Effects of Prepartum Dietary Carbohydrate Source on Metabolism and Performance of Primiparous Holstein Cows during the Periparturient Period

  • Forty-six Holstein heifers were used in a completely randomized design and assigned to 1 of 2 treatments to evaluate the effects of 2 diets varying in ruminal fermentable carbohydrate sources, namely ground corn (GC) and rolled wheat (RW), on metabolism and performance of primiparous cows in the periparturient period. The heifers were fed diets as a total mixed ration (TMR) with similar energy and crude protein content including i) 18.57% GC, or ii) 18.57% RW from -24.13${\pm}$7.73 d relative to expected calving until calving. After calving, all animals received the same lactation diet until 28 d. Animals were group fed from the beginning of the study to -7 d relative to expected calving, fed individually from d -7 to 7 days in milk (DIM), and again group fed to 28 DIM. The pre-partum diets affected (p<0.05) dry matter intake (DMI), energy intake, energy balance (EB) and urinary pH during the last week pre-partum. There was no effect of pre-partum carbohydrate source on overall plasma concentration of glucose, nonesterified fatty acid (NEFA), $\beta$-hydroxybutyrate (BHBA), albumin, triglyceride (TG), cholesterol, aspartate aminotransferase (AST), insulin, and cortisol during the periparturient period. Cows fed the RW diet during the pre-partum period had greater calcium for the first week (p<0.05) and during 28 d (p = 0.08) of lactation compared with heifers fed the GC diet. Primiparous cows fed the RW diet produced greater milk protein content and yield (p<0.05). Primiparous cows fed the RW diet had lower milk urea nitrogen (MUN) and somatic cell count (SCC) than cows fed the GC diet (p<0.05). The results of this study show that feeding pre-partum diets with a rapidly fermentable source of starch but low energy content can improve animal metabolism and performance and smooth the transition of primiparous Holstein cows from gestation to lactation.

  • The problems with unsorted municipal waste are always associated with disposal issues as it requires a large area for landfilling or high energy used for incineration. In recent years, an autoclaving technique has been considered a promising approach which could minimize the volume of organic waste from being directly disposed or incinerated. In this work, an attempt was done to study the saccharification potential of organic residues under elevated temperature Thermal treatment involving hot water bath was applied to treat the organic residue ranging from 60℃ to 100℃ for 30 and 60 minutes. The result obtained showed an increasing trend for the concentration of glucose and carbohydrate. However, the result for lignocellulose content which contains various component includes extractive, holocellulose, hemicellulose, cellulose and lignin show variation. Based on the thermal treatment carried out, the result indicated that the trend of glucose and carbohydrate content. The highest percentage of glucose that can be obtained 978.602 ㎍/ml which could be obtained at 90℃ at 60 minutes. The carbohydrate also shows an increasing trend with 0.234 mg/ml as the highest peak achieved at 80℃ for 30 minutes treatment. However, it was found that the lignocellulose content varies with temperature and time. The statistical analysis was carried out using two-ways ANOVA shows an interaction effect between the independent variables (temperature and contact time) and the saccharification effects on the food wastes. The result shows a variation in the significant effect of independent variables on the changes in the composition of food waste.

Effects of lactic acid bacteria and molasses on fermentation dynamics, structural and nonstructural carbohydrate composition and in vitro ruminal fermentation of rice straw silage

  • Objective: This study was to evaluate the fermentation dynamics, structural and nonstructural carbohydrate composition and in vitro gas production of rice straw ensiled with lactic acid bacteria and molasses. Methods: Fresh rice straw was ensiled in 1-L laboratory silos with no additive control (C), Lactobacillus plantarum (L), molasses (M) and molasses+Lactobacillus plantarum (ML) for 6, 15, 30, and 60 days. After storage, the silages were subjected to microbial and chemical analyses as well as the further in vitro fermentation trial. Results: All additives increased lactic acid concentration, and reduced pH, dry matter (DM) loss and structural carbohydrate content relative to the control (p<0.05). The highest organic acid and residual sugar contents and lignocellulose reduction were observed in ML silage. L silage had the highest V-score with 88.10 followed by ML silage. L and ML silage improved in vitro DM digestibility as compared with other treatments, while in vitro neutral detergent fibre degradability (IVNDFD) was increased in M and ML silage (p<0.05). M silage significantly (p<0.05) increased propionic acid (PA) content and decreased butyric acid content and acetic acid/PA as well as 72-h cumulative gas production. Conclusion: The application of ML was effective for improving both the fermentation quality and in vitro digestibility of rice straw silage. Inclusion with molasses to rice straw could reduce in vitro ruminal gas production.

