Effects of nitrogen and organic carbon sources on growth and lipid production of Chlorella sp. KR-1 in flask cultures (플라스크 배양에서 Chlorella sp. KR-1의 균체 성장 및 지질 생산에 대한 질소원 및 유기탄소원의 영향)

  • Recently microalgae have been proposed as a promising biodiesel feedstock, owing to their higher lipid productivity and non-arable land based cultivation system. Biomass and lipid productivities of microalgae are largely affected by various environmental and nutritional factors. In this study, the effects of nitrogen (nitrate and ammonium) and organic carbon (glucose and glycerol) sources on the cell growth and lipid production of Chlorella sp. KR-1 were examined in flask cultures. Under autotrophic culture conditions for 15 days, overall cell growth and lipid (fatty acid methyl ester, FAME) production with nitrate were better than those of ammonium, resulting in 1.06 g cell/L and 333 mg FAME/L, respectively. Maximal intracellular lipid contents (348 - 352 mg FAME/g cell) were observed at low concentrations of 1 mM for both nitrate and ammonium. In the supply of light, addition of glucose in the range of 1 - 20 g/L showed higher cell densities than the autotrophic cell growth condition. Higher lipid accumulation of 375 mg FAME/g cell could achieved at 5 g glucose/L albeit of relatively short incubation of 7 days. With glycerol, intracellular lipid contents were ~1.9 times lower than glucose cases although similar cell growths were observed for both carbon sources.

Comparison of Carbon Sequestration Potential of Winter Cover Crop Cultivation in Rice Paddy Soil

  • BACKGROUND: Cultivation of winter cover crops is strongly recommended to increase land utilization efficiency, animal feeding material self-production, and to improve soil and environmental quality. METHODS AND RESULTS: Four major winter crops (barley, Chinese milk vetch, hairy vetch, and rye) having different C/N ratio were seeded in silt loam paddy soil in the November 2007 and the aboveground biomass was harvested on the late May 2008 to evaluate its effectiveness as green manure, and root biomass distribution was characterized at the different depth (0-60 cm) to study its effect on physical properties and carbon sequestration in soil. During this experiment, the naturally growing weed in the rice paddy soil in Korea, short awn foxtail (Alopecurus aequalis Sobol), was considered as control treatment. Above-ground biomass of all cover crops selected was significantly higher than that of the control treatment (2.8 Mg/ha). Comparatively higher above-ground biomass productivity of rye and barley (15.8 and 13.5 Mg/ha, respectively) suggested that these cover crops possibly had the highest potential as a green manure and animal feeding material. Root biomass production of different cover crops followed the same trend as that for their above ground biomass. Rye (Secale cereal) might have the highest potential for soil C accumulation (7893 C kg/ha) by root biomass development, and then followed by barley (6985 C kg/ha), hairy vetch (6467 C kg/ha), Chinese milk vetch (6671 C kg/ha), and control (5791 C kg/ha). CONCLUSION(s): Cover crops like rye and barley having high biomass productivity might be the most effective winter cover crops to increase organic carbon distribution in different soil aggregates which might be beneficial to improve soil structure, aeration etc. and C sequestration.

  • The liquefied petroleum gas(LPG), mixture of propane and butane, has the potential to reduce toxic hydrocarbon emissions and inhibit ozone formation due to its chemical composition. Conventional mixer systems, however, have problems in meeting the future lower emission standards because of the difficulty in controlling air-fuel ratio precisely according to mileage tar accumulation. Liquid Phase LPG injection(LPLi) system has several advantages in more precise fuel metering and higher engine performance than those of the conventional mixer type. On the other hands, leakage problem of LPLi system at the injector tip is a main obstacle for meeting more stringent future emission regulations because these phenomena might cause excessive amount of THC emission during cold and hot restart phase. The main focus of this paper is the development of a leaked fuel recirculation system, which can eliminate the leaked fuel at the intake system with the activated carbon canister. Leaked fuel level was evaluated by using a fast response THC analyzer and gas chromatography. The result shows that THC concentration during cold and hot restart stage decreases by over 60%, and recirculation system is an effective method to meet the SULEV standard of the LPLi engine.

  • To increase the high production of 1-deoxynojirimycin (DNJ) from Streptomyces sp. SID9135, the effect of various carbon sources, nitrogen sources, cationic metal ions, the initial pH of the medium, and agitation speed were investigated. The most effective carbon and nitrogen sources were found to be lactose 2.5% (w/v) and soybean meal 2.0% (w/v), respectively. None of the cationic metal ions examined had any detectable stimulating effect on DNJ production except $Fe^{+2}$ ion. The initial optimum pH for DNJ production ranged from 6-8 and agitation speed was most effective at 400 rpm. In the jar fermentor experiments under optimal culture conditions, the accumulation of DNJ reached about $640{\mu}g$/ml after 5 days of cultivation and the level remained the same for a further two days.

Implication of the Change in Overturning Circulation to the LGM CO2 Budget

  • The observational proxy estimates suggest that the North Atlantic overturning stream function associated with the North Atlantic Deep Water (NADW) production and outflow was substantially weaker during the last glacial maximum (LGM) than that observed under present conditions. The impact of the changes in overturning circulation on the glacial carbon budget is investigated using a box model. The carbon box model reveals that the atmospheric $CO_2$ concentration is more sensitive to change in the overturning circulation of the North Atlantic than that of the Southern Ocean, especially when North Atlantic overturning becomes weaker. For example, when the strength of the North Atlantic overturning circulation is halved, the atmospheric $CO_2$ concentration is reduced by 50ppm of that associated with the accumulation of $CO_2$ in the deep ocean. This result implies that a weaker North Atlantic overturning circulation may play an important role in the lowering of LGM atmospheric $CO_2$ concentration.

