Association Study Analysis of Phospholipase C Zeta (PLCz) Gene Polymorphism (g.158T>C) for Duroc Boar Post-Thawed Semen Motility and Kinematic Characteristics (PLCz 유전자의 유전적 다형성(g.158T>C)과 두록 동결정액의 운동학적 특성과의 연관성 분석)

  • Cryopreservation of boar semen is continually researched in reproductive technologies and genetic resource banking in breed conservation. For evaluating the boar semen quality, sperm motility (MOT) is an important parameter because the movement of spermatozoa indicates active metabolism, membrane integrity and fertilizing capacity. Various researches have been trying to improve the quality of semen post-thawed in boar. Recently, polymorphism (g.158T>C) of phospholipase C zeta (PLCz) gene reported to be significant association with MOT. This study was conducted to evaluate the PLCz gene as a positional controlling for motility and kinematic characteristics of post-thawed boar semen. To results, The g.158 T>C SNP of PLCz was significantly associated with frozen semen motility and kinematic characteristics. g.158 T>C SNP was high significantly associated with MOT, VCL, VSL and VAP (p<0.0001, p=0.0002, p<0.0001 and p<0.0001, respectively). Therefore, we suggest that the intron region of the porcine PLCz, may be used as a molecular marker for Duroc boar post-thawed semen quality, although its functional effect was not defined yet. Whether the association is due to the candidate gene or not require further verification. Thus, it will be of interest to continue association studies in the regions surrounding those genes.

Debaryomyces hansenii Strains from Valle De Los Pedroches Iberian Dry Meat Products: Isolation, Identification, Characterization, and Selection for Starter Cultures

  • Yeasts, filamentous fungi, and bacteria colonize the surface of fermented sausages during the ripening process. The source of this microbiota is their surrounding environment, and is influenced by the maturing conditions and starter cultures. Debaryomyces hansenii was previously isolated from several dry-cured meat products and associated with the lipolytic and proteolytic changes that occur in these products, influencing their taste and flavor. Therefore, this study isolated the yeast microbiota present in the casing from different meat products ("lomo," "chorizo," and "$salchich{\acute{o}}n$") from the Valle de los Pedroches region in southern Spain. D. hansenii was by far the most abundant species in each product, as all 22 selected isolates were identified as D. hansenii by biochemical and/or molecular methods. In contrast, no yeasts were found in the meat batter. These data constitute the first study of the yeasts present in "lomo" sausages and particularly the highly appreciated Valle de los Pedroches "lomo" sausages. Furthermore, the resistance of these isolates to different pHs, temperatures, and saline stress was studied, together with their catabolic characteristics. Based on the results, certain isolates are proposed as valuable candidate starter cultures that could improve both the manufacture and the flavor of such dry-cured meat products, and provide an understanding of new mechanisms involved in stress tolerance. Applied medium-scale industrial tests are currently in progress.

A Color Flame Region Segmentation Method Using Temperature Distribution Characteristics of Flame (화염의 온도 분포 특성을 이용한 컬러화염 영역분할 방법)

  • This paper propose a method to sort flame regions and non-flame regions in a color image based on temperature Characteristics of flame. The traditional algorithms simply detect flame regions those are colored between yellow and red and there are lot of false detection in this method. But the colors of real flame are fallen between white and red and flame color variation over the flame. In this paper, it reduce false detection by separating colors according to temperature Characteristics of flame. The proposed method firstly finds a color model to express the temperature Characteristics of fire and then the color model is non-linearly quantized based on color values and analyzed using histogram and finally detect the candidate flame regions. The proposed method has 71.8% of matching rate and if it is compared with non-matching rate of traditional algorithms, the non-matching rate is improved by 27 times than others.

  • This paper propose a novel method for tracking moving object based on covariance matrix and Riemannian Manifolds. With image backgrounds continuously changed, we use the covariance matrices to extract features for tracking nonrigid object undergoing transformation and deformation. The covariance matrix can make fusion of different types of features and has its small dimension, therefore we enable to handle the spatial and statistical properties as well as the component correlation. The proposed method can estimate the position of the moving object by employing the covariance matrix of object region as a feature vector and comparing the candidate regions. Rimannian Geometry is efficiently adapted to object deformation and change of shape and improve the accuracy by using geodesic distance to predict the estimated position with the minimum distance. The experimental results have shown that the proposed method correctly tracked the moving object.

  • In this paper, we propose a face recognition system by using the CCD color image. We first get the face candidate image by using YCbCr color model and adaptive skin color information. And we use it initial curve of active contour model to extract face region. We use the Eye map and mouth map using color information for extracting facial feature from the face image. To obtain center point of Log-polar image, we use extracted facial feature from the face image. In order to obtain feature vectors, we use extracted coefficients from DCT and wavelet transform. To show the validity of the proposed method, we performed a face recognition using neural network with BP learning algorithm. Experimental results show that the proposed method is robuster with higher recogntion rate than the conventional method for the rotation and scale variant.