Comparison of time course changes in blood glucose, insulin and lipids between high carbohydrate and high fat meals in healthy young women

  • Few studies have examined short tenn responses to the different contents of carbohydrate or fat in the meal, although long tenn effects of the high fat meal have been considered as compound risk factor for metabolic disorders. The aim of this study was to investigate the postprandial changes of plasma glucose, insulin and lipids upon intakes of high carbohydrate or high fat meal in young healthy women. Subjects were randomly assigned to either the high carbohydrate meal (HCM, 75% carbohydrate, n=13) or the high fat meal (HFM, 60% fat, n=12) groups. The meals were prepared as isocaloric typical Korean menu. Blood samples were obtained prior to and 30, 60, 90, 120, 180 and 240 minute after the meal. There were no significant differences on fasting blood parameters including glucose, insulin, lipids concentrations between the groups prior to the test. The HCM had higher blood glucose and insulin concentrations, reached the peak at 30 min and maintained for 240 min compared to the HFM (P<0.05). The HFM had higher plasma triglyceride (TG) and free fatty acid (FFA) concentrations, reached the peak at 120 min and maintained for 240 min compared to the HCM (P<0.05). It is concluded that macronutrients content in the meal may be an important determinant of postprandial substrate utilization in healthy women.

  • Fifteen naked barley cultivars including radiation breeding lines from three places were analized for crude protein, carbohydrates, P, K, Ca, Mg and tested protein by dye binding method and biuret method. Their content and simple correlation analyses among them were as follows. 1. Protein content was 7.67 for average (max. 10.3 in Baegdong, min. 6.0 in Bangju) that was lower than in milled barley and had significant (at p=0.01) correlation with dye binding capacity (r=0.769) and biuret absorbance (r=0.616). 2. Protein content also had significant correlation with $P_2O_5$(r=0.607, p=0.01) and with MgO(r=0.498, p=0.05). 3. There was great difference in protein content among radiation breeding lines(max. 8.40, min. 6.75%). 4. Naked barley appeared to be lower in carbohydrate content but higher in crude ash to compare with milled barley. 5. There was significant correlation(r=0.560, p=0.01) between Ca and K, indicating competition in uptake or translocation to grain. 6. Carbohydrate content showed the highest negative correlation with protein content but it was not significant. 7. The low protein variety (Bangju) showed higher yield than the high protein one (Baegdong) both with (16%) and without (48%) fertilizers.

Antioxidant Activity of Peanut Flours with Germination and Roasting (볶음 및 발아처리한 땅콩분말의 항산화 활성)

  • The purpose of this study was to evaluate the antioxidant activity of roasted and germinated peanut flours. This study also aims to utilize it as a functional material to be applied to processed foods. The moisture, crude protein, crude fat and ash carbohydrate contents of the common peanut powder used in this study were 1.27, 25.63, 42.19, 2.38, 28.20 g / 100 g, respectively. The moisture content, crude protein, crude fat and ash carbohydrate in germinated peanut powder were 1.47, 25.86, 42.86, 2.25 and 26.66 g / 100 g, respectively. 26.52, 45.02, 2.33, 24.70, g / 100 g, and the dietary fiber content of peanut, roasted peanut and germinated peanut powder was 12.27, 13.05 and 14.22 g / 100g, respectively. The antioxidants and radical scavenging ability of polyphenols and flavonoids in peanut powder treated with germination and germination compared to ordinary peanuts. Resverasterol content was high in the germinated peanut powder. Especially, germinated peanut powder can act as a natural antioxidant.

Quality characteristics and antioxidant activity of roasted yakgwa according to the addition ratio of mealworm

  • The proximate composition, quality characteristics, antioxidant activity, and sensory evaluation scores of yakgwa added with mealworm powder (MP) were examined. MP contained 5.83 wt% moisture, 55.70 wt% crude protein, 35.96 wt% crude fat, 3.70 wt% crude ash, and 2.43 wt% carbohydrate and feature total polyphenol and flavonoid contents of 406.52 mg GAE/100 g and 21.18 mg NE/100 g, respectively. The DPPH and ABTS•+ radical scavenging activities and the reducing power of MP were determined as 90.25%, 44.06%, and 1.74, respectively. Except for moisture and carbohydrate content, the proximate composition of mealworm yakgwa (MY) increased with the amount of MP increased. The pH of the dough increased with the addition of MP, whereas the expansion degree tended to decrease. Sugar content was highest at MP contents of 0 wt% and 12 wt% (FM4 group), and hardness was lowest in the FM4 group. With the increasing MP content, the L, b values and antioxidant activity increased, whereas a value decreased. The sensory evaluation scores for the overall preference, appearance, color, and taste were lowest in the FM4 group. These results suggested that MP contents of 6-9 wt% were optimal for mealworm-based yakgwa.