Extracts of Codonopsis lanceolata Decrease $CCl_4$-Induced Hepatotoxicity on High Fat Diet in Rats

  • This study was designed to investigate the effect of Codonopsis lanceolata extracts on carbon tetrachloride (CCl₄)­induced hepatotoxicity in rats fed high, medium or low fat diet. The liver weight was increased not by the level of fat in diet but by administration of CCl₄. Activities of serum aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were significantly increased by CCl₄, the activities of AST and alanine aminotransferase (ALT) were significantly decreased in case of CCl₄ treatment with C. lanceolata extracts. In high fat diet with CCl₄ administration, the content of total lipid in liver was significantly increased, which was significantly decreased by the administration of C. lanceolata extracts. Administration of C. lanceolata water extracts remarkably prevented the tissue injury such as lipid degeneration, infiltration of inflanunatory cells and necrosis caused by CCl₄, but those methanol extracts did not affect histopathological changes of the liver as water extract. The data suggest that intake of C. lanceolata extracts would prevent lipid accumulation and hepatotoxicity induced by high fat diet and CCl₄ abministratim, and the water extract of C. lanceolata is supposed to be more effective than those methanol extract.

  • In order to investigate central metabolic changes in Beijerinckia indica, cells were grown on different carbon sources and intracellular flux distributions were studied under varying concentrations of nitrogen. Metabolic fluxes were estimated by combining material balances with extracellular substrate uptake rate, biomass formation rate, and exopolysaccharide (EPS) accumulation rate. Thirty-one metabolic reactions and 30 intracellular metabolites were considered for the flux analysis. The results revealed that most of the carbon source was directed into the Entner-Doudoroff pathway, followed by the recycling of triose-3-phosphate back to Hexose­6-phosphate. The pentose phosphate pathway was operated at a minimal level to supply the precursors for biomass formation. The different metabolic behaviors under varying nitrogen concentrations were observed with flux analysis.

  • The European Iron Fertilization Experiment EIFEX studied the growth and decline of a phytoplankton bloom stimulated by fertilising $10km^2$ in the core of a mesoscale $(80{\times}120km)$ cyclonic eddy south of the Antarctic Polar Front with about 2 times 7 tonnes of iron sulphate. The phytoplankton accumulation induced by iron fertilization did not exceed $3{\mu}g\;chl\;a\;l^{-1}$ despite a draw down of $5{\mu}M$ of nitrate that should have resulted in at least double to triple the amount of phytoplankton biomass assuming regular Redfield-ratios for draw down after phytoplankton growth in the Southern Ocean. During EIFEX the fertilized core of the mesoscale eddy evolved to a hotspot for a variety of small and medium sized mesozooplankton copepods. In contrast to copepods, the biomass of salps (Salpa thompson)) that dominated zooplankton biomass before the onset of our experiment decreased to nearly extinction. Most of the species of the rnosozooplankton community showed extremely hiか feeding rates compared to literature values from Southern Ocean summer communities. At the end of the experiment, massive phytoplankton sedimentation reached the sea floor at about 3800m water depth.

Electrical Properties and Self-poling Mechanism of CNT/PVDF Piezoelectric Composite Films Prepared by Spray Coating Method

  • Carbon nanotubes (CNT) / polyvinylidene fluoride (PVDF) piezoelectric composite films for nanogenerator devices were fabricated by spray coating method. When the CNT/PVDF mixture solution passes through the spray nozzle with small diameter by the compressed nitrogen gas, electric charges are generated in the liquid by a triboelectric effect. Then randomly distributed ${\beta}$ phase PVDF film could be re-oriented by the electric field resulting from the accumulated electrical charges, and might be resulted in extremely one-directionally aligned ${\beta}$ phase PVDF film without additional electric field for poling. X-ray diffraction patterns were used to investigate crystal structure of the CNT/PVDF composite films. It was confirmed that they revealed extremely large portion of the ${\beta}$ phase PVDF crystalline in the film. Therefore we could obtain the poled CNT/PVDF piezoelectric composite films by the spray coating method without additional poling process. Charge accumulation and resulting electric field generation mechanism by spray coating method were shown in Fig. 1. The capacitance of the CNT/PVDF films increased by adding CNTs into the PVDF matrix, and finally saturated. However, the I-V curves didn't show any saturation effect in the CNT concentration range of 0~4 wt%. Therefore we can control the performance of the devices fabricated from the CNT/PVDF composite film by adjusting the current level resulted from the CNT concentration with the uniform capacitance value.

  • Radionuclides, particularly radioactive cesium (Cs), are a concern of human health in some nuclear power accidents. It could lead to a high level of intracellular accumulation due to its high radioactivity and long half-life. Therefore, it is imperative to develop a method to remove Cs from wastewater. Herein, we synthesized activated carbon fibers (ACFs) doped with Prussian blue (PB) via in situ methods. We classified samples by their preparation method as either physical (PB-ACF-A) or physicochemical (PB-ACF-B) syntheses for comparison. The PB-ACF-B sample showed a significant surface loss compared to PB-ACF-A but a better 133Cs adsorption capacity. All samples fit well to Langmuir isotherms and the values of qmax were directly correlated to the amount of PB on the surface of the ACFs. Adsorption characteristics were further confirmed by the calculated free energy, enthalpy, and entropy.
