes of horticultural products during air shipment can be found. By the stand point of keeping fresh quality, investigations on the actual condition of air shipments were carried out to improve the technique during the distribution process of fresh horticultural products. Temperature, humidity, atmospheric pressure, carbon dioxide, ethylene, impacts, and changes in quality during the air shipment of snapbeans, okras and chrysanthemums were measured. Temperature was measured by recording thermometers, relative humidity by recording hygrometers, atmospheric pressure by a strain -guage type pressure sensor, carbon dioxide by testing tubes, ethylene by sampling bags and a gaschromatograph, impacts and vibrations by impact recorders and a 3D accelerometer. Relationships between environmental conditions and quality changes during air shipments were clarified. It was expected from investigations into actual shipments that the ventilation and insulation properties of air freight containers were related to the quality of agricultural products. Aircraft can no directly load and unload trucks into them. The transshipment is inclined to cause shocks and vibrations, and to invite damages within a short time.