  • Detecting wildfire using only color in image information is a very difficult issue. This paper proposes an algorithm to detect forest fire area by analyzing color and motion of the area in the video including forest fire. The proposed algorithm removes the background region using the Gaussian Mixture based background segmentation algorithm, which does not depend on the lighting conditions. In addition, the RGB channel is changed to an HSV channel to extract flame candidates based on color. The extracted flame candidates judge that it is not a flame if the area moves while labeling and tracking. If the flame candidate areas extracted in this way are in the same position for more than 2 minutes, it is regarded as flame. Experimental results using the implemented algorithm confirmed the validity.

  • In PDP industry, the dielectrics and barrier ribs have been required with low dielectric constant, low melting point and Pb-free composition due to the low power consumption, low signal delay time and the environment restriction. We were studied with $B_2O_3-Al_2O_3$-SrO glass systems about optical, thermal and dielectric properties. The glass forming region of the $B_2O_3-Al_2O_3$-SrO glass systems was narrow due to the amount of the glass former $(B_2O_3)$. The glass transition temperature (Tg) of the glasses was at $550{\sim}590^{\circ}C$. The glasses have 6~8 for the dielectric constant. Furthermore, the transmittance of the glasses was over 80% on the range of the visible ray. From the results, the glasses of the $B_2O_3-Al_2O_3$-SrO glass systems should enable to be a good candidate of the PDP devices for information display with low dielectric constant. The aim of this study is to give a fundamental result of new glass system for low dielectric constant in the information display.

  • The method of moments, a Lagrangian scheme, considers the zeroth, first, and second moments of the grid cell spatial distributions of the concentration and then advects the concentration by maintaining conservation of the moments. The reasonable inital description of the first and second moments as well as the mean concentration, the zeroth moments, in grid element is important in the method of moments. In this study, the description methods of each initial moment are reviewed, and the method of moments is extended to overcome the restrictions of Courant number. Its performance is compared with those of available Eulerian and Lagrangian schemes. As the results, the method is successfully extended to overcome the stability restriction and is an accurate scheme for the advection simulation of concentration distribution, especially of which the gradient is steep. In addition, the method is very promising scheme in terms of computational efficiency when the mixing is confined in a relatively small region to the entire domain in two-dimensional problem.

Identification of Korean Native Pork Using Breed-Specific DNA Marker of KIT Gene

  • Accurate methods for the identification of closely related species or breeds in raw and processed meats must be developed in order to protect both consumers and producers from mislabeling and fraud. This paper describes the development of DNA markers for the discrimination and improvement of Korean native pig (KNP) meat. The KIT gene is related to pig coat color and is often used as a candidate marker. A 538 bp fragment comprising intron 19 of the pig KIT gene was amplified by PCR using specific primers, after which the PCR amplicons of a number of meat samples from KNP and three major improved breeds (Landrace, Duroc and Yorkshire) were sequenced in order to find a nucleotide region suitable for PCR-RFLP analysis. Sequence data showed the presence of two nucleotide substitutions, g.276G>A and g.295A>C, between KNP and the improved pig breeds. Digestion of KIT amplicons with AccII enzyme generated characteristic PCR-RFLP profiles that allowed discrimination between meats from KNP and improved pig. KNP showed three visible DNA bands of 264/249, 199, and 75 bp, whereas DNA bands of 249, 199, and 90 bp were detected in the three improved pig breeds. Therefore, the 75 bp DNA fragment was specific only to KNP, whereas the 90 bp DNA fragment was specific to the improved breeds. The breed-specific DNA markers reported here that target the KIT gene could be useful for the identification of KNP meat from improved pig meats, thus contributing to the prevention of falsified breed labeling.

In-vivo Fluorescence Characteristics of Pteridine for Identification of Phytoplankton (Pteridine계 화합물의 in-vivo 형광 특성을 이용한 식물 플랑크톤의 동정에 관한 연구)

  • The steady state and decay characteristics of primary fluorescence of twelve clones of phytoplankton were investigated in vivo. At 380-435nm region, intense fluorescence emission spectra were obtained from the all phytoplankton, examined', The primary fluorescence of phytoplankton in different growth states was examined In order to find out fluorophores for the observed fluorescence, eight different pteridine derivatives in phosphate buffer solution were examined for their fluorescence characteristics and compared with those of phytoplankton. Fluorescence lifetimes $(\tau)$ and decay curves were compared with standard solution of candidate organic compounds. Decay kinetics of observed fluorescence were shown as hi- and tri-exponential decay curves with 430nm cut-off filter for phytoplankton. Comparison between fluorescence characteristics of bacteria and phytoplankton showed distinct differences for their steady state fluorecence spectra and decay kinetics.