Physicochemical Characteristics on Main and Fine root of Ginseng Dried by Various Temperature with Far-Infrared drier (원적외선 건조온도에 따른 백삼의 주근과 세근의 이화학적 특성)

  • To find up using of more efficient white ginseng, white ginseng was dried on various temperature (70, 80, 90,100, 110, 120, 130 and $140^{\circ}C$) with far-infrared drier and analyzed the composition of ginsenoside, carbohydrate, organic acid content and color. The type of ginseng shape was sliced and dried main and fine root, separately. As heating temperature increased, total ginsenoside content increased on main root, its content was the highest at $130^{\circ}C$, while decreased on fine root. Soluble carbohydrate content was the highest at $70^{\circ}C$ both on main and fine root. Increase of Re, Rc and Rb2 content was increased more high at $130^{\circ}C$, especially. But on fine root, content of Rg1, Rg3, Rf and Rb3 was increased and Re, Rc,Rb1 and Rb2 were decreased by the increased of temperature. As heating temperature increased, lightness of both main and fine root were decreased. Redness and yellowness of both main and fine root was increased to $120^{\circ}C$ and $100^{\circ}C$, respectively and decreased over this temperature.

Alleviating Effect of the Application of the Easily Decomposable Carbohydrate on Ammonium Toxicity in Chinese Cabbage (Brassica rapa var. chinensis)

  • An excess application of N fertilizer causes physiological and morphological disorder known as ammonium ($NH_4{^+}$) toxicity in Chinese cabbage and it has been to be an issue for appropriate N fertilizer management. Hence, the pot experiment was conducted in order to evaluate the alleviating effect of the application of the easily decomposable carbohydrate on $NH_4{^+}$ toxicity in Chinese cabbage. Four levels of urea at 0, 160, 320, and $640kg\;N\;ha^{-1}$, represented as T1, T2, T3, and T4, respectively, were applied. In order to evaluate the alleviating effect of the application of the easily decomposable carbohydrate (sucrose) at T3 and T4 where $NH_4{^+}$ toxicity had occurred, five levels of sucrose were applied to meet C/N ratios of 0, 2, 4, 6, and 10, respectively. Our results showed that the $NH_4{^+}$ toxicity was observed at T3 and T4 at 5 days after treatment (DAT). $NH_4{^+}$ toxicity contributed to decrease fresh weight, length of leaves, length of root, and number of leaves significantly (p<0.05). The application of sucrose as a source of mitigating $NH_4{^+}$ toxicity had a good performance at T3 with the alleviating effect as 73 % and reduced in $NH_4{^+}-N$ content in soil at 29 DAT. In the maximum N rate of T4, however, sucrose application recovered it as 32 % only compared to T2 even though the same C/N ratio was treated. Consequently, sucrose as the easily decomposable carbohydrate played crucial role to reduce $NH_4{^+}$ concentration in soil and finally alleviated $NH_4{^+}$ toxicity in plant.

Sedimentation and EPS Production by the Change of Dissolved Oxygen Concentration for the Aeration Tank to treat Wastewater with Bacillus sp. (바실러스 미생물을 이용한 하수처리에서 포기조의 DO농도 변화에 따른 EPS 물질생성과 슬러지 침강성에 관한 연구)

  • The factors affecting on sludge sedimentation are reported as F/M ratio, ingredient, composition of influent substrate, dissolved oxygen concentration, temperature, pH, filamentous bacteria and SRT. Aeration tank applying Bacillus sp. has an important role for maintaining the dominant microorganism species to make steady progress for spore growth affecting sedimentation. This research aims to investigate the affecting factor for the sedimentation in B3 system and RABC system with aeration tank applying tapered aeration. Extracellular polymeric substances(EPS), protein and carbohydrate can be produced for the extreme condition, that is down to 0.2 mg/L of dissolved oxygen in the aeration tank. This research found out the relation between the sedimentation and the EPS production, especially the ratio of protein/carbohydrate. The spore of Bacillus sp. was formed at the low DO then microorganisms produced EPS. The results showed that the production of EPS was 109.95 mgEPS/gSS at 1.6 mg/L of DO, however it was 131.77 mgEPS/gSS at 0.5 mg/L of DO. The sedimentation was affected by protein content in EPS and the ratio of protein and carbohydrate. The settleability of sludge was not affected by the ratio of protein/carbohydrate in B3 process, meanwhile settleability was affected by the ratio of it in RABC process, respectively.
